| 1 | Epigenetic regulation in alcoholic liver disease显示文摘Alcoholic liver disease(ALD)is characterized by steatosis or fat deposition in the liver and inflammation,which leads to cirrhosis and hepatocellular carcinoma.Induction of target genes without involving changes in DNA sequence seems to contribute greatly to liver injury. Chromatin modifications including alterations in histones and DNA,as well as post-transcriptional changes collectively referred to as epigenetic effects are altered by alcohol.Recent studies have pointed to a significant role for epigenetic mechanisms at the nucleosomal level influencing gene expression and disease outcome in ALD. Specifically,epigenetic alterations by alcohol include histone modifications such as changes in acetylation and phosphorylation,hypomethylation of DNA,and alterations in miRNAs.These modifications can be induced by alcoholinduced oxidative stress that results in altered recruitment of transcriptional machinery and abnormal gene expression.Delineating these mechanisms in initiation and progression of ALD is becoming a major area of interest.This review summarizes key epigenetic mechanisms that are dysregulated by alcohol in the liver.Alterations by alcohol in histone and DNA modifications,enzymes related to histone acetylation suchas histone acetyltransferases,histone deacetylases and sirtuins,and methylation enzymes such as DNA methyltransferases are discussed.Chromatin modifications and miRNA alterations that result in immune cell dysfunction contributing to inflammatory cytokine production in ALD is reviewed.Finally,the role of alcohol-mediated oxidative stress in epigenetic regulation in ALD is described. | 3 | Signaling mechanisms in alcoholic liver injury: Role of transcription factors, kinases and heat shock proteins显示文摘Alcoholic liver injury comprises of interactions of various intracellular signaling events in the liver. Innate immune responses in the resident Kupffer cells of the liver, oxidative stress-induced activation of hepatocytes, fibrotic events in liver stellate cells and activation of liver sinusoidal endothelial cells all contribute to alcoholic liver injury. The signaling mechanisms associated with alcoholic liver injury vary based on the cell type involved and the extent of alcohol consumption. In this review we will elucidate the oxidative stress and signaling pathways affected by alcohol in hepatocytes and Kupffer cells in the liver by alcohol. The toll-like receptors and their down-stream signaling events that play an important role in alcohol-induced inflammation will be discussed. Alcohol-induced alterations of various intracellular transcription factors such as NFκB, PPARs and AP-1, as well as MAPK kinases in hepatocytes and macrophages leading to induction of target genes that contribute to liver injury will be reviewed. | 5 | Development and organisation of gonadal steroidogenesis in bony fishes - A review显示文摘Gonadal steroidogenesis is pivotal to synchronize various reproductive stages including sexual development,growth and maturation.In all vertebrates including teleost,steroidogenesis is triggered by the mobilization of cholesterol by steroidogenic acute regulatory protein from outer to inner mitochondrial membrane.Thereafter,the entire process occurs in endoplasmic reticulum or mitochondria wherein various steroidogenic enzyme genes play a crucial role.The onset of steroidogenesis during sexual development in teleost is essentially regulated by differential expression of several transcription as well as steroidogenesis-related factors.More specifically,the role of dmrt,sox9,sox3,other sox forms,ad4bp/sf-1,wt-1,foxl2,ftz-f1,gata4,gsdf,Activator protein-1,fgfs and growth factors and steroidogenic enzymes such as cytochrome P450aromatase(cyp19a1),hydroxysteroid 3β-dehydrogenase(hsd3b),hydroxysteroid 17β-dehydrogenase(hsd17b)have been well characterised.Recently,the role of pax2,THO complex(thoc),pentraxin(ptx)and few signalling molecules like wnt4/5 regulating teleostean steroidogenesis has been reported.In females,cyp19a1 appears to be critical as it converts androgens to estrogens during ovarian differentiation which suggests that estradiol-17βis indispensable for the event.Unlike females,males do not depend on testosterone for testicular determination,yet has a major role along with 11-ketotestosterone in testicular development and growth when compared to early testicular differentiation.In males,onset of sex determining or testis-related genes seems most essential.Considering these,the regulation of steroidogenesis is virtually critical at later stages.In view of this,several steroidogenic motifs pertaining to transcriptional regulation have been analysed in teleost,yet far below than the reports on mammals.In this context,the regulatory influence on HHG axis plays a critical role in teleost.Further,multiple gonadotropin-releasing hormones and duality of gonadotropins play a differential role in gonadal function.As for steroidogenic pathway,the synthesis of sex steroids predominantly usesΔ4,however,Δ5 pathway was also evident.The next aspect is shift in steroidogenesis that usually occurs in the maturing follicles during final oocyte maturation,yet such a mechanism is not clear in males.The review highlights the interactions of steroidogenic enzyme gene regulation in terms of HHG axis and other important transcription factors that are involved in the regulatory pathways as well as the influence of environmental and dietary factors by comparing both sexes to present a holistic view on steroidogenesis onset and organisation during gametogenesis in teleost. | Swathi Tenugu Akanksha Pranoty Sajwan-Khatri Mamta Balasubramanian Senthilkumaran | 2021 | Aquaculture and Fisheries2021,6,3: | 2 |

| 10 | A critical review on Nepal Dock(Rumex nepalensis):A tropical herb with immense medicinal importance显示文摘Rumex nepalensis Spreng.(Polygonaceae) commonly known as Nepal Dock has wide-spectrum therapeutic potencies and is extensively used for centuries in traditional medicine systems. The leaves of this plant are edible and a rich source of natural antioxidants. They act as a possible food supplement and are largely used in pharmaceutical industry. Extracts and metabolites from this plant exhibits pharmacological activities including anti-inflammatory, antioxidant, antibacterial, antifungal, antiviral, insecticidal, purgative, analgesic, antipyretic, anti-algal, central nervous system depressant, genotoxic, wound healing and skeletal muscle relaxant activity. Due to its remarkable biological activities, it has the potential to act as a rich source of drug against life threatening diseases. However, more studies are needed to scientifically validate the traditional uses of this plant, beside isolating and identifying their active principles and characterizing the mechanisms of action.
