| 1 | Significance and relationship between Yes-associated protein and survivin expression in gastric carcinoma and precancerous lesions显示文摘AIM:To analyze the differences and relevance of Yes-associated protein (YAP) and survivin, and to explore the correlation and signifi cance of their expression in gastric carcinoma and precancerous lesions.METHODS: The PV9000 immunohistochemical method was used to detect the expression of YAP and survivin in 98 cases of normal gastric mucosa, 58 intestinal metaplasia (IM), 32 dysplasia and 98 gastric carcinoma.RESULTS: The positive rates of YAP in dysplasia (37.5%) and gastric carcinoma (48.0%) were significantly higher than that in normal gastric mucosa (13.3%), P<0.01. The positive rates of survivin in IM (53.4%), dysplasia (59.4%) and gastric carcinoma (65.3%) were significantly higher than in normal gastric mucosa (11.2%), P<0.01. Survivin expression gradually increased from 41.7% in well differentiated adenocarcinoma through 58.3% in moderately differentiated adenocarcinoma to 75.6% in poorly differentiated adenocarcinoma, with significant Rank correlation, rk=0.279, P<0.01. The positive rate of survivin in gastric carcinoma of diffused type (74.6%) was significantly higher than that in intestinal type (51.3%), P<0.05. In gastric carcinoma with lymph node metastasis (76.9%), the positive rate of survivin was signifi cantly higher than that in the group without lymph node metastasis (41.2%), P<0.01. In 98 cases of gastric carcinoma, the expression of YAP and of survivin were positively correlated, rk=0.246, P<0.01.CONCLUSION: YAP may play an important role as a carcinogenic factor and may induce survivin expression. | 3 | Role and limitation of FMPSPGR dynamic contrast scanning in the follow-up of patients with hepatocellular carcinoma treated by TACE显示文摘AIM:To evaluate the role and limitation of fast multiplanarspoiled gradient-recalled (FMPSPGR) MR dynamiccontrast scanning in the follow-up of patients with HCCtreated by transarterial chemoembolization (TACE).METHODS:Twenty-two patients with 24 HCC lesionsconfirmed by biopsy or surgical resection underwentMR imaging in 4-9wks after TACE with a superconducting1.5 T MR scanner, including SE T1WI, T2WI and FMPSPGRdynamic contrast scanning. The signal intensities of alllesions on SE T1WI, T2WI and the enhancement patternson FMPSPGR dynamic contrast scanning were observed,and the comparison was made between MRI findingsand pathological results in ail the cases.RESULTS:Of the 24 lesions, the signal intensities werevarious on SE T1WI and T2WI. On T1WI, 13 lesionsappeared as hyperintense, 4 lesions were isointenseand the other 7 lesions were hypointensese.Histologically, hyperintense lesions showed on T1WIwere viable tumor or hemorrhage; isointensities werecoagulative necrosis or inflammatory infiltration;hypointensities were tumor, liquified necrosis,coagulative necrosis or inflammatory infiltration. OnT2WI, 15 lesions appeared as hyperintense, 3 lesionswere isointense and the other 6 lesions werehypointensese. Hyperintense lesions showed on T2WIwere residuals of viable tumor, hemorrhage, liquefiednecrosis or inflammatory infiltration; isointense lesionswere residuals of viable tumor or inflammatoryinfiltration; hypointense lesions were coagulativenecrosis. On FMPSPGR dynamic contrast scanning, 18of the 24 lesions enhanced on early-phase dynamicscanning conesponding to residuals of viable tumor andthe other 6 lesions had no enhancement at this phasebecause complete necrosis were seen in the histologicexamination. On delayed-phase dynamic scanning, 6lesions had permanent enhancement appeared asinhomogeneous hyperintensity and both residuals ofviable tumor and inflammatory infiltration were foundby histologic examination. 18 lesions were hypointenseat this phase and 8 of them coexisted with peripheralring-like enhancement of the lesions resulting fromviable tumors or inflammatory infiltration.CONCLUSION: FMPSPGR MR dynamic contrast scanningcan reflect the pathologic changes of HCC treated byTACE. Especially, early-phase dynamic scanning canevaluate accurately residuals of viable tumor andnecrosis in HCC lesions. | 5 | Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. | 8 | In situ optical backpropagation training of diffractive optical neural networks显示文摘Training an artificial neural network with backpropagation algorithms to perform advanced machine learning tasks requires an extensive computational process.This paper proposes to implement the backpropagation algorithm optically for in situ training of both linear and nonlinear diffractive optical neural networlks,which enables the acceleration of training speed and improvement in energy efficiency on core computing modules.We demonstrate that the gradient of a loss function with respect to the weights of diffractive layers can be accurately calculated by measuring the forward and backward propagated optical fields based on light reciprocity and phase conjunction principles.The diffractive modulation weights are updated by programming a high-speed spatial light modulator to minimize the error between prediction and target output and perform inference tasks at the speed of light.We numerically validate the effectiveness of our approach on simulated networks for various applications.The proposed in situ optical learning architecture achieves accuracy comparable to in silico training with an electronic computer on the tasks of object dlassification and matrix-vector multiplication,which further allows the diffractive optical neural network to adapt to system imperfections.Also,the self-adaptive property of our approach facilitates the novel application of the network for all-optical imaging through scattering media.The proposed approach paves the way for robust implementation of large-scale difractive neural networks to perform distinctive tasks all-optically. | TIANKUANG ZHOU LU FANG TAO YAN JIAMIN WU YIPENG LI JINGTAO FAN HUAQIANG WU XING LIN QIONGHAI DA | 2020 | Photonics Research2020,8,6: | 11 |
| 9 | Key nodes of a micro RNA network associated with the integrated mesenchymal subtype of high-grade serous ovarian cancer显示文摘Metastasis is the main cause of cancer mortality. One of the initiating events of cancer metastasis of epithelial tumors is epithelial-to-mesenchymal transition(EMT), during which cells dedifferentiate from a relatively rigid cell structure/morphology to a flexible and changeable structure/morphology often associated with mesenchymal cells. The presence of EMT in human epithelial tumors is reflected by the increased expression of genes and levels of proteins that are preferentially present in mesenchymal cells. The combined presence of these genes forms the basis of mesenchymal gene signatures, which are the foundation for classifying a mesenchymal subtype of tumors. Indeed, tumor classification schemes that use clustering analysis of large genomic characterizations, like The Cancer Genome Atlas(TCGA), have defined mesenchymal subtype in a number of cancer types, such as high-grade serous ovarian cancer and glioblastoma. However, recent analyses have shown that gene expression-based classifications of mesenchymal subtypes often do not associate with poor survival. This 'paradox' can be ameliorated using integrated analysis that combines multiple data types. We recently found that integrating m RNA and micro RNA(mi RNA) data revealed an integrated mesenchymal subtype that is consistently associated with poor survival in multiple cohorts of patients with serous ovarian cancer. This network consists of 8 major mi RNAs and 214 m RNAs. Among the 8 mi RNAs, 4 are known to be regulators of EMT. | 12 | Mycoplasma pneumoniae Macrolide Resistance and MLVA Typing in Children in Beijing,China,in 2016:Is It Relevant?显示文摘Objective The aim of this study is to investigate the macrolide resistance rate and molecular type with multiple-locus variable-number tandem-repeat analysis(MLVA)of Mycoplasma pneumoniae of Beijing in 2016 in pediatric patients.Methods Real-time quantitative polymerase chain reaction(PCR)was used to identify M.pneumoniae,and MLVA was performed.The domain V of the 23 S rRNA was sequenced to detect macrolide-resistant point mutations.We also investigated the activities of antibiotics against M.pneumoniae isolates in vitro.Results The PCR detection rate of M.pneumoniae in children in Beijing was 40%,and the macrolide resistance rate was 66%.The A2063 G mutation in the 23 S rRNA V region is the dominant mutation(137/146,93.84%),whereas the A2064 G mutation is rare(9/146,6.16%).Seventy-three samples were typed successfully by MLVA typing,including 86.3%(63/73)were MLVA type 4-5-7-2,and 13.7%(10/73)were MLVA type 3-5-6-2.No other types were found.No strains were resistant to levofloxacin or tetracycline.Conclusion In 2016,a specific decrease in the macrolide resistance rate occurred in Beijing.The detection rate and macrolide resistance rate of outpatients are lower than those of inpatients.The A2063 G mutants M.pneumoniae have high levels of resistance to erythromycin and azithromycin.The primary MLVA type is 4-5-7-2,followed by 3-5-6-2.No other MLVA types were detected.No strains resistant to tetracycline or levofloxacin were found in vitro. | DOU Hai Wei TIAN Xiu Jun XIN De Li WEI Ran ZHOU Wei WANG Hong QIN Xuan Guang SHAO Jun Yan XU Bao Ping GE Li Xia SHI Da Wei | 2020 | Biomedical and Environmental Sciences2020,33,12: | 9 |
| 13 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 14 | Clinicopathological significance of B-cell-specific Moloney murine leukemia virus insertion site 1 expression in gastric carcinoma and its precancerous lesion显示文摘AIM: To explore the relation between B-cell-specific Moloney murine leukemia virus insertion site 1 (Bmi-1) expression and the clinicopathological features of gastric carcinoma (GC). METHODS: Immunohistochemistry was used to detect the expression of Bmi-1 and ki-67. Double-labeling staining was used to display the distribution of Bcl-2+/ki-67- cells in 162 cases of GC and its matched normal mucosa and precancerous lesion. RESULTS: The positive rate of Bmi-1 expression in GC (52.5%) was significantly higher than that in normal gastric mucosa (21.6%, χ2 = 33.088, P < 0.05). The Bmi-1 expression in GC was closely related with the Lauren's and Borrmann's classification and clinical stage (χ2 = 4.400, 6.122 and 11.190, respectively, P < 0.05). The expression of ki-67 was related to the Borrmann's classification (χ2 = 13.380, P < 0.05). Bcl-2 expression was correlated with the Lauren's classification (χ2 = 4.725, P < 0.05), and the Bmi-1expression both in GC (rk = 0.157, P < 0.05) and in intestinal metaplasia (rk = 0.270, P < 0.05). CONCLUSION: Abnormal Bmi-1 expression in GC may be involved in cell proliferation, apoptosis and cancerization. The present study was designed to establish a more efficient system for producing cloned pigs by somatic cell nuclear transfer (SCNT). Our approach was as follows: SCNT embryos were reconstructed by using fetal fibroblasts of Chinese miniature pig as donors and in vitro matured oocytes of prepubertal gilts as recipients. Reconstructed em- bryos were induced by electrical fusion/activation and cultured in BSA-containing North Carolina State University 23 medium (NCSU-23) or Porcine Zygote Medium (PZM-3) at the gas condition of 5% CO2, 7% O2, 88% N2. A total of 230 cloned embryos were transferred to three surrogate sows, producing three piglets. One of them is apparently healthy. The clonal provenance of the piglet was indicated by its coat color and confirmed by DNA microsatellite analysis. | 20 | An action-based Markov chain modeling approach for predicting the window operating behavior in office spaces显示文摘Reliable energy and performance prediction for building design and planning is important for newly-designed or retrofitted buildings.Window operating behavior has an important influence on the ventilation and energy consumption of these buildings under different realistic scenarios.Therefore,quantitatively describing this behavior and constructing a prediction model are important.In this work,an action-based Markov chain modeling approach for predicting window operating behavior in office spaces was proposed.Two summer measurement data(2016 and 2018)were used to verify the accuracy and validity of the modeling approach.The opening rate,outdoor temperature,time distribution,and on-off curve were proposed as four inspection standards.This study also compared the prediction performance between the action-based Markov chain modeling approach with the state-based Markov chain modeling approach,which is the most popular modeling approach to model occupant window operating behavior.This study proved that the yearly variation of occupants’behavior performed a form of action that remained unchanged during a certain period.Meanwhile,the results also proved that the action-based Markov chain modeling approach can reflect the actual window operating behavior accurately within an open-plan office,which is a beneficial supplement for energy-consumption simulation software in a window-state prediction module.The state-based Markov chain modeling approach showed better stability and accuracy in terms of the opening rate,whereas the action-based Markov chain modeling approach showed good consistency with the measurement data in the on-off curves and in situations with little data.For the on-off curves,the accuracy of action-based modeling approach in the prediction of window open-state is 20%higher. | Xin Zhou Tiance Liu Da Yan Xing Shi Xing Jin | 2021 | Building Simulation2021,14,2: | 7 |
