1Puerarin protects brain tissue against cerebral ischemia/reperfusion injury by inhibiting the inflammatory response显示文摘Puerarin, a traditional Chinese medicine, exerts a powerful neuroprotective effect in cerebral ischemia/reperfusion injury, but its mechanism is unknown. Here, we established rat models of middle cerebral artery ischemia/reperfusion injury using the suture method. Puerarin(100 mg/kg) was administered intraperitoneally 30 minutes before middle cerebral artery occlusion and 8 hours after reperfusion. Twenty-four hours after reperfusion, we found that puerarin significantly improved neurological deficit, reduced infarct size and brain water content, and notably diminished the expression of Toll-like receptor-4, myeloid differentiation factor 88, nuclear factor kappa B and tumor necrosis factor-α in the ischemic region. 3MicroRNA-663 induces immune dysregulation by inhibiting TGF-β1 production in bone marrow-derived mesenchymal stem cells in patients with systemic lupus erythematosus显示文摘Mesenchymal stem cells(MSCs)are critical for immune regulation.Although several microRNAs(miRNAs)have been shown to participate in autoimmune pathogenesis by affecting lymphocyte development and function,the roles of miRNAs in MSC dysfunction in autoimmune diseases remain unclear.Here,we show that patients with systemic lupus erythematosus(SLE)display a unique miRNA signature in bone marrow-derived MSCs(BMSCs)compared with normal controls,among which miR-663 is closely associated with SLE disease activity.MiR-663 inhibits the proliferation and migration of BMSCs and impairs BMSC-mediated downregulation of follicular T helper(Tfh)cells and upregulation of regulatory T(Treg)cells by targeting transforming growth factorβ1(TGF-β1).MiR-663 overexpression weakens the therapeutic effect of BMSCs,while miR-663 inhibition improves the remission of lupus disease in MRL/lpr mice.Thus,miR-663 is a key mediator of SLE BMSC regulation and may serve as a new therapeutic target for the treatment of lupus.Linyu Geng Xiaojun Tang Kangxing Zhou Dandan Wang Shiying Wang Genhong Yao Weiwei Chen Xiang Gao Wanjun Chen Songtao Shi Nan Shen Xuebing Feng Lingyun Sun 2019Cellular & Molecular Immunology2019,16,3:16

5Three dimensional velocity structure and accurate earthquake location in Changning–Gongxian area of southeast Sichuan显示文摘In order to understand the crustal structure and tectonic background of the Changning–Gongxiang area, southeastern Sichuan Province, where a series of moderate-to-strong earthquakes occurred in recent years, we utilized the seismic phase data both from a local dense array and from the regional seismic networks;we used the tomoDD program to invert for the high-resolution three-dimensional velocity structure within the depth range of 0–10 km and for accurate hypocentral locations in this area. We analyzed the seismogenic structures for the events of Xingwen M5.7 in 2018 and Gongxian M5.3 and Changning M6.0 in 2019. The results show that:(1) widespread lateral inhomogeneity exists in the velocity structure of the study area, and the location of the velocity anomaly is largely consistent with known structures. In the range of distinguishable depth, the inhomogeneity decreases with increasing depth, and the velocity structure anomalies in some areas are continuous in depth;(2) earthquakes occurred in clusters, showing the characteristics of zonal folding trends in the NW-SE and NE-SW directions;the focal depth in the area is generally shallow in both the sedimentary cap and the crystalline basement. The seismogenic structures of small earthquake clusters are different in size and occurrence in different sections, and the clusters occurred mostly in regions with high P-or S-wave velocities;(3) synthesis of a variety of data suggests that the seismogenic structures of the Xingwen M5.7 and Changning M6.0 earthquakes are associated with slip faults that trend NW-SE in, respectively, the south wing and the axis of the Changning–Shuanghe anticline, while that of the Gongxian M5.3 earthquake is associated with thrust faults that trend N-S in the Jianwu syncline region. 6The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 7IL-25 stimulates M2 macrophage polarization and thereby promotes mitochondrial respiratory capacity and lipolysis in adipose tissues against obesity显示文摘Obesity and associated metabolic diseases are characterized by a chronic low-grade inflammatory state with the infiltration of many inflammatory cells,especially macrophages.Immune molecules,including some cytokines,have a close relationship with metabolism.Interleukin(IL)-25 is a member of the IL-17 cytokine family that can regulate macrophages and alleviate some metabolic dysfunction;however,its role and mechanisms in lipid metabolism remain to be extensively clarified.Human serum and liver biopsy specimens,high-fat diet-induced obesity mice and DB/DB(Lepr−/−)animal models were used to examine IL-25 expression in obesity and nonalcoholic fatty liver diseases(NAFLD).To observe the role of IL-25 in lipid metabolism,model mice were administered with IL-25 or adoptively transferred with IL-25-educated macrophages in vivo,whereas bone marrow-derived macrophages,the macrophage cell line RAW264.7 and adipocytes differentiated from 3T3-L1 were used in vitro.IL-25 was decreased in NAFLD patients and obese mice.In addition,IL-25 reduced body weight gain and lipid accumulation,enhanced lipid uptake by macrophages and increased the expression of lipolysis andβ-oxidation enzymes via alternatively activating macrophages.IL-25 also promoted lipolysis and suppressed lipogenesis in adipocytes co-cultured with the IL-25-educated macrophages.Furthermore,IL-25 improved the mitochondrial respiratory capacity and oxygen consumption rate of macrophages and produced more NAD+/NADH and ATP.In conclusion,IL-25 can stimulate M2 macrophage polarization and thereby promote lipolysis and mitochondrial respiratory capacity,highlighting the potential for IL-25 to be used as a therapeutic agent against obesity and associated metabolic syndromes.Juan Feng Lingyi Li Zhiying Ou Qiao Li Baoyong Gong Zhenxian Zhao Weiwei Qi Ti Zhou Jun Zhong Weibin Cai Xia Yang Aiping Zhao Guoquan Gao Zhonghan Yang 2018Cellular & Molecular Immunology2018,15,5:10
8A Single-Site Positioning Method Based on TOA and DOA Estimation Using Virtual Stations in NLOS Environment显示文摘This paper presents a single-site positioning method based on the joint estimation of propagation time-of-arrival(TOA) and direction-of-arrival(DOA), with the assist of virtual stations in the typical non-line-of-sight(NLOS) environment. Consider the influence of multipath noise on the positioning performance, the proposed method firstly presents a modified high-resolution estimation technique called Multipath noise Limiting Matrix Pencil(MLMP) algorithm to achieve the TOA/DOA estimations, in which the matrix pencil and matrix enhancement process are implemented to deal with the measurements from the uniform linear array(ULA) receiver. Meanwhile, the subspace dimension estimation is improved via an adaptive threshold, for enhancing the performance of high-resolution techniques in low signal-noise-ration(SNR) situation. Next the proposed method generates virtual stations utilizing the known floor plan of surrounding reflectors, and adopts a weighted Least Square(WLS) position estimator to calculate the required position, combining the TOA/DOA estimations with the location of virtual stations. 9Dek42 encodes an RNA-binding protein that affects alternative pre-m RNA splicing and maize kernel development显示文摘RNA-binding proteins (RBPs) play an important role in post-transcriptional gene regulation. However, the functions of RBPs in plants remain poorly understood. Maize kernel mutant dek42 has small defective kernels and lethal seedlings. Dek42 was cloned by Mutator tag isolation and further confirmed by an independent mutant allele and clustered regularly interspaced short palindromic repeats (CRISPR)-CRISPR-associated protein 9 materials. Dek42 encodes an RRM_RBM48 type RNA-binding protein that localizes to the nucleus. Dek42 is constitutively expressed in various maize tissues. The dek42 mutation caused a significant reduction in the accumulation of DEK42 protein in mutant kernels. RNA-seq analysis showed that the dek42 mutation significantly disturbed the expression of thousands of genes during maize kernel development. Sequence analysis also showed that the dek42 mutation significantly changed alternative splicing in expressed genes, which were especially enriched for the U12-type intron-retained type. Yeast two-hybrid screening identified SF3a1 as a DEK42-interacting protein. DEK42 also interacts with the spliceosome component U1-70K. These results suggested that DEK42 participates in the regulation of pre-messenger RNA splicing through its interaction with other spliceosome components. 10Micro-droplet Digital Polymerase Chain Reaction and Real-Time Quantitative Polymerase Chain Reaction Technologies Provide Highly Sensitive and Accurate Detection of Zika Virus显示文摘The establishment of highly sensitive diagnostic methods is critical in the early diagnosis and control of Zika virus(ZIKV)and in preventing serious neurological complications of ZIKV infection. In this study, we established micro-droplet digital polymerase chain reaction(ddPCR) and real-time quantitative PCR(RT-qPCR) protocols for the detection of ZIKV based on the amplification of the NS5 gene. For the ZIKV standard plasmid, the RT-qPCR results showed that the cycle threshold(Ct) value was linear from 10~1 to 10~8 copy/l L, with a standard curve R^2 of 0.999 and amplification efficiency of 92.203%;however, a concentration as low as 1 copy/l L could not be detected. In comparison with RT-qPCR, the dd PCR method resulted in a linear range of 10~1–10~4 copy/l L and was able to detect concentrations as low as 1 copy/l L. Thus, for detecting ZIKV from clinical samples, RT-qPCR is a better choice for high-concentration samples(above 10~1 copy/l L),while ddPCR has excellent accuracy and sensitivity for low-concentration samples. 13Advances in tumor-endothelial cells co-culture and interaction on microfluidics显示文摘The metastasis in which the cancer cells degrade the extracellular matrix(ECM) and invade to the surrounding and far tissues of the body is the leading cause of mortality in cancer patients. With a lot of advancement in the field, yet the biological cause of metastasis are poorly understood. The microfluidic system provides advanced technology to reconstruct a variety of in vivo-like environment for studying the interactions between tumor cells(TCs) and endothelial cells(ECs). This review gives a brief account of both two-dimensional models and three-dimensional microfluidic systems for the analysis of TCs-ECs coculture as well as their applications to anti-cancer drug screening. 14Emission of intermediate volatility organic compounds from a ship main engine burning heavy fuel oil显示文摘Intermediate volatility organic compounds(IVOCs)are crucial precursors of secondary organic aerosol(SOA).In this study,gaseous IVOCs emitted from a ship main engine burning heavy fuel oil(HFO)were investigated on a test bench,which could simulate the real-world operations and emissions of ocean-going ships.The chemical compositions,emission factors(EFs)and volatility distributions of IVOC emissions were investigated.The results showed that the main engine burning HFO emitted a large amount of IVOCs,with average IVOC EFs of 20.2-201 mg/kg-fuel.The IVOCs were mainly comprised of unspeciated compounds.The chemical compositions of exhaust IVOCs were different from that of HFO fuel,especially for polycyclic aromatic compounds and alkylcyclohexanes.The volatility distributions of IVOCs were also different between HFO exhausts and HFO fuel.The distinctions in IVOC emission characteristics between HFO exhausts and HFO fuel should be considered when assessing the IVOC emission and related SOA formation potentials from ocean-going ships burning HFO,especially when using fuel-surrogate models.Haijun Lou Yuejiao Hao Weiwei Zhang Penghao Su Fan Zhang Yingjun Chen Daolun Feng Yifan Li 2019Journal of Environmental Sciences2019,31,10:5

18Microbial alteration of natural gas in Xinglongtai field of the Bohai Bay Basin, China显示文摘Xinglongtai field has been an important petroleum-producing area of Liaohe Depression for 30 years. Oil exploration and production were the focus of this field, but the gas was ignored. This study examined twenty five gas samples with the purpose of determining the gas genetic types and their geochemical characteristics. Molecular components, stable carbon isotopic compositions and light hydrocarbons were also measured, and they proved that microbial activity has attacked some of the gas components which resulted in unusual carbon isotopic distributions. Propane seems to be selectively attacked during the initial stage of microbial alteration, with abnormally lower con-tent compared to that of butane as well as anomalously heavy carbon isotope. As a consequence, the carbon isotopic distribution among the gas components is partially reverse, as δ13C1<δ13C2<δ13C3>δ13C4. Besides, n-alkanes of C3+ gas components are preferentially attacked during the process of microbial alteration. This is manifested that n-alkanes are more enriched in 13C than corresponding iso-alkanes. As a result, the concentrations of n-alkanes be-come very low, which may be misleading in indentifying the gas genetic types. As to four gas samples, light hydro-carbon compositions display evidence for microbial alteration. The sequence of hexane isomers varies obviously with high content of 2,3-DMC4, which indicates that they have been in the fourth level of extensively bacterial al-teration. 19Preparation of an anti-diethylstilbestrol monoclonal antibody and development of an indirect competitive ELISA to detect diethylstilbestrol in biological samples显示文摘Based on the preparation of an anti-diethylstilbestrol(DES) monoclonal antibody,a simple and convenient indirect competitive enzyme-linked immunosorbent assay(ELISA) method for DES detection has been developed.The monoclonal antibody demonstrated high sensitivity to DES with an IC50 value of 275 pg mL-1 and detection limit(LOD) of 90 pg mL-1.The specificity of the assay was studied by measuring cross-reactivity of the antibody with structurally related compounds of ethinyl estradiol(<7%),estrone(<0.1%),estriol(<0.1%),and diethylstilbestrol benzoate(<0.1%).Chicken,fish,shrimp,urine and bile spiked with different concentration of DES were detected by the developed method,and the recovery rates were greater than 79.5%.Intra-and inter-assay variations were about 6%.This method exhibited high stability with a coefficient of variation less than 10% in buffer and in real samples.The LODs in fish/shrimp,liver,feed and urine spiked with DES were 600,600,4800 and 600 pg mL-1,respectively.These results confirmed that the antibody to DES was successfully produced and could be used to establish ELISA methods for DES detection in food producing animals.LI WeiHua MENG Meng HE FangYang WAN YuPing XUE HuYin LIU Wei YIN WeiWei XU Jing FENG CaiWei WANG ShanLiang LU Xiao LIU JinTing XI RiMo 2011Chinese Science Bulletin2011,56,8:4
20Low-overpotential selective reduction of CO2 to ethanol on electrodeposited CuxAuy nanowire arrays显示文摘Direct electrochemical reduction of CO2 to multicarbon products is highly desirable, yet challenging. Here, we present a potentiostatic pulse-electrodeposition of high-aspect-ratio CuxAuy nanowire arrays (NWAs) as high-performance electrocatalysts for the CO2 reduction reaction (CO2RR). The surface electronic structure related to the Cu:Au ratio in the CuxAuy NWAs could be facilely modulated by controlling the electrodeposition potential and the as-fabricated CuxAuy NWAs could be directly used as the catalytic electrode for the CO2RR. The morphology of the high-aspect-ratio nanowire array significantly lowers the onset potential of the alcohol formation due to the diffusion-induced enhancement of the local pH and CO concentration near the nanowire surface. Besides, the properly adjusted surface electronic structure of the CuxAuy NWA enables the adsorption of CO and facilitates the subsequent CO reduction to ethanol via the C-C coupling pathway. Owing to the synergistic effect of morphology and electronic structure, the optimized CuxAuy NWA selectively reduces CO2 to ethanol at low potentials of -0.5——0.7 V vs. RHE with a highest Faradaic efficiency of 48%.
