1Baicalin prevents LPS-induced activation of TLR4/NF-κB p65 pathway and inflammation in mice via inhibiting the expression of CD14显示文摘Previous studies have shown that baicalin,an active ingredient of the Chinese traditional medicine Huangqin,attenuates LPS-induced inflammation by inhibiting the activation of TLR4/NF-κBp65 pathway,but how it affects this pathway is unknown.It has been shown that CD14 binds directly to LPS and plays an important role in sensitizing the cells to minute quantities of LPS via chaperoning LPS molecules to the TLR4/MD-2 signaling complex.In the present study we investigated the role of CD14 in the anti-inflammatory effects of baicalin in vitro and in vivo.Exposure to LPS(1μg/mL)induced inflammatory responses in RAW264.7 cells,evidenced by marked increases in the expression of MHC II molecules and the secretion of NO and IL-6,and by activation of MyD88/NF-κB p65 signaling pathway,as well as the expression of CD14 and TLR4.These changes were dose-dependently attenuated by pretreatment baicalin(12.5–50μM),but not by baicalin post-treatment.In RAW264.7 cells without LPS stimulation,baicalin dose-dependently inhibit the protein and mRNA expression of CD14,but not TLR4.In RAW264.7 cells with CD14 knockdown,baicalin pretreatment did not prevent inflammatory responses and activation of MyD88/NF-κB p65 pathway induced by high concentrations(1000μg/mL)of LPS.Furthermore,baicalin pretreatment also inhibited the expression of CD14 and activation of MyD88/NF-κB p65 pathway in LPS-induced hepatocyte-derived HepG2 cells and intestinal epithelial-derived HT-29 cells.In mice with intraperitoneal injection of LPS and in DSS-induced UC mice,oral administration of baicalin exerted protective effects by inhibition of CD14 expression and inflammation.Taken together,we demonstrate that baicalin pretreatment prevents LPS-induced inflammation in RAW264.7 cells in CD14-dependent manner.This study supports the therapeutic use of baicalin in preventing the progression of LPS-induced inflammatory diseases.Ya-jun Fu Bo Xu Shao-wei Huang Xia Luo Xiang-liang Deng Shuang Luo Chang Liu Qing Wang Jin-yan Chen Lian Zhou 2021Acta Pharmacologica Sinica2021,42,1:31

3Additive and Over-dominant Effects Resulting from Epistatic Loci Are the Primary Genetic Basis of Heterosis in Rice显示文摘A set of 148 F9 recombinant inbred lines(RILs) was developed from the cross of an indica cultivar 93-11 and japonica cultivar DT713,showing strong F1 heterosis.Subsequently,two backcross F1(BCF1) populations were constructed by backcrossing these 148 RILs to two parents,93-11 and DT713.These three related populations(281BCF1 lines,148 RILs) were phenotyped for six yield-related traits in two locations.Significant inbreeding depression was detected in the population of RILS and a high level of heterosis was observed in the two BCF1 populations.A total of 42 main-effect quantitative trait loci(M-QTLs) and 109 epistatic effect QTL pairs(E-QTLs) were detected in the three related populations using the mixed model approach.By comparing the genetic effects of these QTLs detected in the RILs,BCF1 performance and mid-parental heterosis(HMP),we found that,in both BCF1 populations,the QTLs detected could be classified into two predominant types:additive and over-dominant loci,which indicated that the additive and over-dominant effect were more important than complete or partially dominance for M-QTLs and E-QTLs.Further,we found that the E-QTLs detected collectively explained a larger portion of the total phenotypic variation than the M-QTLs in both RILs and BCF1 populations.All of these results suggest that additive and over-dominance resulting from epistatic loci might be the primary genetic basis of heterosis in rice.Xiaojin Luo Yongcai Fu Peijiang Zhang Shuang Wu Feng Tian Jiayong Liu Zuofeng Zhu Jinshui Yang Chuanqing Sun 2009Journal of Integrative Plant Biology2009,51,4:20
4Concurrent chemoradiotherapy combined with enteral nutrition support:a radical treatment strategy for esophageal squamous cell carcinoma patients with malignant istulae显示文摘Background: Concurrent chemoradiotherapy(CCRT) significantly increases the survival rate of esophageal squa?mous cell carcinoma(ESCC) patients with malignant fistulae. Recent clinical evidence has shown the benefits of enteral nutrition for malnourished cancer patients. In this study, we aimed to validate that, with the support of enteral nutrition, ESCC patients who develop malignant fistulae might be able to complete CCRT and achieve long?term survival.Methods: We reviewed the medical records of 652 patients with ESCC who received definitive CCRT at Sun Yat?sen University Cancer Center between January 2010 and December 2012. Treatment outcome and toxicity were ret?rospectively evaluated in 40 ESCC patients with malignant fistulae. All the 40 patients were treated with CCRT and evaluated by clinical nutritionists using nutrition risk screening(NRS) before, during, and after treatment. Twenty?two patients received a nasogastric tube, and 18 underwent percutaneous endoscopic gastrostomy feeding. The median energy intake was 2166 kcal/day. Treatment response was evaluated at 3 months after the completion of CCRT.Results: With a median follow?up of 18 months(range, 3–39 months), patients' 1?year overall survival(OS) rate was 62.5%, and the estimated OS time was 25.5 months. Univariate analysis showed that the NRS score(P n NRS score(P se to treatment(P < 0.001) were sig= 0.003), increase i= 0.024), fistula closure(P = 0.011), and responnifi?cantly associated with OS. Multivariate analysis showed that tumor response(P = 0.044) and increase in NRS score(P = 0.044) were independent predictors of OS. Grade 3 vomiting was observed in 8 patients(20.0%), grade 3 neutro?penia was observed in 11 patients(27.5%), and grade 3 cough was observed in 13 patients(32.5%); 2 patients(5.0%) died of massive bleeding during treatment.Conclusions: CCRT combined with enteral nutrition support is effective for ESCC patients with malignant fistulae. Patients have an increased potential to be cured, especially those who experience complete response and have an increase in NRS score. 5Recent advances on environmental corrosion behavior and mechanism of high-entropy alloys显示文摘In the past decade, the sudden rise of high-entropy alloys(HEAs) has become a research hotspot in the domain of metal materials. HEAs break through the design concept of traditional single-principal element alloys, and the four core effects, especially the high entropy and cocktail effects, make HEAs exhibit much better corrosion resistance than traditional corrosion-resistant metal materials, e.g., stainless steels, copper-nickel alloys, and high-nickel alloys. Currently, the corrosion resistance of HEAs causes great concern in the field of corrosion research. This article reviews the corrosion behavior and mechanism of HEAs in various aqueous solutions, revealing the correlation among the composition, microstructure and corrosion resistance of HEAs, and elaborates the influence of heat treatment, anodizing treatment and preparation methods on the corrosion behavior of HEAs. This knowledge will benefit the on-demand design of corrosion-resistant HEAs, which is an important trend of future development. 6Simvastatin inhibits apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax显示文摘BACKGROUND: Many studies have showed that apoptosis of endothelial cells plays a curial role in the progress of sepsis. But the role of simvastatin in apoptosis of endothelial cells induced by sepsis is not clear. The present study aimed to investigate the role of simvastatin in apoptosis of endothelial cells induced by sepsis and its mechanism.METHODS: Human umbilical vein endothelial cells(HUVECs) were randomly divided into three groups: control group, sepsis serum intervention group(sepsis group) and simvastatin+sepsis serum intervention group(simvastatin group). After 24-hour incubation with corresponding culture medium, the relative growth rate of HUVECS in different groups was detected by MTT assay; the apoptosis of HUVECs was detected by Hoechst33258 assay and fl ow cytometry; and the expression of the Bcl-2 and Bax genes of HUVECs was detected by PCR.RESULTS: Compared with the sepsis group, HUVECs in the simvastatin group had a higher relative growth rate. Apoptotic HUVECs decreased significantly in the simvastatin group in comparison with the sepsis group. Expression of the Bcl-2 gene in HUVECs decreased obviously, but the expression of the Bax gene increased obviously after 24-hour incubation with sepsis serum; however, the expression of the Bcl-2 and Bax genes was just the opposite in the simvastatin group.CONCLUSIONS: Our study suggests that simvastatin can inhibit apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax. 8Oroxindin inhibits macrophage NLRP3 inflammasome activation in DSS-induced ulcerative colitis in mice via suppressing TXNIP-dependent NF-κB pathway显示文摘Oroxindin is a flavonoid isolated from the traditional Chinese medicine Huang-Qin,which has shown various pharmacological activities including anti-inflammatory,antitumor,antioxidant,etc.Thus far,the effect of oroxindin on colonic inflammation and the underlying mechanism remain unknown.In this study,we investigated the tissue distribution of oroxindin and its therapeutic effects on ulcerative colitis(UC)as well as the underlying mechanisms.UC model was established in mice by administrating dextran sulfate sodium(DSS)in drinking water for 7 d.We first showed that oroxindin was largely absorbed by the colon as an active ingredient after normal mice received Huang-Qin-Tang,a traditional Chinese medicine decoction.UC mice were then treated with oroxindin(12.5,25,50 mg-kg^-1·d^-1,i.g.)for 10 d.We found that oroxindin treatment greatly suppressed massive macrophages infiltration and attenuated pathological changes in colonic tissue.Furthermore,oroxindin treatment significantly inhibited the generation of IL-13 and IL-18 in the colon via inhibiting the nucleotide-binding oligomerization domain-like receptor 3(NLRP3)inflammasome formation and activation.In cultured macrophages,LPS induced NLRp3 inflammasome formation and caspase-1 activation,which were suppressed by oroxindin(12.5-50μM).In LPS-treated macrophages,oroxindin dose-dependently restored the expression of TXNIP protein,leading to suppressing TXNIP-dependent NF-κB activation.In conclusion,these results demonstrate that oroxindin could be absorbed by the colon and attenuate inflammatory responses via inhibiting NLRP3 inflammasome formation and activation,which is related to the inhibitory effect on TXNIP-dependent NF-κB-signaling pathway.Hence,oroxindin has the potential of becoming an effective drug for treating UC.Qi Liu Rui Zuo Kai Wang Fei-fei Nong Ya-jun Fu Shao-wei Huang Zeng-feng Pan Yi Zhang Xia Luo Xiang-liang Deng Xiao-xue Zhang Lian Zhou Yang Chen 2020Acta Pharmacologica Sinica2020,41,6:15
9LY294002 potentiates the anti-cancer effect of oxaliplatin for gastric cancer via death receptor pathway显示文摘AIM:To examine the effects of combined treatment of oxaliplatin and phosphatidylinositol 3'-kinase inhibitor,2-(4-morpholinyl) -8-phenyl-4H-1-benzopyran-4-one(LY294002) for gastric cancer. METHODS:Cell viability was evaluated by 3-(4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide assay.Apoptotic cells were detected by flow cytometric analysis and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling assay.Western blotting and immuno-precipitation were used to examine protein expression and recruitment,respectively.Nuclear factorκB(NFκB) binding activities were investigated using electrophoretic mobility shift assay.Nude mice were used to investigate tumor growth. RESULTS:Treatment with combined oxaliplatin and LY294002 resulted in increased cell growth inhibi-tion and cell apoptosis in vitro,and increased tumor growth inhibition and cell death in the tumor mass in vivo.In MKN45 and AGS cells,oxaliplatin treatment promoted both protein kinase B(Akt) and NFκB activation,while pretreatment with LY294002 significantly attenuated oxaliplatin-induced Akt activity and NFκB binding.LY294002 promoted oxaliplatin-induced Fas ligand(FasL) expression,Fas-associated death domain protein recruitment,caspase-8,Bid,and caspase-3 activation,and the short form of cellular caspase-8/FLICEinhibitory protein(c-FLIPS) inhibition.In vivo,LY294002 inhibited oxaliplatin-induced activation of Akt and NFκB,and increased oxaliplatin-induced expression of FasL,inhibition of c-FLIPS,and activation of caspase-8,Bid,and caspase-3. 10Long-term thermal regimes of the Qinghai-Tibet Railway embankments in plateau permafrost regions显示文摘Ten years of ground temperature data(2003–2013) indicate that the long-term thermal regimes within embankments of the Qinghai-Tibet Railway(QTR) vary significantly with different embankment structures. Obvious asymmetries exist in the ground temperature fields within the traditional embankment(TE) and the crushed-rock basement embankment(CRBE). Measurements indicate that the TE and CRBE are not conducive to maintaining thermal stability. In contrast, the ground temperature fields of both the crushed-rock sloped embankment(CRSE) and the U-shaped crushed-rock embankment(UCRE) were symmetrical. However, the UCRE gave better thermal stability than the CRSE because slow warming of deep permafrost was observed under the CRSE. Therefore, the UCRE has the best long-term effect of decreasing ground temperature and improving the symmetry of the temperature field. More generally, it is concluded that construction using the cooling-roadbed principle meets the design requirements for long-term stability of the railway and for train transport speeds of 100 km h?1. 11Time-domain analysis methodology for large-scale RLC circuits and its applications显示文摘With soaring work frequency and decreasing feature sizes, VLSI circuits with RLC parasitic components are more like analog circuits and should be carefully analyzed in physical design. However, the number of extracted RLC components is typically too large to be analyzed efficiently by using present analog circuit simulators like SPICE. In order to speedup the simulations without error penalty, this paper proposes a novel methodology to compress the time-descritized circuits resulted from numerical integration approximation at every time step. The main contribution of the methodology is the efficient structure-level compression of DC circuits containing many current sources, which is an important complement to present circuit analysis theory. The methodology consists of the following parts: 1) An approach is proposed to delete all intermediate nodes of RL branches. 2) An efficient approach is proposed to compress and back-solve parallel and serial branches so that it is error-free and of linear complexity to analyze circuits of tree topology. 3) The Y toπtransformation method is used to error-free reduce and back-solve the intermediate nodes of ladder circuits with the linear complexity. Thus, the whole simulation method is very accurate and of linear complexity to analyze circuits of chain topology. Based on the methodology, we propose several novel algorithms for efficiently solving RLC-model transient power/ground (P/G) networks. Among them, EQU-ADI algorithm of linear-complexity is proposed to solve RLC P/G networks with mesh-tree or mesh-chain topologies. 16Effect of high glucose levels on the calcification of vascular smooth muscle cells by inducing osteoblastic differentiation and intracellular calcium deposition via BMP-2/Cbfα-1 pathway显示文摘In this paper,we investigate the effect and the possible mechanism of high glucose levels on the calcification of human aortic smooth muscle cells (HASMCs).HASMCs were divided into four groups: normal glucose group (NG),osmolality control group (OC),high glucose group (HG,HASMCs culture medium containing 30 mmol/L glucose),and high glucose plus recombinant human Noggin protein (bone morphogenetic protein-2 (BMP-2) antagonist) group (HN).The mRNA levels and the protein expressions of BMP-2 and core binding factor alpha-1 (Cbfα-1) were measured by real-time quantitative polymerase chain reaction (PCR) and Western blot.After induced by 10 mmol/L β-glycerol phosphoric acid,cells were harvested for assessments of alkaline phosphatase (ALP) activities at Days 1,2,and 3,and intracellular calcium contents at Days 7 and 14,respectively.High glucose levels increased the mRNA levels and the protein expressions of BMP-2 and Cbfα-1 (P<0.05).The expression of Cbfα-1 was partially blocked by Noggin protein (P<0.05),while BMP-2 was not (P>0.05).After being induced by β-glycerol phosphoric acid,high glucose levels increased the ALP activity [(48.63±1.03) vs.(41.42±2.28) U/mg protein,Day 3;P<0.05] and the intracellular calcium content [(2.76±0.09) vs.(1.75±0.07) μmol/mg protein,Day 14;P<0.05] in a time-dependent manner when compared with the NG group,while the ALP activity could not be blocked by Noggin protein [(48.63±1.03) vs.(47.37±0.97) U/mg protein,Day 3;P>0.05].These results show that high glucose levels can evoke the calcification of HASMCs by inducing osteoblastic trans-differentiation and intracellular calcium deposition via the BMP-2/Cbfα-1 pathway,which can be partially blocked by Noggin protein.Fang LIU Hui ZHONG Jing-yuan LIANG Ping FU Zhi-juan LUO Li ZHOU Rong GOU Jun HUANG 2010Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,12:12
17A Solution on Pork Quality Traceability from Farm to Dinner Table in Tianjin City,China显示文摘In order to meet government supervision of pork production safety as well as consumer's right to know what they buy and protect the public safety of pork food,this study adopts animal identification,intelligent personal digital assistant(PDA) reading and writing,general packet radio service(GPRS),and other information technologies,proposes a pork tracking and traceability inferstructure based on pork production substrace flow and data flow,designs the metadata structure and related datatbases for farming,slaughtering,and retailing sector based on intensive pig farming and smallhold pig farming,develops three different data-recording systems,and finally establishes a public network platform for the information inquiry in light of 'the administrative rules on identification and rearing files for animal and poultry' in China.The farming process information system supplies early warning for the usage of drugs and feed additives based on data of every individual pig and timely uploading all events data to remote traceability database when pigs are sold;the PDA data collecting system can collect farming events data for pigs fed by farmers and submit to the center database by GPRS;the web-based Tianjin's pork traceability platform can integrate all identifications and related pork quality data from farming,slaughtering to marketing by online,and achieve pork tracking from product origin to consumption and tracing in the turnover direction.It is feasible to realize pork quality traceability by identification technologies developed and/or integrated,metadata specifications designed,three data-recording systems developed,and web-inquiring platform established.Some individual technical bottlenecks will be resolved with the development of communication technologies.The full implementation in Tianjin,China,will supply technical support for guaranteeing the quality and safety of pork production and meeting consumer's demands.XIONG Ben-hai FU Run-ting LIN Zhao-hui LUO Qing-yao YANG Liang PAN Jia-rong 2010Agricultural Sciences in China2010,9,1:12
18Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 19Morinda officinalis oligosaccharides increase serotonin in the brain and ameliorate depression via promoting 5-hydroxytryptophan production in the gut microbiota显示文摘Morinda officinalis oligosaccharides(MOO) are an oral drug approved in China for the treatment of depression in China. However, MOO is hardly absorbed so that their anti-depressant mechanism has not been elucidated. Here, we show that oral MOO acted on tryptophan → 5-hydroxytryptophan(5-HTP) → serotonin(5-HT) metabolic pathway in the gut microbiota. MOO could increase tryptophan hydroxylase levels in the gut microbiota which accelerated 5-HTP production from tryptophan;meanwhile, MOO inhibited 5-hydroxytryptophan decarboxylase activity, thus reduced 5-HT generation,and accumulated 5-HTP. The raised 5-HTP from the gut microbiota was absorbed to the blood, and then passed across the blood-brain barrier to improve 5-HT levels in the brain. Additionally, pentasaccharide,as one of the main components in MOO, exerted the significant anti-depressant effect through a mechanism identical to that of MOO. 20Virus-induced Gene Silencing in Eggplant(Solanum melongena)显示文摘Eggplant(Solanum melongena)is an economically important vegetable requiring investigation into its various genomic functions.The current limitation in the investigation of genomic function in eggplant is the lack of effective tools available for conducting functional assays.Virus-induced gene silencing(VIGS)has played a critical role in the functional genetic analyses.In this paper,TRV-mediated VIGS was successfully elicited in eggplant.We first cloned the CDS sequence of PDS(PHYTOENE DESATURASE) in eggplant and then silenced the PDS gene.Photo-bleaching was shown on the newly-developed leaves four weeks after agroinoculation,indicating that VIGS can be used to silence genes in eggplant.To further illustrate the reliability of VIGS in eggplant,we selected Chl H,Su and CLA1 as reporters to elicit VIGS using the high-pressure spray method.Suppression of Chl H and Su led to yellow leaves,while the depletion of CLA1 resulted in albino.In conclusion,four genes,PDS,Chl H,Su(Sulfur),CLA1,were down-regulated significantly by VIGS,indicating that the VIGS system can be successfully applied in eggplant and is a reliable tool for the study of gene function.Haiping Liu Daqi Fu Benzhong Zhu Huaxue Yan Xiaoying Shen Jinhua Zuo Yi Zhu Yunbo Luo 2012Journal of Integrative Plant Biology2012,54,6:11
