2Preparation of ChlL-2 and IBDV VP2 Fusion Protein by Baculovirus Expression System显示文摘This study aims to produce an effective subunit vaccine against infectious bursal disease virus (IBDV). The genes of chicken interleukin-2 (ChIL-2) and IBDV viral protein 2 (VP2) were amplified and fused by splice overlap extension-polymerase chain reaction (SOE-PCR). The fusion gene was digested by EcoR I/Kpn I and inserted into pBacPAK8 vector, resulting in recombinant transfer plasmid pBacPakVP2-IL2. The recombinant plasmid was transfected into Sf-9 cells accompanied with hybrid nuclear polyhedrosis virus (HyNPV) genome DNA and lipofectin. Plaque-purification indicated that we had got the recombinant Hy-VP2-IL2. Fusion protein VP2-IL2was expressed effectively both in insect cells and bombyx mori. The expression of fusion protein was confirmed by ELISA, SDS-PAGE and Western blotting assay, respectively. 3The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 4The High Energy X-ray telescope (HE) onboard the Insight-HXMT astronomy satellite显示文摘The Insight-Hard X-ray Modulation Telescope(Insight-HXMT) is a broadband X-ray and γ-ray(1-3000 ke V) astronomy satellite. One of its three main telescopes is the High Energy X-ray telescope(HE). The main detector plane of HE comprises 18 Na I(Tl)/Cs I(Na) phoswich detectors, where Na I(Tl) is used as the primary detector to measure ~ 20-250 ke V photons incident from the field of view(FOV) defined by collimators, and Cs I(Na) is used as the active shielding detector to Na I(Tl) by pulse shape discrimination. Additionally, Cs I(Na) is used as an omnidirectional γ-ray monitor. The HE collimators have a diverse FOV,i.e. 1.1°×5.7°(15 units), 5.7°×5.7°(2 units), and blocked(1 unit). Therefore, the combined FOV of HE is approximately5.7°×5.7°. Each HE detector has a diameter of 190 mm resulting in a total geometrical area of approximately 5100 cm2, and the energy resolution is ~15% at 60 ke V. For each recorded X-ray event by HE, the timing accuracy is less than 10 μs and the deadtime is less than 10 μs. HE is used for observing spectra and temporal variability of X-ray sources in the 20-250 ke V band either by pointing observations for known sources or scanning observations to unveil new sources. Additionally, HE is used for monitoring the γ-ray burst in 0.2-3 Me V band. 6Follistatin-like protein 1 plays a tumor suppressor role in clear-cell renal cell carcinoma显示文摘Background:We previously showed that the expression of follistatin-like protein 1(FSTL1)was significantly down-regulated in metastatic clear-cell renal cell carcinoma(ccRCC).In this study,we aimed to characterize the role of FSTL1 in the development of ccRCC.Methods:The effects of FSTL1 on cell activity and cell cycle were investigated in ccRCC cell lines with altered FSTL1 expression.Gene expression microarray assays were performed to identify the major signaling pathways affected by FSTL1 knockdown.The expression of FSTL1 in ccRCC and its effect on postoperative prognosis were estimated in a cohort with 89 patients.Results:FSTL1 knockdown promoted anchorage-independent growth,migration,invasion,and cell cycle of ccRCC cell lines,whereas FSTL1 overexpression attenuated cell migration.FSTL1 knockdown up-regulated nuclear factor-κB(NF-κB)and hypoxia-inducible factor(HIF)signaling pathways,increased epithelial-to-mesenchymal transition,up-regulated interleukin-6 expression,and promoted tumor necrosis factor-α-induced degradation of NF-κB inhibitor(IκBα)in ccRCC cell lines.FSTL1 immunostaining was selectively positive in epithelial cytoplasm in the loop of Henle,and positive rate of FSTL1 was significantly lower in ccRCC tissues than in adjacent renal tissues(P<0.001).The mul-tivariate Cox regression analysis showed that the intratumoral FSTL1 expression conferred a favorable independent prognosis with a hazard ratio of 0.325(95%confidence interval 0.118-0.894).HIF-2αexpression was negatively cor-related with FSTL1 expression in ccRCC specimens(r=−0.229,P=0.044).Intratumoral expression of HIF-2α,rather than HIF-1α,significantly predicted an unfavorable prognosis in ccRCC(log-rank,P=0.038).Conclusions:FSTL1 plays a tumor suppression role possibly via repressing the NF-κB and HIF-2αsignaling pathways.To increase FSTL1 expression might be a candidate therapeutic strategy for metastatic ccRCC.Yan Liu Xiaojie Tan Wenbin Liu Xi Chen Xiaomei Hou Dan Shen Yibo Ding Jianhua Yin Ling Wang Hongwei Zhang Yongwei Yu Jianguo Hou Timothy C.Thompson Guangwen Cao 2018Cancer Communications2018,38,1:5

12Generation of VP5 deficient mutant of infectious bursal disease virus strain HZ2显示文摘Infectious bursal disease virus (IBDV) is a bi-segmented, dsRNA virus of the Birnaviridae family. The nonstructural protein VP5 has been re- ported to be associated with virus-induced cell apoptosis and pathogenicity, but its role in viral rep- lication has not been unequivocally identified. Based on a PCR introduced mutagenesis strategy, the 33 bp of 96―129 bp located between ORF A1and ORF A2 of genomic segment A of IBDV strain HZ2 were de- leted, and an Nhe I (GcTaGc) site was inserted at 96―102 bp simultaneously. The mutated segment A was ligated into pCI, resulting in pCI-ANhe3. A chi- meric and deficient IBDV strain, named strain ANhe3, was recovered from chicken embryo fibroblast (CEF) cells by co-transfection with pCI-ANhe3 and pCI-mB, derived from the genomic segment B strain HZ2. The indirect fluorescent assay identified that strain ANhe3 could replicate on CEF cells without expression of VP5. Further examination showed that the patho- genesis of strain ANhe3 replicating on SPF chicken embryos was attenuated compared to strain HZ2. This paper provides a new rapid rescue strategy for gene-deleted virus. 16NATURAL HISTORY OBSERVATION FOR ESOPHAGEAL AND CARDIA PRECURSORS BY REPETITIVE ENDOSCOPIC SCREENING OF 301 SUBJECTS IN SHEXIAN显示文摘OBJECTIVE To investigate the natural history of fast developing esopha- geal and cardia precursors. METHODS Repetitive endoscopic screenings were performed among 40-69-year-olds in the high-incidence areas for esophageal cancer in Shexian. RESULTS The initial diagnosis and the lag-time for 7 subsequently identified severe dysplasia(SD)subjects were as follows:in one subject 13 months after a baseline diagnosis of normal epithelium,in another subject 7 months after a baseline diagnosis of base cell hyperplasia(BCH),in four subjects 3,4,4,and 10.5 months after baseline diagnosis of mild dysplasia (mD),and in one subject 12.5 months after a baseline diagnosis of moder- ate dysplasia(MD).The initial diagnosis and the lag-time for 6 subsequently identified carcinomas in situ or intramucosal carcinoma cases were:in one case 48 months after a baseline diagnosis of mD,in 2 cases 4 and 13 months after baseline diagnoses of MD,and in the other 3 cases 3.5,9,and 17.5 months after baseline diagnoses of SD.The initial diagnosis and lag- time for 3 subsequently identified invasive cancer cases were:in one case 50 months after a baseline diagnosis of MD,in 2 cases 14 and 19 months after baseline diagnoses of SD.In addition,during a 4-year-fol ow-up of 18 subjects after endoscopic mucosa resection,9 of them were found to have developed precursors again at other sites,and also additional findings were obtained for 11 of the 16 dysplasia cases by repetitive biopsy in less than 2 months after the initial endoscopy. 17GmSAP5,a soybean A20/AN1 domain-containing stress-associated protein gene activated by GmAREB3,increases drought stress resistance in soybean by mediating ABA signaling显示文摘Drought stress impairs crop growth and productivity.Stress-associated proteins(SAPs),a class of zinc finger proteins containing the A20/AN1 domain,function in various stress responses in plants.However,little is known about the function of SAPs in drought-stress responses in soybean,an oil and protein crop.We report that a GmSAP5 protein confers drought tolerance by increasing sensitivity to abscisic acid(ABA)and reducing stomatal aperture.Overexpression and RNA interference of GmSAP5in soybean hairy roots resulted in elevated resistance and sensitivity to drought stress,respectively.ABA and proline contents increased in GmSAP5-overexpressing plants under water-deficit conditions.Lower water loss rates and higher relative water contents were observed in GmSAP5-overexpressing plants,resulting in increased drought-stress resistance.A yeast one-hybrid assay and luciferase transient transcriptional activity assay showed that GmAREB3,an AREB/ABF transcription factor,could bind to the promoter of GmSAP5 and activate its expression.These results suggest that GmSAP5 acts downstream of GmAREB3 and improves drought-stress resistance by mediating ABA signaling.Zehao Hou Xiangzhan Zhang Yimiao Tang Taifei Yu Lei Zheng Jun Chen Yongbin Zhou Yongwei Liu Ming Chen Zhao-Shi Xu Youzhi Ma 2022The Crop Journal2022,10,6:0
18Simulation of Two-Phase Flowback Phenomena in Shale Gas Wells显示文摘The gas-water two-phaseflow occurring as a result of fracturingfluidflowback phenomena is known to impact significantly the productivity of shale gas well.In this work,this two-phaseflow has been simulated in the framework of a hybrid approach partially relying on the embedded discrete fracture model(EDFM).This model assumes the region outside the stimulated reservoir volume(SRV)as a single-medium while the SRV region itself is described using a double-medium strategy which can account for thefluid exchange between the matrix and the micro-fractures.The shale gas adsorption,desorption,diffusion,gas slippage effect,fracture stress sensitivity,and capillary imbibition have been considered.The shale gas production,pore pressure distribution and water saturation distribution in the reservoir have been simulated.The influences of hydraulic fracture geometry and nonorthogonal hydraulic fractures on gas production have been determined and discussed accordingly.The simulation results show that the daily gas production has an upward and downward trend due to the presence of a large amount of fracturingfluid in the reservoir around the hydraulic fracture.The smaller the angle between the hydraulic fracture and the wellbore,the faster the daily production of shale gas wells decreases,and the lower the cumulative production.Nonplanar fractures can increase the control volume of hydraulic fractures and improve the production of shale gas wells.Yongwei Duan Zhaopeng Zhu Hui He Gaoliang Xuan Xuemeng Yu 2024Fluid Dynamics & Materials Processing2024,20,2:0
19Theoretical investigation on submicron particle penetration through circular tubes inside a porous medium显示文摘The analytical infinite series solution of submicron particle transport in a circular tube bounded by a porous wall,such as a pinhole,is determined under the slip velocity boundary condition,and the solution is verified by using the experimental data in the previous studies for the specific cases.The results show that particle penetration rate increases with the increase of the porous parameter,the axial pressure drop,and the pinhole radius,whereas it decreases with increasing the pinhole length.The penetration rate of nano-particles are more sensitive to the variation of these parameters.However,the differences between the penetrations of particles ranging from 0.3μm to 1μm are not evident because the diffusion becomes weak gradually in this size range.In addition,a further comparison is performed between the analytical solution and the existing studies,and approximate expressions are presented for accurate calculation of particle penetration rate through pinholes appearing in porous materials including filter devices and masks.Yanrong Yu Yongwei Gao Yanming Kang 2022Particuology2022,,11:0
20Touchable cell biophysics property recognition platforms enable multifunctional blood smart health care显示文摘As a crucial biophysical property,red blood cell(RBC)deformability is pathologically altered in numerous disease states,and biochemical and structural changes occur over time in stored samples of otherwise normal RBCs.However,there is still a gap in applying it further to point-of-care blood devices due to the large external equipment(high-resolution microscope and microfluidic pump),associated operational difficulties,and professional analysis.Herein,we revolutionarily propose a smart optofluidic system to provide a differential diagnosis for blood testing via precise cell biophysics property recognition both mechanically and morphologically.Deformation of the RBC population is caused by pressing the hydrogel via an integrated mechanical transfer device.The biophysical properties of the cell population are obtained by the designed smartphone algorithm.Artificial intelligence-based modeling of cell biophysics properties related to blood diseases and quality was developed for online testing.We currently achieve 100%diagnostic accuracy for five typical clinical blood diseases(90 megaloblastic anemia,78 myelofibrosis,84 iron deficiency anemia,48 thrombotic thrombocytopenic purpura,and 48 thalassemias)via real-world prospective implementation;furthermore,personalized blood quality(for transfusion in cardiac surgery)monitoring is achieved with an accuracy of 96.9%.This work suggests a potential basis for next-generation blood smart health care devices.Longfei Chen Yantong Liu Hongshan Xu Linlu Ma Yifan Wang Le Yu Fang Wang Jiaomeng Zhu Xuejia Hu Kezhen Yi Yi Yang Hui Shen Fuling Zhou Xiaoqi Gao Yanxiang Cheng Long Bai Yongwei Duan Fubing Wang Yimin Zhu 2021Microsystems & Nanoengineering2021,7,6:0
