| 1 | microRNA-218 suppresses the proliferation, invasion and promotes apoptosis of pancreatic cancer cells by targeting HMGB1显示文摘Objective: To detect the expression profiles of micro RNA-218(mi R-218) in human pancreatic cancer tissue(PCT) and cells and their effects on the biological features of human pancreatic cancer cell line PANC-1 and observe the effect of mi R-218 on the expression of the target gene high mobility group box 1(HMGB1), with an attempt to provide new treatment methods and strategies for pancreatic cancer.Methods: The expressions of mi R-218 in PCT and normal pancreas tissue as well as in various pancreatic cancer cell lines including As PC-1, Bx PC-3, and PANC-1 were determined with quantitative real-time reverse transcription polymerase chain reaction(q RT-PCR). The change of mi R-218 expression in PANC-1 cells was detected using qR T-PCT after the transfection of miR-218 mimic for 48 h. Cell Counting Kit-8(CCK-8) was applied for detecting the effect of mi R-218 on the activity of PANC-1 cells. The effects of mi R-218 on the proliferation and apoptosis of PANC-1 cells were analyzed using the flow cytometry. The effect of mi R-218 on the migration of PANC-1 cells was detected using the Trans-well migration assay. The HMGB1 was found to be a target gene of mi R-218 by luciferase reporter assay, and the effect of mi R-218 on the expression of HMGB1 protein in cells were determined using Western blotting.Results: As shown by q RT-PCR, the expressions of mi R-218 in PCT and in pancreatic cancer cell line significantly decreased when compared with the normal pancreatic tissue(NPT)(P<0.01). Compared with the control group, the miR-218 expression significantly increased in the PANC-1 group after the transfection of mi R-218 mimic for 48 h(P<0.01). Growth curve showed that the cell viability significantly dropped after the overexpression of mi R-218 in the PANC-1 cells for two days(P<0.05). Flow cytometry showed that the S-phase fraction significantly dropped after the overexpression of mi R-218(P<0.01) and the percentage of apoptotic cells significantly increased(P<0.01). As shown by the Trans-well migration assay, the enhanced mi R-218 expression was associated with a significantly lower number of cells that passed through a Transwell chamber(P<0.01). Luciferase reporter assay showed that, compared with the control group, the relative luciferase activity significantly decreased in the mi R-218 mimic group(P<0.01). As shown by the Western blotting, compared with the control group, the HMGB1 protein expression significantly decreased in the PANC-1 group after the transfection of mi R-218 mimic for 48 h(P<0.01).Conclusions: The mi R-218 expression decreases in human PCT and cell lines. mi R-218 can negatively regulate the HMGB1 protein expression and inhibit the proliferation and invasion of pancreatic cancer cells. | 2 | Thick Miocene eolian deposits on the Huajialing Mountains:The geomorphic evolution of the western Loess Plateau显示文摘The geomorphic evolution of northwestern China during the Cenozoic has been a subject of much geological interest because of its link with the uplift of the Himalayan-Tibetan complex.Much information about these changes is recoverable from the sedimentary sequences of the region.We report here on the thick eolian deposits mantling the Huajialing Mountains,a relatively flat mountain range in the western Loess Plateau.Correlation of magnetic susceptibility stratigraphy with the QA-I Miocene eolian sequence dates a 134.7 m section(NL-VII) for the interval from 18.7 to 11.8 Ma,as confirmed by micro-mammalian fossils.These eolian deposits demonstrate a much wider distribution of the Miocene eolian deposits,and also indicate that the topography contrasts in the western Loess Plateau,including the uplifts of the Huajialing Mountains and the bedrock highlands in the Qinan region,were formed by the early Miocene.The near-continuous Miocene eolian sequence from 18.7 to 11.8 Ma indicates that the substratum of Huajialing had not experienced any intense tectonic changes during this time interval,which suggests further,the relative tectonic stability of the nearby Tibetan Plateau. | ZHAN Tao GUO ZhengTang WU HaiBin GE JunYi ZHOU Xin WU ChunLin ZENG FangMing | 2011 | Science China Earth Sciences2011,54,2: | 7 |
| 3 | microRNA-218 promotes gemcitabine sensitivity in human pancreatic cancer cells by regulating HMGB1 expression显示文摘Objective: The purpose of this study was to examine the effect of gemcitabine(GEM) on micro RNA-218(mi R-218) expression in human pancreatic cancer cells.Methods: Quantitative reverse transcription polymerase chain reaction(q RT-PCR) was performed to examine the differences in mi R-218 expression between the GEM-sensitive Bx PC-3 pancreatic cancer cells and GEM-resistant PANC-1 cells. The effect of GEM on the expression of mi R-218 in PANC-1 cells was also investigated. PANC-1 cells were transfected either with HMGB1 si RNA to knock down the expression of HMGB1 or with the recombinant HMGB1 expression vector(pc DNA3.1-HMGB1) to overexpress HMGB1. The effect of ectopic expression of HMGB1 on the apoptosis of mi R-218-transfected and GEMtreated PANC-1 cells was examined by flow cytometric analysis.Results: The mi R-218 expression level was lower in GEM-resistant PANC-1 cells compared to GEMsensitive Bx PC-3 cells(P<0.05). The percentage of apoptotic PANC-1 cells was significantly increased in the mi R-218 mimic + GEM group compared to the mimic ctrl + GEM group and the normal control group(P<0.01). The HMGB1 expression level was markedly decreased in PANC-1 cells transfected with HMGB1 si RNA but was significantly increased in PANC-1 cells transfected with the recombinant HMGB1 expression vector, pc DNA3.1-HMGB1(P<0.01). | 11 | A Serum Biomarker Model to Diagnose Pancreatic Cancer Using Proteomic Fingerprint Technology显示文摘OBJECTIVE To establish a serum protein pattern model for screening pancreatic cancer. METHODS Twenty-nine serum samples from patients with pancreatic cancer were collected before surgery,and an additional 57 serum samples from age and sex-matched individuals without cancer were used as controls.WCX magnetic beans and a PBS Ⅱ-C protein chip reader (Ciphergen Biosystems Inc) were employed to detect the protein fingerprint expression of all serum samples. The resulting profiles comparing serum from cancer and normal patients were analyzed with the Biomarker Wizard system,to establish a model using the Biomarker Pattern system software. A double-blind test was used to determine the sensitivity and specificity of the model. RESULTS A group of 4 biomarkers(relative molecular weights were 5,705 Da,4,935 Da,5,318 Da,3,243 Da) were selected to set up a decision tree to produce the classification model to effectively screen pancreatic cancer patients.The results yielded a sensitivity of 100%(20/20),specificity of 97.4% (37/38).The ROC curve was 99.7%.A double-blind test used to challenge the model resulted in a sensitivity of 88.9% and a specificity of 89.5%. | 16 | Lipopolysaccharide-activated microglial-induced neuroglial cell differentiation in bone marrow mesenchymal stem cells显示文摘BACKGROUND: Microglia are very sensitive to environmental changes, often becoming activated by pathological conditions. Activated microglia can exert a dual role in injury and repair in various diseases of the central nervous system, including cerebral ischemia, Parkinson’s disease, and Alzheimer’s disease. OBJECTIVE: An immortal microglial cell line, BV2, was treated with varying concentrations of lipopolysaccharide (LPS) to induce a pathological situation. Supernatant was harvested and incubated with bone marrow mesenchymal stem cells and, concomitantly, bone marrow mesenchymal stem cell differentiation was observed. DESIGN: A controlled observation, in vitro experiment. SETTING: Department of Neurology, First Affiliated Hospital of China Medical University. MATERIALS: Five male 2–3-week-old Sprague Dawley rats were purchased from Animal Laboratory Center of China Medical University and included in this study. The protocol was performed in accordance with ethical guidelines for the use and care of animals. The microglial cell line BV2 was produced by Cell Research Institute of Chinese Academy of Sciences. LPS was produced by Sigma Company, USA. METHODS: This study was performed in the Central Laboratory of China Medical University from September 2006 to March 2007. Rat femoral and tibial bone marrow was collected for separation and primary culture of bone marrow mesenchymal stem cells. Bone marrow mesenchymal stem cell cultures were divided into 5 groups: control group, non-activated group, as well as low-, medium-, and high-dose LPS groups. In the control group, bone marrow mesenchymal stem cells were cultured with Dulbecco’s modified Eagle’s medium (DMEM) supplemented with fetal bovine serum (volume fraction 0.1). In the non-activated group, bone marrow mesenchymal stem cells were incubated with non-activated BV2 supernatant. In the low-, medium-, and high-dose LPS groups, bone marrow mesenchymal stem cells were incubated with LPS (0.01, 0.1 and 1 μg/L, respectively)-activated BV2 supernatant. MAIN OUTCOME MEASURES: Expression of glial fibrillary acidic protein (GFAP) and neuron-specific enolase (NSE) in bone marrow mesenchymal stem cells was detected by immunofluorescence staining. RESULTS: GFAP-positive cells were detected in each group; however, the greatest number were found in the high-dose LPS group. The number of GFAP-positive cells was significantly greater in the high- and medium-dose groups, compared to the control, non-activated, and low-dose LPS groups (P < 0.05). However, there was no significant difference between the medium- and high-dose LPS groups (P > 0.05). NSE-positive cells were also detected in each group. However, there was no significant difference between any two groups (P > 0.05). CONCLUSION: Microglia, when activated to some ertent, could induce neuroglial cell differentiation from bone marrow mesenchymal stem cells; however, they did not exhibit the capacity to markedly promote neuronal cell differentiation.
