| 5 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 6 | Disease Risks of Childhood Obesity in China显示文摘Objective To estimate the relative risks of dyslipidemia, hypertension, diabetes mellitus, and metabolic syndromes among overweight and obese Chinese children compared with their normal weight counterparts. Methods Overweight and obesity were defined by age- and sex-specific BMI classification reference for Chinese children and adolescents. Pediatric metabolic syndrome (MetS) and each risk factor for MetS were defined using the criteria for US adolescents. Definition of hyper-TC, LDL, and dyslipidemia for adults was applied as well. General linear model factor analysis and chi-square test were used to compare the difference in metabolic indicators among normal weight, overweight, and obese groups. Multiple logistic regression analysis was performed to estimate the odds ratio of metabolic abnormalities between obesity, overweight, and normal weight children, after adjustment for living area, family economic level, age, sex, and daily exercise time and TV watching time, as well as different dietary indices in the model. Results Significant increases in blood lipids, glucose, and blood pressure were found among overweight and obese children as compared with their counterparts with normal weight. By applying WGOC-recommended BMI classification, the risks for hypertriglyceridemia, low HDL and dyslipidemia among overweight children were 1.9, 1.4, and 1.5 times, and was 3.3, 1.5, and 1.8 times among obese groups compared to their counterparts with normal weight after adjustment for age, sex, region, socioeconomic status, physical activity, and dietary intakes. The overweight and obese children (15-17.9 years) had a high-risk of developing hypertension, which was 2.3 and 2.9times higher than their counterparts with normal weight. Above 90% obese adolescents had abdominal obesity, while less than1% normal weight ones had abdominal obesity. No obese adolescents were free from any risk factors for MetS, while 36.9% of normal weight adolescents were from the risk factors. 83.3% obese boys and all obese girls had metabolic syndrome, while only15.5% normal weight boys and 18.8% normal weight girls had metabolic syndrome. Four risk factors for metabolic syndrome were found in 8.3% obese boys while none in normal weight boys and girls. The prevalence of MetS among normal weight,overweight, and obesity groups was 1.5%, 18.3%, and 38.1% respectively. Conclusion The cardiovascular disease (CVD)risk factors are clustered in obese Chinese children. Wilson) (FF), and birch (Betula platyphylla Sukaczev) (BF) subalpine forests in western China were measured to understand the monthly variations in litter nutrient concentrations and annual and monthly nutrient returns via litterfall. Nutrient concentration in litter showed the rank order of Ca > N > Mg > K > S > P. Monthly variations in nutrient concentrations were greater in leaf litter (LL) than other litter components. The highest and lowest concentrations of N, P, K, and S in LL were found in the growing season and the nongrowing season, respectively, but Ca and Mg were the opposite. Nutrient returns via litterfall showed a marked monthly pattern with a major peak in October and one or two small peaks in February and/or May, varying with the element and stand type, but no marked monthly variations in nutrient returns via woody litter, reproductive litter, except in May for the BF, and moss litter. Not only litter production but also nutrient concentration controlled the annual nutrient return and the monthly nutrient return pattern. In this paper, we provide a detailed introduction to the Taihu Light system. In contrast with other existing heterogeneous supercomputers, which include both CPU processors and PCIe-connected many-core accelerators(NVIDIA GPU or Intel Xeon Phi), the computing power of Taihu Light is provided by a homegrown many-core SW26010 CPU that includes both the management processing elements(MPEs)and computing processing elements(CPEs) in one chip. With 260 processing elements in one CPU, a single SW26010 provides a peak performance of over three TFlops. To alleviate the memory bandwidth bottleneck in most applications, each CPE comes with a scratch pad memory, which serves as a user-controlled cache. To support the parallelization of programs on the new many-core architecture, in addition to the basic C/C++and Fortran compilers, the system provides a customized Sunway Open ACC tool that supports the Open ACC2.0 syntax. | 18 | Prognostic factors of refractory NSCLC patients receiving anlotinib hydrochloride as the third-or further-line treatment显示文摘Objective:Anlotinib hydrochloride is a multitarget tyrosine kinase inhibitor that targets vascular endothelial growth factor receptor,fibroblast growth factor receptor,platelet-derived growth factor receptor,c-Kit,and c-MET;therefore,it exhibits both antitumor and anti-angiogenetic activities.A phase III trial has shown that anlotinib improved progression-free survival(PFS)and overall survival(OS)in patients with advanced non-small cell lung cancer(NSCLC),who presented with progressive disease or intolerance after standard chemotherapy.This study aimed to analyze the characteristics of patients receiving anlotinib treatment to determine the dominant populations who are fit for the treatment.Methods:Data were collected from March 2015 to January 2017 from a randomized,double-blind,placebo-controlled,multicenter,phase III trial of anlotinib(ALTER0303).A total of 437 patients were enrolled and randomly allocated(2:1)to the anlotinib and placebo groups.Kaplan–Meier analysis and log-rank test were performed to compare PFS and OS.Cox proportional hazards model was adopted for multivariate prognostic analysis.Results:Multivariate analysis indicated that high post-therapeutic peripheral blood granulocyte/lymphocyte ratio and elevated alkaline phosphatase levels were independent risk factors for PFS.Meanwhile,elevated thyroid-stimulating hormone,blood glucose,and triglyceride levels;hypertension;and hand–foot syndrome were independent protective factors of PFS.High posttherapeutic peripheral blood granulocyte/lymphocyte ratio,an Eastern Cooperative Oncology Group(ECOG)score≥2,and the sum of the maximal target lesion length at baseline were independent risk factors of OS,and hypertriglyceridemia was an independent protective factor of OS.Conclusions:This study preliminarily explored the possible factors that affected PFS and OS after anlotinib treatment in patients with advanced refractory NSCLC,and the baseline characteristics of the therapeutically dominant populations were then identified. | Jing Wang Yizhuo Zhao Qiming Wang Li Zhang Jianhua Shi Zhehai Wang Ying Cheng Jianxing He Yuankai Shi Hao Yu Yang Zhao Weiqiang Chen Yi Luo Xiuwen Wang Kejun Nan Faguang Jin Jian Dong Baolan Li Zhujun Liu Baohui Han Kai Li | 2018 | Cancer Biology & Medicine2018,15,4: | 46 |

| 20 | Isotopic evidence for the moisture origin and composition of surface runoff in the headwaters of the Heihe River basin显示文摘We investigated the moisture origin and contribution of different water sources to surface runoff entering the headwaters of the Heihe River basin on the basis of NECP/NCAR(National Centers for Environmental Prediction/National Center for Atmospheric Research) re-analysis data and variations in the stable hydrogen and oxygen isotope ratios(δ D and δ 18O) of precipitation,spring,river,and melt water. The similar seasonality in precipitation δ 18O at different sites reveals the same moisture origin for water entering the headwaters of the Heihe River basin. The similarity in the seasonality of δ 18O and d-excess for precipitation at Yeniugou and Urumchi,which showed more positive δ 18O and lower d-excess values in summer and more negative δ 18O and higher d-excess values in winter,indicates a dominant effect of westerly air masses in summer and the integrated influence of westerly and polar air masses in winter. Higher d-excess values throughout the year for Yeniugou suggest that in arid inland areas of northwestern China,water is intensively recycled. Temporal changes in δ 18O,δ D,and d-excess reveal distinct contributions of different bodies of water to surface runoff. For example,there were similar trends for δ D,δ 18O,and d-excess of precipitation and river water from June to September,similar δ 18O trends for river and spring water from December to February,and similar trends for precipitation and runoff volumes. However,there were significant differences in δ 18O between melt water and river water in September.
