1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Simvastatin inhibits apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax显示文摘BACKGROUND: Many studies have showed that apoptosis of endothelial cells plays a curial role in the progress of sepsis. But the role of simvastatin in apoptosis of endothelial cells induced by sepsis is not clear. The present study aimed to investigate the role of simvastatin in apoptosis of endothelial cells induced by sepsis and its mechanism.METHODS: Human umbilical vein endothelial cells(HUVECs) were randomly divided into three groups: control group, sepsis serum intervention group(sepsis group) and simvastatin+sepsis serum intervention group(simvastatin group). After 24-hour incubation with corresponding culture medium, the relative growth rate of HUVECS in different groups was detected by MTT assay; the apoptosis of HUVECs was detected by Hoechst33258 assay and fl ow cytometry; and the expression of the Bcl-2 and Bax genes of HUVECs was detected by PCR.RESULTS: Compared with the sepsis group, HUVECs in the simvastatin group had a higher relative growth rate. Apoptotic HUVECs decreased significantly in the simvastatin group in comparison with the sepsis group. Expression of the Bcl-2 gene in HUVECs decreased obviously, but the expression of the Bax gene increased obviously after 24-hour incubation with sepsis serum; however, the expression of the Bcl-2 and Bax genes was just the opposite in the simvastatin group.CONCLUSIONS: Our study suggests that simvastatin can inhibit apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax. 3First observation of tropospheric nitrogen dioxide from the Environmental Trace Gases Monitoring Instrument onboard the GaoFen-5 satellite显示文摘The Environmental Trace Gases Monitoring Instrument(EMI)is the first Chinese satellite-borne UV–Vis spectrometer aiming to measure the distribution of atmospheric trace gases on a global scale.The EMI instrument onboard the GaoFen-5 satellite was launched on 9 May 2018.In this paper,we present the tropospheric nitrogen dioxide(NO2)vertical column density(VCD)retrieval algorithm dedicated to EMI measurement.We report the first successful retrieval of tropospheric NO_(2) VCD from the EMI instrument.Our retrieval improved the original EMI NO_(2) prototype algorithm by modifying the settings of the spectral fit and air mass factor calculations to account for the on-orbit instrumental performance changes.The retrieved EMI NO_(2) VCDs generally show good spatiotemporal agreement with the satellite-borne Ozone Monitoring Instrument and TROPOspheric Monitoring Instrument(correlation coefficient R of ~0.9,bias<50%).A comparison with ground-based MAX-DOAS(Multi-Axis Differential Optical Absorption Spectroscopy)observations also shows good correlation with an R of 0.82.The results indicate that the EMI NO_(2) retrieval algorithm derives reliable and precise results,and this algorithm can feasibly produce stable operational products that can contribute to global air pollution monitoring.Chengxin Zhang Cheng Liu Ka Lok Chan Qihou Hu Haoran Liu Bo Li Chengzhi Xing Wei Tan Haijin Zhou Fuqi Si Jianguo Liu 2020Light(Science & Applications)2020,9,1:11

5Recent progress on MOF-derived carbon materials for energy storage显示文摘Metal-organic frameworks(MOFs)are of quite a significance in the field of inorganic-organic hybrid crystals.Especially,MOFs have attracted increasing attention in recent years due to their large specific surface area,desirable electrical conductivity,controllable porosity,tunable geometric structure,and excellent thermal/chemical stability.Some recent studies have shown that carbon materials prepared by MOFs as precursors can retain the privileged structure of MOFs,such as large specific surface area and porous structure and,in contrast,realize in situ doping with heteroatoms(eg,N,S,P,and B).Moreover,by selecting appropriate MOF precursors,the composition and morphology of the carbon products can be easily adjusted.These remarkable structural advantages enable the great potential of MOF-derived carbon as high-performance energy materials,which to date have been applied in the fields of energy storage and conversion systems.In this review,we summarize the latest advances in MOF-derived carbon materials for energy storage applications.We first introduce the compositions,structures,and synthesis methods of MOF-derived carbon materials,and then discuss their applications and potentials in energy storage systems,including rechargeable lithium/sodium-ion batteries,lithium-sulfur batteries,supercapacitors,and so forth,in detail.Finally,we put forward our own perspectives on the future development of MOF-derived carbon materials.Jincan Ren Yalan Huang He Zhu Binghao Zhang Hekang Zhu Shenghui Shen Guoqiang Tan Feng Wu Hao He Si Lan Xinhui Xia Qi Liu 2020Carbon Energy2020,2,2:8
6Modulation of expression of P16 and Her2 in rat breast tissues of mammary hyperplasia model by external use of Rupifang Extract显示文摘OBJECTIVE:To observe the effect of Rupifang Extract in external use on expression of proto-oncogenes her2 and tumor suppression genes p16 in rat breast tissues of mammary hyperplasia model.To explore the mechanisms of Rupifang Extract in external use for preventing and treating mammary hyperplasia.METHODS:Thirty virginal female Wistar rats were randomized into 5 groups,6 in each,A:blank control group;B:model group;C:the low dose group of Rupifang;D:the middle dose group of Rupifang;and E:The high dose group of Rupifang.The mammary hyperplasia rat models were produced by injecting estradiol benzoate and progesterone and irritating by tail nipping.Drug intervention was also launched during the model formation.After 30 days,the expression of her2 and p16 in breast tissues of rats in each group were detected by the SP immunohistochemical method.RESULTS:Compared with Blank control group,the expression of her2 in breast tissues in Model group was higher,and the expression of p16 was lower(P<0.05 or P<0.01).After intervention with Rupifang Extract,compared with Model group,the expression of her2 in breast tissues in Rupifang groups was lower,and the expression of p16 higher(P<0.05 or P<0.01).CONCLUSION:The mechanisms of Rupifang Extract in external application for preventing and treating mammary hyperplasia may be reducing the expression of proto-oncogenes her2 and increasing the expression of tumor suppression genes p16.Guijuan Zhang Dehui Li Hui Guo Rui Liao Bizhu Tan Suyi Zhang Yubin Liu Min Ma Xiaoting Zeng Zhuolong Peng Si Huang Yinghui Zheng Yi Ma 2012Journal of Traditional Chinese Medicine2012,32,4:6
7Dihydroartemisinin suppresses renal fibrosis in mice by inhibiting DNA-methyltransferase 1 and increasing Klotho显示文摘Renal fibrosis is an unavoidable end result of all forms of progressive chronic kidney diseases(CKD).Discovery of efficacious drugs against renal fibrosis is in crucial need.In a preliminary study we found that a derivative of artemisinin,dihydroartemisinin(DHA),exerted strong renoprotection,and reversed renal fibrosis in adenine-induced CKD mouse model.In this study we investigated the anti-fibrotic mechanisms of DHA,particularly its specific target in renal cells.Renal fibrosis was induced in mice by unilateral ureteral obstruction(UUO)or oral administration of adenine(80 mg·kg^(-1)),the mice received DHA(30 mg·kg^(-1)·d^(-1),i.g.)for 14 or 21 days,respectively.We showed that DHA administration markedly attenuated the inflammation and fibrotic responses in the kidneys and significantly improved the renal function in both the renal fibrosis mouse models.In adenine-treated mice,DHA was more effective than 5-azacytidine against renal fibrosis.The anti-fibrotic effects of DHA were also observed in TGF-β1-treated HK-2 cells.In order to determine the target protein of DHA,we conducted pull-down technology coupled with shotgun proteomics using a small-molecule probe based on the structure of DHA(biotin-DHA).As a results,DNA methyltransferase 1(DNMT1)was identified as the anti-fibrotic target of DHA in 3 different types of renal cell lines(HK-2,HEK293 and 3T3).We demonstrated that DHA directly bound to Asn 1529 and Thr 1528 of DNMT1 with a Kd value of 8.18μM.In primary mouse renal tubular cells,we showed that DHA(10μM)promoted DNMT1 degradation via the ubiquitin–proteasome pathway.DHA-reduced DNMT1 expression effectively reversed Klotho promoter hypermethylation,which led to the reversal of Klotho protein loss in the kidney of UUO mice.This subsequently resulted in inhibition of the Wnt/β-catenin and TGF-β/Smad signaling pathways and consequently conferred renoprotection in the animals.Knockdown of Klotho abolished the renoprotective effect of DHA in UUO mice.Our study reveals a novel pharmacological activity for DHA,i.e.,renoprotection.DHA exhibits this effect by targeting DNMT1 to reverse Klotho repression.This study provides an evidence for the possible clinical application of DHA in the treatment of renal fibrosis.Wei Zhou Min-min Chen Hui-ling Liu Zi-lin Si Wen-hui Wu Hong Jiang Lin-xiao Wang Nosratola D.Vaziri Xiao-fei An Ke Su Cheng Chen Ning-hua Tan Zhi-hao Zhang 2022Acta Pharmacologica Sinica2022,43,10:4
8Numerical study on the evolution of the shock-accelerated SF_6 interface:Influence of the interface shape显示文摘The early phases of the shock interaction process on two-dimensional interfaces with different shapes are numerically investigated in this study,which are closely related to the shock refraction and reflection,vorticity production and transport.The numerical method employs an adaptive unstructured quadrilateral mesh,which can capture the wave pattern and interface evolution very well.Simulations are carried out under the conditions of an incident shock Mach number of 1.2 and the light/heavy (air/SF 6) interface.Five different shapes are considered in the simulations:rectangle,ellipse,diamond and two kinds of triangle.The results show that the interfacial shapes can influence the wave patterns particularly on the shape and evolution of refracted shock waves.The generation and the distribution of vorticity on the interfaces with five different shapes also have dissimilarities.The circulation deposition on five interfaces is quantitatively investigated and compared with theoretical model.A good agreement is found between the numerical results and the predictions by the theoretical model.Some characteristic scales of the interface are tracked.Under the influence of nonlinear-acoustic effect and vorticity effect,the interfaces present different evolution modes.FAN MeiRu ZHAI ZhiGang SI Ting LUO XiSheng ZOU LiYong TAN DuoWang 2012Science China(Physics,Mechanics & Astronomy)2012,55,2:4
9Mechanism of persistent hyperalgesia in neuropathic pain caused by chronic constriction injury显示文摘Transmembrane member 16 A(TMEM16 A) is involved in many physiological functions, such as epithelial secretion, sensory conduction, nociception, control of neuronal excitability, and regulation of smooth muscle contraction, and may be important in peripheral pain transmission. To explore the role of TMEM16 A in the persistent hyperalgesia that results from chronic constriction injury-induced neuropathic pain, a rat model of the condition was established by ligating the left sciatic nerve. A TMEM16 A selective antagonist(10 μg T16 Ainh-A01) was intrathecally injected at L5–6. For measurement of thermal hyperalgesia, the drug was administered once at 14 days and thermal withdrawal latency was recorded with an analgesia meter. For measurement of other indexes, the drug was administered at 12 days,once every 6 hours, totally five times. The measurements were performed at 14 days. Western blot assay was conducted to analyze TMEM16 A expression in the L4–6 dorsal root ganglion. Immunofluorescence staining was used to detect the immunoreactivity of TMEM16 A in the L4–6 dorsal root ganglion on the injured side. Patch clamp was used to detect electrophysiological changes in the neurons in the L4–6 dorsal root ganglion. Our results demonstrated that thermal withdrawal latency was shortened in the model rats compared with control rats.Additionally, TMEM16 A expression and the number of TMEM16 A positive cells in the L4–6 dorsal root ganglion were higher in the model rats, which induced excitation of the neurons in the L4–6 dorsal root ganglion. These findings were inhibited by T16 Ainh-A01 and confirm that TMEM16 A plays a key role in persistent chronic constriction injury-induced hyperalgesia. Thus, inhibiting TMEM16 A might be a novel pharmacological intervention for neuropathic pain. 13Durable easy-cleaning and antibacterial cotton fabrics using fluorine-free silane coupling agents and CuO nanoparticles显示文摘Multifunctional fabrics of high durability through a scalable and eco-friendly technique remains a great challenge hindering their commercialization.In this work,we report a facile synthesis technique for the fabrication of superhydrophobic antibacterial fabrics by employing fluorine-free silane coupling agents as cross-linkers for enhanced durability.Three silane cross-linkers,Aminoethylaminopropyltrimethoxysilane(AEAPTMS),Aminopropyltriethoxysilane(APTES),and Methacryloyloxypropyltrimethoxysilane(MPTMS),have been investigated.During the fabrication,a low surface energy polymer,polydimethylsiloxane(PDMS)was first deposited on cotton fabrics.Subsequently,antibacterial copper oxide(CuO)nanoparticles were anchored on the PDMS coated fabrics using the silane cross-linkers.The as-prepared fabrics displayed high superhydrophobicity and antibacterial performance with water contact angle(WCA)>153,water shedding angle(WSA)<5,and up to 99%antibacterial efficiency.Additionally,the as-prepared fabrics displayed high durability against abrasion,ultrasonic washing,and soaking in harsh chemical environments.The air permeability and flexibility of the fabric was not compromised after the coating.The above-reported technique is simple,cost-effective and holds tremendous potential for large-scale production of energy-saving clothing and healthcare products.Neha Agrawal Pearlie Sijia Low Jasmine Si Jia Tan Eileen Wen Mei Fong Yuekun Lai Zhong Chen 2020Nano Materials Science2020,2,3:2
14Expression and effect of sodium-potassium-chloride cotransporter on dorsal root ganglion neurons in a rat model of chronic constriction injury显示文摘Sodium-potassium-chloride cotransporter 1 (NKCC1) and potassium-chloride cotransporter 2 (KCC2) are associated with the transmission of peripheral pain.We investigated whether the increase of NKCC1 and KCC2 is associated with peripheral pain transmission in dorsal root ganglion neurons.To this aim,rats with persistent hyperalgesia were randomly divided into four groups.Rats in the control group received no treatment,and the rat sciatic nerve was only exposed in the sham group.Rats in the chronic constriction injury group were established into chronic constriction injury models by ligating sciatic nerve and rats were given bumetanide,an inhibitor of NKCC1,based on chronic constriction injury modeling in the chronic constriction injury + bumetanide group.In the experiment measuring thermal withdrawal latency,bumetanide (15 mg/kg) was intravenously administered.In the patch clamp experiment,bumetanide (10 μg/μL) and acutely isolated dorsal root ganglion neurons (on day 14) were incubated for 1 hour,or bumetanide (5 μg/μL) was intrathecally injected.The Hargreaves test was conducted to detect changes in thermal hyperalgesia in rats.We found that the thermal withdrawal latency of rats was significantly decreased on days 7,14,and 21 after model establishment.After intravenous injection of bumetanide,the reduction in thermal retraction latency caused by model establishment was significantly inhibited.Immunohistochemistry and western blot assay results revealed that the immune response and protein expression of NKCC1 in dorsal root ganglion neurons of the chronic constriction injury group increased significantly on days 7,14,and 21 after model establishment.No immune response or protein expression of KCC2 was observed in dorsal root ganglion neurons before and after model establishment.The Cl^– (chloride ion) fluorescent probe technique was used to evaluate the change of Cl^– concentration in dorsal root ganglion neurons of chronic constriction injury model rats.We found that the relative optical density of N-(ethoxycarbonylmethyl)-6-methoxyquinolinium bromide (a Cl^– fluorescent probe whose fluorescence Cenintensity decreases as Cl– concentration increases) in the dorsal root ganglion neurons of the chronic constriction injury group was significantly decreased on days 7 and 14 after model establishment.The whole-cell patch clamp technique revealed that the resting potential and action potential frequency of dorsal root ganglion neurons increased,and the threshold and rheobase of action potentials decreased in the chronic constriction injury group on day 14 after model establishment.After bumetanide administration,the above indicators were significantly suppressed.These results confirm that CCI can induce abnormal overexpression of NKCC1,thereby increasing the Cl^– concentration in dorsal root ganglion neurons;this then enhances the excitability of dorsal root ganglion neurons and ultimately promotes hyperalgesia and allodynia.In addition,bumetanide can achieve analgesic effects.All experiments were approved by the Institutional Ethics Review Board at the First Affiliated Hospital,College of Medicine,Shihezi University,China on February 22,2017 (approval No.A2017-169-01).Chao-Yang Tan Yan-Ping Wang Yuan-Yuan Han Bi-Han Lu Wei Ji Li-Cang Zhu Yang Wang Wen-Yan Shi Li-Ya Shan Liang Zhang Ke-Tao Ma Li Li Jun-Qiang Si 2020Neural Regeneration Research2020,15,5:2
