1Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases,especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed.Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung,heart and artery,the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging.With age,expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier,cardiomyocytes,and vascular endothelial cells.We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore,treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether,our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly.Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu 2021Cell Research2021,31,4:10
2Geochronology, Geochemistry and Sr-Nd-Pb-Hf Isotopes of No. I Complex from the Shitoukengde Ni–Cu Sulfide Deposit in the Eastern Kunlun Orogen, Western China: Implications for the Magmatic Source, Geodynamic Setting and Genesis显示文摘The Shitoukengde Ni-Cu deposit, located in the Eastern Kunlun Orogen, comprises three mafic–ultramafic complexes, with the No. I complex hosting six Ni-Cu orebodies found recently. The deposit is hosted in the small ultramafic bodies intruding Proterozoic metamorphic rocks. Complexes at Shitoukengde contain all kinds of mafic-ultramafic rocks, and olivine websterite and pyroxene peridotite are the most important Ni-Cu-hosted rocks. Zircon U-Pb dating suggests that the Shitoukengde Ni-Cu deposit formed in late Silurian(426–422 Ma), and their zircons have εHf(t) values of-9.4 to 5.9 with the older T_(DM1) ages(0.80–1.42 Ga). Mafic-ultramafic rocks from the No. I complex show the similar rare earth and trace element patterns, which are enriched in light rare earth elements and large ion lithophile elements(e.g., K, Rb, Th) and depleted in heavy rare earth elements and high field strength elements(e.g., Ta, Nb, Zr, Ti). Sulfides from the deposit have the slightly higher δ^(34)S values of 1.9–4.3‰ than the mantle(0 ± 2‰). The major and trace element characteristics, and Sr-Nd-Pb and Hf, S isotopes indicate that their parental magmas originated from a metasomatised, asthenospheric mantle source which had previously been modified by subduction-related fluids, and experienced significant crustal contamination both in the magma chamber and during ascent triggering S oversaturation by addition of S and Si, that resulted in the deposition and enrichment of sulfides. 3Correlation of Gut Microbiome Between ASD Children and Mothers and Potential Biomarkers for Risk Assessment显示文摘Variation of maternal gut microbiota may increase the risk of autism spectrum disorders(ASDs) in offspring. Animal studies have indicated that maternal gut microbiota is related to neurodevelopmental abnormalities in mouse offspring, while it is unclear whether there is a correlation between gut microbiota of ASD children and their mothers. We examined the relationships between gut microbiome profiles of ASD children and those of their mothers, and evaluated the clinical discriminatory power of discovered bacterial biomarkers. Gut microbiome was profiled and evaluated by 16S ribosomal RNA gene sequencing in stool samples of 59 mother–child pairs of ASD children and 30 matched mother–child pairs of healthy children. Significant differences were observed in the gut microbiome composition between ASD and healthy children in our Chinese cohort. Several unique bacterial biomarkers, such as Alcaligenaceae and Acinetobacter, were identified. Mothers of ASD children had more Proteobacteria, Alphaproteobacteria, Moraxellaceae, and Acinetobacter than mothers of healthy children. There was a clear correlation between gut microbiome profiles of children and their mothers; however, children with ASD still had unique bacterial biomarkers, such as Alcaligenaceae, Enterobacteriaceae, and Clostridium. Candidate biomarkers discovered in this study had remarkable discriminatory power. 4Geochronology, Geochemistry, and Hf Isotopic Compositions of Monzogranites and Mafic-Ultramafic Complexes in the Maxingdawannan Area, Eastern Kunlun Orogen, Western China: Implications for Magma Sources, Geodynamic Setting, and Petrogenesis显示文摘This paper presents zircon U-Pb-Hf isotopic compositions and whole-rock geochemical data for monzogranites and mafic-ultramafic complexes of the Maxingdawannan area in the western end of the east Kunlun orogenic belt, western China. The data are used to determine the ages, petrogenesis, magma sources, and geodynamic setting of the studied rocks. U-Pb zircon dating indicates that monzogranites and gabbros of the complexes were emplaced at 399 and 397 Ma, respectively. The monzogranites are shoshonitic, with high SiO2, Al2 O3 and total-alkali contents, and low TFeO, MgO, TiO2 and P2 O5 contents. The mafic-ultramafic complexes are characterized by low Si O2 contents. The monzogranites display enrichment in light rare-earth elements(LREE) and large-ion lithophile elements(LILE), depletion in heavy REEs(HREE) and high-field-strength elements(HFSE), and negative Eu anomalies(Eu/Eu*=0.36–0.48). The mafic-ultramafic complexes are also enriched in LREEs and LILEs, and depleted in HREEs and HFSEs, with weak Eu anomalies(Eu/Eu*=0.84–1.16). Zircon εHf(t) values for the monzogranites and mafic-ultramafic complexes range from-6.68 to 1.11 and-1.81 to 6.29, with zircon model ages of 1 812–1 319 Ma(TDM2) and 1 087–769 Ma(TDM1), respectively. Hf isotopic data indicate that primary magmas of the monzogranites are originated from partial melting of ancient lower crust during the Paleo-Mesoproterozoic, with a juvenile-crust component. Primitive magmas of the mafic-ultramafic complexes are likely originated from a depleted-mantle source modified by slab-derived fluids and contaminated by crustal components. 5Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. 7RNA-Seq Study Reveals AP2-Domain-Containing Signalling Regulators Involved in Initial Imbibition of Seed Germination in Rice显示文摘A number of internal signals are required for seed germination.However,the precise signalling responses in the initial imbibition of seed germination are not yet fully understood in rice.In this study,the RNA sequencing(RNA-Seq)approach was conducted in 8 h imbibed seeds to understand the signalling responses in the initial imbibition of rice seed germination.A total of 563 differentially expressed genes(DEGs)with at least 4-fold change were identified in 8 h imbibed seeds compared to dry seeds.MapMan analysis revealed that the majority of signalling response-related DEGs were hormone-and transcription factor-related genes,in which the largest number of DEGs belong to the AP2-domain-containing regulators,and their expressions were significantly induced in the initial imbibition of seed germination in rice.Moreover,at least five AP2-domain-containing transcription factor OsDREBs were identified in the initial imbibition of rice seed germination,and the expressions of 251 DEGs were putatively regulated by OsDREBs through the dehydration-responsive element(DRE)cis-element assay.It suggested that the OsDREBs might play important roles in the regulation of initial seed imbibition in rice.The identified genes provide a valuable resource to study the signalling regulation of seed germination in the future.HE Yongqi ZHAO Jia FENG Defeng HUANG Zhibo LIANG Jiaming ZHENG Yufei CHENG Jinping YING Jifeng WANG Zhoufei 2020Rice science2020,27,4:2
8Filtering enhanced tomographic PIV reconstruction based on deep neural networks显示文摘Tomographic particle image velocimetry(Tomo-PIV)has been successfully applied in measuring three-dimensional(3D)flow field in recent years.Such technology highly relies on the reconstruction technique which provides the spatial particle distribution by using images from multiple cameras at different viewing angles.As the most popular reconstruction method,the multiplicative algebraic reconstruction technique(MART)has advantages in high computational speed and high accuracy for low particle seeding reconstruction.However,the accuracy is not satisfactory in the case of dense particle distributions to be reconstructed.To overcome this problem,a symmetric encode-decoder fully convolutional network is proposed in this paper to improve the reconstruction quality of MART.The input of the neural network is the particle field reconstructed by the MART approach,while the output is the regenerated image with the same resolution.Numerical evaluations indicate that those blurred or irregular particles can be significantly refined by the trained neural network.Most of the ghost particles can also be removed by this filtering method.The reconstruction accuracy can be improved by more than 10%without increasing the computational cost.Experimental evaluations indicate that the trained neural network can also provide similar satisfactory reconstruction and improved velocity fields.Jiaming Liang Shengze Cai Chao Xu Jian Chu 2020IET Cyber-Systems and Robotics2020,2,1:2

11Optimization of Extraction Process and Determination of Antioxidant Activity of Myrica ruba Leaf Pigment by Orthogonal Design显示文摘[Objectives] To determine the optimum conditions and antioxidant activity of Myrica ruba leaf pigment,and provide reference for further development and use of Myrica ruba leaves. [Methods]Pigment of Myrica ruba leaves was extracted by ethanol extraction method. Absorbance was measured at wavelength of 300 nm. Effects of ethanol volume fraction,solid-to-liquid ratio,temperature,and extraction time were studied. 15Porous α-Fe_(2)O_(3)/SnO_(2) nanoflower with enhanced sulfur selectivity and stability for H_(2)S selective oxidation显示文摘Being abundant and active,Fe_(2)O_(3) is suitable for selective oxidation of H_(2)S.However,its practical application is limited due to the poor sulfur selectivity and rapid deactivation.Herein,we report a facile template-free hydrothermal method to fabricate porousα-Fe_(2)O_(3)/SnO_(2) composites with hierarchical nanoflower that can obviously improve the catalytic performance of Fe_(2)O_(3).It was disclosed that the synergistic effect betweenα-Fe_(2)O_(3) and SnO_(2) promotes the physico-chemical properties ofα-Fe_(2)O_(3)/SnO_(2) composites.Specifically,the electron transfer between the Fe^(2+)/Fe^(3+)and Sn^(2+)/Sn^(4+)redox couples enhances the reducibility ofα-Fe_(2)O_(3)/SnO_(2) composites.The number of oxygen vacancies is improved when the Fe cations incorporate into SnO_(2) structure,which facilitates the adsorption and activation of oxygen species.Additionally,the porous structure improves the accessibility of H_(2) S to active sites.Among the composites,Fe1 Sn1 exhibits complete H_(2) S conversion with 100%sulfur selectivity at 220℃,better than those of pureα-Fe_(2)O_(3) and SnO2.Moreover,Fe1 Sn1 catalyst shows high stability and water resistance.Xiaohai Zheng Jiaming Cai Wentao Zhao Shijing Liang Yong Zheng Yanning Cao Lijuan Shen Yihong Xiao Lilong Jiang 2021Chinese Chemical Letters2021,32,7:1

18Deep learning quantified mucus-tumor ratio predicting survival of patients with colorectal cancer using whole-slide images显示文摘Background:In colorectal cancer(CRC),mucinous adenocarcinoma differs from other adenocarcinomas in gene-phenotype,morphology,and prognosis.However,mucinous components are present in a large number of adenocarcinomas,and the prognostic value of mucus proportion has not been investigated.Artificial intelligence provides a way to quantify mucus proportion on whole-slide images(WSIs)accurately.We aimed to quantify mucus proportion by deep learning and further investigate its prognostic value in two CRC patient cohorts.Methods:Deep learning was used to segment WSIs stained with hematoxylin and eosin.Mucus-tumor ratio(MTR)was defined as the proportion of mucinous component in the tumor area.A training cohort(N=419)and a validation cohort(N=315)were used to evaluate the prognostic value of MTR.Survival analysis was performed using the Cox proportional hazard model.Result:Patients were stratified tomucus-low andmucus-high groups,with 24.1%as the threshold.In the training cohort,patients with mucus-high had unfavorable outcomes(hazard ratio for high vs.low 1.88,95%confidence interval 1.18–2.99,P=0.008),with 5-year overall survival rates of 54.8%and 73.7%in mucus-high and mucus-lowgroups,respectively.The resultswere confirmed in the validation cohort(2.09,1.21–3.60,0.008;62.8%vs.79.8%).The prognostic value of MTR was maintained in multivariate analysis for both cohorts.Conclusion:The deep learning quantified MTR was an independent prognostic factor in CRC.With the advantages of advanced efficiency and high consistency,our method is suitable for clinical application and promotes precision medicine development.Ke Zhao Lin Wu Yanqi Huang Su Yao Zeyan Xu Huan Lin Huihui Wang Yanting Liang Yao Xu Xin Chen Minning Zhao Jiaming Peng Yuli Huang Changhong Liang Zhenhui Li Yong Li Zaiyi Liu 2021Precision Clinical Medicine2021,4,1:0
19Multifunctional polyeugenol-based nanoparticles with antioxidant and antibacterial properties显示文摘Over the past few decades,extensive scientific has been dedicated to polymer synthesis employing renewable resources.In this study,we devised and synthesized multifunctional polyeugenol-based nanoparticles,exhibiting remarkable antioxidant and antibacterial properties.The grafting of eugenol onto the nanoparticle surface was achieved via a thiol-ene chemical reaction with a grafting rate of 3.5%.To comprehend the properties of the synthesized nanoparticles,we employed Fourier transform infrared spectroscopy,X-ray photoelectron spectroscopy and pyrolysis-gas chromatography mass spectrometry.Subsequent scanning electron microscopic and transmission electron microscopic analyses revealed the presence of a crosslinked structure within the polyeugenol-based nanoparticles,as well as a heteroge-neous microsphere structure on the surface.Due to the inherent crosslinking structure,the polyeugenol-based nanoparticles demonstrated robust the resistance to solvents,as ascertained through thermog-ravimetric analysis and solvent resistance tests.Notably,nitrogen adsorption/desorption studies confirmed the adsorption capacity of the polyeugenol-based nanoparticles,rendering them potentially suitable for drug transport applications.Moreover,the assays for assessing cytocompatibility and reactive oxygen species scavenging activities demonstrated better performance of the eugenol-based nano-particles compared to eugenol.Furthermore,the polyeugenol-based nanoparticles exhibited certain bactericidal activities against Escherichia coli,Staphylococcus aureus,and Fusobacterium nucleatum.Consequently,these observations indicated the nontoxic nature and expansive application prospects of the polyeugenol-based nanoparticles in the domains of medicine and food preservation.This work presented a pioneering concept for the development of antioxidant and antibacterial multifunctional polymer materials derived from eugenol.Qiaoqiao Lan Xuying Zhang Jiaming Liang Wei Wang Leili Lv Jiamin He Zefeng Wang Chunsheng Qu 2024Particuology2024,87,4:0
