1Intestinal lysozyme liberates Nod1 ligands from microbes to direct insulin trafficking in pancreatic beta cells显示文摘Long-range communication between intestinal symbiotic bacteria and extra-intestinal organs can occur through circulating bacterial signal molecules, through neural circuits, or through cytokines or hormones from host cells. Here we report that Nod1 ligands derived from intestinal bacteria act as signal molecules and directly modulate insulin trafficking in pancreatic beta cells. The cytosolic peptidoglycan receptor Nod1 and its downstream adapter Rip2 are required for insulin trafficking in beta cells in a cell-autonomous manner. Mechanistically, upon recognizing cognate ligands, Nod1 and Rip2 localize to insulin vesicles, recruiting Rab1a to direct insulin trafficking through the cytoplasm. Importantly, intestinal lysozyme liberates Nod1 ligands into the circulation, thus enabling long-range communication between intestinal microbes and islets. The intestine-islet crosstalk bridged by Nod1 ligands modulates host glucose tolerance. 6A YAP/TAZ-CD54 axis is required for CXCR2-CD44-tumor-specific neutrophils to suppress gastric cancer显示文摘As an important part of tumor microenvironment,neutrophils are poorly understood due to their spatiotemporal heterogeneity in tumorigenesis.Here we defined,at single-cell resolution,CD44-CxCR2-neutrophils as tumor-specific neutrophils(tsNeus)in both mouse and human gastric cancer(GC).We uncovered a Hippo regulon in neutrophils with unique YAP signature genes(e.g.,ICAM1,CD14,EGR1)distinct from those identified in epithelial and/or cancer cells.Importantly,knockout of YAP/TAz in neutrophils impaired their differentiation into CD54+tsNeus and reduced their antitumor activity,leading to accelerated GC progression.Moreover,the relative amounts of CD54+tsNeus were found to be negatively associated with GC progression and positively associated with patient survival.Interestingly,GC patients receiving neoadjuvant chemotherapy had increased numbers of CD54+tsNeus.Furthermore,pharmacologically enhancing YAP activity selectively activated neutrophils to suppress refractory GC,with no significant inflammation-related side effects.Thus,our work characterized tumor-specific neutrophils in GC and revealed an essential role of YAP/TAZ-CD54 axis in tsNeus,opening a new possibility to develop neutrophil-based antitumor therapeutics.Pingping Nie Weihong Zhang Yan Meng Moubin Lin Fenghua Guo Hui Zhang Zhenzhu Tong Meng Wang Fan Chen Liwei An Yang Tang Yi Han Ruixian Yu Wenjia Wang Yuanzhi Xu Linxin Wei Zhaocai Zhou Shi Jiao 2023Protein & Cell2023,14,7:0
7Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 8High performance flexible energy storage device based on copper foam supported NiMoO4 nanosheets-CNTs-CuO nanowires composites with core–shell holey nanostructure显示文摘Because of the intensified electrochemical activities,mixed metal oxides as a representative for pseudocapacitive materials play a key role for high performance supercapacitor electrodes.Nevertheless,low ion and electron transfer rate and poor cycling performance in the electrode practically restrict further promotion of their electrochemical performance.In order to offset the defect,a novel copper(Cu)foamsupported nickel molybdate nanosheet decorated carbon nanotube wrapped copper oxide nanowire array(NiMoO4 NSs-CNTs-CuO NWAs/Cu foam)flexible electrode is constructed.The as-prepared electrode demonstrates a unique core-shell holey nanostructure with a large active surface area,which can provide a large number of active sites for redox reactions.Besides,the CNTs networks supply improved conductivity,which can hasten electron transport.Through this simple and efficient design method,the spatial distribution of each component in the flexible electrode is more orderly,short and fast electron transport path with low intrinsic resistance.As a result,the NiMoO4 NSs-CNTs-CuO NWAs/Cu foam as an adhesiveless supercapacitor electrode material exhibits excellent ene rgy storage perfo rmance with high specific areal capacitance of 23.40 F cm^(-2)at a current density of 2 mA cm^(-2),which outperforms most of the flexible electrodes re ported recently.The assembled asymmetric supercapacitor demonstrates an energy density up to 96.40 mW h cm^(-3)and a power density up to 0.4 W cm^(-3)under a working voltage window of 1.7 V.In addition,outstanding flexibility of up to 100°bend and good cycling stability with the capacitance retention of 82.53%after 10,000 cycles can be obtained.Pingping Yao Chenyang Li Jiali Yu Shuo Zhang Meng Zhang Huichao Liu Muwei Ji Guangtao Cong Tao Zhang Caizhen Zhu Jian Xu 2021Journal of Materials Science & Technology2021,,26:0
9Neuritin affects the activity of neuralized-like 1 by promoting degradation and weakening its affinity for substrate显示文摘Neuritin plays a key role in neural development and regeneration by promoting neurite outgrowth and synapse maturation.Our previous research revealed the mechanism by which neuritin inhibits Notch signaling through interaction with neuralized-like 1(Neurl1)to promote neurite growth.However,how neuritin regulates Notch signaling through Neurl1 has not been elucidated.Here,we first confirm that neuritin is an upstream regulator of Neurl1 and inhibits Notch signaling through Neurl1.Neurl1 is an E3 ubiquitin ligase that can promote ubiquitination and endocytosis of the Notch1 ligand Jagged1.Therefore,we observe the effect of neuritin on the ligase activity of Neurl1.The results indicate that neuritin inhibits Neurl1 activity by reducing the ubiquitination level and endocytosis of the target protein Jagged1.Moreover,we find that decreased activity of Neurl1 results in reduced expression of Notch receptor Notch intracellular domain(NICD)and downstream target gene hairy and enhancer of split-1(HES1).Furthermore,we investigate how neuritin affects Neurl1 enzyme activity.The results show that neuritin not only weakens the affinity between Neurl1 and Jagged1 but also promotes the degradation of Neurl1 by the 26S proteasome pathway.Taken together,our results suggest that neuritin negatively regulates Notch signaling by inhibiting the activity of Neurl1,promoting the degradation of Neurl1 and weakening the affinity of Neurl1 for Jagged1.Our study clarifies the molecular mechanisms of neuritin in regulating the Notch signaling pathway and provides new clues about how neuritin mediates neural regeneration and plasticity.Jingling Zhu Yu Li Chen Zhong Meiyi Zhu Yan Zheng Anying Xiong Pingping Meng Liya Shan Yang Li Jin Huang 2023Acta Biochimica et Biophysica Sinica2023,55,10:0
10Spatiotemporally co-delivery of triple therapeutic drugs via HA-coating nanosystems for enhanced immunotherapy显示文摘There is growing empirical evidence that certain types of chemotherapy and phototherapy trigger immunogenic cell death and enhance the therapeutic anticancer efficacy of genetic immunotherapy. However, the main challenge is spatiotemporally co-delivering different drugs to maximize the therapeutic index of the combination therapy. In this study, a drug delivery system(HTCP-Au/shPD-L1/DOX) was designed with a polysaccharide-wrapped shell and a condensed DNA core. To construct the HTCP-Au vector, dodecyl side chains with a polyethylenimine(PEI) head were grafted onto hyaluronic acid, and Au NPs were grafted via Au-S bonds. During drug loading, PEI arrested shRNA plasmid DNA targeting programmed cell death ligand 1(shPD-L1) via electrostatic interactions. It also formed a PEI-DNA core that was automatically enclosed when aliphatic hydrocarbons pulled the hyaluronic acid backbone. A hydrophobic interlayer consisting of dodecyl bridge chains between the PEI-DNA core and the hyaluronic acid shell was required to accommodate hydrophobic doxorubicin. In vitro and in vivo assays demonstrated that this core-shell drug delivery system could efficiently load and transport three different drugs and effectively target tumors.
