| 2 | A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide. | Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen | 2019 | Molecular Plant2019,12,5: | 18 |
| 3 | Relationship between the Flow Blockage of Tip Leakage Vortex and its Evolutionary Procedures inside the Rotor Passage of a Subsonic Axial Compressor显示文摘The near casing flow fields inside the rotor passage of a 1.5 stage axial compressor with different blade-loading levels and tip gap sizes were measured by using stereoscopic particle image velocimetry(SPIV). Based on a carefully defined blockage extracting method, the variations of blockage parameter inside the blade passage were analyzed. It was found that the variation of blockage parameter appeared as a non-monotonic behavior inside the blade passage in most cases. This non-monotonic behavior became much more remarkable as the blade loading increases or mass flow rate decreases.The variations of the blockage parameter inside the blade passage had close relation to the evolutionary procedures of the tip leakage vortex(TLV). The destabilization of the TLV caused a rapid increasing of the blockage parameter. After the TLV lost the features of a concentrated streamwise vortex,the blockage parameter usually got a peak value. | 11 | Standard pancreatoduodenectomy versus extended pancreatoduodenectomy with modified retroperitoneal nerve resection in patients with pancreatic head cancer:a multicenter randomized controlled trial显示文摘Background:The extent of pancreatoduodenectomy for pancreatic head cancer remains controversial,and more high-level clinical evidence is needed.This study aimed to evaluate the outcome of extended pancreatoduodenectomy(EPD)with retroperitoneal nerve resection in pancreatic head cancer.Methods:This multicenter randomized trial was performed at 6 Chinese highvolume hospitals that enrolled patients between October 3,2012,and September 21,2017.Four hundred patients with stage I or II pancreatic head cancer and without specific pancreatic cancer treatments(preoperative chemotherapy or chemoradiation)within three months were randomly assigned to undergo standard pancreatoduodenectomy(SPD)or EPD,with the latter followed by dissection of additional lymph nodes(LNs),nerves and soft tissues 270◦on the right side surrounding the superior mesenteric artery and celiac axis.The primary endpoint was overall survival(OS)by intention-to-treat(ITT).The secondary endpoints were disease-free survival(DFS),mortality,morbidity,and postoperative pain intensity.Results:TheR1 ratewas slightly lower with EPD(8.46%)thanwith SPD(12.56%).The morbidity and mortality rates were similar between the two groups.The median OS was similar in the EPD and SPD groups by ITT in the whole study cohort(23.0 vs.20.2 months,P=0.100),while the median DFS was superior in the EPD group(16.1 vs.13.2 months,P=0.031).Patients with preoperative CA19–9<200.0 U/mL had significantly improved OS and DFS with EPD(EPD vs.SPD,30.8 vs.20.9 months,P=0.009;23.4 vs.13.5 months,P<0.001).The EPD group exhibited significantly lower locoregional(16.48%vs.35.20%,P<0.001)andmesenteric LNrecurrence rates(3.98%vs.10.06%,P=0.022).The EPD group exhibited less back pain 6 months postoperation than the SPD group.Conclusions:EPD for pancreatic head cancer did not significantly improve OS,but patients with EPD treatment had significantly improved DFS.In the subgroup analysis,improvements in bothOS and DFS in the EPD armwere observed in patients with preoperative CA19–9<200.0 U/mL.EPD could be used as an effective surgical procedure for patients with pancreatic head cancer,especially those with preoperative CA19–9<200.0 U/mL. | Qing Lin Shangyou Zheng Xianjun Yu Meifu Chen Yu Zhou Quanbo Zhou Chonghui Hu Jing Gu Zhongdong Xu LinWang Yimin Liu Qingyu Liu MinWang Guolin Li He Cheng Dongkai Zhou Guodong Liu Zhiqiang Fu Yu Long Yixiong Li Weilin Wang Renyi Qin Zhihua Li Rufu Chen | 2023 | Cancer Communications2023,43,2: | 3 |

| 13 | The Dynamic Experiment on Treating Acid Mine Drainage with Iron Scrap and Sulfate Reducing Bacteria Using Biomass Materials as Carbon Source显示文摘The study is aimed at the problem of high content of Cr^(6+),Cr^(3+)and SO_(4)^(2-)is high and low pH value in acid mine drainage(AMD).Moreover,treatment of AMD by sulfate reducing bacteria(SRB)requires the addition of carbon source,while the treating effectiveness is not good enough on its own.The sugarcane slag,the corn cob and the sunflower straw were selected as the SRB carbon source cooperating with iron scrap to construct the dynamic columns 1,2 and 3.The mechanism of removing Cr^(6+),Cr^(3+),SO_(4)^(2-)and H+and the regularity of sustained release of carbon source and TFe release was studied in AMD.The removal efficiency of heavy metal ions,the ability of sustained release of carbon source,and the ability of adjusting acid by the three dynamic columns were compared.The result shows that the average removal rates of Cr^(6+),Cr^(3+)and SO_(4)^(2-)in effluent of dynamic column 1,filled by sugarcane slag,iron scrap and SRB,were 96.9%,67.1%and 54.3%.The average release of TFe and chemical oxygen demand(COD)were 4.4 and 287.3 mg/L.Its average pH was 6.98.Compared with the performance of dynamic columns 1,2 and 3,dynamic column 1 performed best in removing Cr^(6+),Cr^(3+)and SO_(4)^(2-)from AMD and controlling the release of COD and TFe,adjusting the pH of the solution.The study is of significance in treatment of AMD by taking for biomass materials as SRB carbon source in cooperation with iron scrap. | Xianjun Wang Junzhen Di Yanrong Dong Yu Yang Bing Liang Fankang Meng Tingting Wang Wenbo An Zengxin Li Jianzhi Guo | 2021 | Journal of Renewable Materials2021,9,1: | 2 |
| 14 | Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. | 16 | Using tandem blades to break loading limit of highly loaded axial compressors显示文摘It is confirmed that tandem-blade configurations have potential to enlarge the flow turning in two-dimension(2D) studies. However, the potential of tandem blades to enlarge the design space for highly loaded axial compressors was rarely investigated in open literatures. The present work aims to show the capability of tandem blades to break the loading limit of conventional blades for highly loaded compressors. The 2D models of the maximum static pressure rise derived in previous work were validated by a large amount experimental data, which showed a good agreement. An E parameter was defined to evaluate the stall margin of compressor based on the theoretical models, which indicated that the tandem blade was able to increase the loading limit of axial compressors. A single-blade stage with a loading coefficient of 0.46(based on the blade tip rotating speed) was designed as the baseline case under the guidance of the E parameter. A tandem-blade stage was then designed by ensuring that the velocity triangles were similar to the single-blade stage. The performances of both stages were investigated experimentally. The results showed that the maximum efficiency of the tandem-blade stage was 92.8%, 1% higher than the single;the stall margin increased from 16.9% to 22.3%. | 17 | FGFBPI-mediated crosstalk between fibroblasts and pancreatic cancer cells via FGF22/FGFR2 promotes invasion and metastasis of pancreatic cancer显示文摘Fibroblast growth factor-binding protein 1(FGFBP1)promotes fibroblast growth factor(FGF)activity by releasing FGFs from extracellular matrix storage.We previously reported that the tumor suppressor F-box and WD repeat domain-containing 7 suppresses FGFBP1 by reducing expression of c-Myc,which inhibits the proliferation and migration of pancreatic cancer cells.However,the potential mechanism by which FGFBP1 facilitates pancreatic ductal adenocarcinoma(PDAC)remains unexplored.In this study,we focused on the function of FGFBP1 in the interplay between cancer-associated fibroblasts(CAFs)and pancreatic cancer cells(PCCs).Decreased FGF22 expression was detected in CAFs co-cultured with PCCs with FGFBP1 abrogation,which was verified in the cell culture medium by enzyme-linked immunosorbent assay.Active cytokine FGF22 significantly facilitated the migration and invasion of PANC-1 and Mia PaCa-2 cells.The number of penetrating PCCs cocultured with CAFs with FGF22 abrogation was significantly less than that of the control group.Interestingly,higher expressions of FGF22 and fibroblast growth factor receptor 2(FGFR2)were associated with worse prognosis of patients with PDAC and FGFR2,an independent prognostic marker of PDAC.The PANC-1 and Mia PaCa-2 cells with silenced FGFR2 showed weaker invasion and metastasis,even if these cells were simultaneously treated with cytokine FGF22.These results revealed that FGFBPI-mediated interaction between CAFs and PCCs via FGF22/FGFR2 facilitates the migration and invasion of PCCs.FGFR2 could act as a prognostic marker for patients with PDAC. | Zheng Zhang Yi Qin Shunrong Ji Wenyan Xu Mengqi Liu Qiangsheng Hu Zeng Ye Guixiong Fan Xianjun Yu Wensheng Liu Xiaowu Xu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,8: | 2 |
