| 7 | Effects of biological soil crusts and drought on emergence and survival of a Patagonian perennial grass in the Monte of Argentina显示文摘Biological soil crusts are widely distributed in arid and semiarid regions. They have an important ecological role, especially by modifying physical and chemical properties of soils. Biological crusts may also modify seed germination and seedling establishment. The effects vary widely according to the type of crust and the vascular plant species. The objective of this study was to determine the effect of moss-dominated biological soil crusts on the emergence, biomass and survival of Poa ligularis Nees ex Steud. under different irrigation regimes. We collected seeds of P. ligularis and biological soil crusts composed of two species of mosses: Syntrichia princeps(De Not.) Mitt and Ceratodon purpureus(Hedw.) Brid. from an area in the Monte of Argentina. The result showed that seedling emergence of P. ligularis was higher in treatments with bare soil than in soil covered by crusts, and also in those with watering to field capacity. Mean emergence time was higher in treatments with bare soil and watering to field capacity. Seedling biomass also showed significant differences between treatments. | 14 | Complete loss of RNA editing from the plastid genome and most highly expressed mitochondrial genes of Welwitschia mirabilis显示文摘Comparative genomics among gymnosperms suggested extensive loss of mitochondrial RNA editing sites from Welwitschia mirabilis based on predictive analysis. However, empirical or transcriptome data to confirm this massive loss event are lacking,and the potential mechanisms of RNA site loss are unclear. By comparing genomic sequences with transcriptomic and reversetranscription PCR sequencing data, we performed a comprehensive analysis of the pattern of RNA editing in the mitochondrial and plastid genomes(mitogenome and plastome, respectively) of W. mirabilis and a second gymnosperm, Ginkgo biloba. For W.mirabilis, we found only 99 editing sites located in 13 protein-coding genes in the mitogenome and a complete loss of RNA editing from the plastome. The few genes having high editing frequency in the Welwitschia mitogenome showed a strong negative correlation with gene expression level. Comparative analyses with G. biloba, containing 1,405 mitochondrial and 345 plastid editing sites, revealed that the editing loss from W. mirabilis is mainly due to the substitution of editable cytidines to thymidines at the genomic level, which could be caused by retroprocessing. Our result is the first study to uncover massive editing loss from both the mitogenome and plastome in a single genus. | 15 | Proteome alterations in aqueous humour of primary open angle glaucoma patients显示文摘AIM: To unravel the primary open angle glaucoma(POAG) related proteomic changes in aqueous humour(AH).METHODS: Totally 35 patients listed for cataract surgery(controls: n=12, age: 67.4±13.6 y) or trabeculectomy for POAG(n=23, age: 72.5±8.3 y) were included. AH samples of those patients were obtained during cataract surgery or trabeculectomy. AH samples were subsequently pooled into the experimental groups under equal contribution in terms of protein amount of each individual patient. Protein samples were analyzed by a linear trap quadrupol Orbitrap Mass Spectrometry device with an upstream liquid chromatography system. The obtained raw data were analyzed using the Maxquant proteome software and compared. Proteins with a fold-change ratio higher than a cut-off of 2 were considered as noticeably altered.RESULTS: A total number of 175 proteins could be identified out of the AH from POAG and cataract by means of quantitative mass spectrometric analysis.
