Discrimination of False Negative Results in RT-PCR Detection of SARS-CoV-2 RNAs in Clinical Specimens by Using an Internal Reference显示文摘Reverse transcription-polymerase chain reaction(RT-PCR)is an essential method for specific diagnosis of SARS-CoV-2 infection.Unfortunately, false negative test results are often reported. In this study,we attempted to determine the principal causes leading to false negative results of RT-PCR detection of SARS-CoV-2 RNAs in respiratory tract specimens.Multiple sputum and throat swab specimens from 161 confirmed COVID-19 patients were tested with a commercialfluorescent RT-PCR kit targeting the ORF1 ab and N regions of SARS-CoV-2 genome. The RNA level of a cellular housekeeping gene ribonuclease P/MRP subunit p30(RPP30)in these specimens was also assessed by RT-PCR.Data for a total of 1052 samples were retrospectively re-analyzed and a strong association between positive results in SARS-CoV-2 RNA tests and high level of RPP30 RNA in respiratory tract specimens was revealed.By using the ROC-AUC analysis,we identified Ct cutoff values for RPP30 RT-PCR which predicted false negative results for SARS-CoV-2 RT-PCR with high sensitivity(95.03%–95.26%)and specificity(83.72%–98.55%)for respective combination of specimen type and ampli-fication reaction.Using these Ct cutoff values, false negative results could be reliably identified. Therefore, the presence of cellular materials, likely infected host cells, are essential for correct SARS-CoV-2 RNA detection by RT-PCR in patient specimens.RPP30 could serve as an indicator for cellular content,or a surrogate indicator for specimen quality. In addition, our results demonstrated that false negativity accounted for a vast majority of contradicting results in SARS-CoV-2 RNA test by RT-PCR. | Yafei Zhang Changtai Wang Mingfeng Han Jun Ye Yong Gao Zhongping Liu Tengfei He Tuantuan Li Mengyuan Xu Luping Zhou Guizhou Zou Mengji Lu Zhenhua Zhang | 2020 | Virologica Sinica2020,35,6: | 5

Progress in graphene-based magnetic hybrids towards highly efficiency for microwave absorption显示文摘Nowadays, the yearning for microwave absorption materials(MAMs) are more and more urgent for dealing with the increasingly serious electromagnetic pollution and the demand of modern military security.Among potential candidates, the graphene(GE) based magnetic hybrids have advantages in structural controllable and designing flexibility, providing opportunities for achieving highly efficiency of microwave absorption(MA). Thus, the structural regulation and MA performances of GE-based magnetic hybrids arouse great attention in related fields. In this review, we summarize the recently progress in MA performance of GE-based magnetic hybrids. Typical absorption process and corresponding mechanism are firstly introduced, for guiding the design of GE-based magnetic MAMs. Then, the magnetic components, synthesis methods, structural features and regulation strategies of these GE-related magnetic materials are reviewed, and their influences on MA performances have also been discussed. Challenges, and prospects of the GE-based magnetic MAMs are suggested. This review provides a brief but systematic introduction to GE-based magnetic MAMs, which may pave the way for the design of MAMs with highly efficient MA performances. | Fuxi Peng Mingfeng Dai Zhenyu Wang Yifan Guo Zuowan Zhou | 2022 | Journal of Materials Science & Technology2022,,11: | 3
Tocilizumab in patients with moderate or severe COVID-19: a randomized, controlled, open-label, multicenter trial显示文摘Tocilizumab has been reported to attenuate the“cytokine storm”in COVID-19 patients. We attempted to verify the effectiveness and safety of tocilizumab therapy in COVID-19 and identify patients most likely to benefit from this treatment. We conducted a randomized,controlled,open-label multicenter trial among COVID-19 patients. The patients were randomly assigned in a 1:1 ratio to receive either tocilizumab in addition to standard care or standard care alone. The cure rate, changes of oxygen saturation and interference, and inflammation biomarkers were observed.Thirty-three patients were randomized to the tocilizumab group, and 32 patients to the control group. The cure rate in the tocilizumab group was higher than that in the control group, but the difference was not statistically significant(94.12%vs.87.10%,rate difference 95%CI−7.19%–21.23%,P=0.4133).The improvement in hypoxia for the tocilizumab group was higher from day 4 onward and statistically significant from day 12(P=0.0359).In moderate disease patients with bilateral pulmonary lesions, the hypoxia ameliorated earlier after tocilizumab treatment, and less patients(1/12,8.33%)needed an increase of inhaled oxygen concentration compared with the controls(4/6,66.67%;rate difference 95%CI−99.17%to−17.50%,P=0.0217).No severe adverse events occurred.More mild temporary adverse events were recorded in tocilizumab recipients(20/34,58.82%)than the controls(4/31,12.90%).Tocilizumab can improve hypoxia without unacceptable side effect profile and significant influences on the time virus load becomes negative. For patients with bilateral pulmonary lesions and elevated IL-6 levels, tocilizumab could be recommended to improve outcome. | Dongsheng Wang Binqing Fu Zhen Peng Dongliang Yang Mingfeng Han Min Li Yun Yang Tianjun Yang Liangye Sun Wei Li Wei Shi Xin Yao Yan Ma Fei Xu Xiaojing Wang Jun Chen Daqing Xia Yubei Sun Lin Dong Jumei Wang Xiaoyu Zhu Min Zhang Yonggang Zhou Aijun Pan Xiaowen Hu Xiaodong Mei Haiming Wei Xiaoling Xu | 2021 | Frontiers of Medicine2021,15,3: | 2

Overexpression of OsSPL9 enhances accumulation of Cu in rice grain and improves its digestibility and metabolism显示文摘Copper(Cu)is an essential trace mineral element for all forms of life, and is an important structural component and co-factor for a variety of metalloenzymes(Pen^a et al.,1999;Bertinato and L'Abbe,2004).In humans,Cu deficiency is not common because of the ubiquitous occurrence of Cu and ease of gastrointestinal absorption(Zidar et al.,1977;Uauy et al.,1998).However,because of the low Cu content in most sources of feed, the use of Cu as a growth promoter to maximize animal production has been well documented in animal feeding(Braude,1945;Lu et al.,2010).Pre- | Mingfeng Tang Chuanshe Zhou Lu Meng Donghai Mao Can Peng Yuxing Zhu Daoyou Huang Zhiliang Tan Caiyan Chen Chengbing Liu Dechun Zhang | 2016 | Journal of Genetics and Genomics2016,43,11: | 2

DFT Study of H_2 Dissociation on Mo_xS_y Clusters显示文摘A DFT study of H2 dissociation on a series of Mo x S y clusters was performed, including homolytic and heterolytic dissociation. The preference for the two pathways on these models show much difference, as the Mo coordination number increases, the homolytic dissociation becomes easier, whereas the heterolytic dissociation becomes more difficult. Furthermore, frontier orbital theory was used to analyze the dissociation mechanisms of these two pathways. It was found that the symmetry and energy gap of Mo x S y's HOMO and H2's LUMO are the decisive factors in H2 activation. | Wang Wei Zhao Xiaoguang Li Huifeng Zhou Han Li Mingfeng | 2015 | China Petroleum Processing & Petrochemical Technology2015,17,1: | 1

Differences in olfactory functional connectivity in early-onset depression and late-onset depression显示文摘Background:Late-onset depression(LOD)and early-onset depression(EOD)exhibit different pathological mechanisms and clinical phenotypes, including different extents of olfactory dysfunction. However, the brain abnormalities underlying the differences in ol-factory dysfunction between EOD and LOD remain unclear. Objective:The aim of this study was to compare the functional connectivity(FC)patterns of olfactory regions between EOD patients and LOD patients and examine their relationship with cognitive function. Methods:One hundred and five patients with EOD,101 patients with LOD and 160 normal controls(NCs)were recruited for the present study.Participants underwent clinical assessment, olfactory testing, cognitive assessments, and magnetic resonance imaging.Eight regions of the primary and secondary olfactory regions were selected to investigate olfactory FC.Results:Patients with LOD exhibited decreased odor identification(OI)compared with patients with EOD and NCs.The LOD group exhibited decreased FC compared with the EOD and NC groups when primary and secondary olfactory regions were selected as the regions of interest(the piriform cortex, lateral entorhinal cortex, and orbital-frontal cortex).Additionally, these abnormal olfactory FCs were associated with decreased cognitive function scores and OI,and the FC between the left orbital-frontal cortex and left amygdala was a partial mediator of the relationship between global cognitive scores and OI.Conclusion:Overall, patients with LOD exhibited decreased FC in both the primary and secondary olfactory cortices compared with patients with EOD,and abnormal olfactory FC was associated with OI dysfunction and cognitive impairment. The FC between the orbital-frontal cortex and amygdala mediated the relationship between global cognitive function and OI. | Ben Chen Mingfeng Yang Meiling Liu Qiang Wang Huarong Zhou Min Zhang Le Hou Zhangying Wu Si Zhang Gaohong Lin Xiaomei Zhong Yuping Ning | 2023 | Psychoradiology2023,3,1: | 0
SCGN deficiency is a risk factor for autism spectrum disorder显示文摘Autism spectrum disorder(ASD)affects 1-2%of all children and poses a great social and economic challenge for the globe. As a highly heterogeneous neurodevelopmental disorder, the development of its treatment is extremely challenging.Multiple pathways have been linked to the pathogenesis of ASD,including signaling involved in synaptic function, oxytocinergic activities, immune homeostasis, chromatin modifications, and mitochondrial functions. Here,we identify secretagogin(SCGN),a regulator of synaptic transmission,as a new risk gene for ASD.Two heterozygous loss-of-function mutations in SCGN are presented in ASD probands.Deletion of Scgn in zebrafish or mice leads to autism-like behaviors and impairs brain development.Mechanistically,Scgn deficiency disrupts the oxytocin signaling and abnormally activates inflammation in both animal models. Both ASD probands carrying Scgn mutations also show reduced oxytocin levels.Importantly,we demonstrate that the administration of oxytocin and anti-inflammatory drugs can attenuate ASD-associated defects caused by SCGN deficiency.Altogether,we identify a convergence between a potential autism genetic risk factor SCGN,and the pathological deregulation in oxytocinergic signaling and immune responses, providing potential treatment for ASD patients suffering from SCGN deficiency. Our study also indicates that it is critical to identify and stratify ASD patient populations based on their disease mechanisms, which could greatly enhance therapeutic success. | Zhe Liu Shuai Tan Lianyu Zhou Li Chen Mingfeng Liu Wang Wang Yingying Tang Qin Yang Sensen Chi Peiyan Jiang Yue Zhang Yonghua Cui Junhong Qin Xiao Hu Shenglong Li Qi Liu Lu Chen Song Li Ezra Burstein Wei Li Xiaohu Zhang Xianming Mo Da Jia | 2023 | Signal Transduction and Targeted Therapy2023,8,2: | 0
Simultaneously enhancing capacity and security in free-space optical chaotic communication utilizing orbital angular momentum显示文摘Optical chaotic signals emitted from an external-cavity feedback or injected laser diode enable small-signal information concealment in a noise-like carrier for secure optical communications.Due to the chaotic bandwidth limitation resulting from intrinsic relaxation oscillation frequency of lasers, multiplexing of optical chaotic signal, such as wavelength division multiplexing in fiber,is a typical candidate for high-capacity secure applications. However,to our best knowledge, the utilization of the spatial dimension of optical chaos for free-space secure communication has not yet been reported. Here,we experimentally demonstrate a free-space all-optical chaotic communication system that simultaneously enhances transmission capacity and security by orbital angular momentum(OAM)multiplexing.Optical chaotic signals with two different OAM modes totally carrying 20 Gbps on-off keying signals are secretly transmitted over a 2 m free-space link, where the channel crosstalk of OAM modes is less than-20 d B,with the mode spacing no less than 3.The receiver can extract valid information only when capturing approximately 92.5% of the OAM beam and correctly demodulating the corresponding mode.Bit error rate below the 7%hard-decision forward error correction threshold of 3.8×10^(-3)can be achieved for the intended recipient. Moreover,a simulated weak turbulence is introduced to comprehensively analyze the influence on the system performance, including channel crosstalk, chaotic synchronization, and transmission performance. Our work may inspire structured light application in optical chaos and pave a new way for developing future high-capacity free-space chaotic secure communication systems. | YIQUN ZHANG MINGFENG XU MINGBO PU MENGJIE ZHOU JIAZHENG DING SHUANGCHENG CHEN KUN QIU NING JIANG XIANGANG LUO | 2023 | Photonics Research2023,11,12: | 0

Growth hormone promotes the reconstruction of injured axons in the hypothalamo-neurohypophyseal system显示文摘Previous studies have shown that growth hormone can regulate hypothalamic energy metabolism, stress, and hormone release. Therefore, growth hormone has great potential for treating hypothalamic injury. In this study, we established a specific hypothalamic axon injury model by inducing hypothalamic pituitary stalk electric lesions in male mice. We then treated mice by intraperitoneal administration of growth hormone. Our results showed that growth hormone increased the expression of insulin-like growth factor 1 and its receptors, and promoted the survival of hypothalamic neurons, axonal regeneration, and vascular reconstruction from the median eminence through the posterior pituitary. Altogether, this alleviated hypothalamic injury-caused central diabetes insipidus and anxiety. These results suggest that growth hormone can promote axonal reconstruction after hypothalamic injury by regulating the growth hormone-insulin-like growth factor 1 axis. | Kai Li Zhanpeng Feng Zhiwei Xiong Jun Pan Mingfeng Zhou Weizhao Li Yichao Ou Guangsen Wu Mengjie Che Haodong Gong Junjie Peng Xingqin Wang Songtao Qi Junxiang Peng | 2024 | Neural Regeneration Research2024,19,10: | 0
