| 1 | Effects of emodin on treating murine nonalcoholic fatty liver induced by high caloric laboratory chaw显示文摘AIM: To investigate the effects of emodin on the treatment of non-alcoholic fatty liver in rats induced by high caloric laboratory chaw.METHODS: Non-alcoholic fatty liver model was successfully established by feeding with high caloric laboratory chaw for 12 wk. Then the model rats were randomly divided into 3 groups, namely model control group, emodin group and dietary treatment group. The rats in emodin group in othergroups were given distilled water of the same volume. The rats in model control group were fed with high caloric laboratory chaw while animals in other groups were fed with normal diet. Four weeks later, liver index (liver/body weight ratio), serum activities of liver-associated enzymes, blood lipid, fasting blood glucose, fasting plasma insulin, HOMA insulin resistance index (HOMA-IR), hepatic triglyceride content and histology features of all groups were assayed. The expression of hepatic peroxisomal proliferator activated receptor (PPAR) gamma was determined by RT-PCR.RESULTS: The body weight, liver index, serum activities of alanine aminotransferase (ALT), blood lipid, hepatic triglyceride content of model control group were significantly elevated, with moderate to severe hepatocyte steatosis.The expression of hepatic PPAR gamma mRNA was obviously reduced in model control group. Compared with model control group, the body weight, liver index, serum activities of ALT, blood lipids and hepatic triglyceride of emodin group significantly decreased and hepatic histology display was also greatly improved. Meanwhile, the expression of hepatic PPAR gamma mRNA was elevated.However, high serum activities of ALT and hyperlipidemia were persisted in dietary treatment group although liver index was decreased and liver histology was somewhat improved.CONCLUSION: It is suggested that emodin might be effective in the treatment of non-alcoholic fatty liver in rats. | 3 | Dynamic immune profiling identifies the stronger graft-versus-leukemia (GVL) effects with haploidentical allografts compared to HLA-matched stem cell transplantation显示文摘Haploidentical stem cell transplantation(haplo-SCT)achieves superior or at least comparable clinical outcomes to HLA-matched sibling donor transplantation(MSDT)in treating hematological malignancies.To define the underlying regulatory dynamics,we analyzed time courses of leukemia burden and immune abundance of haplo-SCT or MSDT from multiple dimension.First,we employed two nonirradiated leukemia mouse models which carried human AML-ETO or MLL-AF9 fusion gene to establish haplo-identical and major histocompatibility(MHC)-matched transplantation models and investigated the immune cell dynamic response during leukemia development in vivo.We found that haplo-matching the MHCs of leukemia cells with recipient mouse T cells prolonged leukemic mice survival and reduced leukemia burden.The stronger graft-versus-leukemia activity in haplo-SCT group mainly induced by decreased apoptosis and increased cytotoxic cytokine secretion including tumor necrosis factor–α,interferon-γ,pore-forming proteins and CD107a secreted by T cells or natural killer cells.Furthermore,we conducted a prospective clinical trial which enrolled 135 patients with t(8;21)acute myeloid leukemia that displayed minimal residual disease before transplantation and underwent either haplo-SCT or MSDT.The results showed that the haplo-SCT slowed the kinetics of the leukemia burden in vivo and reduced the cumulative incidence of relapse compared with MSDT.Ex vivo experiments showed that,1 year after transplantation,cytotoxic T lymphocytes from the haplo-SCT group had higher cytotoxicity than those from the MSDT group during the same period.Our results unraveled the role of immune cells in superior antileukemia effects of haplo-SCT compared with MSDT. | Huidong Guo Ying-Jun Chang Yan Hong Lan-Ping Xu Yu Wang Xiao-Hui Zhang Ming Wang Huan Chen Yu-Hong Chen Feng-Rong Wang Wei-Han Yu-Qian Sun Chen-Hua Yan Fei-Fei Tang Xiao-Dong Mo Kai-Yan Liu Xiao-Jun Huang | 2021 | Cellular & Molecular Immunology2021,18,5: | 11 |
| 4 | Homozygosity for Pro of p53 Arg72Pro as a potential risk factor for hepatocellular carcinoma in Chinese population显示文摘AIM: Codon 72 exon 4 polymorphism (Arg72Pro) of the p53 gene has been implicated in cancer risk. Our objective was to investigate the possible association between p53Arg72Pro polymorphism and susceptibility to hepatocellular carcinoma (HCC) among Chinese population.METHODS: The p53 Arg72Pro genotypes were determined by PCR-based restriction fragment length polymorphism (RFLP) analysis in 507 HCC cases and 541 controls. Odds ratios (ORs) for HCC and 95% confidence intervals (CIs)from unconditional logistic regression models were used to evaluate relative risks. Potential risk factors were included in the logistic regression models as covariates in the multivariate analyses on genotype and HCC.RESULTS: The frequencies for Pro and Arg alleles were 44.5%, 55.5% in HCC cases, and 40.3% and 59.7% in controls, respectively. The Pro allele was significantly associated with the presence of HCC (P = 0.05) and had a higher risk for HCC (OR = 1.19, 95% CI 1.00-1.41) as compared with the Arg allele. After adjusted for potential risk factors, Arg/Pro heterozygotes had an 1.21-fold increased risk (95% CI 0.82-1.78, P = 0.34) of HCC compared with Arg homozygotes, whereas the risk for Pro homozygotes was 1.79 (95% CI 1.06-3.01, P = 0.03) times higher than that for Arg homozygotes. Pro-allele carriers had a higher relative risk of HCC than the Arg-only carriers (adjusted OR = 1.33, 95% CI 0.92-1.92, P = 0.13), although the difference was not statistically significant.CONCLUSION: Homozygosity for Pro of p53 Arg72Pro is potentially one of the genetic risk factors for HCC in Chinese population. | 6 | A high-throughput model for screening anti-tumor agents capable of promoting polymerization of tubulin in vitro显示文摘AIM: To establish a high-throughput model for screening anti-tumor agents capable of promoting the polymeriza- tion of tubulin in vitro. METHODS: Tubulin was prepared in different purity for two screening steps. The first step was a high-throughput screening (HTS) for a set of 1500 samples using the GTP-containing tubulin and the end-reading method. The second step was performed on 119 hits from the first screening by a kinetic assay with GTP-lacking tubulin. RESULTS: The HTS for 1500 samples was accomplished in less than 3 h. From the screening, 108 samples were identified with >20 % promotion activity at 10 mg/L. Five of 108 were further confirmed by the kinetic assay using the purified tubulin subsequently. Three of the hit compounds were Epothilone A or its analogs, the other two compounds had new structures with a common pharmacophore for cytotoxic natural products that stabilize microtubules. In an MTT test, the five selected samples from the screening showed a minimal IC50 at0.28±0.06 nmol/Lto Helacells. We propose a novel multi-label classification approach named MFSM(Multi-task joint feature selection for multi-label classification). In MFSM, we compute the asymmetric label correlation matrix in the label space. The multi-label learning problem can be formulated as a joint optimization problem including two regularization terms,one aims to consider the label correlations and the other is used to select the similar sparse features shared among multiple different classification tasks(each for one label).Our model can be reformulated into an equivalent smooth convex optimization problem which can be solved by the Nesterov's method. | 8 | Selection of Chelated Fe(Ⅲ)/Fe(Ⅱ) Catalytic Oxidation Agents for Desulfurization Based on Iron Complexation Method显示文摘Optimization of factors influencing the experiments on reactions involving 8 different chelating agents and soluble Fe(III)/Fe(II) salts was carried out to yield chelated iron complexes. A combination of optimized influencing factors has resulted in a Fe chelating capacity of the iron-based desulfurization solution to be equal to 6.83—13.56 g/L at a redox potential of 0.185—0.3. The desulfurization performance of Fe(III)/Fe(II) chelating agents was investigated on a simulated sulfur-containing industrial gas composed of H2 S and N2 in a cross-flow rotating packed bed. Test results have revealed that the proposed iron-based desulfurization solution showed a sulfur removal efficiency of over 99% along with a Fe chelating capacity exceeding 1.35 g/L. | 9 | Similar Ability of FbaA with M Protein to Elicit Protective Immunity Against Group A Streptococcus Challenge in Mice显示文摘Group A streptococcus(GAS),an important human pathogen,can cause various kinds of infections including superficial infections and potentially lethal infections,and the search for an effective vaccine to prevent GAS infections has been ongoing for many years. This paper compares the immunogenicity and immunoprotection of FbaA(an Fn-binding protein expressed on the surface of GAS) with that of M protein,the best immunogen of GAS. Assay for immune response showed that FbaA,similar to M protein,could induce protein-specific high IgG titer in BALB/c mice. Furthermore,following GAS challenge,the mice immunized with FbaA showed the same protective rate as those with M protein. Pt-4C4-TPP, Pt-4C6-TPP and Pt-4C8-TPP exhibit similar luminescent properties in solution and solid state. Three protonated platinum (II) porphyrins are assembled with mesoporous silica MCM-48, respec- tively, resulting in assembly materials Pt-4Cn-TPP4+/ MCM-48 (n = 4, 6 and 8). The luminescent intensity of Pt-4Cn-TPP4+/MCM-48 can be extremely quenched by molecular oxygen with high sensitivity (I0/I100 > 9). The Stern-Volmer plots of these assembly materials display considerable linearity within a wide range of oxygen concentration (0 to 100%). | 13 | Relationship between increase rate of human plague in China and global climate index as revealed by cross-spectral and cross-wavelet analyses显示文摘Plague has caused the death of hundreds of millions of people throughout the human history.Today this disease is again re-emerging and hence is again becoming an increasing threat to human health in several parts of the world.However,impacts of global climate variation(e.g.El Nino and Southern Oscillation[ENSO])and global warming on plagues are largely unknown.Using cross-spectral analysis and cross-wavelet analysis,we have analyzed the relationship between increase rate of human plague in China during 1871–2003 and the following climate factors(as measured by the Southern Oscillation Index[SOI],Sea Surface Temperature of east Pacific equator[SST]and air Temperature of the Northern Hemisphere[NHT]).We found in the frequency domain that increase rate of human plague was closely associated with SOI and SST.Cross-spectral analysis reveals that significant coherencies between increase rate of human plague and ENSO were found over short periods(2–3 years),medium periods(6–7 years)and long periods(11–12 years,30–40 years).Cross-wavelet analysis reveals that increase rate of human plague oscillates in phase with SOI,but in anti-phase with SST over periods of 2–4 years and approximately 8 years(6–10 years).These results indicate that ENSO-driven climate variation may be important for occurrences of human plague in China.However,there is a need for a further analysis of the underlying mechanism between human plague in China and ENSO. | Zhibin ZHANG Zhenqing LI Yi TAO Min CHEN Xinyu WEN Lei XU Huidong TIAN Nils Chr.STENSETH | 2007 | Integrative Zoology2007,2,3: | 3 |

| 16 | Novel nanoporous binary Ag-Ni electrocatalysts for hydrazine oxidation显示文摘From an aqueous mixture of Ag(I)-EDTA complex and Ni(II) nitrate, silver and nickel particles were co-deposited on the surface of titanium substrates by the hydrothermal method using hydrazine hydrate as a reduction agent. The prepared titanium-supported nano-scale Ag and Ag-Ni particles (nano Ag/Ti, nano Ag86Ni14/Ti, nano Ag77Ni23/Ti, and nano Ag74Ni26/Ti) exhibit nanoporous 3D network textures. Their electrocatalytic activity towards hydrazine oxidation in alkaline solutions was evaluated by cyclic voltammetry and chronoamperometry. The results show that the four samples present a low onset potential of ca. -0.60 V vs. SCE and considerably high and stable anodic current densities for hydrazine oxidation. | 17 | Influence of Si Content on Properties of Ti1-xSixN Coatings显示文摘To investigate the evolution of microstructure and wear behavior of TiSiN coatings with the variation of Si in targets and lays the foundation for its controllable mass production, Ti1-xSixN composite coatings were deposited onto Si(100) and cemented carbide substrates using TiSi targets with different Si content by cathodic arc ion plating. The influences of Si on the microstructure and mechanical properties were studied. Nano-amorphous composite structure appeared in the Ti1-xSixN coatings when Si content in TiSi target was higher than 5 at%. However, further increase of Si content in TiSi target exhibited a negligible effect on the microstructure of Ti1-xSixN coatings. Hardness and deformation resistance were correlated to the content of Si in TiSi targets. Maximum hardness was obtained as the Si content in target increased up to 20 at%. | 18 | A sensitivity study of the WRF model in offshore wind modeling over the Baltic Sea显示文摘Accurate wind modeling is important for wind resources assessment and wind power forecasting. To improve the WRF model configuration for the offshore wind modeling over the Baltic Sea, this study performed a sensitivity study of the WRF model to multiple model configurations, including domain setup,grid resolution, sea surface temperature, land surface data, and atmosphere-wave coupling. The simulated offshore wind was evaluated against LiDAR observations under different wind directions, atmospheric stabilities, and sea status. Generally, the simulated wind profiles matched observations, despite systematic underestimations. Strengthening the forcing from the reanalysis data through reducing the number of nested domains played the largest role in improving wind modeling. Atmosphere-wave coupling further improved the simulated wind, especially under the growing and mature sea conditions.Increasing the vertical resolution, and updating the sea surface temperature and the land surface information only had a slight impact, mainly visible during very stable conditions. Increasing the horizontal resolution also only had a slight impact, most visible during unstable conditions. | 19 | Microstructure control of organic semiconductors via UV-ozone for high-sensitivity NO2 detection显示文摘Developing high sensitive organic semiconductors(OSCs)in organic thin-film transistors(OTFTs)is the key for OTFT based gas sensors.Herein,we report a simple processing route of highly sensitive OSCs for high performance OTFT based nitrogen dioxide(NO2)sensors,where the active OSC layer is based on ultraviolet-ozone(UVO)treated poly(3-hexylthiophene-2,5-diyl)(P3HT).Compared to conventional P3HT based OTFT sensors,the reported device exhibits a remarkable improvement of the gas response from 350%to 30000%.The studies in morphologies,chemical compositions and microstructures of the UVOtreated films reveal that a large number of carrier traps generated in the P3HT films is the decisive reason for the enhancement of sensing performance.Moreover,the optimized device shows great potential of practical applications on the stand points of sensitivity,selectivity,reusability and the ability of recovery,as well as limit of detection of~7.3 ppb.This simple method provides an innovative understanding for the role of the carrier traps in sensing performance and demonstrates a bright future for developing high performance OTFT gas sensors. | HOU SiHui FAN HuiDong WU MengGe YU XinGe YU JunSheng | 2021 | Science China(Technological Sciences)2021,64,5: | 2 |
| 20 | Experimental study and simulation of a three-phase flow stirred bioreactor显示文摘In order to obtain the reasonable operating conditions and minimize the power consumption in the stirred bioreactor, the hydrodynamic experiments in the stirred bioreactor have been taken to obtain the basic data. Subsequently, an Eulerian model for the gas–liquid–solid three phase flow in the stirred bioreactor has been proposed and the CFD simulation has been conducted. By comparing the results of experiment and simulation, it can be concluded that the simulation results were consistent with the experimental data. The inner relationship between operating variables and indicators could be obtained by comparing the results of just suspension speed, gas holdup, power consumption and operational maps, further the reasonable operating conditions could be also determined under the minimum power consumption.
