| 1 | Macrophage inhibitory cytokine-1/growth differentiation factor-15 in premalignant and neoplastic tumours in a high-risk pancreatic cancer cohort显示文摘BACKGROUND Pancreatic cancer(PC)is a leading cause of cancer related mortality worldwide,with poor survival due to late diagnosis.Currently,biomarkers have limited use in early diagnosis of PC.Macrophage inhibitory cytokine-1 or growth differentiation factor-15(MIC-1/GDF15)has been implicated as a potential serum biomarker in PC and other malignancies.AIM To determine the role of MIC-1/GDF15 in detecting pre-malignant pancreatic lesions and neoplastic tumours in an asymptomatic high-risk cohort part of Australian Pancreatic Cancer Screening Program.METHODS A feasibility prospective single centre cohort study was performed.Participants recruited for yearly surveillance with endoscopic ultrasound(EUS)had serial fasting blood samples collected before EUS for MIC-1/GDF15,C-reactive protein and carbohydrate antigen 19-9.Patients were stratified into five groups based on EUS findings:Normal;pancreatic cysts,branch-duct intraductal papillary mucinous neoplasm;diffuse non-specific abnormalities;and neoplastic tumours.MIC-1/GDF15 serum levels were quantified using ELISA.Participants in whom EUS demonstrated abnormalities but not malignancy were closely followed up with magnetic resonance imaging(MRI)or computed tomography.RESULTS One hundred twenty participants were prospectively recruited from 2011-2018.Forty-seven participants(39.2%)had an abnormal EUS and five participants(4.2%)were diagnosed with neoplastic tumours,three by EUS(two pancreatic and one liver)and two by MRI/computed tomography(breast cancer,bladder cancer),which were performed for follow up of abnormal EUS.Baseline serum MIC-1/GDF15 was a significant predictor of neoplastic tumours on receiver operator characteristic curve analysis[area under curve(AUC)=0.814,P=0.023].Baseline serum MIC-1/GDF15 had moderate predictive capacity for branch-duct intraductal papillary mucinous neoplasm(AUC=0.644)and neoplastic tumours noted on EUS(AUC=0.793),however this was not significant(P=0.188 and 0.081 respectively).Serial serum MIC-1/GDF15 did not demonstrate a significant percentage change between a normal and abnormal EUS(P=0.213).Median baseline MIC-1/GDF15 was greater in those with neoplastic tumours(Median=1039.6,interquartile range=727.0-1977.7)compared to those diagnosed with a benign lesion(Median=570.1,interquartile range=460.7-865.2)on EUS and MRI(P=0.012).CONCLUSION In this pilot study MIC-1/GDF15 has predictive capacity for neoplastic tumours in asymptomatic individuals with a genetic predisposition for PC.Further imagining may be warranted in patients with abnormal EUS and raised serum MIC-1/GDF15.Larger multicentric prospective studies are required to further define the role of MIC-1/GDF15 as a serological biomarker in pre-malignant pancreatic lesions and neoplastic tumours. | Robert Sean O’Neill Sam Emmanuel David Williams Alina Stoita | 2020 | World Journal of Gastroenterology2020,26,14: | 8 |
| 2 | Potential relevance of pig gut content transplantation for production and research显示文摘It is becoming increasingly evident that the gastrointestinal microbiota has a significant impact on the overall health and production of the pig.This has led to intensified research on the composition of the gastrointestinal microbiota,factors affecting it,and the impact of the microbiota on health,growth performance,and more recently,behavior of the host.Swine production research has been heavily focused on assessing the effects of feed additives and dietary modifications to alter or take advantage of select characteristics of gastrointestinal microbes to improve health and feed conversion efficiency.Research on faecal microbiota transplantation(FMT) as a possible tool to improve outcomes in pigs through manipulation of the gastrointestinal microbiome is very recent and limited data is available.Results on FMT in humans demonstrating the transfer of phenotypic traits from donors to recipients and the high efficacy of FMT to treat Clostridium difficile infections in humans,together with data from pigs relating GI-tract microbiota composition with growth performance has likely played an important role in the interest towards this strategy in pig production.However,several factors can influence the impact of FMT on the recipient,and these need to be identified and optimized before this tool can be applied to pig production.There are obvious inherent biosecurity and regulatory issues in this strategy,since the donor's microbiome can never be completely screened for all possible non-desirable microorganisms.However,considering the success observed in humans,it seems worth investigating this strategy for certain applications in pig production.Further,FMT research may lead to the identification of specific bacterial group(s) essential for a particular outcome,resulting in the development of banks of clones which can be used as targeted therapeutics,rather than the broader approach applied in FMT.This review examines the factors associated with the use of FMT,and its potential application to swine production,and includes research on using the pig as model for human medical purposes. | Nuria Canibe Mark O’Dea Sam Abraham | 2019 | Journal of Animal Science and Biotechnology2019,10,3: | 6 |

| 5 | Monitoring dioxins and PCBs in eggs as sensitive indicators for environmental pollution and global contaminated sites and recommendations for reducing and controlling releases and exposure显示文摘This review compiles information on PCDD/F-and PCB-contaminated eggs from 20 years of global egg monitoring around emission sources in four continents conducted by the International Pollutants Elimination Network(IPEN)and Arnika as well as a compilation of data from scientific literature.IPEN monitored 127 pooled egg samples including samples from 113 chicken flocks at potential PCDD/F-and PCB-contaminated sites around priority sources listed in the Stockholm Convention(e.g.waste incinerators,metal industries,cement plants,and open burning).99(88%)of pooled egg samples were above the EU maximum limits for PCDD/Fs(2.5 pg PCDD/F-TEQ/g fat)or the sum of PCDD/Fs and dioxinlike PCBs(5 pg PCDD/F-PCB-TEQ/g fat).Children consuming such eggs exceed the tolerable weekly intake(TWI).This demonstrates that close to 90%of these areas were not safe for the production of free-range eggs.Sixteen out of the 113 egg samples(14%)were contaminated above 50 pg TEQ/g fat and exceeded the EU maximum limit more than 10 times.From the 26 pooled egg samples around incinerators 24(92%)exceeded the limit with a mean of 43.1 pg TEQ/g fat(2.6e234 pg TEQ/g).All 21 egg samples around metal industries(4.4e112.6 pg TEQ/g fat)were above limits with mean concentration of 26.0 pg TEQ/g fat.Also all 7 egg samples measured at e-waste recycling sites were above limits(mean 308 pg TEQ/g fat).In 58(51%)pooled egg samples the PCB-TEQ was above 5 pg TEQ/g fat exceeding the EU maximum limit with dioxin-like PCBs alone.This highlights the role of commercial PCBs for global contamination with dioxin-like compounds.It was discovered that around metal industries,shredder plants,open burning sites of e-waste and dump sites,a high share of contamination was caused by dl-PCBs.This clearly shows severe PCB release from the end-of-life management of PCB-containing equipment in developing countries.Also highly contaminated eggs were found at many sites where plastic was incinerated.The highest contaminated egg sample ever measured came from an e-waste site in Ghana and had 856 pg TEQ/g fat plus 300 pg TEQ from brominated dioxins(PBDD/Fs).Other extreme PCDD/F contaminations of eggs were found at a chlor-alkali site(514 pg TEQ/g fat),Agent Orange contaminated areas in Vietnam(490,249 and 246 pg TEQ/g fat)and e-waste sites(568 and 520 pg TEQ/g fat).Where DR CALUX®bioassay revealed higher TEQ compared to measured PCDD/F-PCB-TEQ in IPEN studies,polybrominated PBDD/F were also measured and detected up to 300 pg TEQ/g fat at e-waste sites.One positive outcome from the IPEN studies is that all 10 pooled supermarket eggs in developing countries were below regulatory limit.Policy recommendations are made including:a systematic assessment of areas around PCDD/Fs and PCBs sources;measures for reduction of exposures of populations;urgent control of emission sources including PCB equipment,the open burning of plastic,and the use of plastic as fuel in boilers/incinerators in developing countries without air pollution control.Furthermore,soil limits need to be re-assessed and lowered for free-range poultry. | Jindrich Petrlik Lee Bell Joe DiGangi Serge Molly Allo'o Allo'o Gilbert Kuepouo Griffins Ochieng Ochola Valeriya Grechko Nikola Jelinek Jitka Strakova Martin Skalsky Yuyun Ismawati Drwiega Jonathan N.Hogarh Eric Akortia Sam Adu-Kumi Akarapon Teebthaisong Maria Carcamo Bjorn Beeler Peter Behnisch Claudia Baitinger Christine Herold Roland Weber | 2022 | Emerging Contaminants2022,8,1: | 2 |
