| 10 | ERK1/2-mediated Cytoplasmic Accumulation of hnRNPK Antagonizes TRAIL-induced Apoptosis through Upregulation of XIAP in H1299 Cells显示文摘Objective Tumor necrosis factor-related apoptosis-inducing ligand(TRAIL) resistance greatly limits the clinical therapeutic efficacy of TRAIL. Elucidating the molecular mechanism underlying TRAIL resistance will be fundamental to resolving this problem. Methods Nuclear and cytoplasmic protein extraction and immunofluorescence(IF) assay were used to detect changes in heterogeneous nuclear ribonucleoprotein K(hn RNPK) localization in H1299 cells. The evaluation of cell apoptosis in cells transfected with GFP-hn RNPK, GFP-hn RNPK S284/353 A, or GFP-hn RNPK S284/353 D mutant was performed using cleaved caspase-3 antibody. The gene expression of XIAP was tested by quantitative RT-PCR. Results Previously, we reported that hn RNPK antagonized TRAIL-induced apoptosis through inhibition of PKC-mediated GSK3β phosphorylation. In this study, we further demonstrate that TRAIL treatment induces cytoplasmic accumulation of hn RNPK in H1299 cells. The hn RNPK localized in the cytoplasm has a higher capacity to antagonize TRAIL-induced apoptosis. Both ERK1/2 signaling inhibitor U0126 and ERK-phosphoacceptor-site mutant(GFP-hn RNPK S284/353A) diminish cytoplasmic accumulation of hn RNPK induced by TRAIL. Moreover, we show that XIAP is involved in hn RNPK-mediated TRAIL resistance in H1299 cells. | 16 | Improving the Local Sheep in Gansu via Crossing with Introduced Sheep Breeds Dorset and Borderdale显示文摘In order to improve the meat performance of local sheep in Gansu Province,Dorset and Borderdale were introduced to crossbreed with local sheep which were Tan sheep,Small-tail Han sheep and Mongolia sheep. The offspring under different crossbreeding combinations were sampled randomly at the different growing stage to measure their growth traits so as to select optimize the crossbreeding mode. The results indicated that,for the same crossbreeding mode,the growth rate of progeny was in order F3> F2> F1; for the F3 progeny,the combinations Dorset- Borderdale- Small tail Han sheep and Dorset- Borderdale- Mongolia sheep gave a higher growth rate,with a body weight of 1. 57%,3. 17%,8. 23%,1. 15% higher in male and female individuals than the counterparts of Dorset and Tan sheep and Small tail Han sheep; for the F2 progeny,the combinations Dorset- Borderdale- Small tail Han sheep and Dorset- Borderdale- Mongolia sheep also gave a higher growth rate,with a body weight of 2. 15%,4. 53%,9. 21% and 2. 75% higher in male and female individuals than the counterparts of Dorset and Tan sheep and Small tail Han sheep; for the F1 progeny,the combination Borderdale and Small tail Han sheep assumed a higher growth rate,with a body weight of 3. 23%,6. 07%,7. 42% and 8. 66% higher in male and female individuals than the counterparts of Borderdale- Mongolia sheep and Tan sheep- Small tail Han sheep,respectively. Therefore,in the Small-tail Han sheep and Mongolia sheep producing regions,the F2 or F3progeny bred by using Dorset or Borderdale sheep as male parent to cross with local breeds,or the hybrid lambs of Small-tail Han sheep and Borderdale sheep as highly qualified commodity,would produce significant economic benefit. | 17 | Role of laparoscopy in the diagnosis and treatment of adnexal masses显示文摘Background Laparoscopy has been accepted for years as a management of benign ovarian tumors. The aim of this study was to estimate the feasibility and safety of laparoscopy in diagnosis and management of adnexal masses. Methods A total of 2083 patients with benign adnexal mass were treated by laparoscopy at Peking Union Medical College Hospital from January 2000 to December 2003. Their clinical data were reviewed retrospectively. All the adnexal masses suspicious of malignancy at the time of laparoscopy were sent for frozen section evaluation intraoperatively. The rates of unexpected intracystic vegetation and low malignant potential (LMP) tumor or malignancy were investigated. The sensitivity, specificity, positive predictive value, and negative predictive value of laparoscopic diagnosis for LMP or ovarian malignancies were calculated. The ratios were compared by Chi-square test and the continuous variables were tested using two-tailed t test. Results Of the 2083 patients, 16 had LMP or invasive tumors (0.77%), among which 14 were diagnosed histologically intraoperatively and 2 postoperatively. Fifty-five (2.6%) of the 2083 patients had unexpected intracystic vegetations. Their frozen sections showed benign tumors in 41 (74.5%), LMP tumors in 8 (14.5%), and focal invasive ovarian cancers (stage Ic) in 6 (10.9%). The final pathological diagnosis were benign tumors in 41 (74.5%), LMP tumors 7 (12.7%), and focal invasive ovarian cancers (stage Ic) in 7 (12.7%). Laparoscopy achieved a sensitivity of 87.5%, specificity of 98%, positive predictive value of 25.5%, and negative predictive value of 99.9% in the diagnosis of ovarian malignancies. 2067 cases with benign adnexal masses underwent laparoscopy successfully. No conversion to laparotomy, or intra- and postoperative complications in this series. Of the 16 patients with LMP or invasive ovarian cancer, seven underwent laparoscopic surgery including immediate staging laparoscopy in 3. The mean follow-up was 17.3 months for the 16 patients. Among them, 1 developed a recurrent LMP tumor in the contralateral ovary 36 months after laparoscopic salpingo-oophorectomy, and received subsequent laparoscopic cystectomy and pelvic lymph node sampling; the others had no evidence of recurrent tumor during the follow-up. | 18 | Amperometric Determination of lndole-3-acetic Acid Based on Platinum Nanowires and Carbon Nanotubes显示文摘Platinum nanowire (PtNW) can be grown by electrodeposition in polycarbonate membrane, with the average diameter of the nanowires about 250 nm. The PtNW and multiwalled carbon nanotubes (CNT) are then dispersed into chitosan (CHIT) solution. The resulting PtNW-CNT-CHIT material brings new capabilities for electrochemical devices by using the synergistic action of the electrocatalytic activity of PtNW and CNT. By dropping the PtNW-CNT-CHIT film onto the glassy carbon (GC) electrode surface, and after evaporation an amperometric sensor for the determination of indole-3-acetic acid (IAA) was developed. The oxidation current of IAA increased significantly at the PtNW-CNT-CHIT film coated GC electrode, in contrast to that at the CNT-CHIT modified GC. | 20 | Using seismic surveys to investigate sediment distribution and to estimate burial fluxes of OC, N, and P in a canyon reservoir显示文摘As a high-precision survey method,seismic surveying has been increasingly applied to inland water research,although its application to artificial reservoirs has remained limited.As a special artificial water body,reservoirs have important effects on the fluvial transport of material from land to ocean,and inevitably have complex terrain which can complicate and distort the results of seismic surveys.Therefore,there are still some problems need to be resolved in the application of seismic surveys in reservoirs with complex terrain.For this study,the Dongfeng Reservoir located in the upper reaches of the Wujiang River was chosen as an example to test the seismic survey method.Our testing showed that(1)because of the complex underwater terrain,the signal-to-noise ratio of the echo signal in canyon reservoir is low,making it difficult to determine sediment layers thicknesses in some areas;and(2)due to the large spatial heterogeneity of sediment distribution,insufficient density of cross-sections can lead to inaccurate interpolation results.To improve the accuracy of calculations,a mathematical method was used.Ultimately,the total burial mass of sediment was estimated at 2.85 x 107 tons,and the average burial rates of total organic carbon,total phosphorus,and total nitrogen were estimated at 0.194,0.011,and 0.014 g cm-2 year-1,respectively.These values were close to the results of previous studies and hydrographic station data,indicating that seismic survey can be a reliable and efficient method for the mapping of reservoirs. | Ningxiao Yu Yong Qin Feng Hao Yunchao Lang Fushun Wang | 2019 | Acta Geochimica2019,38,6: | 3 |
