3Tau Accumulation via Reduced Autophagy Mediates GGGGCC Repeat Expansion-Induced Neurodegeneration in Drosophila Model of ALS显示文摘Expansions of trinucleotide or hexanucleotide repeats lead to several neurodegenerative disorders,including Huntington disease[caused by expanded CAG repeats(CAGr)in the HTT gene],and amyotrophic lateral sclerosis[ALS,possibly caused by expanded GGGGCC repeats(G4C2r)in the C9ORF72 gene],of which the molecular mechanisms remain unclear.Here,we demonstrated that lowering the Drosophila homologue of tau protein(dtau)significantly rescued in vivo neurodegeneration,motor performance impairments,and the shortened life-span in Drosophila expressing expanded CAGr or expanded G4C2r.Expression of human tau(htau4 R)restored the disease-related phenotypes that had been mitigated by the loss of dtau,suggesting an evolutionarily-conserved role of tau in neurodegeneration.We further revealed that G4C2r expression increased tau accumulation by inhibiting autophagosome-lysosome fusion,possibly due to lowering the level of BAG3,a regulator of autophagy and tau.Taken together,our results reveal a novel mechanism by which expanded G4C2r causes neurodegeneration via an evolutionarily-conserved mechanism.Our findings provide novel autophagy-related mechanistic insights into C9ORF72-ALS and possible entry points to disease treatment.Xue Wen Ping An Hexuan Li Zijian Zhou Yimin Sun Jian Wang Lixiang Ma Boxun Lu 2020Neuroscience Bulletin2020,36,12:4

5Oxide transformation and break-up of liquid metal in boiling solutions显示文摘The non-toxic gallium-based liquid metals(LMs) are promising for various newly emerging areas due to having excellent fluidity, high electrical and heat conductivity. However, the oxide layers often affect their applications. In this paper, we disclosed a straightforward boiling strategy to efficiently remove the oxide of LM in deionized water. Meanwhile such method was also demonstrated to be capable of quickly breaking up LM into droplets(mean 0.51 mm, SD 0.20 mm) in Na Cl solution,which has important value for manufacture purpose. According to the force analysis, surface tension of LM dominates the interaction between solution bubbles and LM under the same heating conditions. Then we characterized the surface morphology and compositions of LM droplets boiled(LMDB) using scanning electron microscope(SEM), X-ray diffraction(XRD) and Xray photoelectron spectroscopy(XPS) and tested the dynamic force between a rod and LMDB. It was shown that after being boiled in water, most of Ga2O3 was transformed to scattered α-Ga OOH nanorods which has less restriction to the deformation of LM and large resistance force(0.8 mN) was tested;otherwise, after boiled in Na Cl solution, more Ga2 O3 with contact patch structure reduces the surface resistance of LM(0.2 m N) and makes it to be broken up by bubbles. The different surface products are determined by the inhibition of hydrolysis of Ga3+by Na Cl solution. This study paved a simple way for oxide removal in neutral solution and mass production of LMD. 6Alkyl-chain branched effect on the aggregation and photophysical behavior of polydiarylfluorenes toward stable deep-blue electroluminescence and efficient amplified spontaneous emission显示文摘The control of the condensed superstructure of light-emitting conjugated polymers(LCPs)is a crucial factor to obtain high performance and stable organic optoelectronic devices.Side-chain engineering strategy is an effective platform to tune inter chain aggregation and photophysical behaviour of LCPs.Herein,we systematically investigated the alkyl-chain branched effecton the conformational transition and photophysical behaviour of polydiarylfluorenes toward efficient blue optoelectronic devices.The branched side chain will improve materials solubility to inhibit interchain aggregation in solution according to DLS and optical analysis,which is useful to obtain high quality film.Therefore,our branched PEODPF,POYDPF pristine film present high luminance efficiency of 36.1%and 39.6%,enhanced about 20%relative to that of PODPF.Compared to the liner-type sides'chain,these branched chains also suppress chain planarization and improve film morphological stability effectively.Interestingly,the branched polymer also had excellent stable amplified spontaneous emission(ASE)behaviour with low threshold(4.72μJ/cm2)and a center peak of 465 nm,even thermal annealing at 220℃in the air atmosphere.Therefore,side-chain branched strategy for LCPs is an effective means to control interchain aggregation,film morphology and photophysical property of LCPs.Lili Sun Ning Sun Lubing Bai Xiang An Bin Liu Chen Sun Lixiang Fan Chuanxin Wei Yamin Han Mengna Yu Jinyi Lin Dan Lu Ning Wang Linghai Xie Kang Shen Xinwen Zhang Yanan Xu Juan Cabanillas-Gonzalez Wei Huang 2019Chinese Chemical Letters2019,30,11:1

8Mechanical behaviors of extruded Mg alloys with high Gd and Nd content显示文摘The influence of alloying elements and heat treatment on the microstructure and mechanical behaviors of extruded Mg–Gd–Nd ternary alloys was investigated in this study.The grain sizes dramatically decreased after extrusion,and the particles which distributed in Mg matrix had great effect on the grain size.The grain sizes of extruded alloys decreased from 26 to 5μm with the alloying content increasing.The mechanical test results show that both Gd and Nd had positive effect on the hardness,yield strength and Young’s modulus.The ultimate tensile strength(UTS)was enhanced by Gd content,decreased with Nd content.The elongation of alloys was lower with higher alloying elements.Those extruded alloys were aged for 200 h in 200℃.The Young’s moduli were decreased by ageing treatment.Combined with microstructure study,the part of the reinforcement which identified as Mg5(Gd/Nd)was dissolved in Mg matrix.Nd element obviously has influence on the solubility of Gd in Mg alloys.Yuling Xu Shiwei Wang Yuye Wang Li Chen Lixiang Yang Lu Xiao Li Yang Norbert Hort 2021Progress in Natural Science:Materials International2021,31,4:1

14Microstructural evolution of Mg-14Gd-0.4Zr alloy during compressive creep显示文摘The present work reports the creep behavior and microstructural evolution of the sand-cast Mg-14Gd-0.4Zr alloy(wt.%) prepared by the differential pressure casting machine. Their compressive creep tests at 250 ℃ were performed under various applied stresses(i.e., 60, 80 and100 MPa). Among them, the sand-cast Mg-14Gd-0.4Zr samples examined under 250 ℃/80 MPa for 39 and 95 h, respectively, were chosen to systemically analyze their creep mechanisms using high-angle annular dark field-scanning transmission electron microscopy(HAADF-STEM).The obtained results showed that the enhancement of creep resistance can be mainly attributed to the coherent β' and β'_F phases with an alternate distribution, effectively impeding the basal dislocations movement. However, with the creep time increasing, the fine β'+β'_F precipitate chains coarsened and transformed to semi-coherent β_1 phase and even to large incoherent β phase(surrounded by precipitate-free areas) in grain interiors. The precipitate-free zones(PFZs) at grain boundaries(GBs) were formed, and they could expand during creep deformation. Apart from the main cross-slip of basal and prismatic dislocations, type dislocations were activated and tended to distribute near the GBs. 15Biased heteroplasmy within the mitogenomic sequences of Gigantometra gigas revealed by sanger and high-throughput methods显示文摘Few studies have explored the differences between Sanger and HTS methods in the results of mitogenome sequencing. We used a single individual of insect to study the differences between the sequences given by Sanger and PCR-free HTS methods. Here we provided evidence for biased results of sequencing due to different methods in the mitochondrial genes of atp6, atp8, cox1, cox2, cox3, Cytb, nad2, nad3, nad4, nad5, rrn S, rrnL, trnH, trn I, and control region at various degrees. Especially, in cox1, the differently sequenced nucleotides account for 2.6% of the complete length. Furthermore, the highest value of the intraspecific genetic distance based on K2 P accounts for 2.5% using a barcode fragment size of cox1(651 bp, Sanger), while the maximum distance of the corresponding cox1 fragment obtained by the two sequencing methods was 5.0%. We revealed that the methods of Sanger and HTS may give different sequencing results of mitochondrial genes, which may reflect the heteroplasmy of mitogenomes within an insect individual. 16Study on Quality Standard of Zhuang Medicine Buddlejae Flos显示文摘[Objectives]This study was conducted to explore the quality control method and establish the quality standard of Zhuang medicine Buddlejae Flos.[Methods]The microscopic identification method was adopted to identify the characters and microscopic characteristics of dried Buddlejae Flos in combination with traditional experience.The moisture,ash,acid-insoluble components and extracts of Buddlejae Flos were determined with reference to general rules of Chinese Pharmacopoeia(2015 edition).TLC identification was performed.Liquid chromatography-mass spectrometry(LC-MS)was used to determine the contents of linarin and verbascoside in Buddlejae Flos,and gas chromatography-mass spectrometry(GC-MS)was adopted to detect pesticide residues in Buddlejae Flos.Heavy metal elements As,Cd and Pb were detected by inductively coupled plasma mass spectrometry(ICP-MS).[Results]The established method is simple and accurate.Clear spots were observed on the thin layer chromatograms,and the resolution was good.The average value of moisture content was 12.24%;the average value of ash content was 5.1%;the average value of acid-insoluble content was 7.5%;and the average value of extract content was 27.3%.The regression equation of rutin in Buddlejae Flos was y=11.896x-0.0049,R^(2)=0.9996,and the contents of linarin and verbascoside were 5680 and 2080 mg/kg,respectively.No pesticide residues and heavy metals were detected in the medicinal materials of Buddlejae Flos.[Conclusions]This study can provide reference for the quality standard control of Guangxi Zhuang medicine Buddlejae Flos.Tong LI Zhengsan LING Xin XIE Liyuan LI Haifeng WEI Suoyi HUANG Shiyou ZHOU Lixiang LU Tingting HE Ran CHEN Yue LI Zheng LIU Jinchi LIU 2023Medicinal Plant2023,14,2:0
