3Electroluminescence performances of 1,1-bis(4-(N,N-dimethylamino)phenyl)-2,3,4,5-tetraphenylsilole based polymers in three cathode architectures显示文摘A new silole monomer with two 4-(N,N-dimethylamino)phenyl substitutions on silicon atom as designed and synthesized.Three copolymers PF-N-HPS1,PF-N-HPS10 and PF-N-HPS20 were then obtained by copolymerizations of 2,7-fluorene derivatives with the silole monomer at feed ratios of 1%,10%,and 20%.Their UV-vis absorption,electrochemical,photoluminescent,and electroluminescent (EL) properties were investigated.PF-N-HPS possessed HOMO levels of 5.25-5.58 eV,and showed green emissions.Using PF-N-HPS as the emissive layer,three different polymer light-emitting diodes were fabricated as device A with ITO/PEDOT/PF-N-HPS/Al,device B with ITO/PEDOT/PF-N-HPS/Ba/Al,and device C with ITO/PEDOT/PF-N-HPS/TPBI/Ba/Al.For the device A,PF-N-HPS only showed very low EL efficiency of 0.06-0.33 cd/A,indicating that the Al cathode could not inject electron efficiently to the emissive polymers containing the 4-(N,N-dimethylamino)phenyl groups.For the device B,low work function Ba supplied better electron injections,and the EL efficiency could be improved to 0.85-1.44 cd/A.TPBI with a deep HOMO level of 6.2 eV could enhance electron transport and hole blocking.Thus modified recombinations and largely elevated EL efficiency of 4.56-7.96 cd/A were achieved for the device C.The separation of the emissive layer and metal cathode with the TPBI layer may also suppress exciton quenching at the cathode interface.LIU ZhiTian HU SuJun ZHANG LinHua CHEN JunWu PENG JunBiao CAO Yong 2013Science China Chemistry2013,56,8:2

6IL-33 drives the antitumor effects of dendritic cells via the induction of Tc9 cells显示文摘Dendritic cell(DC)tumor vaccines exert their antitumor effects through the induction of effector T cells.We recently identified Tc9 cells as a new potent antitumor effector T cell subset.However,approaches to direct DCs to preferably prime antitumor Tc9 cells should be further exploited.Here,we demonstrate that the addition of interleukin(IL)-33 potently promotes the induction of Tc9 cells by DCs in vitro and in vivo.IL-33 treatment also drives the cytotoxic activities of DC-induced Tc9 cells.Notably,IL-33 treatment enhances cell survival and proliferation of DC-primed CD8+T cells.More importantly,the addition of IL-33 during in vitro priming of tumor-specific Tc9 cells by DCs increases the antitumor capability of Tc9 cells.Mechanistic studies demonstrated that IL-33 treatment inhibits exhaustive CD8+T cell differentiation by inhibiting PD-1 and 2B4 expression and increasing IL-2 and CD127(IL-7 receptor-α,IL-7Rα)expression in CD8+T cells.Finally,the addition of IL-33 further promotes the therapeutic efficacy of DC-based tumor vaccines in the OT-I mouse model.Our study demonstrates the important role of IL-33 in DC-induced Tc9 cell differentiation and antitumor immunity and may have important clinical implications.Ning Liu Yuxue Jiang Jintong Chen He Nan Yinghua Zhao Xiao Chu Alison Wang Dongjiao Wang Tianxue Qin Sujun Gao Ying Yue Siqing Wang 2019Cellular & Molecular Immunology2019,16,7:2

13Comparative risk of acute kidney injury among cancer patients treated with immune checkpoint inhibitors显示文摘With the development and introduction of immune checkpoint inhibitors(ICIs)in cancer patients,immune-related side effects have increasingly attracted attention.However,the risks of immune-related renal toxicity are poorly characterized.In this study,we performed a network meta-analysis(NMA)of ICI-related randomized clinical trials(RCTs)to elucidate the comparative risk of acute kidney injury(AKI)in cancer patients receiving different ICIs.We also sought to identify other factors potentially affecting the risk of AKI.PubMed and EMBASE were searched for peer-reviewed trial reports published between January 2000 and May 2021.Eligible studies were RCTs studying ICIs in cancer patients and reporting AKI data.We performed a frequentist NMA to evaluate the risk ratios for grade 1-5 and grade 3-5 AKI between the treatment groups.We also assessed the absolute incidence of AKI in the ICI-containing arm using traditional direct meta-analysis.Once significant heterogeneity was detected in a traditional direct meta-analysis,multivariable meta-regression analysis was applied to identify factors that significantly affected the absolute incidence of AKI.A total of 85 RCTs were included in this study.In the NMA for the risk of grade 1-5 and 3-5 AKI,ipilimumab showed a significantly higher risk than avelumab and durvalumab,whereas 1 mg/kg nivolumab plus 3 mg/kg ipilimumab(N1I3)showed a significantly higher risk than other groups.In terms of treatment ranking,durvalumab±low-dose tremelimumab and avelumab were consistently among the top three safest treatments for grade 1-5 or 3-5 AKI,whereas N1I3,ipilimumab and tremelimumab were consistently among the top three treatments with the highest risk for grade 1-5 or 3-5 AKI.Compared with other cancers,renal cell carcinoma and urothelial carcinoma showed a significantly higher risk of AKI.The incidence of AKI was significantly higher with ICI+chemotherapy than with ICI monotherapy.In this NMA involving largescale up-to-date ICI trials,we demonstrated the comparative safety of existing ICI drugs for grade 1-5 and grade 3-5 AKI.Based on data from the ICI arms of these trials,we also revealed several potential risk factors for immune-related AKI,including tumor type and treatment paradigm.Fei Liu ZixianWang Xiaofan Li Zhen Zhang Yue Yang Junquan Chen Dinghua Chen Lingling Wu Xiangyu Liu Sujun Han Fangming Wang Wasilijiang Wahafu Yibo Gao Shancheng Ren Nianzeng Xing Guangyan Cai Xiangmei Chen 2023Cancer Communications2023,43,2:1
14Direct-acting Antiviral-induced Transient Recovery of NK Cells in Early-stage Treatment of Chronic Hepatitis C Patients显示文摘Background and Aims:The rapid clearance of hepatitis C virus induced by direct-acting antivirals(DAAs)affects natural killer(NK)cells,but the reported results are not consistent,and the relative mechanism was unclear.This study focused on the dynamic changes of NK cells during and after DAA treatment and analyzed the reasons.Meth-ods:Peripheral blood from 35 chronic hepatitis C patients who were treated with DAAs were collected at baseline and weeks 1,2,4,12,and post-treatment week-12.The fre-quency,subset,and phenotype of NK cells were assayed by flow cytometry.Lactate dehydrogenase assays were used to evaluate the cytotoxicity of NK cells.Cytokine concentra-tions were measured with Luminex kits.Results:All pa-tients achieved a sustained viral response(SVR),and the NK cell frequencies were not changed significantly during DAA therapy.However,the cytotoxicity of NK cells recov-ered significantly early in week 1,and then continuously decreased below normal levels.The changes of genotypes including NKp30+,NKp46+,and NKG2A+NK cells were par-allel to NK function.The subset of CD56dim NK cells con-tinuously increased and did not return to normal even at 12 weeks after treatment.Interleukin(IL)-2,IL10,IL15,interferon-gamma,and tumor necrosis factor-alpha all in-creased after week 4,peaked at the end of therapy,and then exhibited varying degrees of reduction with time.Con-clusions:DAA treatment led to transient functional recov-ery of NK cells in the early stage of treatment,and then continuously decreased to below normal levels.Alterations of NK subsets,phenotypes,and the microenvironment may be involved in the changes.Xiaohui Zhang Yingying Jiang Shaobin Li Dandan Bian Mei Liu Ming Kong Yu Chen Zhongping Duan Sujun Zheng 2022Journal of Clinical and Translational Hepatology2022,10,6:0
15Performance of Hepatitis Delta Virus(HDV)RNA Testing for the Diagnosis of Active HDV Infection:Systematic Review and Meta-analysis显示文摘Background and Aims:Hepatitis delta virus(HDV)is a defective virus and causes severe liver disease.Several HDV RNA assays have been developed,however the diagnostic efficacy remains unclear.This systematic review and metaanalysis aims to evaluate the diagnostic accuracy of HDV RNA assays to aid in the diagnosis of active hepatitis D.Methods:The PubMed,Embase,and Cochrane Library databases were systematically searched from the beginning to June 31,2022.Information on the characteristics of the literature and data on sensitivity,specificity,and area under curve(AUC)of the receiver operating characteristic(ROC)were extracted.Stata 14.0 was used for meta-analysis of the combined sensitivity,specificity,positive likelihood ratio,and negative likelihood ratio.Results:A total of 10 studies were included in the meta-analysis.The summary sensitivity,specificity,positive likelihood ratio,negative likelihood ratio,and diagnostic odds ratio of HDV RNA assays for HDV diagnosis were 0.92(95%CI:0.87-0.95),0.90(95%CI:0.86-0.93),7.74(95%CI:5.31-11.29),0.10(95%CI:0.06-0.18)and 99.90(95%CI:47.08-211.99),respectively.The AUC of the pooled ROC curve was 0.95(95%CI:0.92-0.96).Conclusions:The results show that HDV RNA assays had high diagnostic performance.However,that is limited by the number and quality of studies.Standard protocols for the development of assays by manufacturers and larger studies on the use of the assays are needed.Sisi Chen Xiangying Zhang Ling Xu Yuan Tian Zihao Fan Yaling Cao Zhenzhen Pan Yao Gao Sujun Zheng Zhongping Duan Mei Liu Feng Ren 2023Journal of Clinical and Translational Hepatology2023,11,6:0
16UGT1A1-related Bilirubin Encephalopathy/Kernicterus in Adults显示文摘Background and Aims:Bilirubin encephalopathy/kernicterus is very rare in adults.This study is aimed to investigate the clinical manifestations and genetic features of two patients with UGT1A1-related kernicterus.Methods:Sanger sequencing analysis was performed to identify UGT1A1 gene mutations in the patients and their families.Bioinformatics analysis was used to predict the potential functional effects of novel missense mutations.Clinical manifestations and biochemical parameters were collected and analyzed.Results:Two patients with Crigler-Najjar syndrome type II(CNS2)developed kernicterus in adulthood.Sanger sequencing identified a compound heterozygous mutation in the UGT1A1 gene in patient 1,which was inherited from his mother(G71R)and his father(c.-3279T>G;S191F).Patient 2 carried three heterozygous mutations,namely G71R,R209W and M391K;among which,the M391K mutation has not been reported before.Multiple prediction software showed that the M391K mutation was pathogenic.Symptoms were relieved in the two patients after phenobarbital and artificial liver support treatment.Patient 1 also underwent liver transplantation.Conclusions:Adults with CNS2 are at risk for kernicterus.Phenobarbital treatment is beneficial for maintaining bilirubin levels and preventing kernicterus.Jie Bai Lu Li Hui Liu Shuang Liu Li Bai Wenyan Song Yu Chen Sujun Zheng Zhongping Duan 2021Journal of Clinical and Translational Hepatology2021,9,2:0
17Influence Mechanism of the Trabecular and Chamfer Radii on the Three-point Bending Properties of Trabecular Beetle Elytron Plates显示文摘To improve the mechanical properties of Trabecular Beetle Elytron Plates(TBEPs,a type of biomimetic sandwich structure inspired by the beetle elytron)under transverse loads,three-point bending tests are performed to investigate the influence of the trabecular and chamfer radii of the core structure on the mechanical performance of TBEPs manufactured by 3D printing technology.The results show that the three-point bending performance of TBEPs can be improved by setting reasonable trabecular and chamfer radii;however,excessive increases in these radii can cause a decline in the mechanical performance.For the reason,these two structural parameters can enhance the deformation stiffness of the whole structure and the connection property between the core and skin,which is also the mechanical reason why Prosopocoilus inclinatus beetle elytra have thick,short trabeculae with a large chamfer radius.However,when these radii increase to a certain extent,the cracks are ultimately controlled between two adjacent trabeculae,and the failure of the plate is determined by the skin rather than the core structure.Therefore,this study suggests a reasonable range for trabecular and chamfer radii,and indicates that TBEPs are better suited for engineering applications that have high compression requirements and general bending requirements.Xiaoming Zhang Xindi Yu Jinxiang Chen Caiqi Zhao Sujun Guan Yaqin Fu 2021Journal of Bionic Engineering2021,18,2:0
18Mesenchymal stem cell exosomes inhibit nucleus pulposus cell apoptosis via the miR-125b-5p/TRAF6/NF-κB pathway axis显示文摘Intervertebral disc degeneration(IVDD)is the pathological basis of a range of degenerative spinal diseases and is the primary cause of lower back pain.Mesenchymal stem cell(MSC)transplantation inhibits IVDD progression.However,the specific mechanisms that underlie these effects remain unclear.In this study,candidate microRNAs(miRNAs)are screened using bioinformatics and high-throughput sequencing.TNF-αis used to induce nucleus pulposus cell(NPC)degeneration.MSC-derived exosomes(MSC-exosomes)are obtained using high-speed centrifugation and identified by transmission electron microscopy(TEM),nanoparticle tracking analysis(NTA)and western blot analysis.Cell viability is determined by CCK-8 assay.Flow cytometry and TUNEL assays are used to detect cell apoptosis.The expression levels of miR-125b-5p are detected by RT-qPCR,and a dual-luciferase gene reporter assay confirms the downstream target genes of miR-125b-5p.Protein expression is determined by western blot analysis.Rat models are used to validate the function of miR-125b-5p in MSC-exosomes.The results show that miR-125b-5p is expressed at low levels in degenerated disc tissues compared with that in normal disc tissues;however,it is highly expressed in MSC-exosomes.Furthermore,MSC-exosomes are efficiently taken up by NPCs while miR-125b-5p is delivered into NPCs;thus,MSC-exosomes act as inhibitors of apoptosis in NPCs.Overexpression of miR-125b-5p downregulates TRAF6 expression and inhibits NF-κB activation.However,TRAF6 overexpression reverses these effects of miR-125b-5p.We demonstrate that MSC-exosomes attenuate IVDD in vivo by delivering miR-125b-5p.MSC-exosomes can deliver miR-125b-5p to target TRAF6,inhibit NF-κB activation,and attenuate the progression of IVDD.Yang Duan Cheng Yu Wenhao Kuang Jianjun Li Sujun Qiu Songjia Ni Zhong Chen 2023Acta Biochimica et Biophysica Sinica2023,55,12:0

20Glioblastoma cell-derived exosomes functionalized with peptides as efficient nanocarriers for synergistic chemotherapy of glioblastoma with improved biosafety显示文摘Glioblastoma(GBM)has been regarded as one of the most deadly and challenging cancers to treat with extremely poor prognosis.The limited efficacy of current chemotherapies might be attributed to the presence of glioma stem cells(GSCs)as well as the difficulties in passing through the blood–brain barrier(BBB)and targeting tumor cells.Tumor-derived exosomes are emerging as novel and promising drug delivery systems.However,great concerns regarding the biosafety and BBB penetrability remain to be addressed.Herein,we have developed a simple and feasible strategy to engineer GBM cell-derived exosomes with improved biosafety termed“Exo@TDPs”to deliver the cargos of chemotherapeutic agents temozolomide(TMZ)and doxorubicin(DOX)into GBM tissues.Exo@TDPs decorated with angiopep-2(Ang-2)and CD133-targeted peptides improve the capacity to penetrate the BBB and target tumor cells.Both in vitro and in vivo studies demonstrate that Exo@TDPs can cross the BBB,target GBM cells,penetrate into deep tumor parenchyma,and release the therapeutic cargos effectively.Synergistic delivery of TMZ and DOX by Exo@TDPs exerts therapeutic effects to suppress the tumor growth and prolong the survival time of orthotopic syngeneic mouse GBM models.These findings suggest that our developed Exo@TDPs loaded with chemotherapeutic drugs may bring new possibilities for the application of tumor cell-derived exosomes for brain tumor treatment.Ying Zhou Long Wang Lufei Chen Wei Wu Zhimin Yang Yuanzhuo Wang Anqi Wang Sujun Jiang Xuzhen Qin Zucheng Ye Zhiyuan Hu Zihua Wang 2023Nano Research2023,16,12:0
