4Autoantibody to MOG suggests two distinct clinical subtypes of NMOSD显示文摘We characterized a unique group of patients with neuromyelitis optica spectrum disorder(NMOSD) who carried autoantibodies of aquaporin-4(AQP4) and myelin-oligodendrocyte glycoprotein(MOG). Among the 125 NMOSD patients, 10(8.0%) were AQP4- and MOG-ab double positive, and 14(11.2%) were MOG-ab single positive. The double-positive patients had a multiphase disease course with a high annual relapse rate(P=0.0431), and severe residual disability(P<0.0001). Of the double-positive patients, 70% had MS-like brain lesions, more severe edematous, multifocal regions on spinal magnetic resonance imaging(MRI), pronounced decreases of retinal nerve fiber layer thickness and atrophy of optic nerves. In contrast, patients with only MOG-ab had a higher ratio of monophasic disease course and mild residual disability. Spinal cord MRI illustrated multifocal cord lesions with mild edema, and brain MRIs showed more lesions around lateral ventricles. NMOSD patients carrying both autoantibodies to AQP4 and MOG existed and exhibited combined features of prototypic NMO and relapsing-remitting form of MS, whereas NMOSD with antibodies to MOG only exhibited an 'intermediate' phenotype between NMOSD and MS. 5QTL editing confers opposing yield performance in different rice varieties显示文摘Grain yield is one of the most important and complex trait for genetic improvement in crops; it is known to be controlled by a number of genes known as quantitative trait loci(QTLs). In the past decade, many yield-contributing QTLs have been identified in crops.However, it remains unclear whether those QTLs confer the same yield performance in different genetic backgrounds. 7Rapid growth cost in “all-fish” growth hormone gene transgenic carp:Reduced critical swimming speed显示文摘Evidence has accumulated that there is a trade-off between benefits and costs associated with rapid growth. A trade-off between growth rates and critical swimming speed (Ucrit) had been also reported to be common in teleost fish. We hypothesize that growth acceleration in the F3 generation of 'all-fish' growth hormone gene (GH) transgenic common carp (Cyprinus carpio L.) would reduce the swimming abilities. Growth and swimming performance between transgenic fish and non-transgenic controls were compared. The results showed that transgenic fish had a mean body weight 1.4―1.9-fold heavier,and a mean specific growth rate (SGR) value 6%―10% higher than the controls. Transgenic fish,however,had a mean absolute Ucrit (cm/s) value 22% or mean relative Ucrit (BL/s) value 24% lower than the controls. It suggested that fast-growing 'all-fish' GH-transgenic carp were inferior swimmers. 8Fingolimod alters inflammatory mediators and vascular permeability in intracerebral hemorrhage显示文摘Intracerebral hemorrhage(ICH) leads to high rates of death and disability. The pronounced infl ammatory reactions that rapidly follow ICH contribute to disease progression. Our recent clinical trial demonstrated that oral administration of an immune modulator fingolimod restrained secondary injury derived from initial hematoma,but the mechanisms remain unknown. In this study,we aim to investigate the effects of fingolimod on inflammatory mediators and vascular permeability in the clinical trial of oral fingolimod for intracerebral hemorrhage(ICH). The results showed that fingolimod decreased the numbers of circulating CD4^+ T,CD8^+ T,CD(19)^+B,NK,and NKT cells and they recovered quickly after the drug was stopped. The plasma ICAM level was decreased and IL-10 was increased by fingolimod. Interestingly,fingolimod protected vascular permeability as indicated by a decreased plasma level of MMP9 and the reduced r T1%. In conclusion,modulation of systemic inflammation by fingolimod demonstrates that it is an effective therapeutic agent for ICH. 9Rapid generation of genetic diversity by multiplex CRISPR/Cas9 genome editing in rice显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated endonuclease 9(CRISPR/Cas9) system has emerged as a promising technology for specific genome editing in many species. Here we constructed one vector targeting eight agronomic genes in rice using the CRISPR/Cas9 multiplex genome editing system. By subsequent genetic transformation and DNA sequencing, we found that the eight target genes have high mutation efficiencies in the T_0 generation. Both heterozygous and homozygous mutations of all editing genes were obtained in T_0 plants. In addition, homozygous sextuple, septuple, and octuple mutants were identified. As the abundant genotypes in T_0 transgenic plants, various phenotypes related to the editing genes were observed. 10Distributed Scheduling Problems in Intelligent Manufacturing Systems显示文摘Currently,manufacturing enterprises face increasingly fierce market competition due to the various demands of customers and the rapid development of economic globalization.Hence,they have to extend their production mode into distributed environments and establish multiple factories in various geographical locations.Nowadays,distributed manufacturing systems have been widely adopted in industrial production processes.In recent years,many studies have been done on the modeling and optimization of distributed scheduling problems.This work provides a literature review on distributed scheduling problems in intelligent manufacturing systems.By summarizing and evaluating existing studies on distributed scheduling problems,we analyze the achievements and current research status in this field and discuss ongoing studies.Insights regarding prior works are discussed to uncover future research directions,particularly swarm intelligence and evolutionary algorithms,which are used for managing distributed scheduling problems in manufacturing systems.This work focuses on journal papers discovered using Google Scholar.After reviewing the papers,in this work,we discuss the research trends of distributed scheduling problems and point out some directions for future studies.Yaping Fu Yushuang Hou Zifan Wang Xinwei Wu Kaizhou Gao Ling Wang 2021Tsinghua Science and Technology2021,26,5:9
11Characterization and Fine Mapping of a Novel Rice Narrow Leaf Mutant nal9显示文摘A narrow leaf mutant was isolated from transgenic rice(Oryza sativa L.)lines carrying a T-DNA insertion.The mutant is characterized by narrow leaves during its whole growth period,and was named nal9(narrow leaf 9).The mutant also has other phenotypes,such as light green leaves at the seedling stage,reduced plant height,a small panicle and increased tillering.Genetic analysis revealed that the mutation is controlled by a single recessive gene.A hygromycin resistance assay showed that the mutation was not caused by T-DNA insertion,so a map-based cloning strategy was employed to isolate the nal9 gene.The mutant individuals from the F2generations of a cross between the nal9 mutant and Longtepu were used for mapping.With 24 F2mutants,the nal9 gene was preliminarily mapped near the marker RM156 on the chromosome 3.New INDEL markers were then designed based on the sequence differences between japonica and indica at the region near RM156.The nal9 gene was fnally located in a 69.3 kb region between the markers V239B and V239G within BAC OJ1212_C05 by chromosome walking.Sequence and expression analysis showed that an ATP-dependent Clp protease proteolytic subunit gene(ClpP)was most likely to be the nal9 gene.Furthermore,the nal9 mutation was rescued by transformation of the ClpP cDNA driven by the 35S promoter.Accordingly,the ClpP gene was identifed as the NAL9 gene.Our results provide a basis for functional studies of NAL9 in future work.Wei Li Chao Wu Guocheng Hu Li Xing Wenjing Qian Huamin Si Zongxiu Sun Xingchun Wang Yaping Fu Wenzhen Liu 2013Journal of Integrative Plant Biology2013,55,11:8
12Microsatellite polymorphisms of Sichuan golden monkeys显示文摘Previous study using protein electrophoresis shows no polymorphism in 44 nuclear loci of Sichuan golden monkey (Rhinopithecus roxellana), which limits our understandings of its population genetic patterns in the nuclear genome. In order to obtain sufficient information, we scanned 14 microsatellite loci in a sample of 32 individuals from its three major habitats (Minshan, Qinling and Shen-nongjia). A considerable amount of polymorphisms were detected. The average heterozygosities in the local populations were all above 0.5. The differentiations among local populations were significant. There was evidence of geneflow among subpopulations, but geneflow between Qinling and Shennongjia local populations was the weakest. Minshan and Qinling populations might have gone through recent bottlenecks. The estimation of the ratio of the effective population sizes among local populations was close to that from census sizes. Comparisons to available mitochondria data suggested that R. roxellana’s social structures played an important role in shaping its population genetic patterns. 13Yellow light dilemma zone researches: a review显示文摘The yellow light dilemma zone is widely known as an area on the high-speed intersection approach,where vehicles neither safely stop before the stop line nor proceed through the intersection during amber interval. Within such an area,a vehicle might be involved in a right-angle crash or rear-end collision. This issue has been extensively discussed over five decades in traffic engineering field,covering fromtheory to practice. However,fewcomprehensive reviewliteratures on the amber signal dilemma zone problems can be found. The objective of this paper is to summarize the evolution of yellow light dilemma zone researches. Basic definition and boundary of dilemma zone followed by driver behavior and dilemma zone hazard measurement are depicted. 14Neural Stem Cells Transfected with Leukemia Inhibitory Factor Promote Neuroprotection in a Rat Model of Cerebral Ischemia显示文摘Leukemia inhibitory factor(LIF) contributes to the neuroprotection by neural stem cells(NSCs) after ischemic stroke. Our aim was to explore whether LIFtransfected NSCs(LIF-NSCs) can ameliorate brain injury and promote neuroprotection in a rat model of cerebral ischemia. To accomplish this goal, we transfected NSCs with a lentivirus carrying the LIF gene to stably overexpress LIF. The LIF-NSCs reduced caspase 3 activation under conditions of oxygen-glucose deprivation in vitro.Transient cerebral ischemia was induced in rats by 2 h of middle cerebral artery occlusion(MCAo), and LIF-NSCs were intravenously injected at 6 h post-ischemia. LIF-NSC treatment reduced the infarction volume and improved neurological recovery. Moreover, LIF-NSCs improved glial cell regeneration and ameliorated white matter injuryin the MCAo rats. 17Beclin-1/LC3-II dependent macroautophagy was uninfluenced in ischemia-challenged vascular endothelial cells显示文摘Autophagy has been extensively studied and occurs in many biological settings.However,a question remains as to whether ischemia enhances Beclin-1/LC3-II-dependent macroautophagy in vascular endothelial cells,as has been previously thought.Furthermore,the effect of the level of autophagy on cell or skin flap survival still requires elucidation.We created a lethal ischemia model in human umbilical vascular endothelial cells(HUVECs),performed quantitative proteomics and bioinformatics analyses,and verified the autophagic status and effect both in vitro and in vivo.The significantly upregulated proteins encoded by autophagy-related genes(ATGs)included ATG2A,ATG3,ATG4B,ATG5,ATG7,ATG9A,ATG12,ATG16,and ATG101.The significantly enhanced lysosomal proteins were cathepsin B,cathepsin D,lysosome-associated membrane protein 1(LAMP1),and LAMP2.However,the differentially expressed proteins excluded Beclin-1,microtubule-associated protein light chain 3(LC3)-I,and LC3-II.Western blot analyses verified that the protein expression levels of Beclin-1,LC3-I,and LC3-II were neither upregulated nor downregulated in ischemia-challenged HUVECs.The autophagic status was not enhanced by rapamycin in ischemic HUVECs but appeared to be inhibited by chloroquine.Our in vivo study on rats showed that a downregulation in autophagic status jeopardized skin flap survival.In conclusion,Ischemia neither enhanced nor inhibited Beclin-1/LC3-II-dependent canonical macroautophagy both in vitro and in vivo,in contradiction to previous studies.An appropriate autophagic homeostasis can minimize cell or skin flap damage.Yaping Ma Chaofan Li Yan He Tiwei Fu Li Song Qingsong Ye Fugui Zhang 2022Genes & Diseases2022,9,2:3

19Young Yum pill inhibits inflammatory mediators and nuclear factor-kappa B signaling in lipopolysaccharide-stimulated RAW 264.7 macrophages显示文摘OBJECTIVE: To investigate the effect of Young Yum pill (YYP) on inflammatory mediators in cultured RAW 264.7 cells and elucidate the nuclear factor- kappa B (NF-κB)-related mechanism behind the action. METHODS: YYP was extracted with 95% ethanol Lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages were used to evaluate the effect of YYP on inflammatory mediators. Production of nitric oxide (NO) and prostaglandin E2 (PGE2) were measured by Griess test and enzyme-linked immunosorbent assay, respectively. The levels of genes and proteins involved in the generation of inflammatory mediators were examined using real-time polymerase chain reaction and Western blotting, respectively. RESULTS: YYP dose-dependently suppressed LPS-induced production of NO, PGE2 and tumor necrosis factor-α(TNF-α), and elevation of mRNA and protein levels of inducible NO synthase and cyclooxygenase- 2 in RAW 264.7 macrophages. These observations were associated with decreased NF-κB p65 phosphorylation and nuclear localization, enhanced Akt (protein kinase B) phosphorylation, as well as reduced inhibitor of κB (IκB)α degradation and IκB kinase α/β phosphorylation. 20Ribonuclease H-like gene SMALL GRAIN2 regulates grain size in rice through brassinosteroid signaling pathway显示文摘Grain size is a key agronomic trait that determines the yield in plants.Regulation of grain size by brassinosteroids(BRs)in rice has been widely reported.However,the relationship between the BR signaling pathway and grain size still requires further study.Here,we isolated a rice mutant,named small grain2(sg2),which displayed smaller grain and a semi-dwarf phenotype.The decreased grain size was caused by repressed cell expansion in spikelet hulls of the sg2 mutant.Using map-based cloning combined with a Mut Map approach,we cloned SG2,which encodes a plant-specific protein with a ribonuclease H-like domain.SG2 is a positive regulator downstream of GLYCOGEN SYNTHASE KINASE2(GSK2)in response to BR signaling,and its mutation causes insensitivity to exogenous BR treatment.Genetical and biochemical analysis showed that GSK2 interacts with and phosphorylates SG2.We further found that BRs enhance the accumulation of SG2 in the nucleus,and subcellular distribution of SG2 is regulated by GSK2 kinase activity.In addition,Oryza sativa OVATE family protein 19(OsOFP19),a negative regulator of grain shape,interacts with SG2 and plays an antagonistic role with SG2 in controlling gene expression and grain size.Our results indicated that SG2 is a new component of GSK2-related BR signaling response and regulates grain size by interacting with Os OFP19.Yunshuai Huang Hui Dong Changling Mou Ping Wang Qixian Hao Min Zhang Hongmin Wu Fulin Zhang Tengfei Ma Rong Miao Kai Fu Yaping Chen Ziyan Zhu Cheng Chen Qikai Tong Zhuoran Wang Shirong Zhou Xi Liu Shijia Liu Yunlu Tian Ling Jiang Jianmin Wan 2022Journal of Integrative Plant Biology2022,64,10:1
