| 2 | Mechanisms of immune checkpoint inhibitormediated liver injury显示文摘The immune checkpoints,cytotoxic T-lymphocyte-associated antigen 4(CTLA-4) and programmed cell death protein-1/ligand-1(PD-1/PD-L1) are vital contributors to immune resulation and tolerance.Recently immune checkpoint inhibitors(ICIs) have revolutionized cancer therapy;however,they come with the cost of immune related adverse events involving multiple organs such as the liver.Due to its constant expo sure to foreign antigens,the liver has evolved a high capacity for immune tolerance,therefore,blockade of the immune checkpoints can result in aberrant immune activation affecting the liver in up to 20% of patients depending on the agent(s) used and underlying factors.This type of hepatotoxicity is termed immune mediated liver injury from checkpoint inhibitors(ILICI) and is more common when CTLA4 and PD-1/PDL1 are used in combination.The underlying mechanisms of this unique type of hepatotoxicity are not fully understood;however,the contribution of CD8^(+) cytotoxic T lymphocytes,various CD4^(+) T cells populations,cytokines,and the secondary activation of the innate immune system leading to liver injury have all been suggested.This review summarizes our current understanding of the underlying mechanisms of liver injury in immunotherapy using animal models of ILICI and available patient data from clinical studies. | Layla Shojaie Myra Ali Andrea Iorga Lily Dara | 2021 | Acta Pharmaceutica Sinica B2021，11，12： | 6 |
| 3 | UFMylation of RPL26 links translocation-associated quality control to endoplasmic reticulum protein homeostasis显示文摘Protein biogenesis at the endoplasmic reticulum(ER)in eukaryotic cells is monitored by a protein quality control system named ER-associated protein degradation(ERAD).While there has been substantial progress in understanding how ERAD eliminates defective polypeptides generated from erroneous folding,how cells remove nascent chains stalled in the translocon during co-translational protein insertion into the ER is unclear.Here we show that ribosome stalling during protein translocation induces the attachment of UFM1,a ubiquitin-like modifier,to two conserved lysine residues near the COOH-terminus of the 60S ribosomal subunit RPL26(uL24)at the ER.Strikingly,RPL26 UFMylation enables the degradation of stalled nascent chains,but unlike ERAD or previously established cytosolic ribosome-associated quality control(RQC),which uses proteasome to degrade their client proteins,ribosome UFMylation promotes the targeting of a translocation-arrested ER protein to lysosomes for degradation.RPL26 UFMylation is upregulated during erythroid differentiation to cope with increased secretory flow,and compromising UFMylation impairs protein secretion,and ultimately hemoglobin production.We propose that in metazoan,co-translational protein translocation into the ER is safeguarded by a UFMylation-dependent protein quality control mechanism,which when impaired causes anemia in mice and abnormal neuronal development in humans. | Lihui Wang Yue Xu Heather Rogers Layla Saidi Constance Tom Noguchi Honglin Li Jonathan Wilson Yewdell Nicholas Raymond Guydosh Yihong Ye | 2020 | Cell Research2020，30，1： | 5 |
| 4 | Polybrominated diphenyl ethers and“novel”brominated flame retardants in floor and elevated surface house dust from Iraq:Implications for human exposure assessment显示文摘Concentrations of polybrominated diphenyl ethers(PBDEs)and selected novel brominated flame retardants(NBFRs)were measured in indoor dust from the living areas of 18 homes in Basrah,Iraq.This is the first report of contamination of the Iraqi environment with these chemicals.To evaluate the implications for human exposure,samples were collected from both the floor and from elevated surfaces like tables,shelves and chairs.When normalised for the organic carbon content of the dust sample,concentrations in elevated surface dust of BDE-99,BDE-209,pentabromoethylbenzene(PBEB),bis(2-ethylhexyl)3,4,5,6-tetrabromophthalate(BEH-TEBP),and decabromodiphenylethane(DBDPE)exceeded significantly(p<0.05)those in floor dust from the same rooms.This suggests that previous studies that base estimates of adult exposure via dust ingestion on floor dust,may underestimate exposure.Such underestimation is less likely for toddlers who are far more likely to ingest floor dust.Concentrations of PBDEs and NBFRs in indoor dust from Basrah,Iraq are at the lower end of levels reported elsewhere.The PBDE contamination pattern in our samples suggests that use in Iraq of the Deca-BDE formulation,exceeds substantially that of Penta-BDE,but that use of the Octa-BDE formulation has been higher in Iraq than in some other regions.Reassuringly,our estimates of exposure to our target BFRs via dust ingestion for the Iraqi population fall well below the relevant health-based limit values. | Layla Salih Al-Omran Stuart Harrad | 2016 | Emerging Contaminants2016，2，1： | 2 |

| 17 | Concentrations of organophosphate esters in drinking water from the United Kingdom:Implications for human exposure显示文摘Data on the presence of organophosphate esters(OPEs)in drinking water and its significance as a pathway of exposure are limited.In this study,we measure for the first time,concentrations of eight OPEs in 50 UK drinking water samples.Arithmetic mean concentrations of ∑8OPEs were:6.4 and 11 ng/L in bottled(n=25)and tap water samples(n=25),respectively.Concentrations of ∑8OPEs in tap water(mean:11 ng/L)exceed significantly those in bottled water(mean:6.4 ng/L)(p˂0.01).Moreover,UK tap water is more contaminated with chlorinated,aryl-,and alkyl-OPEs than bottled water.The predominant OPEs detected were:tris(butoxyethyl)phosphate(TBOEP),tris(2-chloroethyl)phosphate(TCEP),and tris(2-chloroisopropyl)phosphate(TCIPP)with arithmetic mean concentrations in the two water sample types ranging between(3.5e3.8 ng/L),(0.60-3.0 ng/L),and(1.02-2.9 ng/L),respectively.Estimated daily intakes(EDIs)(mean and high-end exposure)via drinking water for different sectors of the UK population were:infants(0.93 and 6.4 ng/kg bw/day)˃toddlers(0.46 and 3.1 ng/kg bw/day)˃children(0.35 and 2.3 ng/kg bw/day)˃adults(0.28 and 2.1 ng/kg bw/day).Based on these data,exposure to ∑8OPEs via drinking water is much lower than via:food,indoor dust ingestion,inhalation,and dermal uptake for adults and toddlers.Reassuringly,our EDIs were lower than relevant reference dose(RfD)values.However,combining our drinking water ingestion data with exposure via other pathways revealed overall exposure to 2-ethylhexyl diphenyl phosphate(EHDPP)and TCIPP to approach health-based limit values for UK toddlers under a high-end exposure scenario. | Muideen Remilekun Gbadamosi Layla Salih Al-Omran Mohamed Abou-Elwafa Abdallah Stuart Harrad | 2023 | Emerging Contaminants2023，9，1： | 1 |
