| 1 | Rapid growth cost in “all-fish” growth hormone gene transgenic carp:Reduced critical swimming speed显示文摘Evidence has accumulated that there is a trade-off between benefits and costs associated with rapid growth. A trade-off between growth rates and critical swimming speed (Ucrit) had been also reported to be common in teleost fish. We hypothesize that growth acceleration in the F3 generation of 'all-fish' growth hormone gene (GH) transgenic common carp (Cyprinus carpio L.) would reduce the swimming abilities. Growth and swimming performance between transgenic fish and non-transgenic controls were compared. The results showed that transgenic fish had a mean body weight 1.4―1.9-fold heavier,and a mean specific growth rate (SGR) value 6%―10% higher than the controls. Transgenic fish,however,had a mean absolute Ucrit (cm/s) value 22% or mean relative Ucrit (BL/s) value 24% lower than the controls. It suggested that fast-growing 'all-fish' GH-transgenic carp were inferior swimmers. | 3 | Insulin-like growth factor 1-induced enolase 2 deacetylation by HDAC3 promotes metastasis of pancreatic cancer显示文摘Enolase 2(ENO2)is a key glycolytic enzyme in the metabolic process of glycolysis,but its potential function in pancreatic ductal adenocarcinoma(PDAC)is unclear.In this study,we observed a significant overexpression of ENO2 in PDAC tissues,and its expression was correlated with metastasis and poor prognosis in PDAC patients.K394 was identified as a major acetylation site in ENO2 that regulates its enzymatic activity,cell metabolism and PDAC progression.Knockdown of ENO2 suppressed tumor growth and liver metastasis in PDAC.Re-expression of wild-type(WT)ENO2,but not the K394 acetylation mimetic mutant,could reverse the decreased tumor malignancy.We further characterized histone deacetylase 3(HDAC3)and P300/CBP-associated factor(PCAF)as the potential deacetylase and acetyltransferase for ENO2,respectively.HDAC3-mediated deacetylation was shown to lead to ENO2 activation and enhancement of glycolysis.Importantly,insulin-like growth factor-1(IGF-1)was found to decrease K394 acetylation and stimulate ENO2 activity in a dose-and time-dependent manner.The PI3K/AKT/mTOR pathway facilitated the phosphorylation of HDAC3 on S424,which promoted K394 deacetylation and activation of ENO2.Linsitinib,an oral small-molecule inhibitor of IGF-1R,could inhibit IGF-1-induced ENO2 deacetylation by HDAC3 and the PI3K/AKT/mTOR pathway.Furthermore,linsitinib showed a different effect on the growth and metastasis of PDAC depending on the overexpression of WT versus K394-mutant ENO2.Our results reveal a novel mechanism by which acetylation negatively regulates ENO2 activity in the metastasis of PDAC by modulating glycolysis.Blockade of IGF-1-induced ENO2 deacetylation represents a promising strategy to prevent the development of PDAC. | Yan Zheng Chao Wu Jimeng Yang Yue Zhao Huliang Jia Min Xue Da Xu Feng Yang Deliang Fu Chaoqun Wang Beiyuan Hu Ze Zhang Tianen Li Shican Yan Xuan Wang Peter J.Nelson Christiane Bruns Lunxiu Qin Qiongzhu Dong | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 3 |

However, substantial quantities of satellite data, especially those derived from low-level(surface-sensitive)channels, are rejected for use because of the difficulty in realistically modeling land surface emissivity and energy budgets.Here, we used an improved land use and leaf area index(LAI) dataset in the WRF-3 DVAR assimilation system to explore the benefit of using improved quality of land surface information to improve rainfall simulation for the Shule River Basin in the northeastern Tibetan Plateau as a case study. The results for July 2013 show that, for low-level channels(e.g., channel 3),the underestimation of brightness temperature in the original simulation was largely removed by more realistic land surface information. In this paper, a new virus propagation model considering the system recovery is proposed first, and then in its steady-state analysis, the virus propagation steady time and steady states are deduced. | 18 | Quantitative Gas-in-place Comparison of Original and Bitumen-free Lacustrine Shale显示文摘Residual bitumen in organic-rich shale of oil windows exists widely, and its effect on the gas storage capacity of shale could be two-fold. Bitumen could occupy and block the nanopores of shale, thereby reducing the gas storage capacity. On the other hand, gas could be dissolved in bitumen in shale gas reservoirs, leading to enhanced gas storage capacity. To quantify the effect of bitumen on the gas-in-place(GIP) estimation of lacustrine organic-rich shale, the micropore characteristics and methane sorption capacity of original and bitumen-free shale from the Triassic Yangchang Formation of the Ordos Basin, combined with the methane dissolution capacity for the isolated bitumen, were analyzed and compared. GIP for the original and bitumen-free shale in the depth range of 500–2500 m was evaluated. The results show that micropores in the shale samples were mainly related to organic matter. Clay mineral-hosted pores contributed slightly to microporosity. Bitumen significantly reduced the micropore surface area and volume of the original shale, with average percentages of 28.09% and 51.26%, respectively. The methane sorption capacity decreased after bitumen removal. When normalized to the original shale mass, the sum of the methane sorption capacity for bitumen-free shale and the methane dissolution capacity for isolated bitumen was similar to the methane sorption capacity of the original shale, indicating that the lack of methane absorbed on bitumen is the main reason for the decrease in methane sorption capacity after bitumen removal. The contribution of absorbed methane on bitumen to sorbed methane in shale could be higher than 36.23%. Dual effects of bitumen on shale GIP were observed.
