1Wnt/b-catenin signaling plays an ever-expanding role in stem cell self-renewal,tumorigenesis and cancer chemoresistance显示文摘Wnt signaling transduces evolutionarily conserved pathways which play important roles in initiating and regulating a diverse range of cellular activities,including cell proliferation,calcium homeostasis,and cell polarity.The role of Wnt signaling in controlling cell proliferation and stem cell self-renewal is primarily carried out through the canonical pathway,which is the best-characterized the multiple Wnt signaling branches.The past 10 years has seen a rapid expansion in our understanding of the complexity of this pathway,as many new components of Wnt signaling have been identified and linked to signaling regulation,stem cell functions,and adult tissue homeostasis.Additionally,a substantial body of evidence links Wnt signaling to tumorigenesis of cancer types and implicates it in the development of cancer drug resistance.Thus,a better understanding of the mechanisms by which dysregulation of Wnt signaling precedes the development and progression of human cancer may hasten the development of pathway inhibitors to augment current therapy.This review summarizes and synthesizes our current knowledge of the canonical Wnt pathway in development and disease.We begin with an overview of the components of the canonical Wnt signaling pathway and delve into the role this pathway has been shown to play in stemness,tumorigenesis,and cancer drug resistance.Ultimately,we hope to present an organized collection of evidence implicating Wnt signaling in tumorigenesis and chemoresistance to facilitate the pursuit of Wnt pathway modulators that may improve outcomes of cancers in which Wnt signaling contributes to aggressive disease and/or treatment resistance.Maryam K.Mohammed Connie Shao Jing Wang Qiang Wei Xin Wang Zachary Collier Shengli Tang Hao Liu Fugui Zhang Jiayi Huang Dan Guo Minpeng Lu Feng Liu Jianxiang Liu Chao Ma Lewis L.Shi Aravind Athiviraham Tong-Chuan He Michael J.Lee 2016Genes & Diseases2016,3,1:70

4长链脂肪乳剂及中/长链脂肪乳剂水解速率的比较研究显示文摘目的 :比较长链脂肪乳剂、物理混合的中 /长链脂肪乳剂及结构脂肪乳剂的水解速率。 方法 :应用 1 1 4 C甘油棕榈酸标记的长链三酰甘油 (LCT)和 2 ,3 3 H甘油辛酸标记的中链三酰甘油 (MCT) ,分别标记Intralipid ,Lipo fundin及Structolipid ,在体外添加缓冲液、脂蛋白酯酶和肝酯酶后 ,置 37℃温箱中水浴、孵育。采用气相色谱仪测定核素标记的各种脂肪酸及三酰甘油浓度 ,游离脂肪酸浓度用放射性核素扫描计数测定 ,通过计算培养液中1 4 C标记的长链脂肪酸及3 H标记的中链脂肪酸分别在1 4 C标记LCT及3 H标记MCT中的比例 ,来计算三种脂肪乳剂的水解率。 结果 :物理混合的中 /长链脂肪乳剂及结构脂肪乳剂的水解程度明显高于长链脂肪乳剂 ,差异有统计学意义。物理混合的中 /长链脂肪乳剂水解程度高于结构脂肪乳剂 ,但在水解程度及水解速度上却无统计学差异。 结论 :物理混合的中吴国豪 Connie Jarstrand Jorgen Nordenstrom 2002肠外与肠内营养2002,9,1:13
5晚期卵巢癌、输卵管癌及原发性腹膜癌腹腔镜肿瘤细胞减灭术的安全性及有效性研究显示文摘目的总结晚期卵巢癌、输卵管癌和腹膜癌患者行腹腔镜初次或间歇性肿瘤细胞减灭术的临床经验。方法回顾分析接受腹腔镜肿瘤细胞减灭术的晚期卵巢癌(FIGOⅡc期以上)、输卵管癌和原发性腹膜癌患者临床资料。结果 32名患者接受腹腔镜评估手术。17例接受全腹腔镜肿瘤细胞减灭术,其中88.2%为满意的肿瘤细胞减灭术;11例腹腔镜评估后转开腹肿瘤细胞减灭术,其中72.7%为满意的肿瘤细胞减灭术;4例仅进行活检和(或)姑息手术。腹腔镜组平均随访时间19.7月9,例无瘤生存6,例带瘤生存2,例因肿瘤死亡。开腹组平均随访时间25.8月,3例无瘤生存,5例带瘤生存,3例因肿瘤死亡。腹腔镜组术中失血量较少,术后住院时间较短(P=0.008和P=0.03),但手术时间及并发症发生率与开腹组相比无统计学差异。中位复发时间,腹腔镜组为31.7月,开腹组为21.5个月(P=0.3)。结论对于经过精心挑选的晚期卵巢癌、输卵管癌和原发性腹膜癌病例,采用腹腔镜进行诊断、分期和肿瘤细胞减灭术,在技术上是可行的。李萌 熊光武 Farr R.Nezhat Shaghayegh M.DeNoble Connie S.Liu Jennifer E.Cho Douglas N.Brown Linus Chuang Herbert Gretz Prakash Saharia 2012中国微创外科杂志2012,12,2:11

8Antimicrobial activity of green tea extract against isolates of methicillin-resistant Staphylococcus aureus and multi-drug resistant Pseudomonas aeruginosa显示文摘Objective:To evaluate antibacterial activity of the Indonesian water soluble green tea extract,Camellia sinensis,against clinical isolates of methicillin-resistantStaphylococcus aureus (S.aureus)(MRSA)and multi-drug resistant Pseudomonas aeruginosa(MDR-P.aeruginosa).Methods:Antimicrobial activity of green tea extract was determined by the disc diffusion method and the minimum inhibitory concentration(MIC)was determined by the twofold serial broth dilutions method.The tested bacteria using in this study were the standard strains and multi-drug resistant clinical isolates of S.aureus and P.aeruginosa,obtained from Laboratory of Clinical Microbiology,Faculty of Medicine,University of Indonesia.Results:The results showed that the inhibition zone diameter of green tea extracts forS.aureus ATCC 25923 and MRSA were(18.970依0.287)mm,and(19.130依0.250)mm respectively.While the inhibition zone diameter forP.aeruginosa ATCC 27853 and MDR-P.aeruginosawere(17.550依0.393)mm and(17.670依0.398)mm respectively.The MIC of green tea extracts againstS.aureus ATCC 25923 and MRSA were 400μg/mL and 400μg/mL,respectively,whereas the MIC for P.aeruginosa ATCC 27853 and MDR-P.aeruginosawere 800μg/mL,and 800μg/mL,respectively.Conclusions:Camellia sinensisleaves extract could be useful in combating emerging drug-resistance caused by MRSA andP.aeruginosa.Maksum Radji Rafael Adi Agustama Berna Elya Conny Riana Tjampakasari 2013Asian Pacific Journal of Tropical Biomedicine2013,3,8:8

15Mechanistic links between gut microbial community dynamics, microbial functions and metabolic health显示文摘Gut microbes comprise a high density, biologically active community that lies at the interface of an animal with its nutritional environment. Consequently their activity profoundly influences many aspects of the physiology and metabolism of the host animal. A range of microbial structural components and metabolites directly interact with host intestinal cells and tissues to influence nutrient uptake and epithelial health. Endocrine, neuronal and lymphoid cells in the gut also integrate signals from these microbial factors to influence systemic responses. Dysregulation of these host-microbe interactions is now recognised as a major risk factor in the development of metabolic dysfunction. This is a two-way process and understanding the factors that tip host-microbiome homeostasis over to dysbiosis requires greater appreciation of the host feedbacks that contribute to regulation of microbial community composition. To date, numerous studies have employed taxonomic profiling approaches to explore the links between microbial composition and host outcomes(especially obesity and its comorbidities), but inconsistent host-microbe associations have been reported. Available data indicates multiple factors have contributed to discrepancies between studies. These include the high level of functional redundancy in host-microbiome interactions combined with individual variation in microbiome composition; differences in study design, diet composition and host system between studies; and inherent limitations to the resolution of r RNA-based community profiling. Accounting for these factors allows for recognition of the common microbial and host factors driving community composition and development of dysbiosis on high fat diets. 18Metronomic chemotherapy for non-metastatic triple negative breast cancer: Selection is the key显示文摘Triple negative breast cancer(TNBC) accounts for 15%-20% of all breast cancer, and is still defined as what it is not. Currently, TNBC is the only type of breast cancer for which there are no approved targeted therapies and maximum tolerated dose chemotherapy with taxanes and anthracycline-containing regimens is still the standard of care in both the neoadjuvant and adjuvant settings. In the last years, metronomic chemotherapy(MC) is being explored as an alternative to improve outcomes in TNBC. In the neoadjuvant setting, purely metronomic and hybrid approaches have been developed with the objective of increasing complete pathologic response(p CR) and prolonging disease free survival. These regimens proved to be very effective achieving pC R rates between 47%-60%, but at the cost of great toxicity. In the adjuvant setting, MC is used to intensify adjuvant chemotherapy and, more promisingly, as maintenance therapy for high-risk patients, especially those with no pC R after neoadjuvant chemotherapy. Considering the dismal prognosis of TNBC, any strategy that potentially improves outcomes, specially being the oral agents broadly available and inexpensive, should be considered and certainly warrants further exploration.
