| 1 | The Sunway TaihuLight supercomputer: system and applications显示文摘The Sunway Taihu Light supercomputer is the world's first system with a peak performance greater than 100 PFlops. In this paper, we provide a detailed introduction to the Taihu Light system. In contrast with other existing heterogeneous supercomputers, which include both CPU processors and PCIe-connected many-core accelerators(NVIDIA GPU or Intel Xeon Phi), the computing power of Taihu Light is provided by a homegrown many-core SW26010 CPU that includes both the management processing elements(MPEs)and computing processing elements(CPEs) in one chip. With 260 processing elements in one CPU, a single SW26010 provides a peak performance of over three TFlops. To alleviate the memory bandwidth bottleneck in most applications, each CPE comes with a scratch pad memory, which serves as a user-controlled cache. To support the parallelization of programs on the new many-core architecture, in addition to the basic C/C++and Fortran compilers, the system provides a customized Sunway Open ACC tool that supports the Open ACC2.0 syntax. | 3 | sLe^(x) expression in invasive micropapillary breast carcinoma is associated with poor prognosis and can be combined with MUC1/EMA as a supplementary diagnostic indicator显示文摘Objective: Mucin 1(MUC1/EMA) and sialyl Lewis X(sLe^(x)) indicate polarity reversal in invasive micropapillary carcinoma(IMPC). The purpose of this study was to evaluate the expression of MUC1/EMA and sLe^(x) and to assess their diagnostic and prognostic value in patients with IMPC.Methods: The expression of sLe^(x) and MUC1/EMA in 100 patients with IMPC and a control group of 89 patients with invasive ductal carcinoma not otherwise specified(IDC-NOS) were analyzed with IHC. Fresh tumor tissues were collected from patients with IMPC or IDC-NOS for primary culture and immunofluorescence analysis.Results: The rate of nodal metastasis was higher in patients with IMPC than those with IDC-NOS, and IMPC cells tended to express more sLe^(x) and MUC1/EMA in the cytomembranes(the stroma-facing surfaces of the micropapillary clusters) than IDC-NOS cells. In IMPC, high cytomembrane expression of sLe^(x), but not MUC1/EMA, indicated poor prognosis. In addition, among the 100 patients with IMPC, 10 patients had sLe^(x)+/EMA-expression patterns, and 8 patients had sLe^(x)-/EMA+ expression patterns. The primary IMPC cells were suspended, non-adherent tumor cell clusters, whereas the primary IDC cells were adherent tumor cells. | 7 | An improved binocular localization method for apple based on fruit detection using deep learning显示文摘Apple picking robot is now being developed as an alternative to hand picking due to a great demand for labor during apple harvest season.Accurate detection and localization of target fruit is necessary for robotic apple picking.Detection accuracy has a great influence on localization results.Although current researches on detection and localization of apples using traditional image algorithms can obtain good results under laboratory conditions,it is difficult to accurately detect and locate objects in natural field with complex environments.With the rapid development of artificial intelligence,accuracy of apple detection based on deep learning has been significantly improved.Therefore,a deep learningbased method was developed to accurately detect and locate the position of fruit.For different localization methods,binocular localization is a widely used localization method for its bionic principle and lower equipment cost.Hence,this paper proposed an improved binocular localization method for apple based on fruit detection using deep learning.First,apples of binocular images were detected by Faster R-CNN.After that,a segmentation based on chromatic aberration and chromatic aberration ratio was applied to segment apple and background pixels in bounding box of detected fruit.Furthermore,template matching with parallel polar line constraint was used to match apples in left and right images.Finally,two feature points on apples were selected to directly calculate three dimensional coordinates of feature points with the binocular localization principle.In this study,Faster R-CNN achieved an AP of 88.12%with an average detection speed of 0.32 s for an image.Meanwhile,standard deviation and localization precision of depth of two feature points on each apple were calculated to evaluate localization.Results showed that the average standard deviation and the average localization precision of 76 groups of datasets were 0.51 cm and 99.64%,respectively.Results indicated that the proposed improved binocular localization method is promising for fruit localization。 | Tengfei Li Wentai Fang Guanao Zhao Fangfang Gao Zhenchao Wu Rui Li Longsheng Fu Jaspreet Dhupia | 2023 | Information Processing in Agriculture2023,10,2: | 1 |
| 8 | Comprehensive Humoral and Cellular Immune Responses to SARS-CoV-2 Variants in Diverse Chinese Population显示文摘The SARS-CoV-2 variants have been emerging and have made great challenges to current vaccine and pandemic control strategies.It is urgent to understand the current immune status of various Chinese populations given that the preexisting immunity has been established by national vaccination or ex posure to past variants.Using sera from 85 individuals(including 21 convalescents of natural infection,15 cases which suffered a breakthrough infection after being fully vaccinated,and 49 healthy vaccinees),we showed signifcantly enhanced neutralizing activities against SRAS-CoV-2 variants in convalescent sera,especially those who had been fully vaccinated.The neut ralizing antibodies against Omicron were detectable in 75%of convalescents and 4.9%of healthy vaccinees(p=0.006),with a GMT of 289.5,180.9-463.3,and 42.6,31.3-59,respetively.However,the neutralizing activities were weaker in young convalescents(aged<18y),with a detectable rate of 50%and a GMT of 46.4 against Omicron.We also examined and found no pan.sarbecovirus neutralizing activities in vaccinated SARS-CoV-I survivors.A booster dose could further increase the breadth and magnitude of neutralization against WT and variants of concern(VOCs)to different degrees.In addition,we showed that COVID-19-inactivated vaccines can elicit Omicron-specific T-cell responses.The positive rates of ELISpot reactions were 26.7%(4/15)and 43.8%(7/16)in the full vaccination group and the booster vaccination group,respectively,although without statistically significant difference.The neutralizing antibody titers declined while T-cell responses remain consistent over 6 months.These findings will inform the optimization of public health vaccination and intervention strategies to protect diverse populations against SARS-CoV-2 variants.Advances.Breakthrough infection significantly boosted neutralizing activities against SARS-CoV-2 variants a compared to booster immunization with inaclivated vaccine.Vaccine-induced vir us-specific T-cell immunity,on the other hand,may compensate for the shortall.Furthermore,the public health system should target the most vulnerable group due to a poorer protective serological response in both infected and vaccinated adolescents. | Jiwei Li Jing Wu Qiuyue Long Yan'an Wu Xiaoyi Hu Yukun He Mingzheng Jiang Jia Li Lili Zhao Shuoqi Yang Xiaoyong Chen Minghui Wang Jianshi Zheng Fangfang Wu Ruiliang Wu Lihong Ren Liang Bu Houzhao Wang Ke Li Lijuan Fu Guojun Zhang Yali Zheng Zhancheng Gao | 2022 | Research2022,,2: | 1 |

| 13 | Plant and animal positive-sense single-stranded RNA viruses encode small proteins important for viral infection in their negative-sense strand显示文摘Positive-sense single-stranded RNA(+ssRNA)viruses,the most abundant viruses of eukaryotes in nature,require the synthesis of negative-sense RNA(-RNA)using their genomic(positive-sense)RNA(+RNA)as a template for replication.Based on current evidence,viral proteins are translated via viral+RNAs,whereas-RNA is considered to be a viral replication intermediate without coding capacity.Here,we report that plant and animal+ssRNA viruses contain small open reading frames(ORFs)in their-RNA(reverse ORFs[rORFs]).Using turnip mosaic virus(TuMV)as a model for plant+ssRNA viruses,we demonstrate that small proteins encoded by rORFs display specific subcellularlocalizations,and confirm the presence of rORF2 in infected cells through mass spectrometry analysis.The protein encoded by TuMV rORF2 forms punctuate granules that are localized in the perinuclear region and co-localized with viral replication complexes.The rORF2 protein can directly interact with the viral RNA-dependent RNA polymerase,and mutation of rORF2 completely abolishes virus infection,whereas ectopic expression of rORF2 rescues the mutant virus.Furthermore,we show that several rORFs in the-RNA of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)have the ability to suppress type l interferon production and facilitate the infection of ve-sicular stomatitis virus.In addition,we provide evidence that TuMV might utilize internal ribosome entry sites to translate these small rORFs.Taken together,these findings indicate that the-RNA of+ssRNA vi-ruses can also have the coding capacity and that small proteins encoded therein play critical roles in viral infection,revealing a viral proteome larger than previously thought. | Pan Gong Qingtang Shen Mingzhen Zhang Rui Qiao Jing Jiang Lili Su Siwen Zhao Shuai Fu Yu Ma Linhao Ge Yaqin Wang Rosa Lozano-Durán Aiming Wang Fangfang Li Xueping Zhou | 2023 | Molecular Plant2023,16,11: | 0 |

| 16 | Silencing of syndecan-binding protein enhances the inhibitory effect of tamoxifen and increases cellular sensitivity to estrogen显示文摘Objective:Tamoxifen is used as a complementary treatment for estrogen receptor(ER)-positive breast cancer(BCa),but many patients developed resistance.The aim of this study was to examine the role of syndecan-binding protein(SDCBP)silencing in ER-positive BCa cells.Methods:In MCF-7/T47D cells,the effects of SDCBP silence/overexpression on cell proliferation and estrogenic response were examined.Cell proliferation was examined using the MTT assay and cell cycle regulators were examined by Western blot.Estrogen response was examined from a luciferase activity and evaluation of transcript levels of p S2 and progesterone receptor(PR)upon estrogen administration.Samples of ER-positive BCa were stained with ERα,PR,and SDCBP antibodies,and their expression correlations were analyzed.Results:We found that SDCBP silencing inhibited the proliferation of ER-positive BCa cells and arrested a greater number of cells in the G1 phase of the cell cycle compared to tamoxifen alone,while SDCBP overexpression limited the anti-cancer effects of tamoxifen.SDCBP silencing and overexpression also enhanced and attenuated the estrogenic response,respectively.Expression of SDCBP was negatively correlated with PR,ERα,and the PR/ERαratio in ER-positive BCa tissue samples.Conclusions:SDCBP may be involved in tamoxifen resistance in ER-positive BCa.Tamoxifen treatment combined with SDCBP silencing may provide a novel treatment for endocrine therapy-resistant BCa. | Jun Zhang Xiaolong Qian Fangfang Liu Xiaojing Guo Feng Gu Li Fu | 2018 | Cancer Biology & Medicine2018,15,1: | 0 |

| 18 | Ginsenoside rg3 reduces body weight by regulating fat content and browning in obese mice显示文摘Objective:To determine the effects of ginsenoside rg3 on the body weight of C57BL/6J obese mice and to investigate its underlying weight loss mechanisms with a focus on white fat browning-related factors.Methods:Eight-week-old C57BL/6J male mice were fed a high-fat diet for 12 successive weeks to construct the obese model.C57BL/6J male mice were fed a standard chow diet to construct normal control group.After 8 weeks of intervention with ginsenoside rg3,the food intake,body weight,body fat mass,blood sugar,and lipid profiles of the mice in each group were detected.Hematoxylin and eosin(HE)staining was used to observe the histological morphology of the adipose tissues.Real-time polymerase chain reaction(RT-PCR)and Western blotting(WB)were applied to detect the gene and protein expression levels of peroxisome proliferators-activated receptor gama(PPARg),Peroxisome proliferatoractivated receptor-gamma coactivator-1alpha(PGC-1a),PR domain containing 16(PRDM16),and uncoupling protein 1(UCP-1).Results:Compared to normal control group mice,the body weight,food intake,body fat composition,and blood lipid levels of model group mice increased significantly.After 8 weeks of intervention with ginsenoside rg3,body weight,body fat composition,food intake,and blood lipid profiles decreased.HE staining showed that ginsenoside rg3 can improve white adipocyte hypertrophy to a certain extent.RTPCR and WB demonstrated that ginsenoside rg3 can increase the mRNA and protein expression levels of PPARg,PGC-1a,PRDM16,and UCP-1 in the adipose tissues of obese mice.Conclusion:The weight reduction effect of ginsenoside rg3 may be related to the promotion of white fat browning. | Qianqian Mu Jiacheng Zuo Dandan Zhao Xiaoshan Zhou Jing Hua Ying Bai Fangfang Mo Xin Fang Min Fu Sihua Gao | 2021 | Journal of Traditional Chinese Medical Sciences2021,8,1: | 0 |

| 20 | The Metasequoia genome and evolutionary relationships among redwoods显示文摘Redwood trees(Sequoioideae),including Metasequoia glyptostroboides(dawn redwood),Sequoiadendron giganteum(giant sequoia),and Sequoia sempervirens(coast redwood),are threatened and widely recognized iconic tree species.Genomic resources for redwood trees could provide clues to their evolutionary relationships.Here,we report the 8-Gb reference genome of M.glyptostroboides and a comparative analysis with two related species.More than 62%of the M.glyptostroboides genome is composed of repetitive sequences.Clade-specific bursts of long terminal repeat retrotransposons may have contributed to genomic differentiation in the three species.The chromosomal synteny between M.glyptostroboides and S.giganteum is extremely high,whereas there has been significant chromosome reorganization in S.sempervirens.Phylogenetic analysis of marker genes indicates that S.sempervirens is an autopolyploid,and more than 48%of the gene trees are incongruent with the species tree.Results of multiple analyses suggest that incomplete lineage sorting(ILS)rather than hybridization explains the inconsistent phylogeny,indicating that genetic variation among redwoods may be due to random retention of polymorphisms in ancestral populations.Functional analysis of ortholog groups indicates that gene families of ion channels,tannin biosynthesis enzymes,and transcription factors for meristem maintenance have expanded in S.giganteum and S.sempervirens,which is consistent with their extreme height.As a wetland-tolerant species,M.glyptostroboides shows a transcriptional response to flooding stress that is conserved with that of analyzed angiosperm species.Our study offers insights into redwood evolution and adaptation and provides genomic resources to aid in their conservation and management. | Fangfang Fu Chi Song Chengjin Wen Lulu Yang Ying Guo Xiaoming Yang Ziqiang Shu Xiaodong Li Yangfan Feng Bingshuang Liu Mingsheng Sun Yinxiao Zhong Li Chen Yan Niu Jie Chen Guibin Wang Tongming Yin Shilin Chen Liangjiao Xue Fuliang Cao | 2023 | Plant Communications2023,4,6: | 0 |
