3Genomic landscape and genetic manipulation of the black soldier fly Hermetia illucens,a natural waste recycler显示文摘The black soldier fly(BSF),Hermetia illucens(Diptera:Stratiomyidae),is renowned for its bioconversion of organic waste into a sustainable source of animal feed.We report a high-quality genome of 1.1 Gb and a consensus set of 16,770 gene models for this beneficial species.Compared to those of other dipteran species,the BSF genome has undergone a substantial expansion in functional modules related to septic adaptation,including immune system factors,olfactory receptors,and cytochrome P450s.We further profiled midgut transcriptomes and associated microbiomes of BSF larvae fed with representative types of organic waste.We find that the pathways related to digestive system and fighting infection are commonly enriched and that Firmicutes bacteria dominate the microbial community in BSF across all diets.To extend its potential practical applications,we further developed an efficient CRISPR/Cas9-based gene editing approach and implemented this to yield flightless and enhanced feeding capacity phenotypes,both of which could expand BSF production capabilities.Our study provides valuable genomic and technical resources for optimizing BSF lines for industrialization.Shuai Zhan Gangqi Fang Minmin Cai Zongqing Kou Jun Xu Yanghui Cao Liang Bai Yixiang Zhang Yongmao Jiang Xingyu Luo Jian Xu Xia Xu Longyu Zheng Ziniu Yu Hong Yang Zhijian Zhang Sibao Wang Jeffery K.Tomberlin Jibin Zhang Yongping Huang 2020Cell Research2020,30,1:9
4A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 5A high-strength self-healing nano-silica hydrogel with anisotropic differential conductivity显示文摘Soft nano electronic materials based on conductive hydrogels have attracted considerable attention due to their exceptional properties.Particle deposition and poor interface compatibility often diminish the mechanical strength and electron transport capabilities of the conductive hydrogel.Mechanical damage can severely impact the performance of the conductive hydrogel and can even damage electronic devices based on the conductive hydrogel.In the current study,a transparent nano-silica hydrogel is prepared by employing an extremely easy-to-operate method.This approach can preclude the deposition of particles via strong mechanical force.In addition,controlling the concentration of the reaction interface makes the hydrogel grow along the mechanical force in the direction with a special directional hole structure formed.The hydrogel is transparent,showing excellent self-healing properties—it can self-heal within 15 seconds.Remarkably,the hydrogel after self-healing maintains its performance.Moreover,it has excellent mechanical properties and can be stretched in length.Up to 1,200%of the original length,the tensile strength of the gel spline can reach 7 MPa.The viscosity of the hydrogel can reach 1.67×10^(8)(MPs).In addition,a large amount of Na+in this hydrogel endow it a conductivity of 389 ps/cm.The conductivity of this hydrogel is adjustable result from the special pore structure.Lastly,the difference between the horizontal and vertical conductivity of the same sample can reach 3-4 times,thus this hydrogel can be used in the field of nano conductive materials.Xingyu Huang Xiaofan Zhou Hao Zhou Yidan Zhong Hui Luo Fan Zhang 2021Nano Research2021,14,8:1
6Cerium and carbon-sulfur codoped mesoporous TiO_(2)nanocomposites for boosting visible light photocatalytic activity显示文摘Ce and C-S codoped mesoporous TiO_(2)nanocomposites were synthesized via a sol-gel method integrated with an evaporation-induced self-assembly approach.The basic physicochemical characteristics of the synthetic samples were analyzed via a series of characterization techniques.The results reveal that C-S and Ce codoping on mesoporous TiO_(2)enhances the photocatalytic activity owing to the synergistic effect caused by narrowing the band gap,enhancing adsorption,trapping and transferring the excited e^(-)/h^(+)pairs and suppressing the recombination of e^(-)/h^(+)pairs.Furthermore,the obtained C,S-TiO_(2)/CeO_(2)materials exhibit large specific surface areas and numerous pores which not only effectively improve the adsorption-enrichment capability,but also supply multi-dimensional mass and electron transfer channels.The photodegradation efficiency of RhB by C,S-TiO_(2)/CeO_(2)within 40 min is nearly 100%,and its degradation efficiency is 6.63 times that of undoped TiO_(2).Recycling experiments show that mesoporous C,S-TiO_(2)/CeO_(2)shows excellent recoverability and stability.Furthermore,by trapping experiments,·O_(2)e^(-)/h^(+)and·OH are the predominant active species and a possible reaction mechanism is proposed.Fei Zheng Faqin Dong Lin Zhou Jieyu Yu Xijie Luo Xingyu Zhang Zhenzhen Lv Luman Jiang Yuheng Chen Mengqing Liu 2023Journal of Rare Earths2023,41,4:1
7An immunoassay based on lab-on-a-chip for simultaneous and sensitive detection of clenbuterol and ractopamine显示文摘Both clenbuterol(CLB)and ractopamine(RAC)areβ-adrenergic agonists.After long-term excessive intake,there will be adverse reactions such as headache,chest tightness,limb numbness,and serious lifethreatening.Simultaneous detection of CLB and RAC in related samples is of great importance for human health.In this work,we outline a microfluidics-based indirect competitive immunoassay(MICI)system that can sensitively detect residual CLB and RAC in pork,swine blood and swine urine.The rapid detection of multiple samples can be achieved in one chip,which greatly improves the detection efficiency.This method has good stability and reproducibility and the microfluidic chips are easy to manufacture.The linear ranges for CLB and RAC detection by MICI are 0.1-2.5 ng/mL and 0.1-5 ng/mL,and the limits of detection(LODs)are 0.094 ng/mL and 0.091 ng/mL,respectively.This straightforward and portable immunoassay system provides a good platform for rapid detection of harmful substances in food samples.Qi Wang Jinqi Deng Yiping Chen Yunjing Luo Xingyu Jiang 2020Chinese Chemical Letters2020,31,7:1
8Relationship between Renying pulse augmentation index and Cunkou pulse condition in different blood pressure groups显示文摘OBJECTIVE: To explore the relationship between Renying pulse (carotid) augmentation index (AI) and Cunkou pulse condition in different blood pressure groups, and the clinical significance of Renying and Cunkou pulse parameters to reflect vascular function.METHODS:Eighty-sixpatients with essential hypertension (EH) and 52 individuals with normal blood pressure(control group) between September 2010 and January 2012 were included in thisstudy.Renying pulse AI was examined by a new diagnostic tool(ALOKA ProSound Alpha 10) — wave intensity (WI) that is calculated as the product of the derivatives of the simultaneously recorded blood pressure changes(dP/dt) and blood-flow-velocity changes(dU/dt), while Cunkou pulse condition was detected by DDMX-100 Pulse Apparatus inboth EH and control groups. A multifactorial correlation analysis was performed for data analysis.RESULTS: After adjustingfor potentialconfoundingvariables,intheEHgroup,AIwaspositivelycorrelated with t5, w2/t(rt5=0.225, P<0.05; rw2/t=0.230, P<0.05)and negatively correlated with h5,h5/h1 and w2(rh5=﹣0.393,P<0.01;rh5/h1=﹣0.444,P<0.01;rw2=﹣0.389,P<0.01). In the control group, AI was positively correlated with t3, t4, t5 and w1(rt3=0.595, P<0.01; rt4=0.292, P<0.05; rt5=0.318, P<0.05; rw1=0.541, P<0.01)and negatively correlated with h1,h2,h3,AdandA(rh1=﹣0.368,P<0.05;rh2=﹣0.330,P<0.05;rh3=﹣0.327, P<0.05; rAd=﹣0.322, P<0.05; rA=﹣0.410, P<0.01). In the total sample group(EH plus control group, n=138), AI was positively correlated with t, t5, w1 and w2/t(rt=0.257,P<0.01;rt5=0.266,P<0.01;rw1=0.184,P<0.05; rw2/t=0.210, P<0.05) and negatively correlated with h5, h5/h1, w2 and Ad(rh5=﹣0.230, P<0.01; rh5/h1=﹣0.218, P<0.05; rw2=﹣0.267, P<0.01; rAd=﹣0.246,P<0.01). Multiple linear regression analysis was carried out to model the relationship(F=7.887, P<0.001).CONCLUSION:Renying pulse AI can effectively predict arterial stiffness in synchrony with the manifestations of Cunkou pulse in elderly patients with hypertension. Cunkou pulse apparatus is a valuable tool for evaluating AI in clinical practice. 10IRE1α regulates the PTHrP-IHH feedback loop to orchestrate chondrocyte hypertrophy and cartilage mineralization显示文摘Cartilage development is controlled by the highly synergistic proliferation and differentiation of growth plate chondrocytes,in which the Indian hedgehog(IHH)and parathyroid hormone-related protein-parathyroid hormone-1 receptor(PTHrP-PTH1R)feedback loop is crucial.The inositol-requiring enzyme 1a/X-box-binding protein-1 spliced(IRE1α/XBP1s)branch of the unfolded protein response(UPR)is essential for normal cartilage development.However,the precise role of ER stress effector IRE1α,encoded by endoplasmic reticulum to nucleus signaling 1(ERN1),in skeletal development remains unknown.Herein,we reported that loss of IRE1α accelerates chondrocyte hypertrophy and promotes endochondral bone growth.ERN1 acts as a negative regulator of chondrocyte proliferation and differentiation in postnatal growth plates.Its deficiency interrupted PTHrP/PTH1R and IHH homeostasis leading to impaired chondrocyte hypertrophy and differentiation.XBP1s,produced by p-IRE1α-mediated splicing,binds and up-regulates PTH1R and IHH,which coordinate cartilage development.Meanwhile,ER stress cannot be activated normally in ERN1-deficient chondrocytes.In conclusion,ERN1 deficiency accelerates chondrocyte hypertrophy and cartilage mineralization by impairing the homeostasis of the IHH and PTHrP/PTH1R feedback loop and ER stress.ERN1 may have a potential role as a new target for cartilage growth and maturation.Mengtian Fan Nana Geng Xingyue Li Danyang Yin Yuyou Yang Rong Jiang Cheng Chen Naibo Feng Li Liang Xiaoli Li Fengtao Luo Huabing Qi Qiaoyan Tan Yangli Xie Fengjin Guo 2024Genes & Diseases2024,11,1:0
11Evaluation of early liquid drinking after radical gastrectomy in gastric cancer:a Chinese multicenter propensity score matching analysis显示文摘Background:Enhanced recovery after surgery is used in gastrointestinal surgery.This study aimed to access the effects of early liquid drinking(ELD)on gastrointestinal function recovery in patients with gastric cancer(GC)who underwent radical gastrectomy,as high-quality evidence on the outcomes of ELD after gastrectomy is currently lacking.Methods:Clinicopathological data of patients with GC from 11 centers were retrospectively analysed.Clinical outcomes were investigated in 555 patients,including 225 who started drinking liquid within 48 h(ELD group)of surgery and 330 who started drinking liquid after flatus resumption(traditional liquid drinking[TLD]group).Propensity score matching(PSM)analysis was performed using a match ratio of 1:1 and 201 patients were selected from each group for the analysis.Primary outcome was time to first passage of flatus.Secondary outcomes included time to first defecation,post-operative hospitalization days,occurrence of short-term post-operative complications,and hospitalization costs.Results:After PSM,baseline characteristics were not significantly different between the two groups.The time to first flatus(2.7261.08 vs 3.3661.39 days),first defecation(4.3461.85 vs 4.7761.61 days),and post-operative hospital stay(8.2764.02 vs 12.9464.43 days)were shorter in the ELD group than in the TLD group(all P<0.05).The ELD group had lower hospitalization costs than the TLD group([7.8362.44 vs 8.7863.41]104 RMB,P=0.041).No significant differences were observed in the incidence of post-operative complications.Conclusions:Compared with TLD,post-operative ELD could promote rapid recovery of gastrointestinal function and reduce hospitalization costs;moreover,ELD does not increase the risk of post-operative complications.Yue Zhang Kaixiong Tao Jinlong Yu Chao Chen Quan Zheng Sanlin Lei Xiaogang Zhong Lixin Liu Wei Wang Qiang Wang En Li Yuwen Luo Guanrong Zhang Xingyu Feng Yong Li Junjiang Wang 2023Gastroenterology Report2023,11,1:0
12Electronic structure of LaIrIn5and f-electron character in its related Ce-115 compounds显示文摘LaIrIn5 is a reference compound of the heavy-fermion superconductor LaIrIn5.The lack of f electrons in LaIrIn5 indicates that there should not be any f electron participating in the construction of its Fermi surface.Thus the electronic structure comparison between LaIrIn5 and LaIrIn5 provides a good platform to study the properties of f electrons.Here angle-resolved photoemission spectroscopy(ARPES)measurements and density functional theory(DFT)calculations are performed to study the electronic structures of LaIrIn5 and LaIrIn5.We find the valence band structures of the two materials are similar to each other,except for the absence of f bands in LaIrIn5.By analyzing the Fermi crossings of the three conduction bands of the two materials quantitatively,we find the volumes of the electron pocketsαandβaround the M′point become larger from LaIrIn5 to LaIrIn5,while the hole pocketγaround theΓ′point becomes smaller.Together with the calculation results,we confirm that this is mainly originated from the f-electron contribution,while the lattice-constant difference between LaIrIn5 and LaIrIn5 only has a finite influence.We also give a summary of the f-electron character in its related Ce-115 heavy fermion compounds.Our results may be essential for the complete microscopic understanding of the 115 compounds and the related heavy-fermion systems.Rui Zhou XueBing Luo ZhaoFeng Ding Lei Shu XingYu Ji ZiHao Zhu YaoBo Huang DaWei Shen ZhengTai Liu ZhongHao Liu Yun Zhang QiuYun Chen 2020Science China(Physics,Mechanics & Astronomy)2020,63,11:0
13Anisotropic magnetoelastic response in the magnetic Weyl semimetal Co_(3)Sn_(2)S_(2)显示文摘Co_(3)Sn_(2)S_(2) is a recently identified magnetic Weyl semimetal in Shandite compounds. Upon cooling, Co_(3)Sn_(2)S_(2) undergoes a ferromagnetic transition with c-axis polarized moments(0.3 μ_(B)/Co) around T_(C)= 175 K, followed by another magnetic anomaly around T_(A)≈ 140 K. A large intrinsic anomalous Hall effect is observed in the magnetic state below TC with a maximum of anomalous Hall angle near T_(A). Here, we report an elastic neutron scattering on the crystalline lattice of Co_(3)Sn_(2)S_(2) in a magnetic field up to 10 T. A strongly anisotropic magnetoelastic response is observed, while only a slight enhancement of the Bragg peaks is observed when B//c. The in-plane magnetic field(B//ab) dramatically suppresses the Bragg peak intensity probably by tilting the moments and lattice toward the external field direction. The in-plane magnetoelastic response commences from T_(C), and as it is further strengthened below T_(A), it becomes nonmonotonic against the field between T_(A) and T_(C) because of the competition from another in-plane magnetic order. 14Determination of Aspartame Content in Liquor by High Performance Liquid Chromatography显示文摘[Objectives]This study was conducted to establish a method for the determination of aspartame in liquor by high performance liquid chromatography.[Methods]The liquor samples having volumes fixed with ultrapure water were filtered by 0.22μm water phase,and then the content of aspartame in liquor was determined by high performance liquid chromatography using methanol(A)and ultrapure water(B)as mobile phase for isocratic elution.[Results]The results showed that a good linearity was obtained for the standard curve(R^(2)=99.98%)in the method,and its quantitative limit was 5.34 mg/kg.Its recovery was 91.26%,91.92%,and 90.55%,respectively.[Conclusions]The method has high sensitivity,high recovery and low quantitative limit,and it was a suitable method for the determination of aspartame in liquor.Zhouli ZHANG Xingyu LUO Ping YONG Lan LIAO Lili YIN Yaoxin LI Min ZHANG Da YI 2022Agricultural Biotechnology2022,11,2:0
15Exploring the nature of black hole and gravity with an imminent merging binary of supermassive black holes显示文摘A supermassive binary black-hole candidate SDSS J1430+2303 reported recently motivates us to investigate an imminent binary of supermassive black holes as potential gravitational wave source, and the radiated gravitational waves at the end of the merger are shown to be in the band of space-borne detectors. We provide a general analysis on the required detecting sensitivity needed for probing such type gravitational wave sources and make a full discussion by considering two typically designed configurations of space-borne antennas. If a source is so close, it is possible to be detected with Taiji pathfinder-plus which is proposed to be an extension for the planned Taiji pathfinder by just adding an additional satellite to the initial two satellites. 17Establishment of highly efficient transgenic system for black soldier fly(Hermetia illucens)显示文摘The black soldier fly(BSF),Hermetia illucens,is a promising insect for miti-gating solid waste problems as its larvae are able to bioconvert organic waste into valuable biomass.We recently reported a high-quality genome assembly of the BSF;analysis of this genome sequence will further the understanding of insect biology and identify genes that can be manipulated to improve efficiency of bioconversion.To enable genetic manip-ulation of the BSF,we have established the first transgenic methods for this economically important insect.We cloned and identified the ubiquitous actin5C promoter(Hiactin5C-p3k)and 3 endogenous U6 promoters(HiU6:1,HiU6:2,and HiU6:3).The Hiactin5C pro-moter was used to drive expression of a hyperactive variant of the piggyBac transposase,which exhibited up to 6-fold improvement in transformation rate when compared to the wild-type transposase.Furthermore,we evaluated the 3 HiU6 promoters using this trans-genic system.HiU6:1 and HiU6:2 promoters provided the highest knockdown efficiency with RNAi and are thus promising candidates for future Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)development.Overall,our findings provide valuable genetic engineering toolkits for basic research and genetic manipulation of the BSF.Zongqing Kou Xingyu Luo Yuguo Jiang Bihui Chen Yu Song Yaohui Wang Jun Xui Jeffery K.Tomberlin Yongping Huang 2023Insect Science2023,30,4:0
18Intersex is required for female sexual development in Hermetia illucens显示文摘Sex-determination pathways are extremely diverse.Understanding the mechanism of sex determination in insects is important for genetic manipulation of the pest population and for breeding of economically valuable insects.Although sex determi-nation has been well characterized in the model species Drosophila melanogaster,little is known about this pathway in Stratiomyidae.In the present study,we first identified the Drosophila intersex(ix)homolog in Hermetia illucens,also known as the black soldier fly,which belongs to the Stratiomyidae family and which is an important insect for the con-version of various organic wastes.Phylogenetic analyses and multiple sequence alignment revealed that Hix is conserved compared with Drosophila.We showed that Hix is highly expressed in internal genitalia.Disruption of the Hix gene using CRISPR/Cas9 resulted in female-specific defects in external genitalia and abnormal and undersized ovaries.Taken together,our study furthers our understanding of sex determination in insects and could facilitatebreedingof H.illucens.Bihui Chen Zongqing Kou Yuguo Jiang Xingyu Luo Peili Li Kaj Sun Weiwei Wang Yongping Huang Yaohui Wang 2023Insect Science2023,30,4:0
19Designing a deep-UV nonlinear optical monofluorophosphate显示文摘As structural variants of famous hexagonal tungsten bronzes,hexagonal tungsten oxides(HTO)represent an important family with fascinating functional properties,such as piezoelectric,ferroelectric,pyroelectric,and nonlinear optical(NLO)properties.However,none of them are transparent in the deep-UV spectral region,which limits their applications.Herein,we report the first HTO-type monofluorophosphate K_(3)Sc_(3)(PO_(4))(PO_(3)F)_(2)F_(5)(I)with deep-UV transparency.Such a monofluorophosphate is NLO-active with a phase-matchable powder second harmonic generation efficiency of 0.9 times that of KH_(2)PO_(4)at 1,064 nm.Importantly,the UV-Vis reflectance spectrum indicates that it is deep-UV transparent down to 200 nm.This work pushes the transparent window of NLO materials with HTO-type structures down to the deep-UV spectral region for the first time and opens up a new door for HTO materials.Qingran Ding Xingyu Zhang Zheshuai Lin Zheyao Xiong Yusong Wang Xifa Long Sangen Zhao Maochun Hong Junhua Luo 2022Science China Chemistry2022,65,9:0
20Ectopic mineralization-inspired cell membrane-based matrix vesicle analogs for in-depth remineralization of dentinal tubules for treating dentin hypersensitivity显示文摘The invasion of etched dentinal tubules(DTs)by external substances induces dentin hypersensitivity(DH).The deep and compact occlusion of DTs is highly desirable for treating DH but still challenging due to the limited penetrability and mineralization capacities of most current desensitizers.Matrix vesicles(MVs)participate in the regulation of ectopic mineralization.Herein,ectopic MV analogs are prepared by employing natural cell membranes to endow mineral precursors with natural biointerfaces and integrated biofunctions for stimulating dentin remineralization.The analogs quickly access DTs(>20μm)in only 5 min and further penetrate deep into the interior of DTs(an extraordinary~200μm)in 7 days.Both in vitro and in vivo studies confirm that the DTs are efficiently sealed by the newly formed minerals(>50μm)with excellent resistance to wear and acid erosion,which is significantly deeper than most reported values.After repair,the microhardness of the damaged dentin can be recovered to those of healthy dentin.For the first time,cell membrane coating nanotechnology is used as a facile and efficient therapy for in-depth remineralization of DTs in treating DH with thorough and long-term effects,which provides insights into their potential for hard tissue repair.Mingjing Li Xiaoran Zheng Zhiyun Dong Yuyue Zhang Wei Wu Xingyu Chen Chunmei Ding Jiaojiao Yang Jun Luo Jianshu Li 2023Nano Research2023,16,5:0
