6A review of composition evolution in Ni-based single crystal superalloys显示文摘Due to the outstanding creep performance, nickel-based single crystal superalloys(Ni-SXs) are extensively applied in modern aero-engine and industrial gas turbine. Apart from the special single crystal structure which is disadvantageous to extension of creep cracks, Ni-SXs derive the creep strength from intrinsic two-phase microstructure(γ phase and γ’ phase). Main microstructural parameters including volume fraction of γ’ phase and the lattice misfit, and the formation and distribution of precipitated phase are determined by the compositions of alloys. Besides, the creep properties are greatly influenced by these microstructural parameters and precipitated phase. This review has summarized the relationships between different alloying elements and microstructures and indicated their influence on creep properties of Ni-SXs. In addition, with the improvements of experimental methods and characterization technique, some recent discoveries have provided additional evidence to support or challenge the pervious creep theories of superalloys. 7Carbon carry capacity and carbon sequestration potential in China based on an integrated analysis of mature forest biomass显示文摘Forests play an important role in acting as a carbon sink of terrestrial ecosystem.Although global forests have huge carbon carrying capacity(CCC)and carbon sequestration potential(CSP),there were few quantification reports on Chinese forests.We collected and compiled a forest biomass dataset of China,a total of 5841 sites,based on forest inventory and literature search results.From the dataset we extracted 338 sites with forests aged over 80 years,a threshold for defining mature forest,to establish the mature forest biomass dataset.After analyzing the spatial pattern of the carbon density of Chinese mature forests and its controlling factors,we used carbon density of mature forests as the reference level,and conservatively estimated the CCC of the forests in China by interpolation methods of Regression Kriging,Inverse Distance Weighted and Partial Thin Plate Smoothing Spline.Combining with the sixth National Forest Resources Inventory,we also estimated the forest CSP.The results revealed positive relationships between carbon density of mature forests and temperature,precipitation and stand age,and the horizontal and elevational patterns of carbon density of mature forests can be well predicted by temperature and precipitation.The total CCC and CSP of the existing forests are 19.87 and 13.86 Pg C,respectively.Subtropical forests would have more CCC and CSP than other biomes.Consequently,relying on forests to uptake carbon by decreasing disturbance on forests would be an alternative approach for mitigating greenhouse gas concentration effects besides afforestation and reforestation.LIU Ying Chun YU Gui Rui WANG Qiu Feng ZHANG Yang Jian XU Ze Hong 2014Science China(Life Sciences)2014,57,12:15
8Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 10Mechanochemical preparation of sulfur-doped nanosized TiO_2 and its photocatalytic activity under visible light显示文摘In order to make nanosized TiO2 photocatalyst responsive to visible light and effectively utilize solar energy, we have, for the first time, prepared S-doped nanosized TiO2 by a mechanochemical method with hydrolysis of TiCl4. The as-prepared S-doped nanosized TiO2 possesses strong ab-sorption for visible light of 400―650 nm and shows high photocatalytic activity for decomposition of methylene blue under irradiation of visible light. The oxidation states of the S atoms incorporated into TiO2 were determined to be S6+ and S4+. 13Monitoring disease progression and treatment efficacy with circulating tumor cells in esophageal squamous cell carcinoma: A case report显示文摘This study investigated whether changes in circulating tumor cell(CTC) numbers reflect tumor progression and treatment efficacy in esophageal squamous cell carcinoma(ESCC). A 47-year-old male patient with ESCC is presented in this case study. The patient was evaluated for a series of serum tumor markers and subjected to radiological examinations before and after surgery and during follow-up over the course of five years. In addition, the CTCs in 7.5 m L of peripheral blood were enriched by magneticactivated cell sorting negative selection and identified by immunofluorescence staining. Serum tumor markers remained within normal ranges and were discordant with imaging scans during the follow-up. Initially, one CTC was detected in the peripheral blood sample, and 14 were observed seven days after the operation. After 12 wk, subcutaneous metastases and bone metastases occurred, and the number of CTCs increased to 84. After 48 wk, lung metastases were noted, and the CTC level was 21. At 104 wk, the number of CTCs was 14, and disease recurrence was detected by positron emission tomography-computed tomography. The CTC counts were in accord with the imaging studies at several time points. 14A novel HfNbTaTiV high-entropy alloy of superior mechanical properties designed on the principle of maximum lattice distortion显示文摘This paper reports a synergistic design of high-performance BCC high-entropy alloy based on the combined consideration of the principles of intrinsic ductility of elements,maximum atomic size difference for solid solution strengthening and the valence electron concentration criterion for ductility.The single-phase BCC HfNbTaTiV alloy thus designed exhibited a high compressive yield strength of 1350 MPa and a high compressive ductility of>45%at the room temperature.This represents a 50%increase in yield strength relative to a HfNbTaTiZr alloy.This is attributed to the maximized solid solution strengthening effect caused by lattice distortion,which is estimated to be 1094 MPa.The alloy was also able to retain 53%of its yield strength and 77%of its ductility at 700℃.These properties are superior to those of most refractory BCC high-entropy alloys reported in the literature.Zibing An Shengcheng Mao Yinong Liu Li Wang Hao Zhou Bin Gan Ze Zhang Xiaodong Han 2021Journal of Materials Science & Technology2021,,20:9
15Cytological and Molecular Characteristics of Pollen Abortion in Lily with Dysplastic Tapetum显示文摘Lily was grown worldwide as a fresh cutting flower because of its colorful petals, but its anther contained a large number of pollen grains that cause serious pollen contamination, however, pollen abortion can effectively reduce the level of pollen pollution. Our analysis aims to use cytological observation to detect the critical stage when pollen abortion occurs and to provide comprehensive gene expression information at the transcriptional level. The result showed that pollen abortion in ‘Little Kiss’ began at the mononuclear stage and the callose that covers the microspores failed to degenerate when young pollens were released from the tetrads. In addition, compared with the normally developed one,the tapetum of ‘Little Kiss’ degraded in advance while the degradation of callose was delayed. Furthermore, 103 differentially expressed genes(DEGs) related to the advance degeneration of tapetum cells and callose were found in the expression levels, including 22 transcription factors(TFs). In particular, two β-glucanase genes(endo-1,3(4)-β-glucanase, exo-β-glucanase) responsible for callose degeneration were significantly down-regulated. These results suggested that pollen abortion may occur at mononuclear stage and that early degeneration of tapetum cells resulted in a significant down-regulation of β-glucanase genes. 16Apolipoprotein E, amyloid-beta, and neuroinflammation in Alzheimer's disease显示文摘Alzheimer's disease(AD) is characterized by the accumulation and deposition of amyloid-beta(Aβ) peptides in the brain. Neuroinflammation occurs in the AD brain and plays a critical role in the neurodegenerative pathology. Particularly, Aβ evokes an inflammatory response that leads to synaptic dysfunction, neuronal death, and neurodegeneration. Apolipoprotein E(ApoE) proteins are involved in cholesterol transport, Aβ binding and clearance, and synaptic functions in the brain. The ApoE4 isoform is a key risk factor for AD, while the ApoE2 isoform has a neuroprotective effect. However, studies have reached different conclusions about the roles of the isoforms; some show that both ApoE3 and ApoE4 have anti-infl ammatory effects, while others show that ApoE4 causes a predisposition to inflammation or promotes an inflammatory response following lipopolysaccharide treatment. These discrepancies may result from the differences in models, cell types, experimental conditions, and inflammatory stimuli used. Further, little was known about the role of ApoE isoforms in the Aβ-induced inflammatory response and in the neuroinflammation of AD. Our recent work showed that ApoE isoforms differentially regulate and modify the Aβ-induced infl ammatory response in neural cells, with ApoE2 suppressing and ApoE4 promoting the response. 17Identification of biomarkers for hepatocellular carcinoma by semiquantitative immunocytochemistry显示文摘AIM:To investigate the expression of key biomarkers in hepatoma cell lines,tumor cells from patients’blood samples,and tumor tissues.METHODS:We performed the biomarker tests in two steps.First,cells plated on coverslips were used to assess biomarkers,and fluorescence intensities were calculated using the NIH Image J software.The measured values were analyzed using the SPSS19.0 software to make comparisons among eight cell lines.Second,eighty-four individual samples were used to assess the biomarkers’expression.Negative enrichment of the blood samples was performed,and karyocytes were isolated and dropped onto pretreated glass slides for further analysis by immunofluorescence staining.Fluorescence intensities were compared among hepatocellular carcinoma(HCC)patients,chronic HBV-infected patients,and healthy controls following methods similar to those used for cell lines.The relationships between the expression of biomarkers and clinical pathological parameters were analyzed by Spearman rank correlation tests.In addition,we studied the distinct biomarkers’expression with three-dimensional laser confocal microscopy reconstructions,and Kaplan-Meier survival analysis was performed to understand the clinical significance of these biomarkers.RESULTS:Microscopic examination and fluorescence intensity calculations indicated that cytokeratin 8/18/19(CK)expression was significantly higher in six of the seven HCC cell lines examined than in the control cells,and the expression levels of asialoglycoprotein receptor(ASGPR)and glypican-3(GPC3)were higher in all seven HCC cell lines than in the control.Cells obtained from HCC patients’blood samples also displayed significantly higher expression levels of ASGPR,GPC3,and CK than cells from chronic HBV-infected patients or healthy controls;these proteins may be valuable surface biomarkers for identifying HCC circulating tumor cells isolated and enriched from the blood samples.The stem cell-like and epithelial-mesenchymal transition-related biomarkers could be detected on the karyocyte slides.ASGPR and GPC3 were expressed at high levels,and thus three-dimensional reconstructions were used to observe their expression in detail.This analysis indicated that GPC3 was localized in the cytoplasm and membrane,but that ASGPR had a polar localization.Survival analyses showed that expression of GPC3 and ASGPR is associated with a patient’s overall survival(OS).CONCLUSION:ASGPR,GPC3,and CK may be valuable HCC biomarkers for CTC detection;the expression of ASGPR and GPC3 might be helpful for understanding patients’OS.Hong Mu Kai-Xuan Lin Hong Zhao Shu Xing Cong Li Fang Liu Hai-Zhen Lu Ze Zhang Yu-Lin Sun Xi-Yun Yan Jian-Qiang Cai Xiao-Hang Zhao 2014World Journal of Gastroenterology2014,20,19:8
18Atomic process of oxidative etching in monolayer molybdenum disulfide显示文摘The microscopic process of oxidative etching of two-dimensional molybdenum disulfide(2D MoS_2) at an atomic scale is investigated using a correlative transmission electron microscope(TEM)-etching study.MoS_2 flakes on graphene TEM grids are precisely tracked and characterized by TEM before and after the oxidative etching. This allows us to determine the structural change with an atomic resolution on the edges of the domains, of well-oriented triangular pits and along the grain boundaries. We observe that the etching mostly starts from the open edges, grain boundaries and pre-existing atomic defects.A zigzag Mo edge is assigned as the dominant termination of the triangular pits, and profound terraces and grooves are observed on the etched edges. 19Three interpenetrated copper(Ⅱ) coordination polymers based on a V-shaped ligand:Synthesis,structures,sorption and magnetic properties显示文摘Three new CuⅡ coordination polymers,[Cu2(oba)2(H2O)(DMF)].3H2O(1),[Cu4(oba)4(DMF)4].4H2O.DMF(2) and [Cu2(oba)(hmp)2](3)(H2oba = 4,4'-oxybis(benzoic acid),Hhmp = 2-pyridine methanol,DMF = N,N'-dimethylformamide) were synthesized by solvothermal reactions and characterized by single-crystal X-ray diffraction.Both complexes 1 and 2 take three-dimensional(3D) three-fold interpenetrating PtS topology with 1D channels.In complex 3,Cu4 clusters were formed by the chelating hmp ligands,and then the clusters were further linked by oba ligands to form a 3D three-fold interpenetrating dia topology network.For 2,N2 sorption shows type I isotherm with BET surface areas of 562 m2 g-1 and Langmuir surface area of 747 m2 g-1,respectively.At 77 and 87 K,the hydrogen uptakes of complex 2 are 0.89 wt% and 0.57 wt%,respectively.Magnetic investigation showed the ferromagnetic coupling between CuII ions and very weak antiferromagnetic interaction between Cu4 clusters in complex 3.ZHANG XiaoFeng YANG Qian ZHAO JiongPeng HU Tong-Liang CHANG Ze BU Xian-He 2011Science China Chemistry2011,54,9:7
20The influence of freeze-thaw cycles on the granulometric composition of Moscow morainic clay显示文摘The freeze-thaw cycling process considerably changes the composition,structure,and properties of soils.Since the grain size is the most important factor in determining soil characteristics,our current research primarily aims to investigate dynamic changes of the soil fraction when exposed to freeze-thaw conditions.We observed two series of Moscow morainic clayey specimens(g QmII):(I) the original series,and(II) the remolded series.We subjected each series of soil specimens to different frequencies of freeze-thaw cycles(3,6,20,and 40 cycles),and we used granulometric tests to analyze both series before and after exposure to freeze-thaw conditions.As a result of our experiments,the granulometric compositions tended to be distributed evenly after 40 freeze-thaw processes(i.e.,content of fraction for 0.1–0.05 mm was increased after 40 freeze-thaw cycles) because the division of coarse grains and the aggregation of fine grains were synchronized during the freeze-thaw process.The soil grains in both series changed bi-directionally.In the original series,changes of the sand grains were conjugated with the clay grains,and in the remolded series,changes of the sand grains were conjugated with the silt grains,because potential energy difference caused the division and aggregation processes to relate to the counteraction process.The even distribution of soil grain size indicated the state of equilibrium or balance.The granulometric compositions were altered the most during the sixth freeze-thaw cycle,because the coefficient of the intensity variation of the grain fineness(Kvar) had its maximum value at that time.Ze Zhang Vadim V.Pendin WenJie Feng ZhongQiong Zhang 2015Research in Cold and Arid Regions2015,7,3:7
