1Relationship between the expression of iNOS,VEGF,tumor angiogenesis and gastric cancer显示文摘AIM:To in vestigate the relationship between the expression of inducible nitric oxide synthase(iNOS),vascular endothelial growth factor(VEGF),the microvascular density(MVD)and the pathological features and clinical staging of gastric cancer.METHODS:Immunohistochemical staining was used for detecting the expression of iNOS and VEGFin46resected specimens of gastric carcinoma;the monoclonal antibody against CD34 was used for displaying vascular endothelial cells,and MVD was detected by counting of CD34-positive vascular endothelial cells.RESULTS:Of 46resected specimens of gastric carcinoma,the rates of expressions of iNOS and VEGF were 58.70%and76.09%,respectively,and MVDaveraged55.59±19.39,Judged by the standard TNM criteria,the rate of expression of iNOS in stageⅣ(84.46%)was higher than those in stageⅠ,Ⅱ,Ⅲ(Fish exact probabilities test,P=0.019,0.023and 0.033,respectively);the rates of expression of VEGFin stage Ⅲ,Ⅳ(76.0%,92.31%,respectively)were higher than those in stageⅠ,Ⅱ(Fis exact probabilities test,P=0.031,0.017,0.022and0.019).MVDs in stageⅢ,Ⅳ(64.72±14.96,67.09±18.29,respectively)were higher than those in stageⅠ,Ⅱ(t\2.378,4.015,2.503and2.450,P<0.05,P<0.001,P<0.001,P<0.05,respectively),In37gastric carcinoma specimens with lymph node metastasis,MVD(68.69±18.07)and the rates of expression of iNOS and VEGF(70.27%,83.78%,respectively)were higher than those in the specimens with absence of metastasis(t=2.205,X^2=6.3587,X^2=6.2584,P<0.01,P<0.05,P<0.05,respectively),MVD and the expressions of iNOS and EGF were not correlated to the location,size or grade of tumor,nor with the depth of invasion of tumor;MVDs in the positive iNOS and VEGF specimens(59.88±18.02,58.39±17.73,repectively)were higher than those in the negative iNOS and VEGF specimens(X^2=6.3587and 6.1574,P<0.05,P<0.05,respectively);thus the expressions of iNOS and VEGF was correlated to MVD,but the expression of iNOS was not correlated to that of VEGF,In addition.of the 46 surviving patients,the 5-year survival rate of patients with positive iNOS or VEGF tumors was significantly less than that of patients with negative iNOS-or VEGF tumors(X^2=4.3842and 5.4073,P<0.05,P<0.05.respectively).CONCLUSION:The expressions of iNOS and VEGF are colosely related to tumor angiogenesis,and are involved in the advancement and the lymph node metastasis;thusMVD and the expressions of iNOS and EGF may serve indexes for evaluating staging of gastric carcinoma and forecasting its risk of metastasis,which will help establish a comprehensive therapeutical measure of post-operative patients and provide a new approach to tumor therapy.Zheng-Jun Song Ping Gong Yu-E Wu Department of Gastroenterology,First Affiliated Hospital of Xi’an Jiaotong University,Xi’an 710061,ShaanXi Province,China 2002World Journal of Gastroenterology2002,8,4:87

3Expression of nuclear factor-KB in hepatocellular carcinoma and its relation with the X protein of hepatitis B virus显示文摘AIM In this study we investigated therelationship of the X protein of HBV and nuclearfactor-KB (NF-κB) and the expression of NF-KB inhuman hepatocellular carcinoma tissues.METHODS Immunohistochemistry SP methodwas used to detect the expression of NF-κB and the X protein of HBV in human hepatocellularcarcinoma tissues of 52 cases. Gene transfectionmediated by lipofectamine was used to transfectthe eukaryotic expression vector pCDNA3. 1-HBXof HBV x gene into human hepatocellularcarcinoma cell line HCC-9204 and NF-κB wasdetected.RESULTS NF-κB was widely expressed inhuman hepatocellular carcinoma tissues in atotal of 52 cases and its expression was relatedto the X protein of HBV. NF-KB was localizedboth in the cytoplasm and . The nuclei ofhepatocellular carcinoma cells in 11 cases whichwere positive for the X protein of HBV while in 41cases negative for the X protein of HBV, NF-Kbwas only localized in the cytoplasm ofhepatocellular carcinoma cells but translocatedto the nuclei of hepatocellular carcinoma cellsafter the eukaryotic expression vectorpCDNA3.1-HBX was transfected into HCC-9204cells.CONCLUSION This study strongly suggeststhat the nuclear factor NF-KB is widely expressedin hepatocellular carcinoma tissues in differentstyles according to the expression of the Xprotein of HBV. NF-κB is abnormally activated inhepatocellular carcinoma, which is probablyrelated to the X protein of HBV. 42018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 6Loess in the Tian Shan and its implications for the development of the Gurbantunggut Desert and drying of northern Xinjiang显示文摘Eolian loess is widely distributed on the various geomorphic surfaces between 700-2400 m a.s.l. on the northern slope of the Tian Shan. It is formed in a synchronous manner with dust transported from the Gurbantunggut Desert in the Junggar Basin. The thickest section of loess was found in the Shawan and Shihezi regions. Paleomagnetic and climatic proxy analyses of over 71 m of a loess-paleosol sequence on the highest terrace of the Qingshui He (River) in the Shawan show that the paleomagnetic Bruhues/Matuy-ama (B/M) boundary lies at the bottom of paleosol S8, at a depth of 69.5 m, and the bottom of the sequence was estimated to be -0.8 Ma. This implies that the extremely dry climatic conditions in the Junggar Basin and the initial Gurbantunggut Desert were present at least by 0.8 Ma. 10Soil Bacterial Communities Under Different Long-Term Fertilization Regimes in Three Locations Across the Black Soil Region of Northeast China显示文摘Although soil bacteria play critical roles in agro-ecosystems, the knowledge of their response to long-term fertilization across the black soil region of Northeast China is limited. In this study, we sequenced 16 S rRNA genes to assess the effects of four long-term fertilization regimes—non-fertilization(NoF), chemical fertilizer(CF), manure(M), and chemical fertilizer plus manure(CFM)—on soil properties and bacterial communities in three locations, the northern, middle, and southern parts, across the black soil region. Results showed that the influence of fertilization regimes on soil properties varied significantly among the three locations. Manure fertilization significantly increased microbial biomass carbon and relative abundance of copiotrophic bacteria. Principal component analysis(PCA)showed that the total bacterial communities were separated into three groups according to the sampling location despite long-term fertilization, and that soil pH was the most important factor in shifting bacterial communities. In addition, similar fertilization regimes resulted in different influences on bacterial community composition, and the most influential soil properties varied among the three locations. 11Pharmacokinetics of traditional Chinese syndrome and recipe:a hypothesis and its verification(Ⅰ)显示文摘AIM To propose a hypothesis defining theabsorption,distribution,metabolism andelimination of traditional Chinese recipe(TCR)-component in blood of healthy subjects andpatients,and estimate its correctness.METHODS The pharmacokinetics(PK)of samedose of drug was studied in the animal model oftraditional Chinese syndrome(S)and healthyanimals.The classification,terminology,concept and significance of the hypothesis wereset forth with evidence provided in the presentstudy.The hypotheses consisted of traditionalChinese syndrome PK(S-PK)and traditionalChinese recipe PK(R-PK).Firstly,the observedtetramethylpyrazine(TMP)PK in healthy,chronically reserpinized rats(rat model ofspleen deficiency syndrome,RMSDS)andRMSDS treated with Sijunzi decoction(SJZD)forconfirmation were used to verify S-PK; secondly,the ferulic acid(FA)PK in healthy andhigh molecular weight dextran(HMWD)-inducedrabbit model with blood stasis syndrome(RDBSS)was also used to verify S-PK;andlastly,TMP PK parameters in serum of healthyrats after orally taken Ligusticum wallichii(LW),LW and Salvia miltiorrhiza(LW&SM)decoctions were compared to verify R-PK.RESULTS The apparent first-order absorption[Ka,(13.61±2.56)h-1,area under the blooddrug concentration-time curve[AUC,(24.88±9.76)μg.h-1mL-1],maximum drug concentration[Cmax,(4.82±1.23)μg·mL-1]of serum TMP inRMSDS were increased markedly(P<0.05)compared with those[Ka=(5.41±1.91)h-1,AUC=(5.20±2.57)μg·h-1·mL-1,Cmax=(2.33±1.77)μg·mL-1]of healthy rats(HR).Theapparent first-order rate constant for α and βdistribution phase[α=(0.38±0.09)h-1,β=(0.06±0.03)h-1,the apparent first-orderintercompartmental transfer rate constants[K10=(0.24±0.07)h-1,K12=(0.11±0.02)h-1,K21=(0.11±0.02)h-1]of serum TMP in RMSDS weredecreased significantly(P<0.01)comparedwith those[K10=(0.88±0.20)h-1,K12=(1.45±0.47)h-1,K21=(0.72±0.22)h-1]of HR.However,no apparent differences occurredbetween HR and RMSDS treated with SJZD.Theserum FA concentration and its AUC[(5.6690±2.3541)μg·h-1·mL-1] in RMBSS were also higherthan those[AUC=(2.7566±0.8232)μg·h-1·mL-1]of healthy rabbits(P<0.05).The Ka(11.51±2.82)h-1,AUC(0.84±0.17)μg·h-1·mL-1of LW & SM-derived TMP in serum weremuch lower(P<0.05)than those[Ka=(19.58±4.14)h-1,AUC=(1.27±0.26)μg·h-1·mL-1]ofLW-derived TMP in serum after oral decoctions.CONCLUSION The SDS and blood stasissyndrome state could affect significantly thepharmacokinetic parameters of drugs and theabnormal SDS pharmacokinetic parameters couldbe normalized by SJZD.The combination ofChinese medicine in TCR could reciprocallyaffect the pharmacokinetic parameters of othercomponents absorbed into the systemiccirculation.These results support the S-and R-PK hypothesis.Xi Huang Ping Ren Ai Dong Wen Li Li Wang Li Zhang Feng Gao Laboratory of Clinical Pharmacology of Chinese Medicine,Xijing Hospital,The Fourth Military Medical University,Xi’an 710032,Shaanxi Province,China Department of Pharmacy,Xijing Hospital,The Fourth Military Medical University,Xi’an 710032,Shaanxi Province,China Department of Physiology,The Fourth Military Medical University,Xi’an 710032,Shaanxi Province,China 2000World Journal of Gastroenterology2000,6,3:16
12Long-chain alkenones in Hotong Qagan Nur Lake sediments and its paleoclimatic implications显示文摘The long-chain alkenones present in the shallow sediment of Hotong Qagan Nur Lake in China are reported. According to the similarity of compositions and distributions of the alkenones determined commonly in the sediment section to those of marine sediment, it is reasonably referred that the alkenones might also originate from Prymnesiophyceae. ln addition, the parameter U37k'of alkenone is in positive correlation to δ18O values of authigenic calcium and magnesium minerals. 13Function and regulation of cholecystokinin-octapeptide, β-endorphin and gastrin in anorexic infantile rats treated with ErBao Granules显示文摘AIM To study the role of cholecystokinin octapeptide ( CCK-8), β-endorphin ( β-EP), and gastrin in an anorexic infantile rat model and no subsequent regulation of nose peptides by the Yunpi complex prescription ErBao Granule.METHODS We fed infantile rats with special prepared forage. A liquid extract of ErBao Granule was administered to the rats daily for 3weeks, CCK-8, β-EP, and gastrin concentrations in hypothalamus, gastric antrum, and plasma of the rats were measured by radioimmunoassay,and were compared with controls.RESULTS Treatment of rats with ErBao Granule inhibited CCK-8 secretion and increased β-EP and gastrin secretion. CCK-8 concentration in hypothalamus and plasma of model control group increased significantly and correlated negatively with food intake of models.respectively. β-EP concentration in gastric antrum and plasma of model control group decreased significantly and showed a positive correlation with food intake of models,respectively. Hypothalamus concentration of β-EP was similar in models and controls. Gastrin concentration in gastric antrum of models was lower than in the blank control group, and correlated positively to food intake of models.Finally, CCK-8 concentrations in plasma of rats showed a positive correlation with plasma β-EP(r- 0.68, P<0.05).CONCLUSION The increased plasma and hypothalamus concentration of CCK-8,decreased gastric antrum and plasma level of β-EP. and decreased gastric antrum concentration of gastrin are associated significantly with the anorexia of infantile anorexic rat models produced by special forage. 14Influence of FasL gene expression on hepatic metastasis of colorectal carcinoma显示文摘BACKGROUND: FasL expression was reported to be asso- ciated with hepatic metastasis of colorectal cancer. The aim of this study was to study FasL gene expression in colorectal carcinoma and its influences on biological behavior and he- patic metastasis of colorectal carcinoma. METHODS: FasL gene expressions were examined with re- verse transcriptase-polymerase chain reaction (RT-PCR) in the primary focus of colorectal carcinoma, adjacent can- cerous mucosae, and metastasized liver focus from colorec- tal cancer. HR-8348 cells of human rectal cancer cell line were transfected with FasL cDNA. Cell growth suppression rate and response to 5-FU and carboplatin were observed and analyzed with the MTT method. RESULTS: FasL gene expression was detected in the prima- ry focus of colorectal cancer ( n = 58), adjacent cancerous mucosae ( n = 58), and metastasized hepatic tumor tissues (n =28). The positive rate of FasL expression was 24% (14/ 58), 8% (5/58), and 100% (58/58) in the primary focus, adjacent cancerous mucosae and metastasized hepatic tumor tissues respectively. FasL expression rate in the me- tastasized hepatic tumor tissues was higher than that in the primary focus (χ2 = 43.49, P<0. 01) and adjacent cance- rous mucosae (χ2=57.66, P<0.01). In a group of patients with hepatic metastasis, the FasL expression rate in primary focus was higher than that in patients without hepatic me- tastasis (χ2=3.96, P <0.05). In vitro study positive expres- sion of FasL was shown in transfected HR-8348 cells. When 5-FU or carboplatin was added, there was a significant difference in growth suppression rate between FasL positive and controlled cancer cells (t=9.02, t = 11.93, P<0.01). Under the same concentration of chemotherapeutic agents, the survival rate of FasL positive HR-8348 cells was higher than that of controlled cells. CONCLUSIONS: FasL positive cancer cells are powerfullyresistant to chemotherapeutic agents. 16Molecular organic geochemical peculiarities of lacustrine core sediments in Fildes Peninsula, King George Island, Antarctica显示文摘The peculiarities of lipids in lacustrine core sediments from Fildes Peninsula, King George Island, Antarctica, are reported. The origin of high-molecular-weight n-alkanes, alkenes, fatty acids, alcohols is supposed to be related to the abundant moss occurring in the sediments. Abundant iso- and ante/so-fatty acids that occur in the sediments indicate a relatively strong bacterial activity in non-ice-cover zone than in other regions in Antarctica. The C18:2 unsaturated fatty acids mainly originate from autochthonous algae in the sediment pool, and the C18:2/C18:0 ratios in the profile are controlled more by paleotemperatures than source assemblies. Long-chain alkenones were identified in freshwater lacustrine sediment in Antarctica for the first time, of which Prymnesiophyceae is assumed to be the source organism. 19Aberrant expression of ether à go-go potassium channel in colorectal cancer patients and cell lines显示文摘AIM: To study the expression of ether à go-go (Eag1) potassium channel in colorectal cancer and the relation- ship between their expression and clinico-pathological features. METHODS: The expression levels of Eag1 protein were determined in 76 cancer tissues with paired non- cancerous matched tissues as well as 9 colorectal adenoma tissues by immunohistochemistry. Eag1 mRNA expression was detected in 13 colorectal cancer tissues with paired non-cancerous matched tissues and 4 colorectal adenoma tissues as well as two colorectal cancer cell lines (LoVo and HT-29) by reverse transcription PCR. RESULTS: The frequency of positive expression of Eag1 protein was 76.3% (58/76) and Eag1 mRNA was 76.9% (10/13) in colorectal cancer tissue. Expression level of Eag1 protein was dependent on the tumor size, lymphatic node metastasis, other organ metastases and Dukes’ stage (P < 0.05), while not dependent on age, sex, site and degree of differentiation. Eag1 protein and mRNA were negative in normal colorectal tissue, and absolutely negative in colorectal adenomas except that one case was positively stained for Eag1 protein. CONCLUSION: Eag1 protein and mRNA are aberrantly expressed in colorectal cancer and occasionally expressed in colorectal adenoma. 20Chinese herbal medicine reduces mortality in patients with severe and critical coronavirus disease 2019: a retrospective cohort study显示文摘This study aimed to evaluate the efficacy of Chinese herbal medicine(CHM)in patients with severe/critical coronavirus disease 2019(COVID-19).In this retrospective study,data were collected from 662 patients with severe/critical COVID-19 who were admitted to a designated hospital to treat patients with severe COVID-19 in Wuhan before March 20,2020.All patients were divided into an exposed group(CHM users)and a control group(non-users).After propensity score matching in a 1:1 ratio,156 CHM users were matched by propensity score to 156 non-users.No significant differences in seven baseline clinical variables were found between the two groups of patients.All-cause mortality was reported in 13 CHM users who died and 36 non-users who died.After multivariate adjustment,the mortality risk of CHM users was reduced by 82.2%(odds ratio 0.178,95%CI 0.076–0.418;P<0.001)compared with the non-users.Secondly,age(odds ratio 1.053,95%CI 1.023–1.084;P<0.001)and the proportion of severe/critical patients(odds ratio 0.063,95%CI 0.028–0.143;P<0.001)were the risk factors of mortality.These results show that the use of CHM may reduce the mortality of patients with severe/critical COVID-19.Guohua Chen Wen Su Jiayao Yang Dan Luo Ping Xia Wen Jia Xiuyang Li Chuan Wang Suping Lang Qingbin Meng Ying Zhang Yuhe Ke An Fan Shuo Yang Yujiao Zheng Xuepeng Fan Jie Qiao Fengmei Lian Li Wei Xiaolin Tong 2020Frontiers of Medicine2020,14,6:10
