1Huangqi Guizhi Wuwu Decoction for treating diabetic peripheral neuropathy:a meta-analysis of 16 randomized controlled trials显示文摘OBJECTIVE:This meta-analysis was performed to systematically assess the efficacy and safety of the Chinese herbal medicine Huangqi Guizhi Wuwu Decoction(HGWWD) for treating diabetic peripheral neuropathy.DATA SOURCES:Six electronic databases,including the Cochrane Library,MEDLINE database,Chinese Biomedical Database,Chinese National Knowledge Infrastructure Database,Chinese Science and Technique Journals Database,and the Wanfang Database,were search ed on the internet for randomized controlled trials published up until 1 December 2015.The search terms included 'Chinese herbal medicine','diabetic peripheral neuropathy' and 'randomized controlled trials' in Chinese and in English.DATA SELECTION:We included randomized controlled trials using HGWWD/modified HGWWD for the treatment group,without restriction for the control group.We assessed literature quality in accordance with the Cochrane Review Handbook.A random or a fixed effects model was used to analyze outcomes using Rev Man 5.2 software.OUTCOME MEASURES:The primary outcomes were changes in symptoms and nerve conduction velocities.The secondary outcomeswere fasting blood glucose and hemorheological indexes.RESULTS:Sixteen randomized controlled trials,with a total of 1,173 patients,were included.Meta-analysis revealed that the efficacy of HGWWD for diabetic peripheral neuropathy was significantly superior compared with the control treatment(i.e.,control group)(risk ratio = 0.36,95% confidence interval(CI):0.29–0.46,Z =8.33,P < 0.00001) Compared with the control group,there was an increase in median motor nerve conduction velocity(mean difference(MD) = 3.46,95%CI:1.88–5.04,Z = 4.30,P < 0.01) and median sensory nerve conduction velocity(MD = 3.30,95%CI:2.04–4.56,Z = 5.14,P < 0.01).There was also an increase in peroneal motor nerve conduction velocity(MD = 3.22,95%CI:2.45–3.98,Z = 8.21,P < 0.01) and peroneal sensory nerve conduction velocity(MD = 3.05,95%CI:2.01–4.09,Z = 5.75,P < 0.01) in the treatment groups.No significant difference in fasting blood glucose was found between the treatment groups and the control groups(MD =-0.12,95%CI:-0.42–0.19,Z = 0.76,P = 0.45).Plasma viscosity was significantly decreased after treatment(MD =-0.11,95%CI:-0.21 to-0.02,Z = 2.30,P = 0.02).No significant difference in fibrinogen was detectable(MD =-0.53,95%CI:-1.28–0.22,Z = 1.38,P = 0.17).Four trials reported that treatment groups experienced no adverse reactions.Adverse events were not mentioned in the other 12 trials.No trial reported the incidence of complications,quality of life outcomes,or health economics.CONCLUSION:HGWWD treatment improves diabetic neurologic symptoms and ameliorates nerve conduction velocities.Our study suggests that HGWWD may have significant therapeutic efficacy for the treatment of diabetic peripheral neuropathy.However,the methodological quality of the randomized controlled trials was generally low.Larger and better-designed randomized controlled trials are required to more reliably assess the clinical effectiveness of HGWWD.Bing Pang Tian-yu Zhao Lin-hua Zhao Fang Wan Ru Ye Qiang Zhou Feng Tian Xiao-lin Tong 2016Neural Regeneration Research2016,11,8:32
2Progress in active compounds effective on ulcerative colitis from Chinese medicines显示文摘Ulcerative colitis(UC), a chronic inflammatory disease affecting the colon, has a rising incidence worldwide. The known pathogenesis is multifactorial and involves genetic predisposition, epithelial barrier defects, dysregulated immune responses, and environmental factors. Nowadays, the drugs for UC include 5-aminosalicylic acid, steroids, and immunosuppressants. Long-term use of these drugs, however, may cause several side effects, such as hepatic and renal toxicity, drug resistance and allergic reactions. Moreover, the use of traditional Chinese medicine(TCM) in the treatment of UC shows significantly positive effects, low recurrence rate, few side effects and other obvious advantages. This paper summarizes several kinds of active compounds used in the experimental research of anti-UC effects extracted from TCM, mainly including flavonoids, acids, terpenoids, phenols, alkaloids, quinones, and bile acids from some animal medicines. It is found that the anti-UC activities are mainly focused on targeting inflammation or oxidative stress, which is associated with increasing the levels of anti-inflammatory cytokine(IL-4, IL-10, SOD), suppressing the levels of pro-inflammatory cytokines(TNF-α, IL-1β, IL-6, IL-8, IL-23, NF-κB, NO), reducing the activity of MPO, MDA, IFN-γ, and iNOS. 5Identification of rice cultivars with low brown rice mixed cadmium and lead contents and their interactions with the micronutrients iron,zinc,nickel and manganese显示文摘Paddy fields in mining areas are usually co-contaminated by a cocktail of mixed toxic heavy metals(e.g.,Cd and Pb in Pb/Zn mines).However,previous studies on rice cultivars screened for effective metal exclusion have mostly focused on individual metals,and have been conducted under pot-trial or hydroponic solution conditions.This study identified rice cultivars with both low Cd and Pb accumulation under Cd-and Pb-contaminated field conditions,and the interactions of the toxic elements Cd and Pb with the micronutrient elements Fe,Zn,Mn and Ni were also studied.Among 32 rice cultivars tested,there were significant differences in Cd(0.06-0.59 mg/kg) and Pb(0.25-3.15 mg/kg) levels in their brown rice,and similar results were also found for the micronutrient elements.Significant decreases in concentrations of Fe and Mn were detected with increasing Cd concentrations and a significant elevation in Fe,Mn and Ni with increasing Pb concentrations.A similar result was also shown by Cd and Ni.Three cultivars were identified with a combination of low brown rice Cd and Pb,high micronutrient and grain yield(Wufengyou 2168,Tianyou 196 and Guinongzhan).Present results suggest that it is possible to breed rice cultivars with low mixed toxic element(Cd,Pb) and high micronutrient contents along with high grain yields,thus ensuring food safety and quality.Bing Li Xun Wang Xiaoli Qi Lu Huang Zhihong Ye 2012Journal of Environmental Sciences2012,24,10:18

7Noise Reduction of an Axial Piston Pump by Valve Plate Optimization显示文摘Current researches mainly focus on the investigations of the valve plate utilizing pressure relief grooves. However,air?release and cavitation can occur near the grooves. The valve plate utilizing damping holes show excellent perfor?mance in avoiding air?release and cavitation. This study aims to reduce the noise emitted from an axial piston pump using a novel valve plate utilizing damping holes. A dynamic pump model is developed,in which the fluid properties are carefully modeled to capture the phenomena of air release and cavitation. The causes of di erent noise sources are investigated using the model. A comprehensive parametric analysis is conducted to enhance the understanding of the e ects of the valve plate parameters on the noise sources. A multi?objective genetic algorithm optimization method is proposed to optimize the parameters of valve plate. The amplitudes of the swash plate moment and flow rates in the inlet and outlet ports are defined as the objective functions. The pressure overshoot and undershoot in the piston chamber are limited by properly constraining the highest and lowest pressure values. A comparison of the various noise sources between the original and optimized designs over a wide range of pressure levels shows that the noise sources are reduced at high pressures. The results of the sound pressure level measurements show that the optimized valve plate reduces the noise level by 1.6 d B(A) at the rated working condition. 8Vascular endothelial growth factor concentration in vitreous humor of patients with severe proliferative diabetic retinopathy after intravitreal injection of conbercept as an adjunctive therapy for vitrectomy显示文摘Background:Proliferative diabetic retinopathy(PDR)is a progressive stage of diabetic retinopathy featured by the formation of neovascular and proliferative membrane.Vascular endothelial growth factor(VEGF)acts as a pivot factor in the development of neovascularization.This study was to investigate the changes of intravitreal VEGF concentrations of severe PDR after intravitreal injection of conbercept(IVC)and its potential advantages to the following vitrectomy.Methods:This was a prospective,interventional,randomized controlled study.Sixty eyes(60 patients)with severe PDR and 20 eyes from 20 patients with rhegmatogenous retinal detachment complicated with proliferative vitreoretinopathy were enrolled in this study.PDR eyes were randomly assigned to three groups by sortation randomization method with 20 eyes in each based on the interval of preoperative IVC(group A:7 days,group B:14 days,group C:non-IVC).Another 20 eyes without diabetes were enrolled as the non-diabetic control group(group D),receiving PPV directly.Vitreous specimens of all 80 patients were collected and evaluated afterwards.The intravitreal VEGF concentration of the four groups,and the total surgical time and the intraoperative bleeding rate of the PDR groups were recorded.Results:The mean intravitreal VEGF concentrations of groups A-D were 66.6±43.3,93.1±52.3,161.4±106.1 and 1.8±1.2 pg/mL,respectively.It increased significantly in PDR patients(groups A,B and C)(P=0.002,<0.001,and<0.001,respectively).PDR patients with preoperative IVC(groups A and B)presented significantly lower VEGF concentrations(P<0.001 and 0.001),intraoperative bleeding rates(P=0.004)and total surgical time(P<0.001,P=0.003)compared with group C.No statistical differences were presented between groups A and B on the three parameters.Conclusion:Seven days and 14 days of preoperative IVC are equally efficient and safe for the vitrectomy of severe PDR patients through decreasing vitreous VEGF concentrations,intraoperative bleeding rate and total surgical times.Bing Li Meng-Da Li Jun-Jie Ye Zhe Chen Zi-Jian Guo Yu Di 2020Chinese Medical Journal2020,,6:14
9Xiaotan Sanjie decoction attenuates tumor angiogenesis by manipulating Notch-1-regulated proliferation of gastric cancer stem-like cells显示文摘AIM: To determine the underlying mechanisms of action and influence of Xiaotan Sanjie(XTSJ) decoction on gastric cancer stem-like cells(GCSCs). METHODS: The gastric cancer cell line MKN-45 line was selected and sorted by FACS using the cancer stem cell marker CD44; the stemness of these cells was checked in our previous study. In an in vitro study, the expression of Notch-1, Hes1, Vascular endothelial growth factor(VEGF), and Ki-67 in both CD44-positive gastric cancer stem-like cells(GCSCs) and CD44-negative cells was measured by Western blot. The effect of XTSJ serum on cell viability and on the above markers was measured by MTT assay and Western blot, respectively. In an in vivo study, the ability to induce angiogenesis and maintenance of GCSCs in CD44-positive-MKN-45- and CD44-negative-engrafted mice were detected by immunohistochemical staining using markers for CD34 and CD44, respectively. The role of XTSJ decoction in regulating the expression of Notch-1, Hes1, VEGF and Ki-67 was measured by Western blot and real-time polymerase chain reaction.RESULTS: CD44+ GCSCs showed more cell proliferation and VEGF secretion than CD44-negative cells in vitro, which were accompanied by the high expression of Notch-1 and Hes1 and positively associated with tumor growth(GCSCs vs CD44-negative cells, 2.72 ± 0.25 vs 1.46 ± 0.16, P < 0.05) and microvessel density(MVD)(GCSCs vs CD44-negative cells, 8.15 ± 0.42 vs 3.83 ± 0.49, P < 0.001) in vivo. XTSJ decoction inhibited the viability of both cell types in a dose-dependent manner in vitro. Specifically, a significant difference in the medium-(82.87% ± 6.53%) and high-dose XTSJ groups(77.43% ± 7.34%) was detected at 24 h in the CD44+ GCSCs group compared with the saline group(95.42% ± 5.76%) and the low-dose XTSJ group(90.74% ± 6.57%)(P < 0.05). However, the efficacy of XTSJ decoction was reduced in the CD44- groups; significant differences were only detected in the high-dose XTSJ group at 48 h(78.57% ± 6.94%) and 72 h(72.12% ± 7.68%) when compared with the other CD44- groups(P < 0.05). Notably, these differences were highly consistent with the Notch-1, Hes1, VEGF and Ki-67 expression in these cells. Similarly, in vivo, XTSJ decoction inhibited tumor growth in a dose-dependent manner. A significant difference was observed in the medium(1.76 ± 0.15) and high-dose XTSJ(1.33 ± 0.081) groups compared with the GCSCs control group(2.72 ± 0.25) and the low-dose XTSJ group(2.51 ± 0.25)(P < 0.05). We also detected a remarkable decrease of MVD in the medium-(7.10 ± 0.60) and high-dose XTSJ(5.99 ± 0.47) groups compared with the GCSC control group(8.15 ± 0.42) and the low-dose XTSJ group(8.14 ± 0.46)(P < 0.05). Additionally, CD44 expression was decreased in these groups [medium-(4.43 ± 0.45) and high-dose XTSJ groups(3.56 ± 0.31) vs the GCSC control(5.96 ± 0.46) and low dose XTSJ groups(5.91 ± 0.38)](P < 0.05). 10Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable.Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun 2020Cellular & Molecular Immunology2020,17,6:13

15Effect of emodin on pancreatic fi brosis in rats显示文摘AIM: To establish the rats model of chronic fibrosing pancreatitis and to prove the anti-fibrotic effect of emodin in chronic pancreatitis with fi brosis. METHODS: Fifty rats were randomly divided into five groups, 10 rats in each group. Trinitrobenzene sulfonic acid (TNBS) was infused into the pancreatic duct to induce chronic pancreatitis in rats (except for normal group). Emodin-treated rats were fed with different doses of emodin (20, 40 and 80 mg/kg body weight) for 28 d, while normal group and control group received 0.9% sodium chloride solution. Serum levels of hyaluronic acid (HA) and laminin (LN) were determined by radioimmunoassay. Histopathological alterations were studied by optical microscopy. Expression of collagen was also examined while transforming growth factor-beta-1 (TGF-β1) was localized by immunochemistry. RESULTS: In emodin-treated rats, the serum levels of HA and LN were decreased significantly (HA, 62.2 ± 19.3 μg/L vs 112.7 ± 26.5 μg/L, P < 0.05; LN 44.3 ± 10.4 μg/L vs 86.2 ± 16.5 μg/L, P < 0.05); the degree of fibrosis was ameliorated observably; the expression of collagen in pancreatic tissue was reduced especially in high-dose emodin-treated group (36% ± 5% vs 42% ± 6%, P < 0.05); with the increased doses of emodin, the expression of TGF-β1 was declined, compared with those in control group. CONCLUSION: Emodin has an anti-fibrotic effect on pancreatic fibrosis in rats. 16Clinicopathological, immunohistochemical, and ultrastructural study of 13 cases of melanotic schwannoma显示文摘Background Melanotic schwannoma is a rare variant of schwannoma composed of melanin-producing cellswith ultrastructural features of schwann cells.The description of the course of the tumors differs somewhat, but itis generally considered as a benign lesion.We investigated the clinicopathologic features, immunophenotypes,and ultrastructural features of 13 patients with nonpsammomatous melanotic schwannoma (NPMS).Methods Tumor specimens of each patient were sectioned and stained with hematoxylin-eosin, Fontana-Masson, Prussian blue, and periodic acid-Schiff (PAS).Immunohistochemical markers such as S-100, Leu-7,HMB-45, Melan-A, CK, EMA, vimentin, GFAP, laminin, collagenⅣ and MIB-1 were detected with theEnvision immunohistochemical staining method.Four of the cases were observed by electron microscopy.Results Of the 13 patients, 8 were male and 5 female, aged from 11 to 92 years (mean, 38.6 years).Thetumor sites included the spinal nerve root (5 patients), cranial nerve (1), greater omentum (1), subcutaneoustissue (3), mesentery (1), bone (1) and mediastinum(1).Eleven patients were followed up for over2 years,with a mean of 5.9 years.One patient (9.1%) with a primary tumor in the greater omentum developed anotherprimary tumor of the same type in the subcutaneous tissue of the abdominal wall after the first operation.Localrecurrence of the tumor was seen in2 patients (18.2%).One patient (9.1%) showed the local recurrence andmetastasis.Seven patients (63.6%) showed no evidence of the recurrence or metastasis.Grossly, all tumorswere well-circumscribed and the gross findings were suggestive of melanin-containing tumors.The tumor wascomposed of spindled and epithelioid cells with abundant intracytoplasmic melanin pigments.Nuclei were roundand contained delicate, evenly distributed chromatins as well as small, distinct nucleoli.In some areas, thenucleoli were large and prominent.Rare mitoses were seen in mostlesions exceptthe larger omentumlesion.Thepigment was shown to be positive for the Fontana-Masson and negative for Prussian blue and PAS.Immunohistochemical staining for S-100, Leu-7, HMB-45, Melan-A, and vimentin were strongly positive.Linear immunoreactions of both laminin and collagenⅣ was detected in all patients.Ultrastructurally, numerouselongated tumor-cell processes, duplicated basement membrane and melanosomes were observed in alldevelopmental stages.Conclusions Histologically, melanotic schwannoma is a rare variant of schwannoma composed of melanin-producing cells with ultrastructural features ofschwann cells.Distinguishing between this tumor andmalignant melanoma is of paramount importance inplanning of management. Immunohistochemically,combined use of laminin and collagenⅣ is valuablein distinguishing melanotic schwannoma frommalignant melanoma. Wide local resection andadditional radiotherapy should be advocated.Furtherstudies including cytogenetic or molecular biology arestill required to better delineate melanoticschwannoma from malignant melanoma. 17The Craniofacial Morphology in Adult Patients with Unoperated Isolated Cleft Palate显示文摘Objective:To address the effect of intrinsic factors on craniofacial growth by analyzing the craniofacial morphology of unoperated isolated cleft palate in Chinese adult.Materials and Methods:This study included 37 nonsyndromic isolated cleft palate and 39 age and gender matched non-clefts.Twenty-six cephalometric measurements were employed to evaluate the facial morphology.Independent samples T test and Mann-Whitney U were used for comparison.Significant difference was defined at 95% level.Results:Data from this study showed patients with unoperated isolated cleft palate have a reduced maxillary sagittal length(ANS-PMP,A-PMP,P<0.05),a smaller ANB angle(ANB,P<0.05) and a retrusive ANS point(S-N-ANS,P<0.05;Ba-N-ANS,P<0.05).Measurements descripted position of maxilla(S-Ptm,P>0.05),depth of bony pharynx(Ba-PMP,P>0.05),anterior and posterior maxillary height(N-ANS,P>0.05;R-PMP,P>0.05) and mandible morphology(including linear measurements and angle measurements) did not show any significant difference between case and control groups.Conclusions:Patients with isolated cleft palate were characterized by maxillary retrusion.Mandible morphology and cranial basal morphology in isolated cleft palate showed no significant difference with nonclefts.Patients with isolated cleft palate are more vulnerable to cross bite than nonclefts.Intrinsic deficiencies did detrimental effect on maxilla sagittal length,but did no detrimental effect on maxilla position,mandible size and position.Zhongtai Ye Xue Xu Adalet Ahmatjian Bing Shi 2013Bone Research2013,1,2:9
18Blood Pressure Associated with Arsenic Methylation and Arsenic Metabolism Caused by Chronic Exposure to Arsenic in Tube Well Water显示文摘Objective The effects of arsenic exposure from drinking water, arsenic metabolism, and arsenic methylation on blood pressure(BP) were observed in this study. Methods The BP and arsenic species of 560 participants were determined. Logistic regression analysis was applied to estimate the odds ratios of BP associated with arsenic metabolites and arsenic methylation capability. Results BP was positively associated with cumulative arsenic exposure(CAE). Subjects with abnormal diastolic blood pressure(DBP), systolic blood pressure(SBP), and pulse pressure(PP) usually had higher urinary i As(inorganic arsenic), MMA(monomethylated arsenic), DMA(dimethylated arsenic), and TAs(total arsenic) than subjects with normal DBP, SBP, and PP. The i As%, MMA%, and DMA% differed slightly between subjects with abnormal BP and those with normal BP. The PMI and SMI were slightly higher in subjects with abnormal PP than in those with normal PP. Conclusion Our findings suggest that higher CAE may elevate BP. Males may have a higher risk of abnormal DBP, whereas females have a higher risk of abnormal SBP and PP. Higher urinary i As may increase the risk of abnormal BP. Lower PMI may elevate the BP. 19Combination of iTRAQ proteomics and RNA-seq transcriptomics reveals multiple levels of regulation in phytoplasma-infected Ziziphus jujuba Mill显示文摘Jujube witches’broom(JWB)is caused by infection with a phytoplasma.A multi-omics approach was taken during graft infection of jujube by JWB-infected scion through the analysis of the plant transcriptome,proteome and phytohormone levels.A high number of differentially expressed genes(DEGs)were identified 37 weeks after grafting(WAG),followed by observation of typical symptoms of JWB at 48 WAG.At 37 WAG,the majority of the upregulated DEGs and differentially expressed proteins(DEPs)were related to flavonoid biosynthesis,phenylalanine metabolism and phenylpropanoid biosynthesis.Two of the four upregulated proteins were similar to jasmonate-induced protein-like.Among the downregulated genes,the two most populated GO terms were plant–pathogen interaction and plant hormone signal transduction(mainly for tryptophan metabolism).Moreover,phytoplasma infection resulted in reduced auxin content and increased jasmonate content,indicating that auxin and jasmonic acid have important roles in regulating jujube responses during the first and second stages of phytoplasma infection.At 48 WAG,the two largest groups of upregulated genes were involved in phenylpropanoid biosynthesis and flavonoid biosynthesis.Both genes and proteins involved in carbon metabolism and carbon fixation in photosynthetic organisms were downregulated,indicating that photosynthesis was affected by the third stage of phytoplasma infection.Xia Ye Huiyu Wang Peng Chen Bing Fu Mengyang Zhang Jidong Li Xianbo Zheng Bin Tan Jiancan Feng 2017Horticulture Research2017,4,1:9
