| 1 | Tumor-expressed B7-H3 mediates the inhibition of antitumor T-cell functions in ovarian cancer insensitive to PD-1 blockade therapy显示文摘Although PD-L1/PD-1 blockade therapy has been approved to treat many types of cancers,the majority of patients with solid tumors do not respond well,but the underlying reason remains unclear.Here,we studied ovarian cancer(OvCa),a tumor type generally resistant to current immunotherapies,to investigate PD-1-independent immunosuppression.We found that PD-L1 was not highly expressed in the tumor microenvironment(TME)of human OvCa.Instead,B7-H3,another checkpoint molecule,was highly expressed by both tumor cells and tumor-infiltrating antigen-presenting cells(APCs),which correlated with T-cell exhaustion in patients.Using ID8 OvCa mouse models,we found that B7-H3 expressed on tumor cells,but not host cells,had a dominant role in suppressing antitumor immunity.Therapeutically,B7-H3 blockade,but not PD-1 blockade,prolonged the survival of ID8 tumorbearing mice.Collectively,our results demonstrate that tumor-expressed B7-H3 inhibits the function of CD8^(+)T cells and suggest that B7-H3 may be a target in patients who are not responsive to PD-L1/PD-1 inhibition,particularly OvCa patients. | Dongli Cai Jiaming Li Dingfeng Liu Shanjuan Hong Qin Qiao Qinli Sun Pingping Li Nanan Lyu Tiantian Sun Shan Xie Li Guo Ling Ni Liping Jin Chen Dong | 2020 | Cellular & Molecular Immunology2020,17,3: | 8 |

| 3 | Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. | 8 | Molecular Cloning and Bioinformatics Analysis of TypeⅢSecretion System Effector Protein Va1686 Gene of Vibrio alginolyticus显示文摘In this study,Va1686 gene was cloned from Vibrio alginolyticus. The total length of the gene is 1 164 bp,and it could encode 387 amino acids. The physicochemical properties,protein structure,genetic evolutionary relationship and antigenic characteristics of the effect protein Va1686 of V. alginolyticus HY9901 type Ⅲ secretion system were studied and analyzed by bioinformatics methods and tools. The results showed that Va1686 is a stable hydrophilic and acidic protein without a transmembrane region and a signal peptide,and secondary structure to α-helix. The evolutionary analysis showed that V. alginolyticus HY9901 and V.harveyi were clustered together,which indicated that the genetic relationship between the two species was the closest. Va1686 contains a Fic superfamily conserved domain associated with cell division. Bioinformatics analysis showed that the B-cell preponderant epitopes of Va1686 might be localized in the regions of 48-49,82-85,125-126,150-153,185-186,236-237 and so on. The 3D structure model of Va1686 subunit was simulated by SWISS-MODEL software and it was found that the vop S of V. parahaemolyticus was similar and the similarity was 89. 46%. | 9 | Filtering enhanced tomographic PIV reconstruction based on deep neural networks显示文摘Tomographic particle image velocimetry(Tomo-PIV)has been successfully applied in measuring three-dimensional(3D)flow field in recent years.Such technology highly relies on the reconstruction technique which provides the spatial particle distribution by using images from multiple cameras at different viewing angles.As the most popular reconstruction method,the multiplicative algebraic reconstruction technique(MART)has advantages in high computational speed and high accuracy for low particle seeding reconstruction.However,the accuracy is not satisfactory in the case of dense particle distributions to be reconstructed.To overcome this problem,a symmetric encode-decoder fully convolutional network is proposed in this paper to improve the reconstruction quality of MART.The input of the neural network is the particle field reconstructed by the MART approach,while the output is the regenerated image with the same resolution.Numerical evaluations indicate that those blurred or irregular particles can be significantly refined by the trained neural network.Most of the ghost particles can also be removed by this filtering method.The reconstruction accuracy can be improved by more than 10%without increasing the computational cost.Experimental evaluations indicate that the trained neural network can also provide similar satisfactory reconstruction and improved velocity fields. | Jiaming Liang Shengze Cai Chao Xu Jian Chu | 2020 | IET Cyber-Systems and Robotics2020,2,1: | 2 |

| 17 | SiRNA-mediated ankyrin-G silence modulates the expression of voltage-gated Na channels in murine hippocampal HT22 cells显示文摘Background:Voltage-gated sodium channels are the targets of many commonly used antiepileptic drugs.NaV1.6,encoded by Scn8a,increased in chronic mesial temporal epilepsy animal models and co-localized with Ankyrin-G,encoded by Ank3.We hypothesized that inhibition of Ank3 transcription by siRNA decrease the expression of NaV1.6.Results:We characterized expression of the target genes in hippocampal neuron HT22 cells by Real time-PCR.The melt peak in the resolution curve of Scn1a,Scn8a and Ank3 were all unique.Ank3 transcription was interfered and the relative Ank3 mRNA levels of the three interfered groups compared to GAPDH were 0.89±0.13,0.52±0.07 and 0.26±0.05 while that of the negative control group was 1.01±0.08(P<0.05).When Ank3 transcription was inhibited by siRNA,the relative mRNA levels of Scn8a decreased in the three groups(0.91±0.09,0.33±0.06 and 0.25±0.05),compared to the negative control group(1.10±0.09).Tested by Western blotting,protein levels of ankyrinG and Nav1.6 decreased after ank3-siRNA.Ankyrin-G in negative control group,group1,group2 and group1+2 were 0.813±0.051,0.744±0.041,0.477±0.055 and 0.351±0.190 respectively(P<0.01)while Nav1.6 were 0.934±0.036,0.867±0.078,0.498±0.070 and 0.586±0.180(P<0.01).The quantity analysis of immunofluorescence showed significant decrease of ankyrin-G and Nav1.6(Student’s test,P=0.046 and 0.016 respectively).Conclusion:We therefore concluded that in HT22 cells the expression of Nav1.6 was down-regulated by Ank3 RNA interference. | Guanzhong Ni Xiaoting Hao Xiaodong Cai Jiaming Qin Liemin Zhou Patrick Kwan Ziyi Chen | 2019 | Acta Epileptologica2019,1,1: | 0 |
| 18 | Influence of edaravone on Notch1 and nuclear factor-kappaB in rats with cerebral ischemia/reperfusion injury显示文摘BACKGROUND: It has been demonstrated that edaravone has a a neuroprotective role, inhibits free radical increase, and reduces cell apoptosis. The Notch pathway is a key factor in neurogenesis and cellular apoptosis. The proinflammatory transcription factor nuclear factor-kappaB (NF-κB) plays an important role in inflam- mation and oxidation. OBJECTIVE: To observe the influence of edaravone on Notch1 and NF-κB mRNA and protein expression in rats with focal cerebral ischemia/reperfusion injury. DESIGN, TIME AND SETTING: This randomized controlled neural and molecular biology experiment was performed at the Department of Neurology, the First Affiliated Hospital of Chongqing Medical Univer- sity, and the Chongqing Key Laboratory of Neurology between July 2007 and May 2008. MATERIALS: Thirty female Wistar rats were used. Edaravone was purchased from Jiangsu Xiansheng Pharmaceutical Limited Company, China. METHODS: Wistar rats were randomly divided into five groups (n = 6). Thread was inserted into the inter- nal carotid artery of the sham operation group but the middle cerebral artery was not ligated. A focal cerebral ischemia/reperfusion model was established by inserting thread into the right middle cerebral artery. The model rats in the edaravone groups were given tail vein injections of edaravone at 3 mg/kg body weight after ischemia for 2 hours and reperfusion for 12 or 24 hours. Ischemia/reperfusion groups (model group) received intravenous infusion of normal saline at the same volume as the edaravone groups after ischemia for 2 hours and reperfusion for 12 or 24 hours. MAIN OUTCOME MEASURES: The volume of the ischemic region was measured by 2,3,5-triphenyltetrazolium chloride staining. Notch1 and NF-κB protein and mRNA expression were meas- ured by immunohistochemistry and RT-PCR. Protein expression was represented by the absorbance value. RESULTS: Edaravone greatly reduced the focal infarct volume. Notch1 and NF-κB protein and mRNA ex- pression were rapidly upregulated following cerebral ischemia/reperfusion injury in model and edaravone groups compared with the sham operation group (P < 0.01). In addition, edaravone treatment significantly upregulated Notch1 expression but down-regulated NF-κB expression compared with model groups (P < 0.01). | 20 | Effect of microbial agent ARC-BBBE demonstration application on peanut production in the Huang-huai-hai area of China显示文摘The microbial agent ARC-BBBE demonstration trials were conducted in four provinces in the main peanutproducing areas of the Huang-huai-hai plain of China.The results revealed that the application of ARC-BBBE led to a 1.09–1.70 fold increase in the number of nodules in the treatment group at the demonstration site compared to the control group.Moreover,the nodule weight in the treatment group was 0.80–3.32 times higher than that of the control group,and nitrogenase activity per plant showed a significant enhancement by 1.00–2.83 fold compared to controls.Additionally,notable improvements were observed in terms of increased fresh weight of whole plants,well-filled pod numbers,and enhanced growth performance;ultimately resulting in a harvest yield increase ranging from 9.46%to 49.04%.The abundance of Aspergillus flavus in rhizosphere soil was determined by the dilution spread plate method,and the inhibition rate was up to 86.7%.The application of ARC-BBBE in the significant peanut-producing areas of Huang-huai-hai has effects of promoting growth,nodulation,and increasing production.At the same time,it has the effect of inhibiting and controlling soil Aspergillus flavus,which provides a new green and low-carbon way to promote the high-quality development of the peanut industry. | Pingping Ji Haohua Gu Mingbo Wen Hang Cai Jiaming Zhu Xiaofeng Yue Qi Zhang Peiwu Li | 2023 | Oil Crop Science2023,8,3: | 0 |
