5Accuracy and responses of genomic selection on key traits in apple breeding显示文摘The application of genomic selection in fruit tree crops is expected to enhance breeding eficiency by increasing prediction accuracy,increasing selection intensity and decreasing generation interval.The objectives of this study were to assess the accuracy of prediction and selection response in commercial apple breeding programmes for key traits.The training population comprised 977 individuals derived from 20 pedigreed fllsib families.Historic phenotypic data were available on 10 traits related to productivity and fruit external appearance and genotypic data for 7829 SNPs obtained with an llumina 20K SNP array.From these data,a genome-wide prediction model was built and subsequently used to calculate genomic breeding values of five application fllsib families.The application families had genotypes at 364 SNPs from a dedicated 512 SNP array,and these genotypic data were extended to the high-density level by imputation.These five families were phenotyped for 1 year and their phenotypes were compared to the predicted breeding values.Accuracy of genomic prediction across the 10 traits reached a maximum value of 0.5 and had a median value of 0.19.The accuracies were strongly affected by the phenotypic distribution and heritability of traits.In the largest family,significant selection response was observed for traits with high heritability and symmetric phenotypic distribution.Traits that showed non-significant response often had reduced and skewed phenotypic variation or low heritability.Among the five application families the accuracies were uncorrelated to the degree of relatedness to the training population.The results underline the potential of genomic prediction to accelerate breeding progress in outbred fruit tree crops that still need to overcome long generation intervals and extensive phenotyping costs.Hélène Muranty Michela Troggio Inès Ben Sadok Mehdi Al Rifaï Annemarie Auwerkerken Elisa Banchi Riccardo Velasco Piergiorgio Stevanato W.Eric van de Weg Mario Di Guardo Satish Kumar François Laurens Marco C.A.M.Bink 2015Horticulture Research2015,2,1:14

9Genotyping-by-sequencing of pear (Pyrus spp.) accessions unravels novel patterns of genetic diversity and selection footprints显示文摘Understanding of genetic diversity and marker-trait relationships in pears(Pyrus spp.)forms an important part of gene conservation and cultivar breeding.Accessions of Asian and European pear species,and interspecific hybrids were planted in a common garden experiment.Genotyping-by-sequencing(GBS)was used to genotype 214 accessions,which were also phenotyped for fruit quality traits.A combination of selection scans and association analyses were used to identify signatures of selection.Patterns of genetic diversity,population structure and introgression were also investigated.About 15000 high-quality SNP markers were identified from the GBS data,of which 25%and 11%harboured private alleles for European and Asian species,respectively.Bayesian clustering analysis suggested negligible gene flow,resulting in highly significant population differentiation(Fst=0.45)between Asian and European pears.Interspecific hybrids displayed an average of 55%and 45%introgression from their Asian and European ancestors,respectively.Phenotypic(firmness,acidity,shape and so on)variation between accessions was significantly associated with genetic differentiation.Allele frequencies at large-effect SNP loci were significantly different between genetic groups,suggesting footprints of directional selection.Selection scan analyses identified over 20 outlier SNP loci with substantial statistical support,likely to be subject to directional selection or closely linked to loci under selection.Satish Kumar Chris Kirk Cecilia Deng Claudia Wiedow Mareike Knaebel Lester Brewer 2017Horticulture Research2017,4,1:8
10Seismic response evaluation of base-isolated reinforced concrete buildings under bidirectional excitation显示文摘This paper reports on an investigation of the seismic response of base-isolated reinforced concrete buildings,which considers various isolation system parameters under bidirectional near-fault and far-fault motions. Three-dimensional models of 4-, 8-, and 12-story base-isolated buildings with nonlinear effects in the isolation system and the superstructure are investigated, and nonlinear response history analysis is carried out. The bounding values of isolation system properties that incorporate the aging effect of isolators are also taken into account, as is the current state of practice in the design and analysis of base-isolated buildings. The response indicators of the buildings are studied for near-fault and far-fault motions weight-scaled to represent the design earthquake(DE) level and the risk-targeted maximum considered earthquake(MCER)level. Results of the nonlinear response history analyses indicate no structural damage under DE-level motions for near-fault and far-fault motions and for MCER-level far-fault motions, whereas minor structural damage is observed under MCERlevel near-fault motions. Results of the base-isolated buildings are compared with their fixed-base counterparts. Significant reduction of the superstructure response of the 12-story base-isolated building compared to the fixed-base condition indicates that base isolation can be effectively used in taller buildings to enhance performance. Additionally, the applicability of a rigid superstructure to predict the isolator displacement demand is also investigated. 12Primary squamous cell carcinoma of the rectum: An update and implications for treatment显示文摘AIM: To provide an update on the aetiology, pathogenesis, diagnosis, staging and management of rectal squamous cell carcinoma(SCC).METHODS: A systematic review was conducted according to the preferred reporting items for Systematic Reviews and Meta-Analyses guidelines. A comprehensive search of Ovid MEDLINE was performed with the reference list of selected articles reviewed to ensure all relevant publications were captured. The search strategy was limited to the English language, spanning from 1946 to 2015. A qualitative analysis was undertaken examining patient demographics, clinical presentation, diagnosis, staging, treatment and outcome. The quantitaive analysis was limited to data extracted on treatment and outcomes including radiological, clinical and pathological complete response where available. The narrative and quantitative review were synthesised in concert.RESULTS: The search identified 487 articles in total with 79 included in the qualitative review. The quantitative analysis involved 63 articles, consisting of 43 case reports and 20 case series with a total of 142 individual cases. The underlying pathogenesis of rectal SCC while unclear, continues to be defined, with increasing evidence of a metaplasia-dysplasia-carcinoma sequence and a possible role for human papilloma virus in this progression. The presentation is similar to rectal adenocarcinoma, with a diagnosis confirmed by endoscopic biopsy. Many presumed rectal SCC's are in fact an extension of an anal SCC, and cytokeratin markers are a useful adjunct in this distinction. Staging is most accurately reflected by the tumour-nodemetastasis classification for rectal adenocarcinoma. It involves examining locoregional disease by way of magnetic resonance imaging and/or endorectal ultrasound, with systemic spread excluded by way of computed tomography. Positron emission tomography is integral in the workup to exclude an external siteof primary SCC with metastasis to the rectum. While the optimal treatment remains as yet undefined, recent studies have demonstrated a global shift away from surgery towards definitive chemoradiotherapy as primary treatment. Pooled overall survival was calculated to be 86% in patients managed with chemoradiation compared with 48% for those treated traditionally with surgery. 13Prediction of genetic value for sweet cherry fruit maturity among environments using a 6K SNP array显示文摘The timing of fruit maturity is an important trait in sweet cherry production and breeding.Phenotypic variation for phenology of fruit maturity in sweet cherry appears to be under strong genetic control,but that control might be complicated by phenotypic instability across environments.Although such genotype-by-environment interaction(G×E)is a common phenomenon in crop plants,knowledge about it is lacking for fruit maturity timing and other sweet cherry traits.In this study,1673 genome-wide SNP markers were used to estimate genomic relationships among 597 weakly pedigree-connected individuals evaluated over two seasons at three locations in Europe and one location in the USA,thus sampling eight‘environments’.The combined dataset enabled a single meta-analysis to investigate the environmental stability of genomic predictions.Linkage disequilibrium among marker loci declined rapidly with physical distance,and ordination of the relationship matrix suggested no strong structure among germplasm.The most parsimonious G×E model allowed heterogeneous genetic variance and pairwise covariances among environments.Narrow-sense genomic heritability was very high(0.60–0.83),as was accuracy of predicted breeding values(>0.62).Average correlation of additive effects among environments was high(0.96)and breeding values were highly correlated across locations.Results indicated that genomic models can be used in cherry to accurately predict date of fruit maturity for untested individuals in new environments.Limited G×E for this trait indicated that phenotypes of individuals will be stable across similar environments.Equivalent analyses for other sweet cherry traits,for which multiple years of data are commonly available among breeders and cultivar testers,would be informative for predicting performance of elite selections and cultivars in new environments.Craig M.Hardner Ben J.Hayes Satish Kumar Stijn Vanderzande Lichun Cai Julia Piaskowski JoséQuero-Garcia JoséAntonio Campoy Teresa Barreneche Daniela Giovannini Alessandro Liverani Gérard Charlot Miguel Villamil-Castro Nnadozie Oraguzie Cameron P.Peace 2019Horticulture Research2019,6,1:5
14Use of Teriparatide to improve fracture healing: What is the evidence?显示文摘Teriparatide is a recombinant form of the biologicallyactive component of Parathyroid hormone. It has been shown to increase bone mass and prevent fractures in osteoporotic bone. It is licensed by the Food and Drug Administration for the treatment of Osteoporosis. Over the last decade, a growing body of evidence has accumulated suggesting a role for Teriparatide in the management of fractures. Studies in both normal and delayed healing models have shown improvement in callus volume and mineralisation, bone mineral content, rate of successful union and strength at fracture sites. However most of these results have been derived from animal studies. The majority of this research on humans has comprised low level evidence, with few randomised controlled trials, many case reports and case series. Nevertheless, the results from these studies seem to support research from animal models. This has led to a growing number of clinicians using Teriparatide 'off license' to treat fractures and non-unions in their patients. 17Modal analysis of prestressed draft pad of freight wagons using finite element method显示文摘The present work intends to investigate dynamic behaviour of draft gear using finite element method. The longitudinal force that the draft gear absorbs usually leads to the failure of its components, especially, the load bearing draft pads. Dynamic behaviour of an individual draft pad and a draft gear is determined and characterized with exciting frequencies and corresponding mode shapes. The effect of compressive prestress load on the dynamic behaviour of an individual draft pad is also determined as the draft pads in assembled state are under constant axial compressive force in the draft gear. The vibration characteristics of individual draft pad are compared with draft pads that are part of draft gear.
