| 5 | Correlation between BRAF^(V600E) mutation and clinicopathological features in pediatric papillary thyroid carcinoma显示文摘In adults, the presence of the BRAF^(V600E) mutation in papillary thyroid cancer(PTC) has been demonstrated to be strongly associated with aggressive cancer-cell characteristics and poor patient prognosis. In contrast, the frequency of this mutation in pediatric PTC has undergone limited study, and the few available estimates range from 0 to 63%. Furthermore, the role of the BRAF^(V600E) mutation in pediatric PTC is controversial; thus, the present study aimed to investigate the prevalence and role of the BRAF^(V600E) mutation in48 pediatric patients with PTC, aged 3–13 years. Of these patients, 41 were diagnosed with classic PTC, five were found to have a follicular variant of PTC, and two to exhibit a diffuse sclerosing PTC variant. The BRAF^(V600E) mutation was identified to be present in 35.4% of the 48 analyzed patients, and in 41.5% of the patients diagnosed with classical PTC. Furthermore, the presence of the BRAF^(V600E) mutation was found to be associated with a patient age at diagnosis of less than ten years(P=0.011), the performance of a thyroidectomy(P=0.03), exhibited tumor multifocality(P=0.02) and/or extra-thyroidal invasion(P=0.003), and both a low MACIS(Metastases, Age, Completeness of resection, Invasion, Size)(P=0.036) and AMES(Age, Metastasis, Extent of tumor,Size)(P=0.001)score. | 10 | Multiplex detection of KRAS and BRAF mutations using cationic conjugated polymers显示文摘The mutation detections of KRAS and BRAF genes are of significant importance to predict the responses to anti-cancer therapy and develop new drugs. In this paper, we developed a multi-step fluorescence resonance energy transfer (FRET) assay for multiplex detection of KRAS and BRAF mutations using cationic conjugated polymers (CCP). The newly established detection system could detect as low as 2% mutant DNAs in DNA admixtures. By triggering the emission intensity change of CCP and the dyes labeled in the DNA, four possible statuses (three mutations and one wildtype) can be differentiated in one extension reaction. The detection efficiency of this new method in clinical molecular diagnosis was validated by determining KRAS and BRAF mutations of 51 formalin-fixed paraffin-embedded (FFPE) ovary tissue samples. Furthermore, the result of the CCP-based multi-step FRET assay can be directly visualized under UV light so that no expensive instruments and technical expertise are needed. As search engines continuously update their indexes, the query results in long-lived cache entries may become stale. It is important to provide the refresh mechanism to enhance the degree of freshness of cached results. We present a pre-judgment approach to improve the freshness of the result cache and design an incomplete allocation algorithm. We introduce the queryTime-to-live(TTL) and term-TTL structure to pre-judge the result cache. The query-TTL is used to pre-check the likelihood of a cache hit and term-TTL is applied to maintain all terms of the latest posting list. For the cache structure, we design a Queue-Hash structure and develop the corresponding incomplete allocation algorithm. In this work, we construct the graphene-based bilayers BG/X that consists by the CoN_(4) embedded graphene as the upper layer and the X modified graphene as the bottom layer(X = Si, P, S). The interfacial bonding between CoN_(4) site and the X dopant is spontaneously formed due to the strong pd hybridization, which changes the Co ligand from the planar-four N_(4) coordination into spatial-five N_(4)+X one. The additive glue atom weakens too strong adsorptions of the ORR intermediates on CoN_(4) site and thereby improves the ORR activities in comparison with the monolayer counterpart. From the free energy profiles, the overpotentials η are 0.47, 0.49 and 0.45 V for BG/Si_(a), BG/P_(a) and BG/S_(a), respectively,being comparable to that of state-of-the-art Pt material. Besides, the kinetic barriers for the bilayers are less than 0.75 eV, an indicative of the room temperature activity. Furthermore, the combination of thermodynamic and kinetic analysis ensures the preference of 4e^(-)-OOH associative mechanism over 2e^(-)-H_(2)O_(2) mechanism, being beneficial for membrane stability against the H_(2)O_(2) corrosion.
