1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Dabrafenib,an inhibitor of RIP3 kinase-dependent necroptosis,reduces ischemic brain injury显示文摘Ischemic brain injury triggers neuronal cell death by apoptosis via caspase activation and by necroptosis through activation of the receptor-interacting protein kinases(RIPK) associated with the tumor necrosis factor-alpha(TNF-α)/death receptor. Recent evidence shows RIPK inhibitors are neuroprotective and alleviate ischemic brain injury in a number of animal models, however, most have not yet undergone clinical trials and safety in humans remains in question. Dabrafenib, originally identified as a B-raf inhibitor that is currently used to treat melanoma, was later revealed to be a potent RIPK3 inhibitor at micromolar concentrations. Here, we investigated whether Dabrafenib would show a similar neuroprotective effect in mice subjected to ischemic brain injury by photothrombosis. Dabrafenib administered intraperitoneally at 10 mg/kg one hour after photothrombosis-induced focal ischemic injury significantly reduced infarct lesion size in C57BL6 mice the following day, accompanied by a markedly attenuated upregulation of TNF-α. However, subsequent lower doses(5 mg/kg/day) failed to sustain this neuroprotective effect after 4 days. Dabrafenib bl ocked lipopolysaccharides-induced activation of TNF-α in bone marrow-derived macrophages, suggesting that Dabrafenib may attenuate TNF-α-induced necroptotic pathway after ischemic brain injury. 3Epithelial ovarian cancer:An overview显示文摘Ovarian cancer is the second most common gyneco-logical cancer and the leading cause of death in the United States. In this article we review the diagnosis and current management of epithelial ovarian cancer which accounts for over 95 percent of the ovarian malignancies. We will present various theories about the potential origin of ovarian malignancies. We will discuss the genetic anomalies and syndromes that may cause ovarian cancers with emphasis on Breast cancer type 1/2 mutations. The pathology and pathogenesis of ovarian carcinoma will also be presented. 4Anti-anxiety effect of Valeriana jatamansi Jones extract via regulation of the hypothalamus-pituitary-adrenal axis显示文摘BACKGROUND:Hypothalamus-pituitary-adrenal (HPA) axis dysfunction has been closely linked to anxiety.Previous studies have shown that Valeriana jatamansi Jones extract exhibits clear anxio-lytic effects, but it is unclear about the mechanism underlying regulation of the HPA axis dysfunction in these anxiolytic effects.OBJECTIVE:To observe the effects of Valeriana jatamansi Jones (Zhizhu Xiang) extract on HPA axis function in a rat model of anxiety, and to compare these effects with positive control estazolam.DESIGN, TIME AND SETTING:Randomized, controlled, animal experiment was performed at Chengdu University of Traditional Chinese Medicine, China, between February and September in 2006.MATERIALS:Estazolam was purchased from Shanghai Jiufu Pharmaceutical, China; Valeriana jatamansi Jones was purchased from the Lotus Pond Market for Chinese Herbal Medicine in Chengdu.It consisted of iridoids and flavonoid components.METHODS:A total of 72 Sprague Dawley rats, aged 2 months, were randomly assigned to 6 groups:low-, medium-, and high-dose Valeriana jatamansi Jones groups intragastrically injected with 0.3, 0.6, and 0.9 g/kg per day Valeriana jatamansi Jones extract, respectively; estazolam group intragastrically injected with 1.5 mg/kg per day estazolam; model and normal groups administered 5 mL physiological saline.Anxiety was established in all groups, except the normal group, through the use of elevated plus-maze test at 7 days following drug administration.MAIN OUTCOME MEASURES:Blood β-endorphin and corticosterone levels were determined using enzyme-linked immunosorbent assay following treatment with Valeriana jatamansi Jones extract.Expressions of HPA axis-related genes were measured by cDNA microarray.RESULTS:Blood β-endorphin and corticosterone levels were significantly greater in the model group than in the normal group.Compared with the model group, levels decreased with Valeriana jatamansi Jones extract or estazolam treatment, particularly in the low-dose Valeriana jatamansi Jones group (P<0.01).cDNA microarray results demonstrated that corticotropin-releasing hormone and Orexin, which are associated with HPA axis function, were differentially expressed; expression increased in the model group, but decreased in rats treated with Valeriana jatamansi Jones extract.CONCLUSION:Valeriana jatamansi Jones extract plays a role in regulating HPA axis function in a rat model of anxiety, and this effect was superior to estazolam.Zhiyong Yan Tian'e Zhang Ting Xiao Lingzhen Pan Jinzhi Qin Zhanping Zhang Changying Zuo 2010Neural Regeneration Research2010,5,14:9
5Residual Fatigue Life Prediction of Ball Bearings Based on Paris Law and RMS显示文摘Paris law can reflect the failure mechanism of materials and is usually used to be a method to predict fatigue life or residual fatigue life.But the variable which can represent the health of machine is hardly measured on line.To a degree,the difficulty of on-line application restricts the scope of application of Paris law.The relationship between characteristic values of vibration signals and the variable in the Paris equation which can describe the health of machine is investigated by taking ball bearings as investigative objects.Based on 6205 deep groove ball bearings as a living example,historical lives and vibration signals are analyzed.The feasibility of describing that variable in the Paris equation by the characteristic value of vibration signals is inspected.After that vibration signals decomposed by empirical mode decomposition(EMD),root mean square(RMS) of intrinsic mode function(IMF) involving fault characteristic frequency has a consistent trend with the diameter of flaws.Based on the trend,two improved Paris models are proposed and the scope of application of them is inspected.These two Paris Models are validated by fatigue residual life data from tests of rolling element bearings and vibration signals monitored in the process of operation of rolling element bearings.It shows that the first improved Paris Model is simple and plain and it can be easily applied in actual conditions.The trend of the fatigue residual life predicted by the second improved Paris model is close to the actual conditions and the result of the prediction is slightly greater than the truth.In conclusion,after the appearance of detectable faults,these improved models based on RMS can predict residual fatigue life on line and a new approach to predict residual fatigue life of ball bearings on line without disturbing the machine running is provided.XU Dong HUANG Jin'e ZHU Qin CHEN Xun XU Yongcheng WANG Shuang 2012Chinese Journal of Mechanical Engineering2012,25,2:6
6The m^(6)A methylome of SARS-CoV-2 in host cells显示文摘The newly identified Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2)has resulted in a global health emergency because of its rapid spread and high mortality.The molecular mechanism of interaction between host and viral genomic RNA is yet unclear.We demonstrate herein that SARS-CoV-2 genomic RNA,as well as the negative-sense RNA,is dynamically N6-methyladenosine(m^(6)A)-modified in human and monkey cells.Combined RIP-seq and miCLIP analyses identified a total of 8 m^(6)A sites at single-base resolution in the genome.Especially,epidemic strains with mutations at these identified m^(6)A sites have emerged worldwide,and formed a unique cluster in the US as indicated by phylogenetic analysis.Further functional experiments showed that m^(6)A methylation negatively regulates SARS-CoV-2 infection.SARS-CoV-2 infection also triggered a global increase in host m^(6)A methylome,exhibiting altered localization and motifs of m^(6)A methylation in mRNAs.Altogether,our results identify m^(6)A as a dynamic epitranscriptomic mark mediating the virus–host interaction.Jun’e Liu Yan-Peng Xu Kai Li Qing Ye Hang-Yu Zhou Hanxiao Sun Xiaoyu Li Liu Yu Yong-Qiang Deng Rui-Ting Li Meng-Li Cheng Bo He Jia Zhou Xiao-Feng Li Aiping Wu Chengqi Yi Cheng-Feng Qin 2021Cell Research2021,31,4:5
7Distribution of aluminum species and the characteristics of structure of poly-aluminum-chloride-sulfate(PACS)显示文摘A series of poly\|aluminum\|chloride\|sulfate (PACS), which has different basicities (γ) and Al 3+ /SO 2- 4 molar ratio, has been prepared and dried at 105℃ and 65℃, respectively. The distribution of aluminum species of PACS was examined, and the effect of γ value, Al 3+ /SO 2- 4 molar ratio, dilution on the distribution of aluminum species of PACS was also investigated by using Al\|ferron timed complex colorimetric method. The IR spectroscopy and X \|ray diffraction were used to study the effect of γ value, Al 3+ /SO 2- 4 molar ratio and the drying temperature on the structure of PACS. The experimental results show that Al 3+ /SO 2- 4 molar ratio has a great effect on the distribution of aluminum species, but the dilution has a little effect on the distribution of aluminum species. The lower the Al 3+ /SO 2- 4 molar ratio, the higher the proportions of the polymer and colloidal species in PACS. The polymeric degree of PACS was related to γ value and Al 3+ /SO 2- 4 molar ratio. 9A Novel Model of Predicting Archie's Cementation Factor from Nuclear Magnetic Resonance(NMR) Logs in Low Permeability Reservoirs显示文摘The resistivity experimental measurements of core samples drilled from low permeability reservoirs of Ordos Basin,Northwest China,illustrate that the cementation factors are not agminate,but vary from 1.335 to 1.749.This leads to a challenge for the estimation of water and hydrocarbon saturation.Based on the analysis of Purcell equation and assumption that rock resistivity is determined by the parallel connection of numerous capillary resistances,a theoretical expression of cementation factor in terms of porosity and permeability is established.Then,cementation factor can be calculated if the parameters of porosity and permeability are determined.In the field application,porosity can be easily obtained by conventional logs.However,it is a tough challenge to estimate permeability due to the strong heterogeneity of low permeability reservoirs.Thus,the Schlumberger Doll Research(SDR)model derived from NMR logs has been proposed to estimate permeability.Based on the analysis of the theoretical expressions of cementation factor and SDR model,a novel cementation factor prediction model,which is relevant to porosity and logarithmic mean of NMR T2spectrum(T2lm),is derived.The advantage of this model is that all the input information can be acquired from NMR logs accurately.In order to confirm the credibility of the novel model,the resistivity and corresponding laboratory NMR measurements of 27 core samples are conducted.The credibility of the model is confirmed by comparing the predicted cementation factors with the core analyzed results.The absolute errors for all core samples are lower than 0.071.Once this model is extended to field application,the accuracy of water and hydrocarbon saturation estimation will be significantly improved.Liang Wang Zhiqiang Mao Yujiang Shi Qin'e Tao Yumei Cheng Yong Song 2014Journal of Earth Science2014,25,1:4
10Parkin deficiency accentuates chronic alcohol intake-induced tissue injury and autophagy defects in brain,liver and skeletal muscle显示文摘Alcoholism leads to organ injury including mitochondrial defect and apoptosis with evidence favoring a role for autophagy dysregulation in alcoholic damage.Parkin represents an autosomal recessive in herited gene for Parkins on's disease and an important member of selective autophagy for mitoch on dria.The associati on between Parkins on's disease and alcoholic injury remains elusive.This study aimed to examine the effect of parki n deficiency on chr onic alcohol in takeinduced organ injury in brain,liver and skeletal muscle(rectus femoris muscle).Adult parkinknockout(PRK-/-)and wild-type mice were placed on Liber-De Carli alcohol liquid diet(4%)for 12 weeks prior to assessment of liver enzymes,intraperitoneal glucose toleranee,protein carbonyl content apoptosis,hematoxylin and eosin morphological staining,and mitochondrial respiration(cytochrome c oxidase,NADH:cytochrome c reductase and succinate:cytochrome c reductase).Autophagy protein markers were monitored by western blot analysis.Our data revealed that chronic alcohol intake imposed liver injury as evidenced by elevated aspartate aminotransferase and alanine transaminase,glucose intoleranee,elevated protein carbonyl formation,apoptosis,focal inflammation,necrosis,microvesiculation,autophagy/mitophagy failure and dampened mitochondrial respiration(complexⅣ,complexesⅠandⅢ,and complexesⅡandⅢ)in the brain,liver and rectus femoris skeletal muscle.Although parkin ablation itself did not generate any notable effects on liver enzymes,insulin sensitivity,tissue carbonyl damage,apoptosis,tissue morphology,autophagy or mitochondrial respiration,it accentuated alcohol intake-induced tissue damage,apoptosis,morphological change,autophagy/mitophagy failure and mitochondrial injury without affecting insulin sensitivity.These data suggest that parkin plays an integral role in the preservation against alcohol-induced organ injury,apoptosis and mitochondrial damage.Hu Peng Xing Qin Sainan Chen Asli E Ceylan Maolong Dong Zhaofen Lin Jun Ren 2020Acta Biochimica et Biophysica Sinica2020,52,6:3
