| 2 | Switchgrass as a bioenergy crop in the Loess Plateau, China:Potential lignocellulosic feedstock production and environmental conservation显示文摘A large portion of the Loess Plateau of China is characterized as 'marginal' with serious land degradation and desertification problems. Consequently, two policies, Grain for Green and Western Development Action were established by the Chinese government in response to the demand for ecological protection and economic development in the Loess Plateau. These policies are designed to increase forest cover, expand farmlands, and enhance soil and water conservation, while creating sustainable vegetation restoration. Perennial grasses have gained attention as bioenergy feedstocks due to their high biomass yields, low inputs, and greater ecosystem services compared to annual crops. Moreover, perennial grasses limit nutrient runoff and reduce greenhouse gas emissions and soil losses while sequestering carbon. Additionally, perennial grasses can generate economic returns for local farmers through producing bioenergy feedstock or forage on marginal lands. METHODS: A retrospective chart review of patients with a diagnosis of HIV-related PCNSL at one of five university hospitals between 1987 and 1998 was performed. Clinical details including antiretroviral agent use, brain imaging scan results, RT use, and survival outcomes were recorded. RESULTS: One hundred eleven patients with HIV-related PCNSL were identified. | 6 | Cryptic diversity and significant cophylogenetic signal detected by DNA barcoding the rust fungi (Pucciniaceae) of Cyperaceae-Juncaceae显示文摘Plants play important roles as habitat and food for a tremendous diversity of specialist animals and fungi.The disappearance of any plant species can lead to extinction cascades of its associated biota.In consequence,documenting the diversity and specificity of plant-associated organisms is of high practical relevance in biodiversity conservation.Here,we present the first large-scale molecular investigation into the diversity,host specificity,and cophylogenetic congruence of an especially rich plant-fungal association,the rust fungi(Pucciniaceae)of Cyperaceae and Juncaceae.Using the largest rust fungi DNA barcoding dataset published to date(252 sequences,82 taxa),we reject the presence of a global ITS2-28S barcode gap,but find a local gap in Cyperaceae-Juncaceae rusts,and suggest the existence of many cryptic species in North America,with some broadly circumscribed species possibly corresponding to>10 cryptic species.We test previous hypotheses of correlations between the phylogenies of rust fungi and their Cyperaceae-Juncaceae hosts using a combination of global-fit and event-based cophylogenetic methods.A significant cophylogenetic signal is detected between rusts and their hosts,but the small number of cospeciations argues for preferential host jumps as the driving process behind these correlations.In addition,temporal congruence between the origin of major Carex clades and their rusts suggests that host diversification may have promoted parasite diversification.Finally,we discuss the relevance of rust infection patterns to the systematics of Cyperaceae,highlight some taxonomic problems uncovered by the analyses,and call attention to the promise of DNA barcoding for bridging knowledge gaps in poorly studied plant-associated microorganisms. | Étienne Léveillé-Bourret Quinn Eggertson Sarah Hambleton Julian R.Starr | 2021 | Journal of Systematics and Evolution2021,59,4: | 4 |
| 7 | Habitat suitability and herbivores determine reintroduction success of an endangered legume显示文摘Reintroductions of rare plants require detailed knowledge of habitat requirements, species interactions,and restoration techniques. Thus, incremental experimentation over many years may be required to develop adequate knowledge and techniques for successful reintroduction. To determine drivers of extinction in historical reintroductions of a federally endangered perennial(Astragalus bibullatus), we developed a reintroduction experiment to disentangle the relative importance of habitat quality, herbivores, and restoration technique on reintroduction success. In a factorial design, we manipulated access to vertebrate herbivores across different habitat types(mesic ecotone vs. xeric barren), and used founder populations comprised of more transplants and genetic sources than previous reintroduction attempts.In mesic ecotones where historical reintroductions failed, excluding herbivores, thinning woody encroachment to improve habitat quality, outplanting across a greater array of microhabitats, and increasing founder population size did not improve demographic rates over previous attempts.Compared to mesic ecotones, transplant survival rates and cumulative fruit production were more than two and ten times greater, respectively, in a xeric barren ecotone characterized by open, grassy, and dry microenvironmental conditions. Across all sites, herbivores decreased probabilities of survival and flowering of larger adult plants. Flowering rates were 80% greater inside relative to outside herbivore exclusion cages. Over a four-year period, only a single uncaged plant produced fruit. Our study demonstrates that habitat quality and vertebrate herbivory are key drivers of long-term persistence in rare plant reintroductions. | 8 | Bioactive hydrogel microcapsules for guiding stem cell fate decisions by release and reloading of growth factors显示文摘Human pluripotent stem cells(hPSC)hold considerable promise as a source of adult cells for treatment of diseases ranging from diabetes to liver failure.Some of the challenges that limit the clinical/translational impact of hPSCs are high cost and difficulty in scaling-up of existing differentiation protocols.In this paper,we sought to address these challenges through the development of bioactive microcapsules.A co-axial flow focusing microfluidic device was used to encapsulate hPSCs in microcapsules comprised of an aqueous core and a hydrogel shell.Importantly,the shell contained heparin moieties for growth factor(GF)binding and release.The aqueous core enabled rapid aggregation of hPSCs into 3D spheroids while the bioactive hydrogel shell was used to load inductive cues driving pluripotency maintenance and endodermal differentiation.Specifically,we demonstrated that one-time,1 h long loading of pluripotency signals,fibroblast growth factor(FGF)-2 and transforming growth factor(TGF)-β1,into bioactive microcapsules was sufficient to induce and maintain pluripotency of hPSCs over the course of 5 days at levels similar to or better than a standard protocol with soluble GFs.Furthermore,stem cell-carrying microcapsules that previously contained pluripotency signals could be reloaded with an endodermal cue,Nodal,resulting in higher levels of endodermal markers compared to stem cells differentiated in a standard protocol.Overall,bioactive heparin-containing core-shell microcapsules decreased GF usage five-fold while improving stem cell phenotype and are well suited for 3D cultivation of hPSCs. | Kihak Gwon Hye Jin Hong Alan M.Gonzalez-Suarez Michael Q.Slama Daheui Choi Jinkee Hong Harihara Baskaran Gulnaz Stybayeva Quinn P.Peterson Alexander Revzin | 2022 | Bioactive Materials2022,7,9: | 3 |
| 9 | California Simulation of Evapotranspiration of Applied Water and Agricultural Energy Use in California显示文摘The California Simulation of Evapotranspiration of Applied Water (Cal-SIMETAW) model is a new tool developed by the California Department of Water Resources and the University of California, Davis to perform daily soil water balance and determine crop evapotranspiration (ET c ), evapotranspiration of applied water (ET aw ), and applied water (AW) for use in California water resources planning. ET aw is a seasonal estimate of the water needed to irrigate a crop assuming 100% irrigation efficiency. The model accounts for soils, crop coefficients, rooting depths, seepage, etc. that influence crop water balance. It provides spatial soil and climate information and it uses historical crop and land-use category information to provide seasonal water balance estimates by combinations of detailed analysis unit and county (DAU/County) over California. The result is a large data base of ET c and ET aw that will be used to update information in the new California Water Plan (CWP). The application uses the daily climate data, i.e., maximum (T x ) and minimum (T n ) temperature and precipitation (P cp ), which were derived from monthly USDA-NRCS PRISM data (PRISM Group 2011) and daily US National Climate Data Center (NCDC) climate station data to cover California on a 4 km×4 km change grid spacing. The application uses daily weather data to determine reference evapotranspiration (ET o ), using the Hargreaves-Samani (HS) equation (Hargreaves and Samani 1982, 1985). Because the HS equation is based on temperature only, ET o from the HS equation were compared with CIMIS ET o at the same locations using available CIMIS data to determine correction factors to estimate CIMIS ET o from the HS ET o to account for spatial climate differences. Cal-SIMETAW also employs near real-time reference evapotranspiration (ET o ) information from Spatial CIMIS, which is a model that combines weather station data and remote sensing to provide a grid of ET o information. A second database containing the available soil water holding capacity and soil depth information for all of California was also developed from the USDA-NRCS SSURGO database. The Cal-SIMETAW program also has the ability to generate daily weather data from monthly mean values for use in studying climate change scenarios and their possible impacts on water demand in the state. The key objective of this project is to improve the accuracy of water use estimates for the California Water Plan (CWP), which provides a comprehensive report on water supply, demand, and management in California. | 11 | Conferring receptors on recipient cells with extracellular vesicles for targeted drug delivery显示文摘Triple negative breast cancer(TNBC)is a heterogeneous subset of breast cancer characterized by its lack of estrogen receptor,progesterone receptor,and human epidermal growth factor receptor 2(HER2),which altogether prevents TNBC from being treated effectively.For many years,the treatment paradigms and overall survival of patients with TNBC have remained largely stagnant.Recent attempts to convert cold tumors to hot tumors by promoting antigen presentation have shown increased T cell infiltration and significantly induced immune responses for tumor killing.Inspired by this concept,the expression of specific targetable antigens on TNBC cells may further benefit relevant targeted drug delivery.In this study,we successfully conferred sufficient HER2 on the surface of TNBC MDA-MB-231 cells via simple EV-plasma membrane fusion with HER2+extracellular vesicles(EV)derived from HER2 overexpressing BT-474 cells.Subsequently,anti-HER2 antibody conjugated paclitaxel-loaded liposomes were used for HER2-targeted drug delivery.Our findings demonstrated this HER2 grafting,in conjunction with targeted drug delivery,can improve the treatment efficacy in vitro and in vivo.This novel approach represents a facile method of altering cell membrane antigen presentation via convenient EVs uptake and may pave the way for the burgeoning wave of targeted therapy and/or immunotherapy. | Zachary Quinn Wenjun Mao Yiqiu Xia Rhea John Yuan Wan | 2021 | Bioactive Materials2021,6,3: | 3 |
| 12 | Modulation of tau phosphorylation by environmental copper显示文摘Background:The transition metal copper enhances amyloidβaggregation and neurotoxicity,and in models of concomitant amyloid and tau pathology,copper also promotes tau aggregation.Since it is not clear if the effects of environmental copper upon tau pathology are dependent on the presence of pathological amyloidβ,we tested the effects of copper overload and complexing in disease models which lack pathological amyloidβ.Methods:We used cell culture and transgenic murine models to test the effects of environmental copper on tau phosphorylation.We used oral zinc acetate as a copper lowering agent in mice and examined changes in blood and brain metals through inductively coupled plasma mass spectroscopy.Behavioral effects of copper lowering were assessed with Morris water maze and novel object recognition tasks.Changes in tau phosphorylation were examined by phosphorylation specific antibodies on Western blots.Results:In human neuroblastoma cells,excess copper promoted tau phosphorylation and a copper complexing agent,tetrathiomolybdate,attenuated tau phosphorylation.In a transgenic mouse model expressing wild type human tau,copper-lowering by oral zinc suppressed plasma and brain levels of copper,and resulted in a marked attenuation of tau phosphorylation.No significant changes in behavior were observed with copper lowering,but a trend to improved recognition of the novel object was observed in zinc acetate treated mice.Conclusions:We propose that reduction of brain copper by blocking uptake of copper from the diet may be a viable strategy for modulating tau pathology in Alzheimer’s disease.The potential benefits of this approach are tempered by the absence of a behavioral benefit and by the health risks of excessive lowering of copper. | Kellen Voss Christopher Harris Martina Ralle Megan Duffy Charles Murchison Joseph F Quinn | 2014 | Translational Neurodegeneration2014,3,1: | 3 |
