Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance, essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders. The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells, along with the signaling mechanisms governing stem cell self-renewal and differentiation. In this review,we first summarize the biological characteristics of seven types of dental stem cells, including dental pulp stem cells, stem cells from apical papilla, stem cells from human exfoliated deciduous teeth, dental follicle precursor cells, periodontal ligament stem cells, alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva. We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways. We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications. Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade. | Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji | 2016 | Genes & Diseases2016,3,4: | 16
The wonders of BMP9:From mesenchymal stem cell differentiation, angiogenesis, neurogenesis, tumorigenesis, and metabolism to regenerative medicine显示文摘Although bone morphogenetic proteins(BMPs)initially showed effective induction of ectopic bone growth in muscle,it has since been determined that these proteins,as members of the TGF-b superfamily, play a diverse and critical array of biological roles. These roles include regulating skeletal and bone formation, angiogenesis, and development and homeostasis of multiple organ systems.Disruptions of the members of the TGF-b/BMP superfamily result in severe skeletal and extra-skeletal irregularities, suggesting high therapeutic potential from understanding this family of BMP proteins.Although it was once one of the least characterized BMPs,BMP9 has revealed itself to have the highest osteogenic potential across numerous experiments both in vitro and in vivo, with recent studies suggesting that the exceptional potency of BMP9 may result from unique signaling pathways that differentiate it from other BMPs.The effectiveness of BMP9 in inducing bone formation was recently revealed in promising experiments that demonstrated efficacy in the repair of critical sized cranial defects as well as compatibility with bone-inducing bio-implants,revealing the great translational promise of BMP9.Furthermore,emerging evidence indicates that, besides its osteogenic activity,BMP9 exerts a broad range of biological functions, including stem cell differentiation, angiogenesis, neurogenesis, tumorigenesis, and metabolism. This review aims to summarize our current understanding of BMP9 across biology and the body. | Sami Mostafa Mikhail Pakvasa Elam Coalson Allen Zhu Alex Alverdy Hector Castillo Jiaming Fan Alex Li Yixiao Feng Di Wu Elliott Bishop Scott Du Mia Spezia Alissa Li Ofir Hagag Alison Deng Winny Liu Mingyang Li Sherwin S·Ho Aravind Athiviraham Michael J·Lee Jennifer Moriatis Wolf Guillermo A·Ameer Hue H·Luu Rex C·Haydon Jason Strelzow Kelly Hynes Tong-Chuan He Russell R·Reid | 2019 | Genes & Diseases2019,6,3: | 15
Highly expressed BMP9/GDF2 in postnatal mouse liver and lungs may account for its pleiotropic effects on stem cell differentiation, angiogenesis, tumor growth and metabolism显示文摘Bone morphogenetic protein 9(BMP9)(or GDF2)was originally identified from fetal mouse liver cDNA libraries.Emerging evidence indicates BMP9 exerts diverse and pleiotropic functions during postnatal development and in maintaining tissue homeostasis. However, the expression landscape of BMP9 signaling during development and/or in adult tissues remains to be analyzed. Here,we conducted a comprehensive analysis of the expression landscape of BMP9 and its signaling mediators in postnatal mice.By analyzing mouse ENCODE transcriptome datasets we found Bmp9 was highly expressed in the liver and detectable in embryonic brain, adult lungs and adult placenta. We next conducted a comprehensive qPCR analysis of RNAs isolated from major mouse tissues/organs at various ages.We found that Bmp9 was highly expressed in the liver and lung tissues of young adult mice, but decreased in older mice.Interestingly,Bmp9 was only expressed at low to modest levels in developing bones.BMP9-associated TGFβ/BMPR type I receptor Alk1 was highly expressed in the adult lungs.Furthermore, the feedback inhibitor Smads Smad6 and Smad7 were widely expressed in mouse postnatal tissues. However, the BMP signaling antagonist noggin was highly expressed in fat and heart in the older age groups,as well as in kidney, liver and lungs in a biphasic fashion. Thus, our findings indicate that the circulating BMP9 produced in liver and lungs may account for its pleiotropic effects on postnatal tissues/organs although possible roles of BMP9 signaling in liver and lungs remain to be fully understood. | Wei Liu Zhongliang Deng Zongyue Zeng Jiaming Fan Yixiao Feng Xi Wang Daigui Cao Bo Zhang Lijuan Yang Bin Liu Mikhail Pakvasa William Wagstaff Xiaoxing Wu Huaxiu Luo Jing Zhang Meng Zhang Fang He Yukun Mao Huiming Ding Yongtao Zhang Changchun Niu Rex C.Haydon Hue H.Luu Jennifer Moriatis Wolf Michael J.Lee Wei Huang Tong-Chuan He Yulong Zou | 2020 | Genes & Diseases2020,7,2: | 9
Deep-learning-enabled dual-frequency composite fringe projection profilometry for single-shot absolute 3D shape measurement显示文摘Single-shot high-speed 3D imaging is important for reconstructions of dynamic objects. For fringe projection profilometry(FPP),however,it is still challenging to recover accurate 3D shapes of isolated objects by a single fringe image. In this paper,we demonstrate that the deep neural networks can be trained to directly recover the absolute phase from a unique fringe image that involves spatially multiplexed fringe patterns of different frequencies. The extracted phase is free from spectrum-aliasing problem which is hard to avoid for traditional spatial-multiplexing methods.Experiments on both static and dynamic scenes show that the proposed approach is robust to object motion and can obtain high-quality 3D reconstructions of isolated objects within a single fringe image. | Yixuan Li Jiaming Qian Shijie Feng Qian Chen Chao Zuo | 2022 | Opto-Electronic Advances2022,5,5: | 8

Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. No difference was shown in sex, age, gastrectomy type, tumor location, and clinical T, N and M stage. Conclusions: Our study found that the IPA originates from GDA, ASPDA and RGEA in similar proportions.Laparoscopic surgery may be more helpful in dissection of the IPA than open surgery. | Rulin Miao Jianjun Qu Zhengrong Li Daguang Wang Jiang Yu Weidong Zang Yong Li Fenglin Liu Jian Zhang Wu Song Kai Ye Su Yan Wei Wang Shuangyi Ren Lu Zang Changqing Jing Li Zhang Kuan Wang Weihua Fu Lin Fan Bin Liang Gang Zhao Jun Cai Li Yang Jiaming Zhu Jun You Kun Yang Qingxing Huang Zhaojian Niu Ning Ning Xingfeng Qiu Gang Ji Feng Liang Hua Huang Chao Gao Fei Shan Shuangxi Li Yongning Jia Lianhai Zhang Xiangji Ying Yan Zhang Zhaode Bu Xiangqian Su Gang Zhao Ziyu Li Jiafu Ji | 2018 | Chinese Journal of Cancer Research2018,30,5: | 5
RNA-Seq Study Reveals AP2-Domain-Containing Signalling Regulators Involved in Initial Imbibition of Seed Germination in Rice显示文摘A number of internal signals are required for seed germination. However, the precise signalling responses in the initial imbibition of seed germination are not yet fully understood in rice.In this study, the RNA sequencing(RNA-Seq)approach was conducted in 8 h imbibed seeds to understand the signalling responses in the initial imbibition of rice seed germination.A total of 563 differentially expressed genes(DEGs)with at least 4-fold change were identified in 8 h imbibed seeds compared to dry seeds.MapMan analysis revealed that the majority of signalling response-related DEGs were hormone-and transcription factor-related genes,in which the largest number of DEGs belong to the AP2-domain-containing regulators, and their expressions were significantly induced in the initial imbibition of seed germination in rice. Moreover,at least five AP2-domain-containing transcription factor OsDREBs were identified in the initial imbibition of rice seed germination, and the expressions of 251 DEGs were putatively regulated by OsDREBs through the dehydration-responsive element(DRE)cis-element assay. It suggested that the OsDREBs might play important roles in the regulation of initial seed imbibition in rice. The identified genes provide a valuable resource to study the signalling regulation of seed germination in the future. | HE Yongqi ZHAO Jia FENG Defeng HUANG Zhibo LIANG Jiaming ZHENG Yufei CHENG Jinping YING Jifeng WANG Zhoufei | 2020 | Rice science2020,27,4: | 2
Integrated bioinformatics identifies the dysregulation induced by aberrant gene methylation in colorectal carcinoma显示文摘Colorectal carcinoma(CRC)is one of the most common cancers, and is associated with a poor dlinical outcome. The key genes and potential prognostic markers in colorectal carcinoma remain to be identfed and explored for clinical application.DNA expression/methyl-ation profles were downloaded from the Gene Expression Omnibus(GEO)database to identify differentially expressed/methylated genes(DEGs and DEMs).A total of 255 genes and 372 genes were identified as being up-regulated and down-regulated,respectively,in GSE113513,GSE81558,and GSE89076.There were a total of 3350 hypermethylated genes and 443 hypomethylated genes identifed in GSE 48684.Twenty genes were found to be hypermethylated as well as down-regulated,and a functional enrichment analysis revealed that these genes were mainly involved in cancer-related pathways.Among these 20 genes,GPM6A,HAND2 and C2orf40 were related to poor outcomes in cancer patients based on a survival analysis.Concurrent decreases of GPM6A,HAND2 and C2orf40 protein expression were observed in highly-dif ferentiated colorectal carcinoma tissues, and higher expression levels were found in undifferentiated or minimally-differentiated colorectal carcinoma tissues. In conclusion,20 genes were found to be downregulated and hypermethylated in CRC,among which GPM6A,HAND2 and C2orf40 were explored for their potential prognostic value. | Zhenyu Ye Yecheng Li Jiaming Xie Zhenyu Feng Xiaodong Yang Yong Wu Yuwei Pu Jiawei Gao Xiangrong Xu Zhaobi Zhu Wei Li Wei Chen Chungen Xing | 2021 | Genes & Diseases2021,8,4: | 2
The nucleocapsid protein of rice stripe virus in cell nuclei of vector insect regulates viral replication显示文摘Rice stripe virus(RSV)transmitted by the small brown planthopper causes severe rice yield losses in Asian countries.Although viral nuclear entry promotes viral replication in host cells, whether this phenomenon occurs in vector cells remains unknown. Therefore,in this study,we systematically evaluated the presence and roles of RSV in the nuclei of vector insect cells. We observed that the nucleocapsid protein(NP)and viral genomic RNAs were partially transported into vector cell nuclei by utilizing the importin a nuclear transport system. When blocking NP nuclear localization, cytoplasmic RSV accumulation significantly increased. In the vector cell nuclei,NP bound the transcription factor YY1 and affected its positive regulation to FAIM.Subsequently, decreased FAIM expression triggered an antiviral caspase-dependent apoptotic reaction. Our results reveal that viral nuclear entry induces completely different immune effects in vector and host cells, providing new insights into the balance between viral load and the immunity pressure in vector insects. | Wan Zhao Junjie Zhu Hong Lu Jiaming Zhu Fei Jiang Wei Wang Lan Luo Le Kang Feng Cui | 2022 | Protein & Cell2022,13,5: | 2
Biomarkers of aging显示文摘Aging biomarkers are a combination of biological parameters to(i)assess age-related changes,(ii)track the physiological aging process, and(iii)predict the transition into a pathological status.Although a broad spectrum of aging biomarkers has been developed, their potential uses and limitations remain poorly characterized. An immediate goal of biomarkers is to help us answer the following three fundamental questions in aging research:How old are we?Why do we get old?And how can we age slower? This review aims to address this need. Here,we summarize our current knowledge of biomarkers developed for cellular, organ, and organismal levels of aging, comprising six pillars:physiological characteristics, medical imaging, histological features, cellular alterations, molecular changes, and secretory factors. To fulfill all these requisites,we propose that aging biomarkers should qualify for being specific, systemic, and clinically relevant. | Aging Biomarker Consortium Hainan Bao Jiani Cao Mengting Chen Min Chen Wei Chen Xiao Chen Yanhao Chen Yu Chen Yutian Chen Zhiyang Chen Jagadish K Chhetri Yingjie Ding Junlin Feng Jun Guo Mengmeng Guo Chuting He Yujuan Jia Haiping Jiang Ying Jing Dingfeng Li Jiaming Li Jingyi Li Qinhao Liang Rui Liang Feng Liu Xiaoqian Liu Zuojun Liu Oscar Junhong Luo Jianwei Lv Jingyi Ma Kehang Mao Jiawei Nie Xinhua Qiao Xinpei Sun Xiaoqiang Tang Jianfang Wang Qiaoran Wang Siyuan Wang Xuan Wang Yaning Wang Yuhan Wang Rimo Wu Kai Xia Fu-Hui Xiao Lingyan Xu Yingying Xu Haoteng Yan Liang Yang Ruici Yang Yuanxin Yang Yilin Ying Le Zhang Weiwei Zhang Wenwan Zhang Xing Zhang Zhuo Zhang Min Zhou Rui Zhou Qingchen Zhu Zhengmao Zhu Feng Cao Zhongwei Cao Piu Chan Chang Chen Guobing Chen Hou-Zao Chen Jun Chen Weimin Ci Bi-Sen Ding Qiurong Ding Feng Gao Jing-Dong JHan Kai Huang Zhenyu Ju Qing-Peng Kong Ji Li Jian Li Xin Li Baohua Liu Feng Liu Lin Liu Qiang Liu Qiang Liu Xingguo Liu Yong Liu Xianghang Luo Shuai Ma Xinran Ma Zhiyong Mao Jing Nie Yaojin Peng Jing Qu Jie Ren Ruibao Ren Moshi Song Zhou Songyang Yi Eve Sun Yu Sun Mei Tian Shusen Wang Si Wang Xia Wang Xiaoning Wang Yan-Jiang Wang Yunfang Wang Catherine CL Wong Andy Peng Xiang Yichuan Xiao Zhengwei Xie Daichao Xu Jing Ye Rui Yue Cuntai Zhang Hongbo Zhang Liang Zhang Weiqi Zhang Yong Zhang Yun-Wu Zhang Zhuohua Zhang Tongbiao Zhao Yuzheng Zhao Dahai Zhu Weiguo Zou Gang Pei Guang-Hui Liu | 2023 | Science China(Life Sciences)2023,66,5: | 2

pH- and H_(2)O_(2)-sensitive drug delivery system based on sodium xanthate:Dual-responsive supramolecular vesicles from one functional group显示文摘Nano-drug delivery systems with multiple stimulus-responsive capabilities have superior response performance and efficient drug release.Nevertheless,it is sophisticated to construct multiple stimulus-responsive systems where the two or more functional groups need to be introduced simultaneously.Xanthate, one functional group with pH and H2O2 stimulus responsiveness, has significant potential applications for building dual-responsive drug delivery system.Herein,we present a novel dual stimuli-responsive supramolecular drug delivery system by using sodium xanthate derivative(SXD)as guest molecule and quaternary ammonium capped pillar[5]arene(QAP5)as host molecule through host-guest interaction on the basis of electrostatic interaction. The amphiphile QAP5⊃SXD could self-assemble into vesicles to efficiently load the anti-cancer drug DOX.The experimental results showed that QAP5⊃SXD nanoparticles could achieve efficient drug delivery and controlled release in the tumor microenvironment.Cytotoxicity experiments proved that DOX@QAP5⊃SXD nanoparticles could significantly improve the anticancer efficiency of free DOX on cancer cells. The present study provides an efficient strategy to develop supramolecular nanocarriers with dual-responsiveness in one functional group for controlled drug release. | Ziyan Shen Ning Ma Feng Wang Jiaming Ren Chenxi Hou Shuang Chao Yuxin Pei Zhichao Pei | 2022 | Chinese Chemical Letters2022,33,10: | 1

Luminescence interference-free lifetime nanothermometry pinpoints in vivo temperature显示文摘Luminescence nanothermometry makes non-invasive and real-time temperature readings possible in living animals. However, the spectral fluctuation in tissues and fluids,as well as the interaction between fluorophores and environment hinders accuracy of the thermometry. Here,we report a luminescence lifetime-based nanothermometry which specifically addresses this problem.A temporal based calibration(lifetime sensing)in the NIR range,an endogenous thermal response as well as a polymer encapsulation evading environmental factors, altogether help to pinpoint temperature in vivo. Thanks to the highly condensed NdYb ions in a well-protected tiny core-shell nanocrystal(overall 11 nm),a temperature sensitivity about 2.07%K^(-1)(with 5%Yb^(3+)doped nanoparticles)and an accuracy of 0.27 K(with 25%Yb^(3+)doped nanoparticles)in biological fluids are achieved.Hopefully, combining thermally activated energy transfer nanothermometer with anti-interference lifetime thermometry would provide a more accurate temperature measurement for biological and preclinical studies. | Mengya Kong Yuyang Gu Yingjie Chai Jiaming Ke Yulai Liu Xincheng Xu Zhanxian Li Wei Feng Fuyou Li | 2021 | Science China Chemistry2021,64,6: | 1
Optimization of microbial cell factories for astaxanthin production: Biosynthesis and regulations, engineering strategies and fermentation optimization strategies显示文摘The global market demand for natural astaxanthin is rapidly increasing owing to its safety, the potential health benefits, and the diverse applications in food and pharmaceutical industries. The major native producers of natural astaxanthin on industrial scale are the alga Haematococcus pluvialis and the yeast Xanthopyllomyces dendrorhous. However, the natural production via these native producers is facing challenges of limited yield and high cost of cultivation and extraction.Alternatively, astaxanthin production via metabolically engineered non-native microbial cell factories such as Escherichia coli,Saccharomyces cerevisiae and Yarrowia lipolytica is another promising strategy to overcome these limitations. In this review we summarize the recent scientific and biotechnological progresses on astaxanthin biosynthetic pathways, transcriptional regulations, the interrelation with lipid metabolism, engineering strategies as well as fermentation process control in major native and non-native astaxanthin producers. These progresses illuminate the prospects of producing astaxanthin by microbial cell factories on industrial scale. | Mostafa Basiony Liming Ouyang Danni Wang Jiaming Yu Liming Zhou Mohan Zhu Xuyuan Wang Jie Feng Jing Dai Yijie Shen Chengguo Zhang Qiang Hua Xiuliang Yang Lixin Zhang | 2022 | Synthetic and Systems Biotechnology2022,7,2: | 1
