| 1 | Output Characteristics of a Series Three-port Axial Piston Pump显示文摘Driving a hydraulic cylinder directly by a closed-loop hydraulic pump is currently a key research area in the field of electro-hydraulic control technology,and it is the most direct means to improve the energy efficiency of an electro-hydraulic control system.So far,this technology has been well applied to the pump-controlled symmetric hydraulic cylinder.However,for the differential cylinder that is widely used in hydraulic technology,satisfactory results have not yet been achieved,due to the asymmetric flow constraint.Therefore,based on the principle of the asymmetric valve controlled asymmetric cylinder in valve controlled cylinder technology,an innovative idea for an asymmetric pump controlled asymmetric cylinder is put forward to address this problem.The scheme proposes to transform the oil suction window of the existing axial piston pump into two series windows.When in use,one window is connected to the rod chamber of the hydraulic cylinder and the other is linked with a low-pressure oil tank.This allows the differential cylinders to be directly controlled by changing the displacement or rotation speed of the pumps.Compared with the loop principle of offsetting the area difference of the differential cylinder through hydraulic valve using existing technology,this method may simplify the circuits and increase the energy efficiency of the system.With the software SimulationX,a hydraulic pump simulation model is set up,which examines the movement characteristics of an individual piston and the compressibility of oil,as well as the flow distribution area as it changes with the rotation angle.The pump structure parameters,especially the size of the unloading groove of the valve plate,are determined through digital simulation.All of the components of the series arranged three distribution-window axial piston pump are designed,based on the simulation analysis of the flow pulse characteristics of the pump,and then the prototype pump is made.The basic characteristics,such as the pressure,flow and noise of the pumps under different rotation speeds,are measured on the test bench.The test results verify the correctness of the principle.The proposed research lays a theoretical foundation for the further development of a new pump-controlled cylinder system. | ZHANG Xiaogang QUAN Long YANG Yang WANG Chengbin YAO Liwei | 2012 | Chinese Journal of Mechanical Engineering2012,25,3: | 10 |
| 2 | Targeted Mutagenesis of NAC Transcription Factor Gene,OsN AC041,Leading to Salt Sensitivity in Rice显示文摘Salinity is a major abiotic stress factor that seriously affects plant growth. Many genes are involved in the response to salt stress with various metabolism pathways. A number of plant transcription factor family genes have been found to be involved in the salt stress response, and NAM, ATAF and CUC(NAC) transcription factors are thought to act as active regulators during abiotic stress, especially salt stress. In this study, we detected a rice NAC transcription factor coding gene, OsNAC041, and confirmed that it influenced the germination of seeds under salt stress and salt tolerance of plants. OsNAC041 was primarily expressed in the leaves and located in the nucleus. Furthermore, the CRISPR/Cas9 method was used to obtain a targeted osnac041 mutant, of which the plant height was higher than that of the wild-type, showing increased salt sensitivity. Moreover, RNA-seq analysis revealed a number of differentially expressed genes(DEGs) involved in several important signaling pathways in the osnac041 mutant. Subsequently, Kyoto Encyclopedia of Genes and Genomes annotation also revealed differential expression of DEGs associated with mitogen-activated protein kinase signaling, peroxisome, eukaryotictype ABC transporters, photosynthesis and plant hormones, which are involved in stress-related signaling pathways. Overall, our study suggested that OsNAC041 was involved in the salt stress response in rice. | 5 | One-step synthesis of monodispersed Pt nanoparticles anchored on 3D graphene foams and its application for electrocatalytic hydrogen evolution显示文摘Although platinum-based materials are regarded as the state-of-the-art electro-catalysts for hydrogen evolution reaction(HER),high cost and quantity scarcity hamper their scale-up utilization in industrial deployment.Herein,a one-step strategy was developed to synthesize multi-walled carbon nanotubes and reduced graphene oxide supported Pt nanoparticle hydrogel(PtNP/rGO-MWCNT),in which only ascorbic acid was used as the reductant for one-pot reduction of both GO and chloroplatinic acid.The hydrogel can be directly used as a flexible binder-free catalytic electrode to achieve high performance of HER.Compared to conventional strategies,the current strategy not only significantly reduces the Pt loading to 3.48 wt%,simplifies the synthesis process,but also eliminates the use of any polymer binders,thus decreasing the series resistance and improving catalytic activity.An overpotential of only 11 mV was achieved on as-prepared PtNP/rGO-MWCNT to drive a geometrical current density of 10 mA/cm2 in0.5 mol/L H2 SO4,with its catalytic activity being kept over 15 h.In acidic medium,the HER activity of the PtNP/rGO-MWCNT catalyst exceeds most of the reported Pt-based electro-catalysts and is 3-fold higher than that obtained on commercial Pt/C electrode. | Dan Zhou Bei Jiang Rui Yang Xiandeng Hou Chengbin Zheng | 2020 | Chinese Chemical Letters2020,31,6: | 3 |
| 6 | Biodegradable nanocarriers for small interfering ribonucleic acid (siRNA) co-delivery strategy increase the chemosensitivity of pancreatic cancer cells to gemcitabine显示文摘Ribonucleic acid (RNA) interference (RNAi) therapies are promising cancertreatment modalities that can specifically target abnormal proto-oncogenes, thusimproving the therapeutic effect. For the treatment of pancreatic cancer, targetingone mutant proto-oncogene by RNAi usually does not yield the desired therapeuticefficiency. Both K-ras gene mutations and Notch1 overexpression are commonsymptoms in pancreatic cancer patients, and play a crucial role in pancreaticcancer cell drug resistance. In this study, biodegradable charged polyester-basedvectors (BCPVs) were synthesized for the co-delivery of K-ras and Notch1 smallinterfering ribonucleic acid (siRNA) into MiaPaCa-2 cells (pancreatic cancercell line) to overcome drug resistance to gemcitabine (GEM), a first-line chem-otherapeutic drug used in the clinic. BCPVs could effectively absorb negative siRNAto form a capsule-like structure, prevent siRNA from nuclease digestion in theserum, and promote effective siRNA cell internalization and endosomal escape.Through K-ras and Notch1 gene silencing in MiaPaCa-2 cells, BCPV-siRNAKsiRNAN^tcm nanocomplexes effectively reversed the epithelia-mesenchymaltransition (EMT) in MiaPaCa-2 cells, thereby greatly enhancing the sensitivity ofMiaPaCa-2 cells to GEM. MiaPaCa-2 cell proliferation, migration, and invasionwere effectively inhibited, and cell apoptosis was also significantly enhanced bythe synergistic antitumor effect of BCPV-siRNAK siRNAN^t~l nanocomplexesand GEM. These results suggest that this combination RNAi therapy can be usedto improve cancer cell sensitivity to chemotherapeutic drugs. | 7 | Mercury in alpine fish from four rivers in the Tibetan Plateau显示文摘As a global pollutant, high levels of mercury(Hg) have been found in remote ecosystem due to the long range atmospheric transport. In this study, a total of 60 fish samples were collected from four rivers across the Tibetan Plateau to study the accumulation of Hg in remote and high-altitude aquatic environment. The total Hg(THg) and methylmercury(MeHg) in fish muscles ranged from 11 to 2097 ng/g dry weight(dw)(average: 819 ng/g dw)and from 14 to 1960 ng/g dw(average: 756 ng/g dw), respectively. Significantly positive linear relationships were observed between the THg(r = 0.591, p < 0.01, n = 36) and MeHg concentrations(r = 0.473, p < 0.01, n = 36) with the trophic level of fish from Lhasa River,suggesting trophic transfer and biomagnification of Hg in this aquatic ecosystem. Moreover,the THg levels in fish had significantly positive correlations with the length(r = 0.316,p < 0.05, n = 60) and weight(r = 0.271, p < 0.05, n = 60) of fish. The high levels of Hg were attributed to the slow growth and long lifespan of the fish under this sterile and cold environment. | 9 | Conservation and characterization of unique porcine interstitial telomeric sequences显示文摘Telomeres are composed of TTAGGG repeats and located at the ends of chromosomes. Telomeres protect chromosomes from instability in mammals, including mice and humans. Repetitive TTAGGG sequences are also found at intrachromosomal sites, where they are named as interstitial telomeric sequences (ITSs). Aberrant ITSs are implicated in chromosomal instability and found in cancer cells. Interestingly, in pigs, vertebrate telomere sequences TTAGGG (vITSs) are also localized at the centromeric region of chromosome 6, in addition to the end of all chromosomes. Surprisingly, we found that botanic telomere sequences, TTTAGGG (bITSs), also localize with vITSs at the centromeric regions of pig chromosome 6 using telomere fluorescence in situ hybridization (FISH) and by comparisons between several species. Furthermore, the average lengths of vITSs are highly correlated with those of the terminal telomeres (TTS). Also, pig ITSs show a high incidence of telomere doublets, suggesting that pig ITSs might be unstable and dynamic. Together, our results show that pig cells maintain the conserved telomere sequences that are found at the ITSs from of plants and other vertebrates. XYJ6, a newly isolated bacterial strain for biodegrading aniline, was investigated. It showed that biodegradation rate of aniline was increased with the augment of protein concentration in cell-free extract of Delftia sp. XYJ6. The adsorption amount of aniline by multi-walled carbon nanotubes (MWCNTs) was slightly higher than that by single-walled carbon nanotubes (SWCNTs), however the adsorption amount of protein of Delftia sp. XYJ6 by MWCNTs was lower than that by SWCNTs. Much more amount of aniline could be removed by CE of Delftia sp. XYJ6 in the presence of SWCNTs than MWCNTs, which indicated that an efficient reaction between aniline and enzymes of Delftia sp. XYJ6 on the surface of SWCNTs played a key role in the rapid enzymatic biodegradation of aniline. | 18 | Reduction of mercury(Ⅱ)by electrons contained in carbon dots:An environmentally friendly cold vapor generation for mercury analysis显示文摘In this work,the reduction of mercury ions(Hg2+)to elemental mercury(Hg0)was easily achieved using highly reductive carbon dots(r-CDs),which synthesized from sucrose by a simple and cost-effective method.After a careful mechanistic study,the reduction was probably accomplished with the large numbers of electrons contained in r-CDs rather than the oxidation of its functional groups.Additionally,a3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay showed that the r-CDs were nontoxic to wildlife and human beings.Consequently,the r-CDs were used as an alternative to toxic reductants(SnCl2 or NaBH4)for the sensitive and in situ determination of mercury by cold vapor gene ration(CVG)coupled to a miniature point discharge optical emission spectrometer(μPD-OES).Limit of detection of 0.05μg/L was obtained for Hg2+,with relative standard deviation(RSD)less than 5.4%at a concentration of 5μg/L.The accuracy of r-CDs induced CVG-μPD-OES was validated by the determination of mercury in a certified reference material(DOLT-5,dogfish liver)and five natural water samples collected from different rivers and lakes in Chengdu City,Since r-CDs are nontoxic and prepared from abundant and inexpensive sucrose,the r-CDs induced CVG-μPD-OES retains the great potential for the inexpensive and enviro nmentally friendly field analysis of mercury in natural water.The accuracy of the proposed method was validated by the analysis of a certified reference material and several water samples with satisfactory results. | Tao Chen Yao Lin Haochen Li Rui Yang Xiandeng Hou Baozhan Zheng Chengbin Zheng | 2020 | Chinese Chemical Letters2020,31,10: | 0 |
| 19 | Inhibition of MALT1 paracaspase activity improves lesion recovery following spinal cord injury显示文摘Spinal cord injury(SCI) is a devastating traumatic injury that causes persistent, severe motor and sensory dysfunction. Immune responses are involved in functional recovery after SCI. Mucosa-associated lymphoid tissue lymphoma translocation 1(MALT1) has been shown to regulate the survival and differentiation of immune cells and to play a critical role in many diseases, but its function in lesion recovery after SCI remains unclear. In this paper, we generated KI(knock in) mice with a point mutation(C472 G) in the active center of MALT1 and found that the KI mice exhibited improved functional recovery after SCI.Fewer macrophages were recruited to the injury site in KI mice and these macrophages differentiated into anti-inflammatory macrophages. Moreover, macrophages from KI mice exhibited reduced phosphorylation of p65, which in turn resulted in decreased SOCS3 expression and increased pSTAT6 levels.Similar results were obtained upon inhibition of MALT1 paracaspase with the small molecule inhibitor‘‘MI-2' or the more specific inhibitor ‘‘MLT-827'. In patients with SCI, peripheral blood mononuclear cells(PBMC) displayed increased MALT1 paracaspase. Human macrophages showed reduced proinflammatory and increased anti-inflammatory characteristics following the inhibition of MALT1 paracaspase. | 20 | Hallmark guided identification and characterization of a novel immune-relevant signature for prognostication of recurrence in stageⅠ-Ⅲlung adenocarcinoma显示文摘The high risk of postoperative mortality in lung adenocarcinoma(LUAD)patients is principally driven by cancer recurrence and low response rates to adjuvant treatment.Here,A combined cohort containing 1,026 stageⅠ-Ⅲpatients was divided into the learning(n Z 678)and validation datasets(n Z 348).The former was used to establish a 16-mRNA risk signature for recurrence prediction with multiple statistical algorithms,which was verified in the valida-tion set.Univariate and multivariate analyses confirmed it as an independent indicator for both recurrence-free survival(RFS)and overall survival(OS).Distinct molecular characteristics between the two groups including genomic alterations,and hallmark pathways were compre-hensively analyzed.Remarkably,the classifier was tightly linked to immune infiltrations,high-lighting the critical role of immune surveillance in prolonging survival for LUAD.Moreover,the classifier was a valuable predictor for therapeutic responses in patients,and the low-risk group was more likely to yield clinical benefits from immunotherapy.A transcription factor regulato-ry proteineprotein interaction network(TF-PPI-network)was constructed via weighted gene co-expression network analysis(WGCNA)concerning the hub genes of the signature.The con-structed multidimensional nomogram dramatically increased the predictive accuracy.There-fore,our signature provides a forceful basis for individualized LUAD management with promising potential implications. | Yongqiang Zhang Zhao Yang Yuqin Tang Chengbin Guo Danni Lin d Linling Cheng Xun Hu Kang Zhang Gen Li | 2023 | Genes & Diseases2023,10,4: | 0 |
