3Study on magnetic memory signals of medium carbon steel specimens with surface crack precut during loading process显示文摘Static tensile test and tensile-tensile fatigue test of medium carbon steel sheet specimens with surface crack precut were performed on MTS810 hydraulic testing machine to clear the meaning of the point of Hp(y) value zero. Magnetic memory signals were measured during the test process. The results show that only one point of Hp(y) zero value exists in all measured magnetic signal curves during the loading process, which should be a sign of intersection of positive-negative magnetic poles after magnetic ordered state appears and does not indicate the position of surface crack precut. The analysis shows that the surface crack precut can not interrupt the magnetic ordered state occurred during the test completely, hence its Hp(y) value is not zero. 4Effect of electrolyte anions on the cycle life of a polymer electrode in aqueous batteries显示文摘Redox polymers are a class of high-capacity, low-cost electrode materials for electrochemical energy storage, butthe mechanisms governing their cycling stability are not well understood. Here we investigate the effect of anionson the longevity of a p-dopable polymer through comparing two aqueous zinc-based electrolytes. Galvanostaticcycling studies reveal the polymer has better capacity retention in the presence of triflate anions than that withsulfate anions. Based on electrode microstructural analysis and evolution profiles of the cell stacking pressure, theorigin of capacity decay is ascribed to mechanical fractures induced by volume change of the polymer activematerials during repeated cycling. The volume change of the polymer with the triflate anion is 61% less than thatwith the sulfate anion, resulting in fewer cracks in the electrodes. The difference is related to the different anionsolvation structures—the triflate anion has fewer solvated water molecules compared with the sulfate anion,leading to smaller volume expansion. 5Genome and transcriptome of Papaver somniferum Chinese landrace CHM indicates that massive genome expansion contributes to high benzylisoquinoline alkaloid biosynthesis显示文摘Opium poppy(Papaver somniferum)is a source of morphine,codeine,and semisynthetic derivatives,including oxycodone and naltrexone.Here,we report the de novo assembly and genomic analysis of P.somniferum traditional landrace‘Chinese Herbal Medicine’.Variations between the 2.62 Gb CHM genome and that of the previously sequenced high noscapine 1(HN1)variety were also explored.Among 79,668 protein-coding genes,we functionally annotated 88.9%,compared to 68.8%reported in the HN1 genome.Gene family and 4DTv comparative analyses with three other Papaveraceae species revealed that opium poppy underwent two whole-genome duplication(WGD)events.The first of these,in ancestral Ranunculales,expanded gene families related to characteristic secondary metabolite production and disease resistance.The more recent species-specific WGD mediated by transposable elements resulted in massive genome expansion.Genes carrying structural variations and large-effect variants associated with agronomically different phenotypes between CHM and HN1 that were identified through our transcriptomic comparison of multiple organs and developmental stages can enable the development of new varieties.These genomic and transcriptomic analyses will provide a valuable resource that informs future basic and agricultural studies of the opium poppy.Li Pei Baishi Wang Jian Ye Xiaodi Hu Lihong Fu Kui Li Zhiyu Ni Zhenlong Wang Yujie Wei Luye Shi Ying Zhang Xue Bai Mengwan Jiang Shuhui Wang Chunling Ma Shujin Li Kaihui Liu Wanshui Li Bin Cong 2021Horticulture Research2021,8,1:4
6Pulmonary toxicity induced by three forms of titanium dioxide nanoparticles via intra-tracheal instillation in rats显示文摘Titanium dioxide (TiO2) nanoparticles are in wide commercial use worldwide. To evaluate if acute pulmonary toxicity can be induced by nano-TiO2 particles, rats were intra-tracheally instilled with 0.5, 5, or 50 mg/kg of 5, 21, and 50 nm TiO2 primary particles. Toxic effects were determined with the coefficients of lung tissues to body weight, histopathology, biochemical parameters of blood, activity of lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and acid phosphatase (ACP) in tissues, and the phagocytotic ability of alveolar macrophages (AMs). All the indicators were observed in sacrificed rats one week post-exposure. There was a significant difference of coefficients of pulmonary tissues between the high-dose group and the low-or moderate-dose groups with an exposure of 5 nm TiO2. At the same time, 5 nm TiO2 primary particles increased the activity of LDH and ALP when exposure dose was >5 mg/kg. A significant difference in LDH and ALP activity was observed between the 50 mg/kg group and 0.5 or 5 mg/kg group with exposure of 5 nm TiO2. Lung tissues showed increased ALP activity only if treated with 5 and 50 mg/kg of 21 nm TiO2 particles. There was no significant difference in LDH and ALP activity in the 50 nm TiO2 group and control group. Histo-pathologic examination of lung tissues indicated that the pulmonary response to exposure to TiO2 particles in rats manifested as dose-dependent inflammatory lesions, which mainly consisted of inflltration of inflammatory cells and interstitial thickening. Analysis of uptake of neutral red dye showed that 50 nm TiO2 particles significantly increased phagocytotic ability of AMs compared with controls (P<0.05), whereas exposure with 5 nm TiO2 reduced the phagocytotic ability of AMs when the exposure dose was 50 mg/kg. These results suggest that particle size and exposure dose may have important roles in pulmonary toxicity. The toxic effect of TiO2 nanopar-ticles in lung tissue exhibited a dose-response relationship. After exposure with TiO2 particles of >5.0 mg/kg, 5 and 21 nm TiO2 particles induced standing pulmonary lesions; and 5 nm TiO2 particles may suppress the phagocytotic ability of AMs if exposure dose was P50 mg/kg. 9Effects of Root Zone Temperature on Growth and Photosynthetic Parameters of Grafted Cucumber显示文摘Taking Cucurbita maxima and Cucurbita moschata as root stocks,and‘Jinyou No 3'cucumber as scion,the effects of different root zone temperature conditions optimal temperature( CK)( 18-20 ℃),suboptimal temperature( 13-15 ℃) and low temperature( 8-10 ℃) on the growth and photosynthesis indexes were studied. The results showed that,compared with optimal temperature( CK),suboptimal temperature and low temperature produced a significant inhibition of growth on cucumbers. The plant height,stem diameter,leaf area,number of leaves and dry weight of aboveground part were all reduced,dry weight of underground part and root shoot ratio all increased,while the inhibition was more significant at low temperature. Low and suboptimal temperature conditions also reduced SPAD value,net photosynthetic rate,transpiration rate,intercellular CO_2 concentration and stomatal conductance of the grafted cucumber. 10Loss of hnRNP A1 in murine skeletal muscle exacerbates high-fat diet-induced onset of insulin resistance and hepatic steatosis显示文摘Impairment of glucose(Glu)uptake and storage by skeletal muscle is a prime risk factor for the development of metabolic diseases.Heterogeneous nuclear ribonucleoprotein A1(hnRNP Al)is a highly abundant RNA-binding protein that has been implicated in diverse cellular functions.The aim of this study was to investigate the function of hnRNP A1 on muscle tissue insulin sensitivity and systemic Glu homeostasis.Our results showed that conditional deletion of hnRNP Al in the muscle gave rise to a severe insulin resistance phenotype in mice fed a high-fat diet(HFD).Conditional knockout mice fed a HFD showed exacerbated obesity,insulin resistance,and hepatic steatosis.In vitro interference of hnRNP Al in C2C12 myotubes impaired insulin signal transduction and inhibited Glu uptake,whereas hnRNP Al overexpression in C2C12 myotubes protected against insulin resistance induced by supraphysiological concentrations of insulin.The expression and stability of glycogen synthase(gysl)mRNA were also decreased in the absence of hnRNP A l.Mechanistically,hnRNP Al interacted with gys l and stabilized its mRNA,thereby promoting glycogen synthesis and maintaining the insulin sensitivity in muscle tissue.Taken together,our findings are the first to show that reduced expression of hnRNP Al in skeletal muscle affects the metabolic properties and systemic insulin sensitivity by inhibiting glycogen synthesis.Mingxia Zhao Lihong Shen Zijun Ouyang Manru Li Guoliang Deng Chenxi Yang Wei Zheng Lingdong Kong Xuefeng Wu Xudong Wu Wenjie Guo Ye Yin Qiang Xu Yang Sun 2020Journal of Molecular Cell Biology2020,12,4:2

17Detection of Persistent Organochlorine Pollutants in Eggs of Antarctic Seabirds Based on Gas Chromatography and Their Ecological Environment Significance显示文摘By using internal standard method based on gas chromatography-electron capture detector( GC-ECD),the contents of persistent organochlorine pollutants polychorinated biphenyls( PCBs) and organochlorine pesticides( OCPs) in eggs of seabirds( Catharacta lonnbergi,Catharacta maccormicki,Pygoscelis papua and Macronectes giganteus) breeding on King George Island,Antarctica were detected,and their ecological environment significance was discussed. The results showed that the recovery of various compounds ranged from 68. 6% to 90. 8%; relative standard deviation( RSD) was 3. 6%,and the lowest detection limits of PCBs and OCPs were 0. 8- 16 and 2- 12 pg respectively,which could meet the demands for the residue analysis of trace persistent organochlorine pollutants in the eggs of Antarctic seabirds. The contents of PCBs,DDTs,HCB and HCHs in the eggs of Antarctic seabirds were 0. 5- 515. 5,2. 0- 304. 4,0. 5- 70. 5,and 0. 5- 2. 0 ng / g respectively. In addition,the accumulation of PCBs and OCPs in the skuas was enhanced gradually through the food chain. The maximums of persistent organochlorine pollutants appeared in the eggs of C. lonnbergi,followed by the eggs of C. maccomicki.
