| 1 | Polysaccharides from Tricholoma matsutake and Lentinus edodes enhance 5-fluorouracil-mediated H22 cell growth inhibition显示文摘OBJECTIVE: Few studies have investigated the effects produced by combinations of polysaccharides and chemotherapeutic drugs in cancer treatment.We hypothesized that a combinationo fpolysaccha rides(COP) from Lentinus edodes and Tricholoma matsutake would improve the efficacy of 5-fluorouracil(5-FU)-mediated inhibition of H22 cell growth.METHODS: Mice were injected H22 cells and then treated with either 5-FU, polysaccharides from Tricholoma matsutake(PTM), polysaccharides from Lentinus edodes(PL), PTM+PL, 5-FU+PTM, 5-FU+PL, or 5-FU+COP. The tumor weight and volume,and splenic CD4 + and CD8 + T cell frequencies,were determined. Additionally, splenic natural killer(NK) cell and cytotoxic Tlymphocyte (CTL) activities were assessed and the serum levels of tumor necrosis factor-α(TNF-α),Interleukin-2(IL-2),and Interferon-γ (IFN-γ) were measured. 5-FU, PL, PTM, PTM+PL, 5-FU+PL, and 5-FU+PTM groups, mice treated with 5-FU+COP showed:(a)significantly reduced tumor weight and volume(P<0.05);(b) significantly higher serum levels ofTNF-α,IL-2, and IFN-γ(P<0.05);(c) significantly increased CD4+andCD8+Tcell frequencies in the spleen(P<0.05); and(d) significantly increased splenic NK cell and CTL activities(P<0.05). The tumor weight and volume in mice treated with 5-FU+PL or 5-FU+PTM were significantly reduced compared with mice treated with 5-FU alone(P<0.05). | 2 | Correlation between women's sub-health and reproductive diseases with pregnancies and labors显示文摘OBJECTIVE: To investigate whether female sub-health conditions and reproductive diseases areassociated with pregnancies and labors.METHODS: A cross-sectional survey was performed by using a structured questionnaire. A total of 1343 women aged 35 years or younger in six urban areas of Chengdu were included in the study.According to the Screening Criteria of sub-health conditions, these women were categorized into three groups: postpartum healthy group,sub-healthy group, and reproductive disease group. Data were double-entered using EpiData and the nanalyzed by SPSS.RESULTS: Pregnancy and labor were correlated with postpartum sub-health conditions. The number of pregnancies was negatively correlated with women's postnatal health but was positively correlated with the incidence of postpartum reproductive diseases.CONCLUSION: The number of pregnancies and labors is probably an important factor leading to sub-health conditions and the occurrence of reproductive diseases in women. | 3 | TRAP1 Shows Clinical Significance and Promotes Cellular Migration and Invasion through STAT3/MMP2 Pathway in Human Esophageal Squamous Cell Cancer显示文摘Tumor necrosis factor receptor-associated protein 1(TRAP1), an important member of mitochondrial heat shock protein 90 family, is involved in multiple biological processes in several types of tumors. However, its pathological role in esophageal squamous cell cancer(ESCC) remains unknown. Herein, we demonstrated the clinical value of TRAP1, and its role in apoptosis and motility in ESCC. The clinical potential of TRAP1 was investigated through immunohistochemical analysis in 328 ESCC samples, which revealed that strong TRAP1 expression was associated with increased risk of lymph node metastasis, while high TRAP1 expression correlated with poor prognosis. Expression of TRAP1 was found to be an independent prognostic factor for patients with ESCC. Additionally, the upregulation of TRAP1 antagonized cisplatin-induced apoptosis while its downregulation sensitized cells to cisplatin-induced apoptosis. As revealed by the transwell assay, TRAP1 overexpression promoted cellular migration and invasion as compared to the control groups. In contrast,silencing of endogenous TRAP1 expression attenuated the ability of migration and invasion. Finally, the molecular mechanism investigated in the present study demonstrated that TRAP1-mediated migration and invasion occurred through STAT3/MMP2 signaling pathway. | 5 | Silurian A-type granitoids in the southern margin of the Tongbai-Dabieshan: Evidence from SHRIMP zircon geochronology and geochemistry显示文摘SHRIMP U-Pb dating on magmatic zircons extracted from a riebeckite quartz syenite in the Huangyangshan pluton in Suizhou, the southern margin of Tongbai-Dabieshan yielded an age of 439±6Ma. According to the morphology and high Th/U ratios of the zircons, the age is interpreted as crystallization timing of the pluton. The Huangyangshan pluton is composed of peralkaline quartz syenite, alkaline granite and syenite. All of the rocks are characterized by high agpaitic index (A.I.=(Na+K)/Al, molar ratio) and Fe-number [FeOT/(FeOT+MgO)], low CaO and MgO contents, enrichment of high field strength elements (i.e. Nb, Zr, Ga, Y, Hf) and light REEs, evidently negative Eu anomalies and high Ga/Al ratios, which are consistent with anorogenic A-type granitoids. | 7 | Artificial intelligence-assisted automatic and index-based microbial single-cell sorting system for One-Cell-One-Tube显示文摘Identification,sorting,and sequencing of individual cells directly from in situ samples have great potential for in-depth analysis of the structure and function of microbiomes.In this work,based on an artificial intelligence(AI)-assisted object detection model for cell phenotype screening and a cross-interface contact method for single-cell exporting,we developed an automatic and index-based system called EasySort AUTO,where individual microbial cells are sorted and then packaged in a microdroplet and automatically exported in a precisely indexed,“One-Cell-One-Tube”manner.The target cell is automatically identified based on an AI-assisted object detection model and then mobilized via an optical tweezer for sorting.Then,a crossinterface contact microfluidic printing method that we developed enables the automated transfer of cells from the chip to the tube,which leads to coupling with subsequent single-cell culture or sequencing.The efficiency of the system for single-cell printing is>93%.The throughput of the system for single-cell printing is~120 cells/h.Moreover,>80%of single cells of both yeast and Escherichia coli are culturable,suggesting the superior preservation of cell viability during sorting.Finally,AI-assisted object detection supports automated sorting of target cells with high accuracy from mixed yeast samples,which was validated by downstream single-cell proliferation assays.The automation,index maintenance,and vitality preservation of EasySort AUTO suggest its excellent application potential for single-cell sorting. | Zhidian Diao Lingyan Kan Yilong Zhao Jingyun Song Chen Wang Yang Liu Fengli Zhang Teng Xu Rongze Chen Xixian Wang Xiaoyan Jing Jian Xu Yuandong Li Bo Ma | 2022 | mLife2022,1,4: | 2 |

| 10 | Effect of naloxone on level of plasma beta-endorphin in neonates with severe asphyxia显示文摘BACKGROUND: β-endorphin is the most actively endogenous substance of cerebral endorphin. When combined with opiate receptor specially,it manifests a strong morphine-like activity and can decrease sensitivity of central nervous system to carbon dioxide so as to inhibit breath. OBJECTIVE: To observe the changes of content of plasma β-endorphin in neonates with severe asphyxia after naloxone treatment in a large dosage. DESIGN: Randomized controlled observation. SETTINGS: Department of Pediatrics,Shenzhen Shajing People's Hospital; Center of Pediatrics,Guangzhou Zhujiang Hospital. PARTICIPANTS: A total of 97 neonates with severe asphyxia including 57 boys and 40 girls were selected from Neonatal Intensive Care Unit,Department of Pediatrics,Shenzhen Shajing People's Hospital from January 2004 to November 2005. Their gestational age was (38±3) weeks,body mass was (3.2±1.7) kg,and hospitalization duration was (2.8±2.3) hours. All neonates met the diagnostic criteria of with severe asphyxia and all their parents provided the confirmed consent. METHODS: All neonates were treated with inspired oxygen,sedation,stopping terror,decreasing cranial pressure,maintaining a well blood perfusion and normal level of blood glucose (about 5.0 mmol/L). After hospitalization,0.1 mg/(kg·d) naloxone hydrochloride (Beijing Sihuan Pharmaceutical Technology Co.,Ltd.; certification: H10900021; bullet preparation; 0.4 mg/ampoule) was intravenously dribbled into neonates for 4–6 hours,14 days in total. 2 mL blood was collected from radial artery in neonates at the beginning of hospitalization and at 3 days after naloxone treatment,put in aprotinin-pre-cool tube,mixed evenly,and centrifuged at hypothermia. Plasma was maintained in refrigerator at –70 ℃. The kit was provided by Neurobiology Department of Shanghai Second Military Medical University of Chinese PLA. Concentration of plasma β-endorphin was measured by using radio-immunity assay.All data were expressed as Mean± SD and results were compared with paired t test. MAIN OUTCOME MEASURE: Concentration of plasma β-endorphin. RESULTS: All 97 neonates were involved in the final analysis. Concentration of plasma β-endorphin in neonates with severe asphyxia was lower after treatment as compared with that before treatment,and there was significant difference (t =10.31,P < 0.01). (RP) plant extract exhibits protective effects on cerebral ischemia. This effect is reflected in altered ischemic neuronal apoptosis and associated protein expression. OBJECTIVE: To explore the neuroprotective mechanism of RP after cerebral ischemia injury. DESIGN, TIME AND SETTING: Randomized control experiment of cellular, molecular, and protein levels. The experiment was completed at Chongqing Medical University at the School of Pharmacy Laboratories and Basic Medical Institute from October 2005 to January 2006. MATERIALS: Twenty-four adult, male, Wistar rats, weighing (28 ± 20) g. RP extract, which was a product of ethanol extraction, was provided by the Laboratory of Pharmaceutical Analysis, Chongqing Medical University. RP was dissolved in distilled water to a concentration of 10 mg/mL. All rats were randomly assigned into four groups: 5 g/kg RP, 10 g/kg RP, model, and sham-surgery, with 6 rats in each group. METHODS: In the 5 and 10 g/kg RP groups, as well as the model group, the middle cerebral artery was occluded (MCAO) for 60 minutes, resulting in focal cerebral ischemia, followed by reperfusion for 24 hours. Rats in the 5 and 10 g/kg RP groups received 5 and 10 g/kg RP, respectively. The RP treatment group received RP intragastrically (once a day) for 3 days. One hour after the last dose, rats were subjected to MCAO. The same surgical procedure was performed in the sham-surgery group, except the suture was introduced into the external carotid artery, but not advanced. Rats in the model group were subjected to MCAO. The sham-surgery and model groups received intragastrically administered normal saline once per day for 3 days. One hour after the last dose, the rats were subjected to surgery. MAIN OUTCOME MEASURES: TUNEL labeling and immunohistochemical methods were used to in- vestigate changes in neuronal apoptosis and expression of the apoptosis-related proteins, Bax and Bcl-2, on the ischemic hemisphere, as well as the contralateral hemisphere, after administration of RP or normal saline. RESULTS: All 24 Wistar rats were included in the final analysis, without any loss. Compared with the sham-surgery group, the number of the TUNEL-positive cells in the model group, as well as the 5 and 10 g/kg RP treatment groups, were significantly increased in the ischemic hemisphere (P < 0.05). Compared with the sham-surgery group, the number of Bcl-2-positive and Bax-positive cells increased significantly in the model group (P < 0.01). The number of Bcl-2-positive cells increased, and the number of Bax-positive cells decreased in the model group, compared to the 5 and 10 g/kg RP treatment groups (P < 0.05-0.01). CONCLUSION: RP can prevent neuronal apoptosis following cerebral ischemic-reperfusion of rats. | 19 | Fabrication of the beam splitters for soft X-ray laser application显示文摘The soft X-ray interferometry is completed by the Mach-Zehnder interferometer using a soft X-ray laser, and it is also an important method to measure the electron densities of a laser-produced plasma near the critical surface. It is apparently demonstrated in this paper that the incident angle of each optical element in the soft X-ray Mach- Zehnder interferometer should be near normal incidence based on the polarized characteristics of the soft X-ray mul-tilayers, and the product of reflectivity and transmission of the beam splitter should be taken as a standard of design according to the structure of the soft X-ray Mach-Zehnder interferometer. The beam splitters used in the soft X-ray interferometry at 13.9 nm are fabricated using the ion beam sputtering.
