2激光诱导击穿光谱技术对钢中缺陷的快速表征显示文摘激光诱导击穿光谱技术(LIBS)作为一种快速而简单地检查钢表面缺陷的技术,可直接观测到激光脉冲被聚焦于样品之上而感生的等离子体发射物。一个平凸透镜把斑直径大约为1 mm的光量开关Nd:YAG激光器(脉宽:12 ns;重复频率:10 Hz)辐照聚焦于样品表面,用来烧蚀样品的一部分,形成一个微等离子体。等离子体的发射由光纤传送至帕邢-龙校格装置多色仪(焦距:500 mm)。在已被流入和流出的氩气排空的空间里,样品被安装在一个二维移动台上进行两点分析——常规分析和缺陷分析。通过比较两个分析结果,可检测到缺陷部分有显著信号强度的元素。由不同类型的夹杂物可以检测出典型的元素,由氧化铝可检测出铝,由保护渣可检测出铝、钙、镁、硅和钠,由矿渣可检测出铝、钙和镁。经证实,由LIBS分析得到的结果与EPMA(电子探针)研究得到的结果一致。因此,导致缺陷的夹杂物的类型都能通过LIBS技术确定属性。当涂层被激光烧蚀去掉后,镀锌钢也可以被直接分析出来。包括制样,该项技术评定时间不到半小时,因此在炼钢过程中,可以迅速采取合适措施。KONDO Hiroyuki AIMOTO Michihiro YAMAMURA Hideaki TOH Takehiko 2009冶金分析2009,29,1:13

5Capecitabine with radiation is an effective adjuvant therapy in gastric cancers显示文摘AIM:To analyze the outcome of patients who received concurrent capecitabine(Xeloda) and radiation(XRT) compared to the established concurrent 5-fluorouracil(5-FU) with radiation(5FU-RT) and fluoropyrimidine-based chemotherapy alone as adjuvant treatment in gastric cancers.METHODS:All patients with gastric cancers who received adjuvant treatment at the National Cancer Centre Singapore between 1996 and 2006 were reviewed.Treatment outcomes of patients who received XRT were compared with those who had 5FU-RT or chemotherapy alone as adjuvant therapy for gastric cancers.RESULTS:A total of 108 patients were reviewed.Median age at diagnosis was 60.The majority of the patients(64.8%) had advanced stage Ⅲ and Ⅳ disease(with no distant metastasis).All except 4 patients had D2 gastrectomy.Twenty one patients(19.4%) had positive surgical resection margins.Thirty three patients received XRT compared with 52 who had 5FU-RT and 23 who received chemotherapy alone.For the patients in the chemotherapy-only group,all had fluoropyrimidine-based therapy,with added cisplatin in 7 patients and epirubicin in 2 patients.Median recurrence-free survival was longer for the XRT group(52 mo) compared to the 5FU-RT(35 mo) and chemotherapy-only groups(25 mo)(P=0.48).The patients in the XRT group achieved similar median overall survival(53 mo) as the 5FU-RT(54 mo) and the chemotherapy-only groups(44 mo)(P=0.5).CONCLUSION:Capecitabine with concurrent radiation was as effective as concurrent 5FU with radiation or fluoropyrimidine-based chemotherapy alone when used as adjuvant treatment in patients with gastric cancers.Chee Kian Tham Su Pin Choo Donald Yew Hee Poon Han Chong Toh Simon Yew Kuang Ong Sze Huey Tan Michael Lian Chek Wang Kian Fong Foo 2010World Journal of Gastroenterology2010,16,29:6
6Distribution of pericellular matrix molecules in the temporomandibular joint and their chondroprotective effects against inflammation显示文摘The objectives of this study were to(1) determine the distribution and synthesis of pericellular matrix(PCM) molecules(collagen VI, collagen IV and laminin) in rat temporomandibular joint(TMJ) and(2) investigate the effects of PCM molecules on chondrocytes against inflammation in osteoarthritis. Four zones(fibrous, proliferating, mature and hypertrophic) of condylar cartilage and three bands(anterior, intermediate and posterior) of disc were analysed by immunohistochemistry for the presence of PCM molecules in rat TMJs. Isolated chondrocytes were pre-treated with PCM molecules before being subjected to interleukin(IL)-1β treatment to stimulate inflammation. The responses of the chondrocytes were analysed using gene expression, nitric oxide release and matrix metalloproteinase(MMP)-13 production measures. Histomorphometric analyses revealed that the highest areal deposition of collagen VI(67.4%), collagen IV(45.7%) and laminin(52.4%) was in the proliferating zone of TMJ condylar cartilage. No significant difference in the distribution of PCM molecules was noted among the three bands of the TMJ disc. All three PCM molecules were expressed intracellularly by chondrocytes cultured in the monolayer. Among the PCM molecules, pre-treatment with collagen VI enhanced cellular proliferation, ameliorated IL-1β-induced MMP-3, MMP-9, MMP-13 and inducible nitric oxide synthase gene expression, and attenuated the downregulation of cartilage matrix genes, including collagen I, aggrecan and cartilage oligomeric matrix protein(COMP). Concurrently, collagen VI pretreatment inhibited nitric oxide and MMP-13 production. 8Review of imaging and endovascular intervention of iliocaval venous compression syndrome显示文摘Iliocaval venous compression syndrome(ICS)is the extrinsic compression of the common iliac vein by the overlying iliac artery against the vertebra.Chronic compression can lead to venous stenosis and stasis,which manifests as chronic venous disease and treatment resistance.Therefore,early recognition of ICS and prompt treatment are essential.Clinical presentations of ICS can be ambiguous and diagnosis requires a high index of suspicion with the relevant imaging studies.The initial imaging test is typically a Duplex ultrasound for vessel assessment and pelvic ultrasound to exclude a compressive mass,which is followed by computed tomography(CT)or magnetic resonance(MR)venography.CT and MRI can identify the anatomical causes for venous compression.In patients with high clinical suspicion for ICS,negative findings on CT and MR venography would still warrant further investigations.Definitive diagnosis can be established using catheter-based venography complemented with intravascular ultrasonography but the nature of their invasiveness limits its utility as a routine imaging modality.In this review paper,we will discuss the evidence,utility and limitations of the existing imaging modalities and endovascular intervention used in the management of ICS.Ming Ren Toh Tjun Yip Tang Han Hui Mervin Nathan Lim Nanda Venkatanarasimha Karthikeyan Damodharan 2020World Journal of Radiology2020,12,3:4
9Liposomes as sterile preparations and limitations of sterilisation techniques in liposomal manufacturing显示文摘Liposomes have been widely researched as a delivery system and there have been many manufacturing techniques used in the production of liposomal preparations,the most common being extrusion method which will be introduced in this paper.However because of the unique properties of liposomes and their susceptibility to chemical and physical degradation,sterilisation remains an unresolved issue in the manufacturing of liposomebased formulations.It is especially pertinent in the pharmaceutical industry where liposomes are commonly prepared for intravenous administration.Currently,filtration and aseptic manufacturing are recommended for the preparation of sterile liposomal products.Newer aseptic manufacturing techniques such as dense gas techniques have been devised to eliminate the need for terminal sterilisation.This paper will highlight the limitations of the conventional techniques that are specific to the liposome preparation under the respective sterilisation conditions specified by the 2011 British Pharmacopoeia to achieve 106 Sterility Assurance Level,as well as modifications incorporated in the newer sterilisation technologies to overcome these limitations.This paper will introduce these techniques in brief,including their advantages and limitations.Ming-Ren Toh Gigi N.C.Chiu 2013Asian Journal of Pharmaceutical Sciences2013,8,2:4

11Awakening immunity against cancer:a 2017 primer for clinicians显示文摘Cancer immunotherapy has finally joined the pillars of cancer treatment-surgery, radiation, chemotherapy, hormonal therapy, and targeted therapy-in improving cancer patient lives. In the last 5 years, the development of immune checkpoint inhibitor and T cell therapy, particularly chimeric antigen receptor(CAR) T-cell therapy, has been remarkable for the speed, scale, and number of drug approvals. Still, these treatments may also bring unusual adverse effects and clinical outcomes including unprecedented long-term survival. Interrogating the tumor microenvironment and identifying better biomarkers hold the key to improving cancer immunotherapies. CAR T-cell therapy has dramatic effect on leukemias and lymphomas with significant cure rates, but has yet to show comparable effect on solid tumors. Cutting-edge technology will improve both processing and clinical effect of such therapies. Asia has the largest, most rapidly aging population and the largest number of cancer patients in the world. 13Extracellular histones stimulate collagen expression in vitro and promote liver fibrogenesis in a mouse model via the TLR4-MyD88 signaling pathway显示文摘BACKGROUND Liver fibrosis progressing to liver cirrhosis and hepatic carcinoma is very common and causes more than one million deaths annually.Fibrosis develops from recurrent liver injury but the molecular mechanisms are not fully understood.Recently,the TLR4-MyD88 signaling pathway has been reported to contribute to fibrosis.Extracellular histones are ligands of TLR4 but their roles in liver fibrosis have not been investigated.AIM To investigate the roles and potential mechanisms of extracellular histones in liver fibrosis.METHODS In vitro,LX2 human hepatic stellate cells(HSCs)were treated with histones in the presence or absence of non-anticoagulant heparin(NAHP)for neutralizing histones or TLR4-blocking antibody.The resultant cellular expression of collagen I was detected using western blotting and immunofluorescent staining.In vivo,the CCl4-induced liver fibrosis model was generated in male 6-week-old ICR mice and in TLR4 or MyD88 knockout and parental mice.Circulating histones were detected and the effect of NAHP was evaluated.RESULTS Extracellular histones strongly stimulated LX2 cells to produce collagen I.Histone-enhanced collagen expression was significantly reduced by NAHP and TLR4-blocking antibody.In CCl4-treated wild type mice,circulating histones were dramatically increased and maintained high levels during the duration of fibrosisinduction.Injection of NAHP not only reduced alanine aminotransferase and liver injury scores,but also significantly reduced fibrogenesis.Since the TLR4-blocking antibody reduced histone-enhanced collagen I production in HSC,the CCl4 model with TLR4 and MyD88 knockout mice was used to demonstrate the roles of the TLR4-MyD88 signaling pathway in CCl4-induced liver fibrosis.The levels of liver fibrosis were indeed significantly reduced in knockout mice compared to wild type parental mice.CONCLUSION Extracellular histones potentially enhance fibrogenesis via the TLR4–MyD88 signaling pathway and NAHP has therapeutic potential by detoxifying extracellular histones.Zhi Wang Zhen-Xing Cheng Simon T Abrams Zi-Qi Lin ED Yates Qian Yu Wei-Ping Yu Ping-Sheng Chen Cheng-Hock Toh Guo-Zheng Wang 2020World Journal of Gastroenterology2020,26,47:3
14Biodegradable nanocarriers for small interfering ribonucleic acid (siRNA) co-delivery strategy increase the chemosensitivity of pancreatic cancer cells to gemcitabine显示文摘Ribonucleic acid (RNA) interference (RNAi) therapies are promising cancertreatment modalities that can specifically target abnormal proto-oncogenes, thusimproving the therapeutic effect. For the treatment of pancreatic cancer, targetingone mutant proto-oncogene by RNAi usually does not yield the desired therapeuticefficiency. Both K-ras gene mutations and Notch1 overexpression are commonsymptoms in pancreatic cancer patients, and play a crucial role in pancreaticcancer cell drug resistance. In this study, biodegradable charged polyester-basedvectors (BCPVs) were synthesized for the co-delivery of K-ras and Notch1 smallinterfering ribonucleic acid (siRNA) into MiaPaCa-2 cells (pancreatic cancercell line) to overcome drug resistance to gemcitabine (GEM), a first-line chem-otherapeutic drug used in the clinic. BCPVs could effectively absorb negative siRNAto form a capsule-like structure, prevent siRNA from nuclease digestion in theserum, and promote effective siRNA cell internalization and endosomal escape.Through K-ras and Notch1 gene silencing in MiaPaCa-2 cells, BCPV-siRNAKsiRNAN^tcm nanocomplexes effectively reversed the epithelia-mesenchymaltransition (EMT) in MiaPaCa-2 cells, thereby greatly enhancing the sensitivity ofMiaPaCa-2 cells to GEM. MiaPaCa-2 cell proliferation, migration, and invasionwere effectively inhibited, and cell apoptosis was also significantly enhanced bythe synergistic antitumor effect of BCPV-siRNAK siRNAN^t~l nanocomplexesand GEM. These results suggest that this combination RNAi therapy can be usedto improve cancer cell sensitivity to chemotherapeutic drugs. 16Cytotoxic CD8+ T cells and tissue resident memory cells in colorectal cancer based on microsatellite instability and BRAF status显示文摘BACKGROUND Recent studies in non-colorectal malignancy have associated T resident memory(T_(RM)) cells with improved patient survival. It is unknown if T_(RM) plays a role in colorectal cancer(CRC).AIM To examine the potential role of T_(RM) cells in providing immunogenicity in CRC stratified by microsatellite instability(MSI) and BRAF status.METHODS Patients with known MSI and BRAF mutation status were eligible for inclusion in this study. CRC tumour sections stained with haematoxylin and eosin were microscopically reviewed and the images scanned prior to assessment for location of invading edge and core of tumour. Sequential sections were prepared for quantitative multiplex immunohistochemistry(IHC) staining. Opal Multiplex IHC staining was performed with appropriate positive and negative controls and imaged using a standard fluorescent microscope fitted with a spectral scanning camera(Mantra) in conjunction with Mantra snap software. Images were unmixed and annotated in in Form 2.2.0. Statistical analysis was performed using Graphpad Prism Version 7 and Stata Version 15.RESULTS Seventy-two patients with known MSI and BRAF status were included in the study. All patients were assessed for MSI by IHC and high resolution capillary electrophoresis testing and 44 of these patients successfully underwent quantitative multiplex IHC staining. Overall, there was a statistically significant increase in CD8+ T_(RM) cells in the MSI(BRAF mutant and wild type) group over the microsatellite stable(MSS) group. There was a statistically significant difference in CD8+ T_(RM) between high level MSI(MSI-H):BRAF mutant [22.57, 95% confidence interval(CI): 14.31-30.84] vs MSS [8.031(95%CI: 4.698-11.36)], P = 0.0076 and MSI-H:BRAF wild type [16.18(95%CI: 10.44-21.93)] vs MSS [8.031(95%CI: 4.698-11.36)], P = 0.0279. There was no statistically significant difference in CD8 T cells(both CD8+CD103-and CD8+CD103+T_(RM)) between MSI-H: BRAF mutant and wild type CRC.CONCLUSION This study has shown that CD8+ T_(RM) are found in greater abundance in MSI-H CRC, both BRAF mutant and MSI-H:BRAF wild type, when compared with their MSS counterpart. CD8+ T_(RM) may play a role in the immunogenicity in MSI-H CRC(BRAF mutant and BRAF wild type). 17Indications and surgical options for small bowel, large bowel and perianal Crohn's disease显示文摘Despite advancements in medical therapy of Crohn's disease(CD), majority of patients with CD will eventually require surgical intervention, with at least a third of patients requiring multiple surgeries. It is important to understand the role and timing of surgery, with the goals of therapy to reduce the need for surgery without increasing the odds of emergency surgery and its associated morbidity, as well as to limit surgical recurrence and avoid intestinal failure. The profile of CD patients requiring surgical intervention has changed over the decades with improvements in medical therapy with immunomodulators and biological agents. The most common indication for surgery is obstruction from stricturing disease, followed by abscesses and fistulae. The risk of gastrointestinal bleeding in CD is high but the likelihood of needing surgery for bleeding is low. Most major gastrointestinal bleeding episodes resolve spontaneously, albeit the risk of re-bleeding is high. The risk of colorectal cancer associated with CD is low. While current surgical guidelines recommend a total proctocolectomy for colorectal cancer associated with CD, subtotal colectomy or segmental colectomy with endoscopic surveillance may be a reasonable option. Approximately 20%-40% of CD patients will need perianal surgery during their lifetime.
