3Exercise training in hypertension:Role of microRNAs显示文摘Hypertension is a complex disease that constitutes an important public health problem and demands many studies in order to understand the molecular mechanisms involving his pathophysiology. Therefore, an increasing number of studies have been conducted and new therapies are continually being discovered. In this context, exercise training has emerged as an important non-pharmacological therapy to treat hypertensive patients, minimizing the side effects of pharmacological therapies and frequently contributing to allow pharmacotherapy to be suspended. Several mechanisms have been associated with the pathogenesis of hypertension, such as hyperactivity of the sympathetic nervous system and renin-angiotensin aldosterone system,impaired endothelial nitric oxide production, increased oxygen-reactive species, vascular thickening and stiffening, cardiac hypertrophy, impaired angiogenesis, and sometimes genetic predisposition. With the advent of microRNAs(miRNAs), new insights have been added to the perspectives for the treatment of this disease, and exercise training has been shown to be able to modulate the miRNAs associated with it. Elucidation of the relationship between exercise training and miRNAs in the pathogenesis of hypertension is fundamental in order to understand how exercise modulates the cardiovascular system at genetic level. This can be promising even for the development of new drugs. 6金雀异黄素对脂多糖诱导RAW264.7细胞分泌功能的影响显示文摘目的:研究金雀异黄素对脂多糖(LPS)诱导巨噬细胞RAW264.7分泌功能的影响,探讨金雀异黄素抗炎作用及其机制,为临床应用提供理论基础。方法:用脂多糖诱导巨噬细胞株Raw264.7建立体外细胞炎症模型,金雀异黄素与巨噬细胞株Raw264.7温孵1h后,加入100ng·mL-1脂多糖以诱导巨噬细胞的活化,采用Griess试剂检测细胞培养液中NO的含量,通过双抗体夹心酶联分析法和细胞转染技术观察不同浓度金雀异黄素对脂多糖刺激巨噬细胞分泌细胞因子TNF-α、IL-6和IL-10含量及对核转录因子NF-κB和AP-1表达的影响。结果:金雀异黄素(4~111μmolL-1)能明显抑制脂多糖刺激巨噬细胞生成一氧化氮产生(P<0.05);显著抑制脂多糖诱导巨噬细胞分泌细胞因子TNF-α、IL-6和IL-10(P<0.01),并能抑制核转录因子NF-κB和AP-1的表达。结论:本实验证明了金雀异黄素有抗炎作用,其能显著降低脂多糖诱导巨噬细胞分泌NO及细胞因子TNF-α、IL-6和IL-10的释放,通过抑制这些炎症因子的释放来达到抗炎作用,金雀异黄素抑制核转录因子NF-κB和AP-1的表达来实现的。崔树娜 姜俊杰 李彧 牛建昭 Ursula Bilitewski 2010中医药学报2010,38,2:7

8Perioperative challenges in management of diabetic patients undergoing non-cardiac surgery显示文摘Prediabetes and diabetes are important disease processes which have several perioperative implications.About one third of the United States population is considered to have prediabetes.The prevalence in surgical patients is even higher.This is due to the associated micro and macrovascular complications of diabetes that result in the need for subsequent surgical procedures.A careful preoperative evaluation of diabetic patients and patients at risk for prediabetes is essential to reduce perioperative mortality and morbidity.This preoperative evaluation involves an optimization of preoperative comorbidities.It also includes optimization of antidiabetic medication regimens,as the avoidance of unintentional hypoglycemic and hyperglycemic episodes during the perioperative period is crucial.The focus of the perioperative management is to ensure euglycemia and thus improve postoperative outcomes.Therefore,prolonged preoperative fasting should be avoided and close monitoring of blood glucose should be initiated and continued throughout surgery.This can be accomplished with either analysis in blood gas samples,venous phlebotomy or point-of-care testing.Although capillary and arterial whole blood glucose do not meet standard guidelines for glucose testing,they can still be used to guide insulin dosing in the operating room.Intraoperative glycemic control goals may vary slightly in different protocols but overall the guidelines suggest a glucose range in the operating room should be between 140 mg/dL to 180 mg/dL.When hyperglycemia is detected in the operating room,blood glucose management may be initiated with subcutaneous rapid-acting insulin,with intravenous infusion or boluses of regular insulin.Fluid and electrolyte management are other perioperative challenges.Notably diabetic ketoacidosis and hyperglycemic hyperosmolar nonketotic state are the two most serious acute metabolic complications of diabetes that must be recognized early and treated.Ursula Galway Praveen Chahar Marc T Schmidt Jorge A Araujo-Duran Jeevan Shivakumar Alparslan Turan Kurt Ruetzler 2021World Journal of Diabetes2021,12,8:5
9Methane and nitrous oxide emissions from a subtropical coastal embayment(Moreton Bay, Australia)显示文摘Surface water methane(CH4) and nitrous oxide(N2O) concentrations and fluxes were investigated in two subtropical coastal embayments(Bramble Bay and Deception Bay,which are part of the greater Moreton Bay, Australia). Measurements were done at 23 stations in seven campaigns covering different seasons during 2010–2012. Water–air fluxes were estimated using the Thin Boundary Layer approach with a combination of wind and currents-based models for the estimation of the gas transfer velocities. The two bays were strong sources of both CH4 and N2O with no significant differences in the degree of saturation of both gases between them during all measurement campaigns. Both CH4 and N2O concentrations had strong temporal but minimal spatial variability in both bays.During the seven seasons, CH4 varied between 500% and 4000% saturation while N2O varied between 128 and 255% in the two bays. Average seasonal CH4 fluxes for the two bays varied between 0.5 ± 0.2 and 6.0 ± 1.5 mg CH24/(m·day) while N2 O varied between 0.4 ± 0.1 and1.6 ± 0.6 mg N2O/(m2·day). Weighted emissions(t CO2-e) were 63%–90% N2 O dominated implying that a reduction in N2 O inputs and/or nitrogen availability in the bays may significantly reduce the bays' greenhouse gas(GHG) budget. Emissions data for tropical and subtropical systems is still scarce. 11Mobilization of hematopoietic progenitor cells in patients with liver cirrhosis显示文摘AIM:To test the hypothesis that liver cirrhosis is associated with mobilization of hematopoietic progenitor cells. METHODS:Peripheral blood samples from 72 patients with liver cirrhosis of varying etiology were analyzed by flow cytometry.Identified progenitor cell subsets were immunoselected and used for functional assays in vitro. 13金雀异黄素对巨噬细胞RAW 264.7增殖、形态和细胞周期的影响显示文摘目的研究金雀异黄素对巨噬细胞RAW264.7增殖、细胞形态及细胞周期的影响,以探讨其对免疫系统的作用。方法以鼠源巨噬细胞系RAW264.7为细胞模型,将金雀异黄素用二甲基亚砜(DMSO)溶解,再用培养液稀释成所需终浓度为12.5、25、50、100μmol·L-1的4组及溶剂对照组(DMSO体积含量比低于0.1%)。通过WST-1细胞增殖实验、倒置相差显微镜及流式细胞仪分别检测金雀异黄素对巨噬细胞的增殖、细胞形态及细胞周期的影响。结果金雀异黄素对巨噬细胞的影响呈时间和剂量依赖关系。50~100μmol·L-1浓度的金雀异黄素作用巨噬细胞24h和48h后均能抑制细胞增殖,细胞增殖率分别为77%~81%、58%~73%(P<0.01);并能影响细胞形态改变,表现为细胞体积增大,并形成较长伪足。流式细胞结果显示,该浓度金雀异黄素可将巨噬细胞阻滞在S-G2/M期。结论金雀异黄素能抑制巨噬细胞细胞增殖,影响巨噬细胞形态,可能与干扰细胞周期有关。崔树娜 姜俊杰 艾浩 李健 Ursula Bilitewski 牛建昭 2010北京中医药2010,29,3:4
14Modulating effects of acyl-CoA synthetase 5-derived mitochondrial Wnt2B palmitoylation on intestinal Wnt activity显示文摘AIM:To investigate the role of acyl-CoA synthetase 5(ACSL5)activity in Wnt signaling in intestinal surface epithelia.METHODS:Several cell lines were used to investigate the ACSL5-dependent expression and synthesis of Wnt2B,a mitochondrially expressed protein of the Wnt signaling family.Wnt activity was functionally assessed with a luciferase reporter assay.ACSL5-related biochemical Wnt2B modifications were investigatedwith a modified acyl-exchange assay.The findings from the cell culture models were verified using an Apcmin/+mouse model as well as normal and neoplastic diseased human intestinal tissues.RESULTS:In the presence of ACSL5,Wnt2B was unable to translocate into the nucleus and was enriched in mitochondria,which was paralleled by a significant decrease in Wnt activity.ACSL5-dependent S-palmitoylation of Wnt2B was identified as a molecular reason for mitochondrial Wnt2B accumulation.In cell culture systems,a strong relation of ACSL5 expression,Wnt2B palmitoylation,and degree of malignancy were found.Using normal mucosa,the association of ACSL5 and Wnt2B was seen,but in intestinal neoplasias the mechanism was only rudimentarily observed.CONCLUSION:ACSL5 mediates antiproliferative activities via Wnt2B palmitoylation with diminished Wnt activity.The molecular pathway is probably relevant for intestinal homeostasis,overwhelmed by other pathways in carcinogenesis.Christina Klaus Ursula Schneider Christian Hedberg Anke K Schütz Jürgen Bernhagen Herbert Waldmann Nikolaus Gassler Elke Kaemmerer 2014World Journal of Gastroenterology2014,20,40:4

16藏药“雪莲健骨灵”冲剂治疗成人大骨节病的效果研究显示文摘目的研究藏药'雪莲健骨灵'冲剂治疗成人大骨节病的效果。方法对324例成人大骨节病患者口服'雪莲健骨灵'冲剂6~10个疗程(3~5年),以国家'大骨节病治疗效果判定标准(Ws/T79-1996)'和国际通用的'视觉疼痛判定标准(VAS)'来观察治疗效果。结果 '雪莲健骨灵'冲剂治疗324例成人大骨节病患者6~10个疗程后,症状和体征改善有效率为60.8%~88.8%,关节功能改善有效率为44.0%~50.9%,不但患者的病情得到了很好的控制,而且止痛效果显著,关节功能恢复良好,所有治疗对象均未发生不良反应。对实验点内外环境硒含量检测,病区处于低硒水平,病区人体处于低硒营养状态;'雪莲健骨灵'冲剂硒含量很高。结论 '雪莲健骨灵'冲剂对成人大骨节病治疗效果显著,未发现毒副作用,经济实惠,是目前较好的一种药物治疗方法。其机理既有藏医药补'梅坠'的作用,亦与该药硒含量高有关。索朗次仁 王治伦 Ursula Rechbach 尼玛次仁 罗布 2008中国地方病防治2008,23,2:3
17脾酪氨酸激酶抑制剂对脂多糖诱导巨噬细胞炎症反应的影响及机制显示文摘目的研究脾酪氨酸激酶(Syk)在LtX3诱导的Toll样受体4(TLR4)信号转导通路的作用。方法用100ng/mL脂多糖和不同浓度Syk抑制剂分别处理RAW264.7巨噬细胞后,采用Alarmar blue方法检测细胞增殖;采用Griess试剂检测细胞培养液中NO的含量;Westernblot法检测诱导型一氧化氮合成酶(iNOS)的蛋白表达;ELISA检测巨噬细胞中TNF-α和IL-6含量,RAW—Blue巨噬细胞检测核因子KB(NF—KB)和激活子蛋白-1(AP-1)的活化。结果Syk抑制剂作用于RAW264.7巨噬细胞24h,(0.5~14.0)μmol/L的Syk抑制剂对细胞增殖无明显影响,但当剂量超过1.5μmol/L时,能显著抑制LPS刺激的RAW264.7巨噬细胞NO的产生和iNOS的蛋白表达,抑制TNF-α和IL-6的产生(P〈0.01)以及NF—KB和AP-1的激活。结论Syk抑制剂降低LPS诱导的巨噬细胞分泌NO和iNOS的蛋白表达及TNF-α和IL-6的释放,通过下调Syk下游TLR4受体信号传导通路的蛋白,进而显著降低转录因子NF-KB和AP-1的激活有关。崔树娜 Ursula Bilitewski 2013细胞与分子免疫学杂志2013,29,10:3

20Beta-7 integrin controls enterocyte migration in the small intestine显示文摘AIM:To hypothesize that beta-7 integrin affects cellularmigration of both,lymphocytes and enterocytes.METHODS:The nucleoside analog Brd U was ip injected in beta-7-deficient mice(C57BL/6-Itgbtmlcgn/J)of male gender and age-matched male C57BL/J J mice(wild type)4,20,or 40 h before analysis.The total small intestine was isolated,dissected,and used for morphometrical studies.Brd U-positive epithelial cells were numbered in at least 15 hemi-crypts per duodenum,jejunum,and ileum of each animal.The outer most Brd U-positive cell(cellmax)was determined per hemi-crypt,numerically documented,and statistically analysed.RESULTS:Integrins containing the beta-7-chain were exclusively expressed on leukocytes.In the small intestinal mucosa of beta-7 integrin-deficient mice the number of intraepithelial lymphocytes was drastically decreased.Moreover,the Peyer’s patches of beta-7integrin-deficient mice appeared hypoplastic.In beta-7integrin-deficient mice the location of cellmax was found in a higher position than it was the case for the controls.The difference was already detected at 4 h after Brd U application,but significantly increased with time(40 h after Brd U injection)in all small intestinal segments investigated,i.e.,duodenum,jejunum,and ileum.Migration of small intestinal enterocytes was different between the experimental groups measured by cellmax locations.CONCLUSION:The E-cadherin beta-7 integrin pathway probably controls migration of enterocytes within the small intestinal surface lining epithelial layer.Elke Kaemmerer Paula Kuhn Ursula Schneider Thomas Clahsen Min Kyung Jeon Christina Klaus Julia Andruszkow Michael Hrer Sabine Ernst Angela Schippers Norbert Wagner Nikolaus Gassler 2015World Journal of Gastroenterology2015,21,6:3
