1A novel hepatitis B virus genotyping system by using restriction fragment length polymorphism patterns of S gene amplicons显示文摘AIM: Traditional hepatitis B virus (HBV) genotyping methods using restriction fragment length polymorphism (RFLP) can reliably identify genotypes A to F. As HBV genotypes G and H have been recently identified, this study was to establish an accurate and simple genotyping method for all eight HBV genotypes (A to H).METHODS: Two hundred and forty HBV small S sequences obtained from GeneBank were analysed for restriction enzyme sites that would be genotype-specific. Restriction patterns following digestion with restriction enzymes BsrⅠ, StyⅠ, DpnⅠ, HpaⅡ, and EaeⅠ, were determined to identify all eight HBV genotypes. Mixed genotype infections were confirmed by cloning and further RFLP analysis.RESULTS: The new genotyping method could identify HBV genotypes A to H. Genotypes B and C could be determined by a single step digestion with BsrI and StyI in parallel. This was particularly useful in the Far East where genotypes B and C are predominant. Serum samples from 187 Chinese HBV carders were analysed with this genotyping system, and the genotype distribution was 1.1% (2), 51.9% (97), 40.6% (76) and 4.8% (9) for genotypes A, B, C, and D, respectively. Mixed genotypes were found in only 3 patients (1.6%). Sequence data analysis confirmed the validity of this new method.CONCLUSION: This HBV genotyping system can identify all eight HBV genotypes. 4miR-503-3p promotes epithelialemesenchymal transition in breast cancer by directly targeting SMAD2 and E-cadherin显示文摘Although progress in clinical and basic research has significantly increased our understanding of breast cancer, little is known about the molecular mechanism underlying breast cancer metastasis. Identification of effective therapeutic targets to prevent breast cancer metastasis is urgently needed. The function of mi R-503-3p has been investigated in other cancers, but its role in breast cancer remains undefined.Here, we found that mi R-503-3p was overexpressed in breast cancer tissue and plasma compared with adjacent normal breast tissue and with plasma from healthy individuals. Moreover, we identified mi R-503-3p to be an oncogene of breast cancer cell proliferation, migration and invasion. Upregulation of mi R-503-3p in breast cancer cells inhibited expression of epithelialemesenchymal transition(EMT)-related protein SMAD2 and the epithelial marker protein E-cadherin by directly binding to their m RNA30 untranslated region, whereas increased expression of mesenchymal marker proteins, including vimentin and N-cadherin. 5Origin and phylogenetic analysis of Tibetan Mastiff based on the mitochondrial DNA sequence显示文摘At present,the Tibetan Mastiff is the oldest and most ferocious dog in the world. However,the origin of the Tibetan Mastiff and its phylogenetic relationship with other large breed dogs such as Saint Bernard are unclear. In this study,the primers were designed accord-ing to the mitochondrial genome sequence of the domestic dog,and the 2,525 bp mitochondrial sequence,containing the whole sequence of Cytochrome b,tRNA-Thr,tRNA-Pro,and control region of the Tibetan Mastiff,was obtained. Using grey wolves and coyotes as out-groups,the Tibetan Mastiff and 12 breeds of domestic dogs were analyzed in phylogenesis. Tibetan Mastiff,domestic dog breeds,and grey wolves were clustered into a group and coyotes were clustered in a group separately. This indicated that the Tibetan Mastiff and the other domestic dogs originated from the grey wolf,and the Tibetan Mastiff belonged to Carnivora,Canidae,Canis,Canis lupus,Canis lupus familiaris on the animal taxonomy. In domestic dogs,the middle and small breed dogs were clustered at first; German Sheepdog,Swedish Elkhound,and Black Russian Terrier were clustered into one group,and the Tibetan Mastiff,Old English Sheepdog,Leonberger,and Saint Bernard were clustered in another group. This confirmed the viewpoint that many of the famous large breed dogs worldwide such as Saint Bernard possibly had the blood lineage of the Tibetan Mastiff,based on the molecular data. 7Cadmium accumulation and tolerance of two castor cultivars in relation to antioxidant systems显示文摘To investigate the effects of Cd on tolerance and antioxidant activities of castor, two different castor(Ricinus communis L.) cultivars(Zibo No. 5 and Zibo No. 8) were used for a hydroponic experiment(0, 1 and 2 mg/L Cd) and a pot experiment using Cd-contaminated soil(34 mg/kg) with the addition of ethylenedinitrilotetraacetic acid(EDTA). The results indicated that there were significant differences between the two cultivars with respect to Cd uptake in shoots(113–248 mg/kg for Zibo No. 5 and 130–288 mg/kg Zibo No. 8), biomass tolerance indexes(64.9%–74.6% for Zibo No. 5 and 80.1%–90.9% for Zibo No. 8) in the hydroponic experiment and survival rates(0% for Zibo No. 5 and 100% for Zibo No. 8)determined by the addition of EDTA in the pot experiment, suggesting that Zibo No. 8 has higher tolerance than Zibo No. 5. Moreover, the castor cultivars have low bioconcentration factors(4.80% for Zibo No. 5 and 5.43% for Zibo No. 8) and low translocation factors(<1%).Consequently, Zibo No. 8 can participate in Cd phytostabilization in highly Cd-polluted areas. The results indicated that glutathione(GSH) as a non-enzymatic antioxidant, and antioxidant enzymes including superoxide dismutase(SOD), catalase(CAT) and guaiacol peroxidase(GPX), were cultivar- and dose-dependent. 8Experimental investigation on color variation mechanisms of structural light in Papilio maackii ménétriès butterfly wings显示文摘The body color in animals results from billions of years of their natural evolution in order to evade natural enemies, catch quarries or display themselves beauty, in-vestigation on mechanisms of structural light is an important aspect of bionics. Based on the phenomenon of Papilio maackii ménétriès’ blue scales changing into green ones immediately after dropping some alcohol aqua on the underwing sur-face and soon returning back to the original color, the relationship between micro-structure, optics characteristic of scales and changing color effect were studied using the Olympus Stereomicroscope, Scanning Electron Microscopy (SEM), Transmission Electron Microscopy (TEM) and Ultraviolet (UV)-Visible Spectropho-tometer. The color variation mechanisms of blue scales of Papilio maackii mé-nétriès in Chinese Northeast were revealed in this paper. When visible lights trav-eled through the concaver structure with multilayer reflector and the filled medium with different refractive indices, the reflected lights in definite wavelengths pro-duced interference and color at that wavelength came into being. 9Application of Pb isotopic tracing technique to constraining the source of Pb in the West Lake Longjing tea显示文摘This paper dealt with the Pb contents and Pb isotopic composition of the West Lake Longjing tea. The results showed that in the tea leaves, from young leaf →old leaf →tea limb, the Pb contents tend to increase gradu-ally from 1.63 →4.84 →6.07×10-6, wich revealed that the Pb was accumulated gradually in the tea. After cleaned, the Pb contents of tea leaves were significantly reduced. This indicated that the deposits on the surface of tea leaves made a great contributuion to Pb contents. The survey results for soils in the relevant tea gardens showed that soil from the Longjing tea garden has higher Pb contents, with an average level of 49.6×10-6, two times those of common soils (24×10-6) in China. Results of the systematic analysis for tea, tea garden soil and the samples with the relevant background of the Pb isotopic composition displayed that the Pb isotopic ratio of tea is 206Pb/207Pb=1.164±0.005 (2σ). The ratio of 206Pb/207Pb for the soil gradually decreased from residue phase, soil dilute acid extract phase, and then to urban topsoil, i.e., 1.175 →1.171 →1.170. The 208Pb/(206Pb+207Pb) ratios also show a similar variation trend. 206Pb/207Pb ratios in the samples with the relevant background were: vehicle exhaust, 1.124; coal-combustion, 1.156; atmosphere, 1.168; and water, 1.166. Comparative studies have shown that Pb pollution is popular in the environ-mental media (soil, atmosphere, water) in Hangzhou. With the aggravation of Pb pollution, the Pb isotopic composi-tion gradually changed from the natural background (soil residues) to the direction of automobile exhaust. This phe-nomenon could illustrate that the pollution source was the vehicle exhaust, while the coal-combustion contributed little to environmental pollution in Hangzhou. The Pb of the Longjing tea came mostly from soluble phase Pb in the polluted soil. 10A Spore-Pollen Assemblage from the Damoguaihe Formation in the Tamutsag Basin, Mongolia and Its Geological Implication显示文摘Based on the palynological data from Well Ta-19-36 and Well Ta-19-37 in the Ta'nan Sag, and Well Ta-22-1 and Well Ta-21-1 in the Bayin Gobi Sag, a spore-pollen assemblage from the Damoguaihe Formation is named as Cicatricosisporites minutaestriatus- Aequitriradites spinulosus-Protopinus sp. assemblage in the Tamutsag Basin, Mongolia. The assemblage is characterized by abundant gymnosperm pollen and diverse fern spores, counted 46.35%–65.57% and 34.43%–52.58% in percentage respectively. Among the gymnosperm pollen, Pinuspollenites sp.(2.66%–16.94%), Protopinus sp.(0–11.38%) and Protopicea sp.(0–10.81%) are dominant; and Alisporites sp., Cerebropollenites sp., Podocarpidites sp., and Abietineaepollenites sp. are common. Some important elements, such as Chasmatosporites sp., Callialasporites prominulus, Parvisaccites sp., Parcisporites sp., Jiaohepollis cf. annulatus, J. bellus, J. verus, Polycingulatisporites sp., Schizaeoisporites certus, Classopollis annulatus are seen. Within the fern spores, Osmundacidites wellamanii(0.85%–4.93%), Appendicisporites sp.(0–4.45%), Baculatisporites comaumensis(0.80%–2.87%), Cicatricosisporites sp.(0.51%–2.66%), C. minor(0–2.14%), Foraminisporis asymmetricus(0–2.40%), Aequitriradites sp.(0– 2.19%) and Cyathidites minor(0–2.13%) are dominant; and some specie of Densoisporites, Cooksonites, Impardecispora, Pilosisporites, Schizaeosprites, Fixisporites, Leptolepidites, Trilobosporites, Kuylisporites, Klukisporites, Hsuisporites, Couperisporites, Maculatisporites are seen. The angiosperm pollen are rare in the assemblage, characterized by Clavatipollenites sp.(0–0.80%) and Songipollis sp.(0–0.27%). The geological age of the Damoguaihe Formation is assigned to Hauterivian – Barremian of Early Cretaceous according to the palynological data, due to the fact that there existed a lot of diverse spores of the family Schizaeaceae and prosperous gymnosperm saccat pollen which the corpus and sacci are differentiated completely, and the presence of early angiosperm pollen of Clavatipollenites. However, the zircon U-Pb isotopic dating of the volcanic rocks in the Nantun Formation indicated that the overlying strata of the Nantun Formation, i.e. the Damoguaihe Formation must be younger than 127.0±2.0–137.9±1.5 Ma. This is consistent with the palynological data. Furthermore, the vegetation reconstructed on the palynological data of the Damoguaihe Formation is conifer forest with shrubs and grassland, belonging to the semi-humid or humid middle to south subtropical climate. 11RAB31 marks and controls an ESCRT-independent exosome pathway显示文摘Exosomes are generated within the multivesicular endosomes(MVEs)as intraluminal vesicles(ILVs)and secreted during the fusion of MVEs with the cell membrane.The mechanisms of exosome biogenesis remain poorly explored.Here we identify that RAB31 marks and controls an ESCRT-independent exosome pathway.Active RAB31,phosphorylated by epidermal growth factor receptor(EGFR),engages flotillin proteins in lipid raft microdomains to drive EGFR entry into MVEs to form ILVs,which is independent of the ESCRT(endosomal sorting complex required for transport)machinery.Active RAB31 interacts with the SPFH domain and drives ILV formation via the Flotillin domain of flotillin proteins.Meanwhile,RAB31 recruits GTPase-activating protein TBC1D2B to inactivate RAB7,thereby preventing the fusion of MVEs with lysosomes and enabling the secretion of ILVs as exosomes.These findings establish that RAB31 has dual functions in the biogenesis of exosomes:driving ILVs formation and suppressing MVEs degradation,providing an exquisite framework to better understand exosome biogenesis.Denghui Wei Weixiang Zhan Ying Gao Liyan Huang Run Gong Wen Wang Ruhua Zhang Yuanzhong Wu Song Gao Tiebang Kang 2021Cell Research2021,31,2:12
12Estimation of the Effects of Maize Straw Return on Soil Carbon and Nutrients Using Response Surface Methodology显示文摘Straw incorporation is generally considered an effective agricultural management practice that improves nutrient cycling and maintains soil fertility. To study the interactive effects of straw returning factors on soil organic carbon and available nutrients, a17-month(May 6, 2016 to October 6, 2017) experiment was conducted on straw incorporation by using response surface methodology under a three-factor(straw length, amount, and burying depth), five-level quadratic orthogonal rotation experimental design. Weight was assigned to each indicator for soil carbon and nutrients and then a comprehensive indicator was established. Then, a second-order polynomial model of the three straw returning factors was established using response surface methodology. Results indicated that17 months after straw incorporation, straw amount and burying depth had significant effects on the comprehensive indicator of soil carbon and nutrients. Straw length and the interactions of straw amount and burying depth showed no significant effects on the comprehensive indicator of soil carbon and nutrients. 13Fluorination vs. chlorination: a case study on high performance organic photovoltaic materials显示文摘Halogenation is a very efficient chemical modification method to tune the molecular energy levels, absorption spectra and molecular packing of organic semiconductors. Recently, in the field of organic solar cells(OSCs), both fluorine-and chlorinesubstituted photovoltaic materials, including donors and acceptors, demonstrated their great potentials in achieving high power conversion efficiencies(PCEs), raising a question that how to make a decision between fluorination and chlorination when designing materials. Herein, we systemically studied the impact of fluorination and chlorination on the properties of resulting donors(PBDB-T-2 F and PBDB-T-2 Cl) and acceptors(IT-4 F and IT-4 Cl). The results suggest that all the OSCs based on different donor and acceptor combinations can deliver good PCEs around 13%–14%. Chlorination is more effective than fluorination in downshifting the molecular energy levels and broadening the absorption spectra. The influence of chlorination and fluorination on the crystallinity of the resulting materials is dependent on their introduction positions. 15Spatial and Temporal Changes of Arable Land Driven by Urbanization and Ecological Restoration in China显示文摘Since the industrial revolution, human activities have both expanded and intensified across the globe resulting in accelerated land use change. Land use change driven by China’s development has put pressure on the limited arable land resources, which has affected grain production. Competing land use interests are a potential threat to food security in China. Therefore, studying arable land use changes is critical for ensuring future food security and maintaining the sustainable development of arable land. Based on data from several major sources, we analyzed the spatio-temporal differences of arable land among different agricultural regions in China from 2000 to 2010 and identified the drivers of arable land expansion and loss. The results revealed that arable land decreased by 5.92 million ha or 3.31%. Arable land increased in the north and decreased in the south of China. Urbanization and ecological restoration programs were the main drivers of arable land loss, while the reclamation of other land cover types(e.g., forest, grassland, and wetland) was the primary source of the increased arable land. The majority of arable land expansion occurred in the Northwest, but the centroid for grain production moved to northeast, which indicated that new arable land was of poor quality and did not significantly contribute to the grain production capacity. 17Glass-forming ability and crystallization of Mg-Ni amorphous alloys with Y addition显示文摘Mg86.33Ni13.67-xYx(x=0, 1, 3, 6, 10) amorphous alloys were obtained by single-roller melt-spinning technique and the effect of Y addition on the glass forming ability(GFA), crystallization and micro-hardness of Mg-Ni alloys were studied. The results show that the GFA of Mg86.33Ni13.67-xYx(x=0, 1, 3, 6, 10) is improved successfully with the Y addition. The highest GFA appeares at x=6, while the reduced glass transition temperature (Trg) is 0.5225 and the supercooled liquid region(ΔTx) is 42.06 K; the position of the main diffraction halo is different for the alloys, and the maximum of the main diffraction halo of alloys with x=0, 1, 3 corresponds to the main peaks of a metastable fcc-Mg6Ni or fcc-Mg6Ni + Ni-Y intermetallic phases, and for the alloys with x=6, 10, it corresponds to Mg-Y and Ni-Y intermetallic phases; the micro-hardness of the alloys is improved with Y additions, and the highest micro-hardness is obtained at x=6 at.%, which is 960 MPa.Liu Liyan Teng Xinying Wang Yongjie Liu Teng 2012Rare Metals2012,31,3:8
18Analysis of genetic architecture and favorable allele usage of agronomic traits in a large collection of Chinese rice accessions显示文摘Genotyping and phenotyping large natural populations provide opportunities for population genomic analysis and genome-wide association studies(GWAS). Several rice populations have been re-sequenced in the past decade;however, many major Chinese rice cultivars were not included in these studies. Here, we report large-scale genomic and phenotypic datasets for a collection mainly comprised of 1,275 rice accessions of widely planted cultivars and parental hybrid rice lines from China. The population was divided into three indica/Xian and three japonica/Geng phylogenetic subgroups that correlate strongly with their geographic or breeding origins. We acquired a total of 146 phenotypic datasets for 29 agronomic traits under multi-environments for different subpopulations. With GWAS, we identified a total of 143 significant association loci, including three newly identified candidate genes or alleles that control heading date or amylose content. Our genotypic analysis of agronomically important genes in the population revealed that many favorable alleles are underused in elite accessions, suggesting they may be used to provide improvements in future breeding efforts. 19TRAP1 Shows Clinical Significance and Promotes Cellular Migration and Invasion through STAT3/MMP2 Pathway in Human Esophageal Squamous Cell Cancer显示文摘Tumor necrosis factor receptor-associated protein 1(TRAP1), an important member of mitochondrial heat shock protein 90 family, is involved in multiple biological processes in several types of tumors. However, its pathological role in esophageal squamous cell cancer(ESCC) remains unknown. Herein, we demonstrated the clinical value of TRAP1, and its role in apoptosis and motility in ESCC. The clinical potential of TRAP1 was investigated through immunohistochemical analysis in 328 ESCC samples, which revealed that strong TRAP1 expression was associated with increased risk of lymph node metastasis, while high TRAP1 expression correlated with poor prognosis. Expression of TRAP1 was found to be an independent prognostic factor for patients with ESCC. Additionally, the upregulation of TRAP1 antagonized cisplatin-induced apoptosis while its downregulation sensitized cells to cisplatin-induced apoptosis. As revealed by the transwell assay, TRAP1 overexpression promoted cellular migration and invasion as compared to the control groups. In contrast,silencing of endogenous TRAP1 expression attenuated the ability of migration and invasion. Finally, the molecular mechanism investigated in the present study demonstrated that TRAP1-mediated migration and invasion occurred through STAT3/MMP2 signaling pathway. 20Purifi cation, characterization and hypoglycemic activity of glycoproteins obtained from pea (Pisum sativum L.)显示文摘This study aimed to isolate and characterize the structures of glycoproteins from peas and determine their hypoglycemic activity.The crude pea glycoproteins(PGP)were extracted by hot water and purified by diethylaminoethyl(DEAE)-Sepharose chromatography and Sephadex G-100 size-exclusion chromatography in sequence.Then three main fractions were obtained,namely PGP1,PGP2 and PGP3,with molecular weights of 897615,846740 and 1194692 Da,respectively.The physical and chemical properties of the three fractions were evaluated and compared by Fourier transform infrared spectroscopy(FT-IR),nuclear magnetic resonance(NMR),scanning electron microscope(SEM),high performance liquid chromatography(HPLC)and other analytical techniques.The fraction PGP2 with the highest hypoglycemic activity,was screened using the Caco-2 monolayer cell model.It can inhibit the uptake of glucose in the small intestine,as well as the activities of maltase and sucrase.After simulated gastrointestinal digestion,PGP2 signifi cantly enhanced the inhibitory effect of α-glucosidase,and slightly reduced the inhibitory ability ofα-amylase.In summary,PGP2 possessed strong hypoglycemic activity after digestion.These results indicated that PGP2 has the potential to be developed into a functional food or natural medicine for the treatment of type 2 diabetes mellitus.Gaoyixin Qin Wu Xu Junping Liu Liyan Zhao Guitang Chen 2021Food Science and Human Wellness2021,10,3:8
