1Anatomical and chemical characteristics associated with lodging resistance in wheat显示文摘Anatomical and chemical characteristics of stems affect lodging in wheat(Triticum aestivum L.) cultivars. Traits associated with lodging resistance, such as plant height, stem strength, culm wall thickness, pith diameter, and stem diameter, were extensively investigated in earlier studies. However, the solid stem trait was rarely considered. In this study, we measured a range of anatomical and chemical characteristics on solid and hollow stemmed wheat cultivars. Significant correlations were detected between resistance to lodging and several anatomical features, including width of mechanical tissue, weight of low internodes, and width of stem walls. Morphological features that gave the best indication of improved lodging resistance were increased stem width, width of mechanical tissue layer, and stem density. Multiple linear regression analysis showed that 99% of the variation in lodging resistance could be explained by the width of the mechanical tissue layer, suggesting that solid stemmed wheat has several anatomical features for increasing resistance to lodging. In addition, microsatellite markers GWM247 and GWM340 were linked to a single solid stem QTL on chromosome 3BL in a population derived from the cross Xinongshixin(solid stem)/Line 3159(hollow stem). 2Highly efficient heritable genome editing in wheat using an RNA virus and bypassing tissue culture.显示文摘Genome editing provides novel strategies for improving plant traits,but relies on current genetic transformation and plant regeneration procedures,which can be inefficient.We have engineered a barley stripe mosaic virus(BSMV)-based sgRNA delivery vector(BSMV-sg)that is effective in performing heritable genome editing in Cas9-transgenic wheat plants.Mutated progenies were present in the next generation at frequencies ranging from 12.9%to 100%in three different wheat varieties,and 53.8%to 100%of mutants were virus-free.We also achieved multiplex mutagenesis in progeny using a pool of BSMV-sg vectors harboring different sgRNAs.Furthermore,we devised a virus-induced transgene-free editing procedure(VITF-Edit)to generate Cas9-free wheat mutants by crossing BSMV-infected Cas9-transgenic wheat pollen with wild-type wheat.Our study provides a robust,convenient and tissue culture-free approach for genome editing in wheat through virus infection.Tingdong Li Jiacheng Hu Yu Sun Boshu Li Dingliang Zhang Wenli Li Jinxing Liu Dawei Li Caixia Gao Yongliang Zhang Yanpeng Wang 2021Molecular Plant2021,14,11:10
3An initial discussion on major controlling factors on formation of coal-formed large-medium sas fields显示文摘The so-called 'large-medium gas field' means a gas field with proven reserves equal to or larger than 100×108 m3. Up to 1997, 23 coal-formed great-medium fields are discovered in China, excluding two in Taiwan Province. Their proven reserves take 50.9% in total natural gas reserves. Therefore, the coal-formed great-medium gas field plays a decisive role in natural gas reserves and the study of their controlling factors is very important both in theory and in practice. 4Three-dimensional reconstruction of Picea wilsonii Mast. pollen grains using automated electron microscopy显示文摘Three-dimensional electron microscopy(3 D-EM) has attracted considerable attention because of its ability to provide detailed information with respect to developmental analysis. However, large-scale high-resolution 3 D reconstruction of biological samples remains challenging. Herein, we present a 3 D view of a Picea wilsonii Mast. pollen grain with 100 nm axial and38.57 nm lateral resolution using AutoCUTS-SEM(automatic collector of ultrathin sections-scanning electron microscopy). We established a library of 3,127 100 nm thick serial sections of pollen grains for preservation and observation, demonstrating that the protocol can be used to analyze large-volume samples. After obtaining the SEM images, we reconstructed an entire pollen grain comprising 734 serial sections. The images produced by 3D reconstruction clearly revealed the main components of the P.wilsonii pollen grain, i.e., two sacci and pollen corpus, tube cell, generative cell, and two degenerated prothallial cells, and their internal organization. In addition, we performed a quantitative analysis of the different pollen grain cells, including sacci, and found that there were 202 connections within a saccus SEM image. 6Transparent,stretchable,temperature-stable and self-healing ionogel-based triboelectric nanogenerator for biomechanical energy collection显示文摘A flexible and stable power supply is essential to the rapid development of wearable electronic devices.In this work,a transparent,flexible,temperature-stable and ionogel electrode-based self-healing triboelectric nanogenerator(IS-TENG)was developed.The ionogel with excellent stretchability(1,012%),high ionic conductivity(0.3 S·m^(−1))and high-temperature stability(temperature range of−77 to 250℃)was used as the electrode of the IS-TENG.The IS-TENG exhibited excellent transparency(92.1%)and stability.The output performance did not decrease when placed in a 60℃oven for 48 h.In addition,the IS-TENG behaved like a stable output in the range of−20 to 60℃.More importantly,the IS-TENG could also achieve self-healing of electrical performance at temperatures between−20 and 60℃and its output can be restored to its original state after healing.When the single-electrode IS-TENG with an area of 3 cm×3 cm was conducted under the working frequency of 1.5 Hz,the output values for open-circuit voltage,short-circuit current,short-circuit transferred charge,and maximum peak power density were 189 V,6.2μA,57 nC,and 2.17 W·m^(−2),respectively.The IS-TENG enables to harvest biomechanical energy,and drive electronic devices.Furthermore,the application of IS-TENGs as self-driven sensors for detecting human behavior was also demonstrated,showing good application prospects in the field of wearable power technology and self-driven sensing.Weiqiang Liao Xiukun Liu Yuqi Li Xu Xu Jinxing Jiang Shaorong Lu Dequan Bao Zhen Wen Xuhui Sun 2022Nano Research2022,15,3:2
7NIR-to-NIR UCL/T_1-weighted MR/CT multimodal imaging by NaYbF_4:Tm@NaGdF_4:Yb-PVP upconversion nanoparticles显示文摘Multimodal imaging nanoprobes are urgently sought because they can integrate different imaging function into individual nanoplatform and provide more comprehensive and accurate information for the diagnosis of early-stage tumor.Lanthanide-based upconversion nanoparticles(UCNPs) are regarded as promising nanoplatforms to fabricate these probes.Herein,we firstly developed the active core-active shell structured NaYbF_4:Tm@NaGdF_4:Yb-PVP UCNPs with the average diameter of 13.23±0.96 nm as multimodal imaging probes.These water-dispersible nanoprobes presented excellent near-infrared to near-infrared(NIR-to-NIR) upconversion luminescence(UCL) performance,which is favorable for optical bioimaging due to deeper tissue penetration and autofluorescence reduction.After coated with the NaGdF_4:Yb active shell,the UCL emission intensity at 800 nm increased by 7.2 times.These nanoprobes exhibited a desirable longitudinal relaxivity(r_1=3.58 L/(mmol s)) and strong X-ray attenuation property(58.84 HU L/g).The cytotoxicity assessment,histology analysis and biodistribution study revealed that NaYbF_4:Tm@NaGdF_4:Yb-PVP UCNPs had relatively low cytotoxicity and negligible organ toxicity.These UCNPs were applied for NIR-to-NIR UCL imaging in vivo.More importantly,the detection of small tumor was successfully achieved under T_1-weighted MRI and CT imaging modalities after intravenous injection of these UCNPs.These results revealed that NaYbF_4:Tm@NaGdF_4:Yb-PVP UCNPs could serve as promising NIR-to-NIR UCL/MRI/CT trimodal imaging probes.Tao Wang Min Yang Jiahui Huang Yanzhi Zhao Honglei Wang Shuang Leng Jinxing Chen Guoying Sun Jianhua Liu 2017Science Bulletin2017,62,13:2
