1The linkage maps of Dendrobium species based on RAPD and SRAP markers显示文摘Dendrobium plants are used commonly as tonic herbs and health food in many Asian countries,especially in China.Here we report the genetic map construction of two Dendrobium species with a double pseudo-testcross strategy using random amplified polymorphic DNA(RAPD) and sequence-related amplified polymorphism(SRAP) markers.A F1 mapping population of 90 individuals was developed from a cross between D.officinale and D.hercoglossum.A total of 307 markers,including 209 RAPD and 98 SRAP,were identified and used for genetic linkage group(LG) analysis.The D.officinale linkage map consisted of 11 major linkage groups and 3 doublets,which covered 629.4 cM by a total of 62 markers with an average locus distance of 11.2 cM between two adjacent markers.The D.hercoglossum linkage map contained 112 markers mapped on 15 major and 4 minor linkage groups,spanning a total length of 1,304.6 cM with an average distance of 11.6 cM between two adjacent markers.The maps constructed in this study covered 92.7% and 82.7% of the D.hercoglossum and D.officinale genomes respectively,providing an important basis for the mapping of horticultural and medicinal traits and for the application of marker-assisted selection in Dendrobium breeding program.Dawei Xue Shangguo Feng Hongyan Zhao Hua Jiang Bo Shen Nongnong Shi Jiangjie Lu Junjun Liu Huizhong Wang 2010Journal of Genetics and Genomics2010,37,3:26

3The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 5THEORETICAL MODEL OF EFFECTIVE STRESS COEFFICIENT FOR ROCK/SOIL-LIKE POROUS MATERIALS显示文摘Physical mechanisms and influencing factors on the effective stress coeffcient for rock/soil-like porous materials are investigated,based on which equivalent connectivity index is proposed.The equivalent connectivity index,relying on the meso-scale structure of porous ma-terial and the property of liquid,denotes the connectivity of pores in Representative Element Area(REA).If the conductivity of the porous material is anisotropic,the equivalent connec-tivity index is a second order tensor.Based on the basic theories of continuous mechanics and tensor analysis,relationship between area porosity and volumetric porosity of porous materials is deduced.Then a generalized expression,describing the relation between effective stress coeff-cient tensor and equivalent connectivity tensor of pores,is proposed,and the expression can be applied to isotropic media and also to anisotropic materials.Furthermore,evolution of porosity and equivalent connectivity index of the pore are studied in the strain space,and the method to determine the corresponding functions in expressions above is proposed using genetic algorithm and genetic programming.Two applications show that the results obtained by the method in this paper perfectly agree with the test data.This paper provides an important theoretical support to the coupled hydro-mechanical research.Kai Zhang Hui Zhou Dawei Hu Yang Zhao Xiating Feng 2009Acta Mechanica Solida Sinica2009,22,3:5
6New insights into the associations among feed efficiency, metabolizable efficiency traits and related QTL regions in broiler chickens显示文摘Background: Improving the feed efficiency would increase profitability for producers while also reducing the environmental footprint of livestock production. This study was conducted to investigate the relationships among feed efficiency traits and metabolizable efficiency traits in 180 male broilers. Significant loci and genes affecting the metabolizable efficiency traits were explored with an imputation-based genome-wide association study. The traits measured or calculated comprised three growth traits, five feed efficiency related traits, and nine metabolizable efficiency traits.Results: The residual feed intake(RFI) showed moderate to high and positive phenotypic correlations with eight other traits measured, including average daily feed intake(ADFI), dry excreta weight(DEW), gross energy excretion(GEE), crude protein excretion(CPE), metabolizable dry matter(MDM), nitrogen corrected apparent metabolizable energy(AMEn), abdominal fat weight(Ab F), and percentage of abdominal fat(Ab P). Greater correlations were observed between growth traits and the feed conversion ratio(FCR) than RFI. In addition, the RFI, FCR, ADFI, DEW,GEE, CPE, MDM, AMEn, Ab F, and Ab P were lower in low-RFI birds than high-RFI birds(P < 0.01 or P < 0.05), whereas the coefficients of MDM and MCP of low-RFI birds were greater than those of high-RFI birds(P < 0.01). Five narrow QTLs for metabolizable efficiency traits were detected, including one 82.46-kb region for DEW and GEE on Gallus gallus chromosome(GGA) 26, one 120.13-kb region for MDM and AMEn on GGA1, one 691.25-kb region for the coefficients of MDM and AMEn on GGA5, one region for the coefficients of MDM and MCP on GGA2(103.45–103.53 Mb), and one 690.50-kb region for the coefficient of MCP on GGA14. Linkage disequilibrium(LD) analysis indicated that the five regions contained high LD blocks, as well as the genes chromosome 26 C6 orf106 homolog(C26 H6 orf106), LOC396098, SH3 and multiple ankyrin repeat domains 2(SHANK2), ETS homologous factor(EHF), and histamine receptor H3-like(HRH3 L), which are known to be involved in the regulation of neurodevelopment, cell proliferation and differentiation, and food intake.Conclusions: Selection for low RFI significantly decreased chicken feed intake, excreta output, and abdominal fat deposition, and increased nutrient digestibility without changing the weight gain. Five novel QTL regions involved in the control of metabolizable efficiency in chickens were identified. 7Domesticated cynomolgus monkey embryonic stem cells allow the generation of neonatal interspecies chimeric pigs显示文摘Blastocyst complementation by pluripotent stem cell(PSC)injection is believed to be the most promising method to generate xenogeneic organs.However,ethical issues prevent the study of human chimeras in the late embryonic stage of development.Primate embryonic stem cells(ESCs),which have similar pluripotency to human ESCs,are a good model for studying interspecies chimerism and organ generation.However,whether primate ESCs can be used in xenogenous grafts remains unclear.In this study,we evaluated the chimeric ability of cynomolgus monkey(Macaca fascicularis)ESCs(cmESCs)in pigs,which are excellent hosts because of their many similarities to humans.We report an optimized culture medium that enhanced the anti-apoptotic ability of cmESCs and improved the development of chimeric embryos,in which domesticated cmESCs(D-ESCs)injected into pig blastocysts differentiated into cells of all three germ layers.In addition,we obtained two neonatal interspecies chimeras,in which we observed tissue-specific D-ESC differentiation.Taken together,the results demonstrate the capability of D-ESCs to integrate and differentiate into functional cells in a porcine model,with a chimeric ratio of 0.001-0.0001 in different neonate tissues.We believe this work will facilitate future developments in xenogeneic organogenesis,bringing us one step closer to producing tissue-specific functional cells and organs in a large animal model through interspecies blastocyst complementation.Rui Fu Dawei Yu Jilong Ren Chongyang Li Jing Wang Guihai Feng Xuepeng Wang Haifeng Wan Tianda Li Libin Wang Ying Zhang Tang Hai Wei Li Qi Zhou 2020Protein & Cell2020,11,2:4
8Co-delivery of resveratrol and docetaxel via polymeric micelles to improve the treatment of drug-resistant tumors显示文摘Co-delivery of anti-cancer drugs is promising to improve the efficacy of cancer treatment.This study was aiming to investigate the potential of concurrent delivery of resveratrol(RES)and docetaxel(DTX)via polymeric nanocarriers to treat breast cancer.To this end,methoxyl poly(ethylene glycol)-poly(D,L-lactide)copolymer(mPEG-PDLA)was prepared and characterized using FTIR and 1H NMR,and their molecular weights were determined by GPC.Isobologram analysis and combination index calculation were performed to find the optimal ratio between RES and DTX to against human breast adenocarcinoma cell line(MCF-7 cells).Subsequently,RES and DTX were loaded in the mPEG-PDLA micelles simultaneously,and the morphology,particle size distribution,in vitro release,pharmacokinetic profiles,as well as cytotoxicity to the MCF-7 cells were characterized.IC50 of RES and DTX in MCF-7 cells were determined to be 23.0μg/ml and 10.4μg/ml,respectively,while a lower IC50 of 4.8μg/ml of the combination of RES and DTX was obtained.The combination of RES and DTX at a ratio of 1:1(w/w)generated stronger synergistic effect than other ratios in the MCF-7 cells.RES and DTX loaded mPEG-PDLA micelles exhibited prolonged release profiles,and enhanced cytotoxicity in vitro against MCF-7 cells.The AUC(0→t)of DTX and RES in mPEG-PDLA micelles after i.v.administration to rats were 3.0-fold and 1.6-fold higher than that of i.v.injections of the individual drugs.These findings indicated that the co-delivery of RES and DTX using mPEG-PDLA micelles could have better treatment of tumors.Xiong Guo Zhiyue Zhao Dawei Chen Mingxi Qiao Feng Wan Dongmei Cun Yi Sun Mingshi Yang 2019Asian Journal of Pharmaceutical Sciences2019,14,1:4
9A HYDRO-MECHANICAL-CHEMICAL COUPLING MODEL FOR GEOMATERIAL WITH BOTH MECHANICAL AND CHEMICAL DAMAGES CONSIDERED显示文摘A general framework of hydro-mechanical-chemical coupling model is proposed for geomaterial subjected to the dual effects of mechanical loading and chemical degradation. Mechanical damage due to microcracks in solid matrix and chemical damage induced by the increase of porosity due to dissolution of matrix minerals as well as their interactions are considered. A special model is proposed for sandstone. The reaction rate is formulated within the framework of mineral reaction kinetics and can thus take into account different dissolution mechanisms of three main mineral compositions under different pH values. The increase of porosity is physically defined by the dissolution of mineral composition and the chemical damage is related to the increase of porosity. The mechanical behavior is characterized by unified plastic damage and viscoplastic damage modeling. The effective stress is used for describing the effect of pore pressure. The elastic parameters and plastic evolution as well as viscoplastic evolution are dependent on chemical damage. The advection, which is coupled with mechanical damage and chemical damage, is considered as the dominant mechanism of mass transfer. The application of model proposed is from decoupled experiments to fully coupled experiment. 12Light Intensity Affects Chlorophyll Synthesis During Greening Process by Metabolite Signal from Mitochondrial Alternative Oxidase in Arabidopsis显示文摘Although mitochondrial alternative oxidase(AOX)has been proposed to play essential roles in high light stress tolerance,the effects of AOX on chlorophyll synthesis are unclear.Previous studies indicated that during greening,chlorophyll accumulation was largely delayed in plants whose mitochondrial cyanide-resistant respiration was inhibited by knocking out nuclear encoded AOX gene.Here we show that this delay of chlorophyll accumulation was more significant under high light condition.Inhibition of cyanide-resistant respiration was also accompanied by the increase of plastid NADPH/NADP^+ratio,especially under high light treatment which subsequently blocked the import of multiple plastidial proteins,such as some components of the photosynthetic electron transport chain,the Calvin-Benson cycle enzymes and malate/oxaloacetate shuttle components.Over expression of AOXla rescued the aoxla mutant phenotype,including the chlorophyll accumulation during greening and plastidial protein import.It thus suggests that light intensity affects chlorophyll synthesis during greening process by a metabolic signal,the AOX-derived plastidial NADPH/NADP^+ratio change.And our results thus revealed a molecular mechanism of chloroplast-mitochondria interactions.ZHANG Dawei YUAN Shu 徐飞 ZHU Feng YUAN Ming YE Huaxun GUO Hongqing LV Xin YIN Yanhai 林宏辉 2015武汉生物工程学院学报2015,,3:2
