| 3 | Extent of liver resection modulates the activation of transcription factors and the production of cytokines involved in liver regeneration显示文摘AIM:To investigate the molecular events involved in liver regeneration following subtotal hepatectomy (SH) as previous studies have largely focused on partial hepatectomy (PH). METHODS: Male Wistar rats were subjected to 70% PH or 90% SH, respectively, and sacrificed at different times after surgery. Untreated and sham-operated animals served as controls. Serum and liver samples were obtained to investigate liver function, apoptosis (TUNEL assay) and transcription factors (NF-κB,Stat3; ELISA) or cytokines (HGF, TNF-a,IL-6,TGF-a,TGF-b; quantitative RT-PCR) involved in liver regeneration. RESULTS:Serum levels of ALT and AST in animals with 70% PH differed significantly from sham-operated and control animals. We found that the peak concentration 12 h after surgery returned to control levels 7 d after surgery. LDH was increased only at 12 h after 70% PH compared to sham. Bilirubin showed no differences between the sham and 70% resection. After PH, early NF-κB activation was detected 12 h after surgery (313.21±17.22 ng/mL), while there was no activation after SH (125.22 ± 44.36 ng/mL) compared to controls (111.43±32.68 ng/mL) at this time point. In SH, however, NF-κB activation was delayed until 24 h (475.56±144.29 ng/mL). Stat3 activation was similar in both groups. These findings correlated with suppressed and delayed induction of regenerative genes after SH (i.e. TNF-a 24 h postoperatively: 2375±1220 in 70% and 88±31 in 90%; IL-6 12h postoperatively:2547±441 in 70% and 173±82 in 90%). TUNEL staining revealed elevated apoptosis rates in SH (0.44% at 24 h;0.63% at 7d) compared to PH (0.27% at 24h; 0.15% at 7d). | 10 | Fabrication of biomimetic placental barrier structures within a microfluidic device utilizing two-photon polymerization显示文摘The placenta is a transient organ,essential for development and survival of the unborn fetus.It interfaces the body of the pregnant woman with the unborn child and secures transport of endogenous and exogenous substances.Maternal and fetal blood are thereby separated at any time,by the so-called placental barrier.Current in vitro approaches fail to model this multifaceted structure,therefore research in the field of placental biology is particularly challenging.The present study aimed at establishing a novel model,simulating placental transport and its implications on development,in a versatile but reproducible way.The basal membrane was replicated using a gelatin-based material,closely mimicking the composition and properties of the natural extracellular matrix.The microstructure was produced by using a high-resolution 3D printing method– the two-photon polymerization(2PP).In order to structure gelatin by 2PP,its primary amines and carboxylic acids are modified with methacrylamides and methacrylates(GelMOD-AEMA),respectively.High-resolution structures in the range of a few micrometers were produced within the intersection of a customized microfluidic device,separating the x-shaped chamber into two isolated cell culture compartments.Human umbilical-vein endothelial cells(HUVEC)seeded on one side of this membrane simulate the fetal compartment while human choriocarcinoma cells,isolated from placental tissue(BeWo B30)mimic the maternal syncytium.This barrier model in combination with native flow profiles can be used to mimic the microenvironment of the placenta,investigating different pharmaceutical,clinical and biological scenarios.As proof-of-principle,this bioengineered placental barrier was used for the investigation of transcellular transport processes.While high molecular weight substances did not permeate,smaller molecules in the size of glucose were able to diffuse through the barrier in a time-depended manner.We envision to apply this bioengineered placental barrier for pathophysiological research,where altered nutrient transport is associated with health risks for the fetus. | Denise Mandt Peter Gruber Marica Markovic Maximillian Tromayer Mario Rothbauer Sebastian Rudi Adam Kratz Syed Faheem Ali Jasper Van Hoorick Wolfgang Holnthoner Severin Mühleder Peter Dubruel Sandra Van Vlierberghe Peter Ertl Robert Liska Aleksandr Ovsianikov | 2018 | International Journal of Bioprinting2018,4,2: | 1 |

| 13 | A glacial control on the eruption rate of Mt Erebus, Antarctica显示文摘Mt Erebus is the most active Antarctic volcano, on the flanks of the world's largest ice sheet. Despite this, the interactions between its eruptions and the ice cover have not been studied in detail. Focusing on the most recent deglaciation, we build a glacial retreat model and compare this to recent lava geochemistry measurements to investigate the processes involved. This analysis exposes a previously unknown link between Antarctic glaciation and eruptions, of vital importance to the understanding of volcanism in this context. We find that deglaciation led to rapid emptying of the shallow magma plumbing system and a resulting peak in eruption rates synchronous with ice retreat. We also find that the present day lavas do not represent steady state conditions, but originate from a source with up to 30% more partial melting than older >4 ka eruptions.
