| 3 | Study of liver cirrhosis over ten consecutive years in Southern China显示文摘AIM: To investigate the etiology and complications of liver cirrhosis(LC) in Southern China. METHODS: In this retrospective, cross-sectional study, we identified cases of liver cirrhosis admitted between January 2001 to December 2010 and reviewed the medical records. Patient demographics, etiologies and complications were collected, and etiological changes were illustrated by consecutive years and within twotime periods(2001-2005 and 2006-2010). All results were expressed as the mean ± SD or as a percentage. The χ2 test or Student's t-test was used to analyze the differences in age, gender, and etiological distribution, and one-way analysis of variance was applied to estimate the trends in etiological changes. We analyzed the relationship between the etiologies and complications using unconditioned logistic regression, and the risk of upper gastrointestinal bleeding(UGIB) and hepatocellular carcinoma(HCC) in the major etiological groups was evaluated as ORs. A P value less than 0.05 was considered significant. Statistical computation was performed using SPSS 17.0 software.RESULTS: In this study, we identified 6719(83.16%) male patients and 1361(16.84%) female patients. The average age of all of the patients was 50.5 years at the time of diagnosis. The distribution of etiological agents was as follows: viral hepatitis, 80.62% [hepatitis B virus(HBV) 77.22%, hepatitis C virus(HCV) 2.80%,(HBV + HCV) 0.58%]; alcohol, 5.68%; mixed etiology, 4.95%; cryptogenic, 2.93%; and autoimmune hepatitis, 2.03%; whereas the other included etiologies accounted for less than 4% of the total. Infantile hepatitis syndrome LC patients were the youngest(2.5 years of age), followed by the metabolic LC group(27.2 years of age). Viral hepatitis, alcohol, and mixed etiology were more prevalent in the male group, whereas autoimmune diseases, cryptogenic cirrhosis, and metabolic diseases were more prevalent in the female group. When comparing the etiological distribution in 2001-2005 with that in 2006-2010, the proportion of viral hepatitis decreased from 84.7% to 78.3%(P < 0.001), and the proportion of HBV-induced LC also decreased from 81.9% to 74.6%(P < 0.001). The incidence of mixed etiology, cryptogenic cirrhosis, and autoimmune diseases increased by 3.1%(P < 0.001), 0.5%(P = 0.158), and 1.3%(P < 0.001), respectively. Alcohol-induced LC remained relatively steady over the 10-year period. The ORs of the development of UGIB between HBV and other major etiologies were as follows: HCV, 1.07; alcohol, 1.89; autoimmune, 0.90; mixed etiology, 0.83; and cryptogenic, 1.76. The ORs of the occurrence of HCC between HBV and other major etiologies were as follows: HCV, 0.54; alcohol, 0.16; autoimmune, 0.05; mixed etiology, 0.58; and cryptogenic, 0.60.CONCLUSION: The major etiology of liver cirrhosis in Southern China is viral hepatitis. However, the proportions of viral hepatitis and HBV are gradually decreasing. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. | 6 | Prevalence and evolution of drug resistance HIV-1 variants in Henan, China显示文摘To understand the prevalence and evolution of drug resistant HIV strains in Henan China after the implementation of free antiretroviral therapy for AIDS patients. 45 drug nave AIDS patients, 118 AIDS patients who received three months antiretroviral therapy and 124 AIDS patients who received six months antiretroviral treatment were recruited in the southern part of Henan province. Information on general condition, antiretroviral medicines, adherence and clinical syndromes were collected by face to face interview. Meanwhile, 14ml EDTA anticoagulant blood was drawn. CD4/CD8 T cell count, viral load and genotypic drug resistance were tested. The rates of clinical improvement were 55.1% and 50.8% respectively three months and six months after antiretroviral therapy. The mean CD4 cell count after antiretroviral therapy was significantly higher than in drug nave patients. The prevalence rate of drug resistant HIV strains were 13. 9%, 45.4% and 62.7% in drug nave patients, three month treatment patients and six month treatment patients, respectively. The number of resistance mutation codons and the frequency of mutations increased significantly with continued antiretroviral therapy. The mutation sites were primarily at the 103, 106 and 215 codons in the three-month treatment group and they increased to 15 codon mutations in the six-month treatment group. From this result, the evolution of drug resistant strains was inferred to begin with the high level NNRTI resistant strain, and then develop low level resistant strains to NRTIs. The HIV strains with high level resistance to NVP and low level resistance to AZT and DDI were highly prevalent because of the AZT+DDI+NVP combination therapy. These HIV strains were also cross resistant to DLV, EFV, DDC and D4T. Poor adherence to therapy was believed to be the main reason for the emergence and prevalence of drug resistant HIV strains. The prevalence of drug resistant HIV strains was increased with the continu- ation of antiretroviral therapy in the southern part of Henan province. METHODS: MTT assay was used to determine the cytotoxic effects of SN50 in gastric cancer cell line SGC7901. Hoechst 33258 staining was used to detect apoptosis morphological changes after SN50 treatment. Activation of autophagy was monitored with monodansylcadaverine (MDC) staining after SN50 treatment.Immunofluorescence staining was used to detect the expression of light chain 3 (LC3). Mitochondrial membrane potential was measured using the fluorescent probe JC-1. Western blotting analysis were used to determine the expression of proteins involved in apoptosis and autophagy including p53, p53 upregulated modulator of apoptosis (PUMA), damage-regulated autophagy modulator (DRAM), LC3 and Beclin 1. We detected the effects of p53-mediated autophagy activation on the apoptosis of SGC7901 cells with the p53 inhibitor pifithrin-α. RESULTS: The viability of SGC7901 cells was inhibited after SN50 treatment. Inductions in the expression of apoptotic protein p53 and PUMA as well as autophagic protein DRAM, LC3 and Beclin 1 were detected with Western blotting analysis. SN50-treated cells exhibited punctuate microtubule-associated protein 1 LC3 in immunoreactivity and MDC-labeled vesicles increased after treatment of SN50 by MDC staining. Collapse of mitochondrial membrane potential Δψ were detected for 6 to 24 h after SN50 treatment. SN50-induced increases in PUMA, DRAM, LC3 and Beclin 1 and cell death were blocked by the p53 specific inhibitor pifithrin-α. | 18 | Effects of high pressure on the microstructure and hardness of a Cu-Zn alloy显示文摘The microstructure of a Cu-Zn alloy treated under different high pressures was investigated by means of metallographic, scanning electron microscope (SEM), energy dispersive spectrometer (EDS), and X-ray diffraction (XRD), and the hardness of the Cu-Zn alloy was also measured. The results show that the α phase with a smaller grain size, different shapes, and random distribution appears in the Cu-Zn alloy during the solid-state phase transformation generation in the temperature range of 25-750°C and the pressure range of 0-6 GPa. The amount of residual α phase in the microstructure decreases and then increases with increasing pressure. Under a high pressure of 3 GPa, the least volume fraction of residual α phase was obtained, and under a high pressure of 6 GPa, the changes of the microstructure of the Cu-Zn alloy were not obvious. | 19 | Chinese herb feed additives improved the growth performance, meat quality, and nutrient digestibility parameters of pigs显示文摘Background:Since the use of antibiotics in animal feed has become a critical concern worldwide due to severe threats to human health and environment,we are in need of finding alternatives to antibiotics in pig breeding,maintaining the health of pigs,and getting high-quality pork.As traditional Chinese herbs(TCH)are rich natural resources in China and show great benefits to human health we propose to transfer this abundant resource into animal production industry as additives.Methods:Three groups of Chinese herbs(groups A,B,and C)were used as feed additives in the diet for pigs.In total 32 pigs were arranged in four groups(groups A,B,C,and control group,NC),fed in the same facility,eight pigs(one group)in each colony,free drinking,for 120 days.The feed:gain ratio(F/G),meat quality,total protein,and amino acid concentration of muscle were checked in the experiments.Results:After 120 days of feeding,the feed:gain ratio(F/G)of pigs in groups A,B,and C was decreased 17.56%,9.31%,and 13.86%compared with NC treatment,respectively.The diets supplemented with Chinese herbs improved meat quality,increased loin eye area(especially group A and C showed significant difference,P<.001),the total protein(increased ratio vs NC was A=4.54%,B=0.38%and C=3.53%),amino acid concentration of muscle,increased the villus height:crypt depth ratio,and induced positive effects on serum biochemical parameters and immune function(serum TC and TG concentrations were significantly lower than those in the NC group,P<.05.).Conclusions:The use of Chinese herbal feed additives can reduce the cost of pig breeding and produce high-quality pock.The combination of these effects would contribute to better absorption ability of the intestinal tract and yield a better growth performance. | Zhong-ning Lin Li Ye Zhen-wu Li Xiu-sheng Huang Zheng Lu You-quan Yang Huan-wei Xing Jie-ying Bai Zhao-yang Ying | 2020 | Animal Models and Experimental Medicine2020,3,1: | 17 |
