Multimorbidity patterns and association with mortality in 0.5 million Chinese adults显示文摘Background:Few studies have assessed the relationship between multimorbidity patterns and mortality risk in the Chinese population. We aimed to identify multimorbidity patterns and examined the associations of multimorbidity patterns and the number of chronic diseases with the risk of mortality among Chinese middle-aged and older adults. Methods:We used data from the China Kadoorie Biobank and included 512,723 participants aged 30 to 79 years.Multimorbidity was defined as the presence of two or more of the 15 chronic diseases collected by self-report or physical examination at baseline.Multimorbidity patterns were identified using hierarchical cluster analysis.Cox regression was used to estimate the associations of multimorbidity patterns and the number of chronic diseases with all-cause and cause-specific mortality.Results:Overall,15.8%of participants had multimorbidity. The prevalence of multimorbidity increased with age and was higher in urban than rural participants.Four multimorbidity patterns were identified, including cardiometabolic multimorbidity(diabetes, coronary heart disease, stroke, and hypertension),respiratory multimorbidity(tuberculosis, asthma, and chronic obstructive pulmonary disease),gastrointestinal and hepatorenal multimorbidity(gallstone disease, chronic kidney disease, cirrhosis, peptic ulcer, and cancer),and mental and arthritis multimorbidity(neurasthenia, psychiatric disorder, and rheumatoid arthritis).During a median of 10.8 years of follow-up,49,371 deaths occurred.Compared with participants without multimorbidity, cardiometabolic multimorbidity(hazard ratios[HR]=2.20,95%confidence intervals[CI]:2.14-2.26)and respiratory multimorbidity(HR=2.13,95%CI:1.97-2.31)demonstrated relatively higher risks of mortality, followed by gastrointestinal and hepatorenal multimorbidity(HR=1.33,95%CI:1.22-1.46).The mortality risk increased by 36%(HR=1.36,95%CI:1.35-1.37)with every additional disease. Conclusion:Cardiometabolic multimorbidity and respiratory multimorbidity posed the highest threat on mortality risk and deserved particular attention in Chinese adults. | Junning Fan Zhijia Sun Canqing Yu Yu Guo Pei Pei Ling Yang Yiping Chen Huaidong Du Dianjianyi Sun Yuanjie Pang Jun Zhang Simon Gilbert Daniel Avery Junshi Chen Zhengming Chen Jun Lyu Liming Li On Behalf of the China Kadoorie Biobank Collaborative Group | 2022 | Chinese Medical Journal2022,,6: | 17

Phylogeny and spatio-temporal diversification of Prunus subgenus Laurocerasus section Mesopygeum (Rosaceae) in the Malesian region显示文摘The goals of this study were to reconstruct the phylogeny of Prunus subgenus Laurocerasus section Mesopygeum and to provide a preliminary assessment of its spatio-temporal diversification in the Malesian region. We inferred the phylogeny using nuclear ITS and ETS and plastid psbA-trnH,rps16,rph6,and trnC-petN sequences. Our analyses support the monophyly of sect.Mesopygeum.Within sect.Mesopygeum,we identified four main subclades: (i)Prunus lancilimba from continental Asia;(ii)Prunus ruthii from Malay Peninsula;(iii)a subclade comprising species from areas centered on the Sunda shelf and also a few species from continental Asia and Wallacea;and (iv)a subclade composed of species from areas of the Sahul shelf, with a small number of taxa also from areas of the Sunda shelf, continental Asia, and the Philippines. We estimated that sect.Mesopygeum originated in continental Asia at c.44.71 Mya (95%HPD-31.66-46.90).Nine dispersals between maior geographic areas were inferred. From continental Asia, three and two dispersals were inferred to the 5unda shelf and the Sahul shelf in the mid-Oligocene,respectively.Two dispersals were inferred from the Sahul shelf region to the Sunda shelf in late Oligecene and early Miocene, respectively. There were also two dispersals inferred from the Sunda shelf region, one to the Philippines and one to Wallacea,in the middle and late Miocene, respectively. The diversification in sect.Mesopygeum was likely driven by active geologic events and orogenies in the Neogene in the Malesian region. | Liang Zhao Daniel Potter Yuan Xu Pei-Liang Liu Gabriel Johnson Zhao-Yang Chang Jun Wen | 2018 | Journal of Systematics and Evolution2018,56,6: | 5
Tunable nonlinear refractive index of two-dimensional MoS2, WS2, and MoSe2 nanosheet dispersions [Invited]显示文摘Liquid-phase-exfoliation technology was utilized to prepare layered MoS2, WS2, and MoSe2 nanosheets in cyclohexylpyrrolidone. The nonlinear optical response of these nanosheets in dispersions was investigated by observing spatial self-phase modulation(SSPM) using a 488 nm continuous wave laser beam. The diffraction ring patterns of SSPM were found to be distorted along the vertical direction right after the laser traversing the nanosheet dispersions. The nonlinear refractive index of the three transition metal dichalcogenides dispersions n2 was measured to be 10-7cm2W-1, and the third-order nonlinear susceptibility χ(3)10-9 esu. The relative change of effective nonlinear refractive index Δn2e∕n(2e) of the MoS2, WS2, and MoSe2 dispersions can be modulated 0.012–0.240, 0.029–0.154, and 0.091–0.304, respectively, by changing the incident intensities. Our experimental results imply novel potential application of two-dimensional transition metal dichalcogenides in nonlinear phase modulation devices. | Gaozhong Wang Saifeng Zhang Xiaoyan Zhang Long Zhang Ya Cheng Daniel Fox Hongzhou Zhang Jonathan N.Coleman Werner J.Blau Jun Wang | 2015 | Photonics Research2015,3,2: | 4
Transitional geology and its effects on development and longwall mining in Pittsburgh Seam显示文摘This paper presents the geologic and ground control challenges that were encountered by Consol Energy's coal mining operations in southwestern Pennsylvania,USA. Geologic encounters, such as sandstoneto-limestone geology transition, massive sandstone channels, shale channels, pyritic rich green claystone, laminated roof, and soft floor, have significantly impacted the development and longwall mining in Consol's Pittsburgh Seam coal mines. Experience from different mines shows that,in the sandstone-to-limestone geology transition zone,1.83 m high-tension,fully-grouted primary bolts employed along with 4.88 m center cable bolts at every other strap greatly improved beam building and ensured proper anchorage into the competent roof. Hydraulic fracturing of the massive sandstone was often necessary to enhance caving of the massive sandstone behind the shields to relieve pressure at the face. The presence of soft floor coupled with presence of thick floor coal and deep cover, induced excessive headgate convergence during retreat of the first right hand longwall panel. In all,it is important to explore the roof and in-seam geology in detail to delineate normal and anomalous geologic conditions prior to and during development. With diligent geologic reconnaissance, geotechnical monitoring, and assessment,site-specific geotechnical solutions have been provided to mine operations to improve safety and productivity. | Lu Jun Van Dyke Mark Su Daniel W.H. | 7 | Seed-mediated synthesis of dendritic platinum nanostructures with high catalytic activity for aqueous-phase hydrogenation of acetophenone显示文摘This work reports a facile and efficient seed-mediated method for the synthesis of dendritic platinum(Pt)nanoparticles(NPs) at low temperatures of 55–60 °C in water, using L-ascorbic acid as a reducing agent and sodium citrate as a capping agent. It is found that the dendritic Pt NPs(10–150 nm) are composed of tiny Pt nanocrystals, which nucleate and grow through the introduced smaller Pt seeds with diameters of 3–5 nm.Further investigation shows that the dendritic Pt nanostructures display excellent catalytic performance in an aqueous-phase aromatic ketone hydrogenation reaction, including:(i) acetophenone conversion rate of> 90%, with smaller dendritic Pt NPs(10–46 nm) offering a higher conversion efficiency;(ii) high chemoselectivity toward carbonyl group(90.6%–91.5%), e.g., the selectivity to 1-phenylethanol is ~90.1% with nearly100% acetophenone conversion for 10 nm dendritic Pt NPs within 60 min, under mild reaction conditions(20 °C, 1.5 bar H2 pressure, and 1.5 mol% catalyst). The high catalytic activity, selectivity and stability of the dendritic Pt nanostructures under the organic solvent-free conditions make them promising for many potential applications in green catalytic conversion of hydrophilic biomass derived compounds. | Yuan Yuan Yusuf Valentino Kaneti Minsu Liu Fangzhu Jin Danielle Frances Kennedy Xuchuan Jiang Jun Huang Aibing Yu | 2015 | Journal of Energy Chemistry2015,24,5: | 3
Inhibition of CDK9 by voruciclib synergistically enhances cell death induced by the Bcl-2 selective inhibitor venetoclax in preclinical models of acute myeloid leukemia显示文摘Venetoclax,an FDA-approved Bcl-2 selective inhibitor for the treatment of chronic lymphocytic leukemia and acute myeloid leukemia(AML),is tolerated well in elderly patients with AML and has good overall response rates;however, resistance remains a concern. In this study,we show that targeting CDK9 with voruciclib in combination with venetoclax results in synergistic antileukemic activity against AML cell lines and primary patient samples.CDK9 inhibition enhances venetoclax activity through downregulation of Mcl-1 and c-Myc.However,downregulation of Mcl-1 is transient, which necessitates an intermittent treatment schedule to allow for repeated downregulation of Mcl-1.Accordingly,an every other day schedule of the CDK9 inhibitor is effective in vitro and in vivo in enhancing the efficacy of venetoclax. Our preclinical data provide a rationale for an intermittent drug administration schedule for the clinical evaluation of the combination treatment for AML. | Daniel A.Luedtke Yongwei Su Jun Ma Xinyu Li Steven ABuck Holly Edwards Lisa Polin Juiwanna Kushner Sijana HDzinic Kathryn White Hai Lin Jeffrey WTaub Yubin Ge | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 3
Preventing abnormal NF-κB activation and autoimmunity by Otub1-mediated p100 stabilization显示文摘NF-κB, a family of transcription factors regulating diverse biological processes including immune responses, is activated by canonical and noncanonical pathways based on degradation of IκBα and processing of the IκB-like protein p100, respectively. Although p100 responds to noncanonical NF-κB stimuli for processing, it does not undergo degradation, but rather becomes accumulated, along with canonical NF-κB activation. We show here that the stability of p100 is tightly controlled by a deubiquitinase, Otub1. Otub1 deficiency not only promotes signal-induced p100 processing and noncanonical NF-κB activation but also causes steady-state p100 degradation, leading to aberrant NF-κB activation in the canonical pathway. B-cell-conditional deletion of Otub1 results in B-cell hyperplasia, antibody hyper-production, and lupus-like autoimmunity. Otub1-deficient B cells display aberrantly activated phenotypes and overproduce the cytokine IL-6, contributing to autoimmunity induction. Thus, maintenance of p100 stability by Otub1 serves as an unusual mechanism of NF-κB regulation that prevents autoimmunity. | Yanchuan Li Jin-Young Yang Xiaoping Xie Zuliang Jie Lingyun Zhang Jianhong Shi Daniel Lin Meidi Gu Xiaofei Zhou Haiyan S. | 10 | Antibody response and therapy in COVID-19 patients: what can be learned for vaccine development?显示文摘The newly emerged severe acute respiratory syndrome coronavirus 2(SARS-CoV-2) has infected millions of people and caused tremendous morbidity and mortality worldwide. Effective treatment for coronavirus disease 2019(COVID-19) due to SARSCoV-2 infection is lacking, and different therapeutic strategies are under testing. Host humoral and cellular immunity to SARSCoV-2 infection is a critical determinant for patients’ outcomes. SARS-CoV-2 infection results in seroconversion and production of anti-SARS-CoV-2 antibodies. The antibodies may suppress viral replication through neutralization but might also participate in COVID-19 pathogenesis through a process termed antibody-dependent enhancement. Rapid progress has been made in the research of antibody response and therapy in COVID-19 patients, including characterization of the clinical features of antibody responses in different populations infected by SARS-CoV-2, treatment of COVID-19 patients with convalescent plasma and intravenous immunoglobin products, isolation and characterization of a large panel of monoclonal neutralizing antibodies and early clinical testing, as well as clinical results from several COVID-19 vaccine candidates. In this review, we summarize the recent progress and discuss the implications of these findings in vaccine development. | Ligong Lu Hui Zhang Meixiao Zhan Jun Jiang Hua Yin Danielle JDauphars Shi-You Li Yong Li You-Wen He | 2020 | Science China(Life Sciences)2020,63,12: | 3

Acoustofluidic separation enables early diagnosis of traumatic brain injury based on circulating exosomes显示文摘Traumatic brain injury(TBI)is a global cause of morbidity and mortality.Initial management and risk stratification of patients with TBI is made difficult by the relative insensitivity of screening radiographic studies as well as by the absence of a widely available, noninvasive diagnostic biomarker. In particular,a blood-based biomarker assay could provide a quick and minimally invasive process to stratify risk and guide early management strategies in patients with mild TBI(mTBI).Analysis of circulating exosomes allows the potential for rapid and specific identification of tissue injury.By applying acoustofluidic exosome separation—which uses a combination of microfluidics and acoustics to separate bioparticles based on differences in size and acoustic properties—we successfully isolated exosomes from plasma samples obtained from mice after TBI.Acoustofluidic isolation eliminated interference from other blood components, making it possible to detect exosomal biomarkers for TBI via flow cytometry.Flow cytometry analysis indicated that exosomal biomarkers for TBI increase in the first 24 h following head trauma, indicating the potential of using circulating exosomes for the rapid diagnosis of TBI.Elevated levels of TBI biomarkers were only detected in the samples separated via acoustofluidics;no changes were observed in the analysis of the raw plasma sample. This finding demonstrated the necessity of sample purification prior to exosomal biomarker analysis.Since acoustofluidic exosome separation can easily be integrated with downstream analysis methods,it shows great potential for improving early diagnosis and treatment decisions associated with TBI. | Zeyu Wang Haichen Wang Ryan Becker Joseph Rufo Shujie Yang Brian E.Mace Mengxi Wu Jun Zou Daniel T.Laskowitz Tony Jun Huang | 2021 | Microsystems & Nanoengineering2021,7,2: | 3
Liver resection versus liver transplantation for hepatocellular carcinoma within Milan criteria:a meta-analysis of 18,421 patients显示文摘Background:Outcomes after liver resection(LR)and liver transplantation(LT)for hepatocellular carcinoma(HCC)are heterogenous and may vary by region, over time periods and disease burden. We aimed to compare overall survival(OS)and disease-free survival(DFS)between LT versus LR for HCC within the Milan criteria. Methods:Two authors independently searched Medline and Embase databases for studies comparing survival after LT and LR for patients with HCC meeting the Milan criteria.Meta-analyses and meta-regression were conducted using random-effects models. Results:We screened 2,278 studies and included 35 studies with 18,421 patients.LR was associated with poorer OS[hazard ratio(HR)=1.44;95%confidence interval(CI):1.14-1.81;P<0.01]and DFS(HR=2.71;95%CI:2.23-3.28;P<0.01)compared to LT,with similar findings among intention-to-treat(ITT)studies.In uninodular disease,OS in LR was comparable to LT(P=0.13)but DFS remained poorer(HR=2.95;95%CI:2.30-3.79;P<0.01).By region,LR had poorer OS versus LT in North America and Europe(P≤0.01),but not Asia(P=0.25).LR had inferior survival versus LT in studies completed before 2010(P=0.01),but not after 2010(P=0.12).Cohorts that underwent enhanced surveillance had comparable OS after LT and LR(P=0.33),but cohorts undergoing usual surveillance had worse OS after LR(HR=1.95;95%CI:1.24-3.07;P<0.01).Conclusions:Mortality after LR for HCC is nearly 50%higher compared to LT.Survival between LR and LT were similar in uninodular disease. The risk of recurrence after LR is threefold that of LT. | Jin Hean Koh Darren Jun Hao Tan Yuki Ong Wen Hui Lim Cheng Han Ng Phoebe Wen Lin Tay Jie Ning Yong Mark D.Muthiah Eunice X.Tan Ning Qi Pang Beom Kyung Kim Nicholas Syn Alfred Kow Brian K.P.Goh Daniel Q.Huang | 2022 | Hepatobiliary Surgery and Nutrition2022,11,1: | 3
