1Pharmacologic inducers of the uric acid exporter ABCG2 as potential drugs for treatment of gouty arthritis显示文摘Uric acid is the end product of purine catabolism and its plasma levels are maintained below its maximum solubility in water(6–7 mg/dl).The plasma levels are tightly regulated as the balance between the rate of production and the rate of excretion,the latter occurring in urine(kidney),bile(liver)and feces(intestinal tract).Reabsorption in kidney is also an important component of this process.Both excretion and reabsorption are mediated by specific transporters.Disruption of the balance between production and excretion leads to hyperuricemia,which increases the risk of uric acid crystallization as monosodium urate with subsequent deposition of the crystals in joints causing gouty arthritis.Loss-of-function mutations in the transporters that mediate uric acid excretion are associated with gout.The ATP-Binding Cassette exporter ABCG2 is important in uric acid excretion at all three sites:kidney(urine),liver(bile),and intestine(feces).Mutations in this transporter cause gout and these mutations occur at significant prevalence in general population.However,mutations that are most prevalent result only in partial loss of transport function.Therefore,if the expression of these partially defective transporters could be induced,the increased number of the transporter molecules would compensate for the mutation-associated decrease in transport function and hence increase uric acid excretion.As such,pharmacologic agents with ability to induce the expression of ABCG2 represent potentially a novel class of drugs for treatment of gouty arthritis.Bojana Ristic Mohd Omar Faruk Sikder Yangzom D.Bhutia Vadivel Ganapathy 2020Asian Journal of Pharmaceutical Sciences2020,15,2:13

3Extraordinary evanescent field confinement waveguide sensor for mid-infrared trace gas spectroscopy显示文摘Nanophotonic waveguides are at the core of a great variety of optical sensors.These structures confine light along defined paths on photonic chips and provide light-matter interaction via an evanescent field.However,waveguides still lag behind free-space optics for sensitivity-critical applications such as trace gas detection.Short optical pathlengths,low interaction strengths,and spurious etalon fringes in spectral transmission are among the main reasons why on-chip gas sensing is still in its infancy.In this work,we report on a mid-infrared integrated waveguide sensor that successfully addresses these drawbacks.This sensor operates with a 107%evanescent field confinement factor in air,which not only matches but also outperforms free-space beams in terms of the per-length optical interaction.Furthermore,negligible facet reflections result in a flat spectral background and record-low absorbance noise that can finally compete with free-space spectroscopy.The sensor performance was validated at 2.566μm,which showed a 7 ppm detection limit for acetylene with only a 2 cm long waveguide.Marek Vlk Anurup Datta Sebastián Alberti Henock Demessie Yallew Vinita Mittal Ganapathy Senthil Murugan Jána Jagerská 2021Light(Science & Applications)2021,10,2:9
4The amino acid transporter SLC6A14 in cancer and its potential use in chemotherapy显示文摘Tumor cells have an increased demand for glucose and amino acids to support their rapid growth,and also exhibit alterations in biochemical pathways that metabolize these nutrients.Transport across the plasma membrane is essential to feed glucose and amino acids into these tumor cell-selective metabolic pathways.Transfer of amino acids across biological membranes occurs via a multitude of transporters;tumor cells must upregulate one or more of these transporters to satisfy their increased demand for amino acids.Among the amino acid transporters,SLC6A14 stands out with specific functional features uniquely suited for the biological needs of the tumor cells.This transporter is indeed upregulated in tumors of epithelial origin,including colon cancer,cervical cancer,breast cancer,and pancreatic cancer.Since normal cells express this transporter only at low levels,blockade of this transporter should lead to amino acid starvation selectively in tumor cells,thus having little effect on normal cells.This offers a novel,yet logical,strategy for the treatment of cancers that are associated with upregulation of SLC6A14.In addition,a variety of amino acid-based prodrugs are recognized as substrates by SLC6A14,thus raising the possibility that anticancer drugs can be delivered into tumor cells selectively via this transporter in the form of amino acid prodrugs.This strategy allows exposure of SLC6A14-positive tumor cells to chemotherapy with minimal off-target effects.In conclusion,the amino acid transporter SLC6A14 holds great potential not only as a direct drug target for cancer therapy but also for tumor cell-selective delivery of anticancer drugs.Yangzom D.Bhutia Ellappan Babu Puttur D.Prasad Vadivel Ganapathy 2014Asian Journal of Pharmaceutical Sciences2014,9,6:8

8Investigation of Li-ion transport in Li7P3S11 and solid-state lithium batteries显示文摘The high Li-ion conductivity of the Li7P3S11 sulfide-based solid electrolyte makes it a promising candidate for all-solid-state lithium batteries. The Li-ion transport over electrode-electrolyte and electrolyteelectrolyte interfaces, vital for the performance of solid-state batteries, is investigated by impedance spectroscopy and solid-state NMR experiments. An all-solid-state Li-ion battery is assembled with the Li7P3S11 electrolyte, nano-Li2S cathode and Li-In foil anode, showing a relatively large initial discharge capacity of 1139.5 m Ah/g at a current density of 0.064 m A/cm^ 2 retaining 850.0 m Ah/g after 30 cycles. Electrochemical impedance spectroscopy suggests that the decrease in capacity over cycling is due to the increased interfacial resistance between the electrode and the electrolyte. 1D exchange ^7Li NMR quantifies the interfacial Li-ion transport between the uncycled electrode and the electrolyte, resulting in a diffusion coefficient of 1.70(3) ×10^-14cm^2/s at 333 K and an energy barrier of 0.132 e V for the Li-ion transport between Li2S cathode and Li7P3S11 electrolyte. This indicates that the barrier for Li-ion transport over the electrode-electrolyte interface is small. 9褐海藻中硫酸多糖的抗菌和抗氧化活性(英文)显示文摘AIMS: Sulfated polysaccharide extracted from the brown algae Sargassum swartzii was studied for antioxidant potential. METHODS: The extracted sulfated polysaccharide was analyzed for physico-chemical characteristics, TAC, reducing power, free radical scavenging potentials (DPPH, ABTS, H2O2 radical) and antibacterial properties. RESULTS: The extract showed a high percentage of carbohydrate (7.40 ± 0.63) %, followed by sulfate (5.3 ± 1.54) %. The highest antioxidant activity was observed in ABTS (55 ± 3.61) %, followed by H2O2 (47.23 ± 2.81) % and DPPH (25.33 ± 2.52) %; significant differences were observed at (P ≥ 0.05). Among the ten human pathogenic strains tested, E. coli was the more sensitive. The characterization and mobility of the sulfated polysaccharide was examined by the FT-IR spectrum and assayed by agarose gel electrophoresis which showed highest mobility at higher pH buffer in carbonate-bicarbonate (pH 10) buffer. The molecular weight of the sulfated polysaccharide was determined by gradient PAGE and was found to be 50 KDa. Finally, GC-MS analysis revealed the presence of peaks corresponding to dimethyl-4-nitroaniline (26.34%).
