13D printing of bone tissue engineering scaffolds显示文摘Tissue engineering is promising in realizing successful treatments of human body tissue loss that current methods cannot treat well or achieve satisfactory clinical outcomes.In scaffold-based bone tissue engineering,a high performance scaffold underpins the success of a bone tissue engineering strategy and a major direction in the field is to produce bone tissue engineering scaffolds with desirable shape,structural,physical,chemical and biological features for enhanced biological performance and for regenerating complex bone tissues.Three-dimensional(3D)printing can produce customized scaffolds that are highly desirable for bone tissue engineering.The enormous interest in 3D printing and 3D printed objects by the science,engineering and medical communities has led to various developments of the 3D printing technology and wide investigations of 3D printed products in many industries,including biomedical engineering,over the past decade.It is now possible to create novel bone tissue engineering scaffolds with customized shape,architecture,favorable macro-micro structure,wettability,mechanical strength and cellular responses.This article provides a concise review of recent advances in the R&D of 3D printing of bone tissue engineering scaffolds.It also presents our philosophy and research in the designing and fabrication of bone tissue engineering scaffolds through 3D printing.Chong Wang Wei Huang Yu Zhou Libing He Zhi He Ziling Chen Xiao He Shuo Tian Jiaming Liao Bingheng Lu Yen Wei Min Wang 2020Bioactive Materials2020,5,1:20
2Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance,essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders.The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells,along with the signaling mechanisms governing stem cell self-renewal and differentiation.In this review,we first summarize the biological characteristics of seven types of dental stem cells,including dental pulp stem cells,stem cells from apical papilla,stem cells from human exfoliated deciduous teeth,dental follicle precursor cells,periodontal ligament stem cells,alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva.We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways.We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications.Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade.Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji 2016Genes & Diseases2016,3,4:16
3Geochronology, Geochemistry and Sr-Nd-Pb-Hf Isotopes of No. I Complex from the Shitoukengde Ni–Cu Sulfide Deposit in the Eastern Kunlun Orogen, Western China: Implications for the Magmatic Source, Geodynamic Setting and Genesis显示文摘The Shitoukengde Ni-Cu deposit, located in the Eastern Kunlun Orogen, comprises three mafic–ultramafic complexes, with the No. I complex hosting six Ni-Cu orebodies found recently. The deposit is hosted in the small ultramafic bodies intruding Proterozoic metamorphic rocks. Complexes at Shitoukengde contain all kinds of mafic-ultramafic rocks, and olivine websterite and pyroxene peridotite are the most important Ni-Cu-hosted rocks. Zircon U-Pb dating suggests that the Shitoukengde Ni-Cu deposit formed in late Silurian(426–422 Ma), and their zircons have εHf(t) values of-9.4 to 5.9 with the older T_(DM1) ages(0.80–1.42 Ga). Mafic-ultramafic rocks from the No. I complex show the similar rare earth and trace element patterns, which are enriched in light rare earth elements and large ion lithophile elements(e.g., K, Rb, Th) and depleted in heavy rare earth elements and high field strength elements(e.g., Ta, Nb, Zr, Ti). Sulfides from the deposit have the slightly higher δ^(34)S values of 1.9–4.3‰ than the mantle(0 ± 2‰). The major and trace element characteristics, and Sr-Nd-Pb and Hf, S isotopes indicate that their parental magmas originated from a metasomatised, asthenospheric mantle source which had previously been modified by subduction-related fluids, and experienced significant crustal contamination both in the magma chamber and during ascent triggering S oversaturation by addition of S and Si, that resulted in the deposition and enrichment of sulfides. 4Highly expressed BMP9/GDF2 in postnatal mouse liver and lungs may account for its pleiotropic effects on stem cell differentiation,angiogenesis,tumor growth and metabolism显示文摘Bone morphogenetic protein 9(BMP9)(or GDF2)was originally identified from fetal mouse liver cDNA libraries.Emerging evidence indicates BMP9 exerts diverse and pleiotropic functions during postnatal development and in maintaining tissue homeostasis.However,the expression landscape of BMP9 signaling during development and/or in adult tissues remains to be analyzed.Here,we conducted a comprehensive analysis of the expression landscape of BMP9 and its signaling mediators in postnatal mice.By analyzing mouse ENCODE transcriptome datasets we found Bmp9 was highly expressed in the liver and detectable in embryonic brain,adult lungs and adult placenta.We next conducted a comprehensive qPCR analysis of RNAs isolated from major mouse tissues/organs at various ages.We found that Bmp9 was highly expressed in the liver and lung tissues of young adult mice,but decreased in older mice.Interestingly,Bmp9 was only expressed at low to modest levels in developing bones.BMP9-associated TGFβ/BMPR type I receptor Alk1 was highly expressed in the adult lungs.Furthermore,the feedback inhibitor Smads Smad6 and Smad7 were widely expressed in mouse postnatal tissues.However,the BMP signaling antagonist noggin was highly expressed in fat and heart in the older age groups,as well as in kidney,liver and lungs in a biphasic fashion.Thus,our findings indicate that the circulating BMP9 produced in liver and lungs may account for its pleiotropic effects on postnatal tissues/organs although possible roles of BMP9 signaling in liver and lungs remain to be fully understood.Wei Liu Zhongliang Deng Zongyue Zeng Jiaming Fan Yixiao Feng Xi Wang Daigui Cao Bo Zhang Lijuan Yang Bin Liu Mikhail Pakvasa William Wagstaff Xiaoxing Wu Huaxiu Luo Jing Zhang Meng Zhang Fang He Yukun Mao Huiming Ding Yongtao Zhang Changchun Niu Rex C.Haydon Hue H.Luu Jennifer Moriatis Wolf Michael J.Lee Wei Huang Tong-Chuan He Yulong Zou 2020Genes & Diseases2020,7,2:9
5Deep learning in optical metrology:a review显示文摘With the advances in scientific foundations and technological implementations,optical metrology has become versatile problem-solving backbones in manufacturing,fundamental research,and engineering applications,such as quality control,nondestructive testing,experimental mechanics,and biomedicine.In recent years,deep learning,a subfield of machine learning,is emerging as a powerful tool to address problems by learning from data,largely driven by the availability of massive datasets,enhanced computational power,fast data storage,and novel training algorithms for the deep neural network.It is currently promoting increased interests and gaining extensive attention for its utilization in the field of optical metrology.Unlike the traditional,,physics-basedH approach,deep-learning-enabled optical metrology is a kind of,/data-drivenw approach,which has already provided numerous alternative solutions to many challenging problems in this field with better performances.In this review,we present an overview of the current status and the latest progress of deep-learning technologies in the field of optical metrology.We first briefly introduce both traditional image-processing algorithms in optical metrology and the basic concepts of deep learning,followed by a comprehensive review of its applications in various optical metrology tasks,such as fringe denoising,phase retrieval,phase unwrapping,subset correlation,and error compensation.The open challenges faced by the current deep-learning approach in optical metrology are then discussed.Finally,the directions for future research are outlined.Chao Zuo Jiaming Qian Shijie Feng Wei Yin Yixuan Li Pengfei Fan Jing Han Kemao Qian Qian Chen 2022Light(Science & Applications)2022,11,4:7
6Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. 7Single-nucleus transcriptomic landscape of primate hippocampal aging显示文摘The hippocampus plays a crucial role in learning and memory,and its progressive deterioration with age is functionally linked to a variety of human neurodegenerative diseases.Yet a systematic profiling of the aging effects on various hippocampal cell types in primates is still missing.Here,we reported a variety of new aging-associated phenotypic changes of the primate hippocampus.These include,in particular,increased DNA damage and heterochromatin erosion with time,alongside loss of proteostasis and elevated inflammation.To understand their cellular and molecular causes,we established the first single-nucleus transcriptomic atlas of primate hippocampal aging.Among the 12 identified cell types,neural transiently amplifying progenitor cell(TAPC)and microglia were most affected by aging.In-depth dissection of gene-expression dynamics revealed impaired TAPC division and compromised neuronal function along the neurogenesis trajectory;additionally elevated pro-inflammatory responses in the aged microglia and oligodendrocyte,as well as dysregulated coagulation pathways in the aged endothelial cells may contribute to a hostile microenvironment for neurogenesis.This rich resource for understanding primate hippocampal aging may provide potential diagnostic biomarkers and therapeutic interventions against age-related neurodegenerative diseases.Hui Zhang Jiaming Li Jie Ren Shuhui Sun Shuai Ma Weiqi Zhang Yang Yu Yusheng Cai Kaowen Yan Wei Li Baoyang Hu Piu Chan Guo-Guang Zhao Juan Carlos Izpisua Belmonte Qi Zhou Jing Qu Si Wang Guang-Hui Liu 2021Protein & Cell2021,12,9:5

9Investigation on experimental load spectrum for high and low cycle combined fatigue test显示文摘As the outfield load spectrum is so complicated that it cannot be used directly for test study in laboratory.This paper presents a method to determine load spectrum for high and low cycle combined fatigue of turbine mortise at elevated temperature through experimental and numerical method.First of all,the low cycle load spectrum with duration time is determined through cumulative damage rule.The rain flow counting method is applied to obtain main cycles and sub cycles,and then the stress cycle is converted into pulsation cycle(stress ratio=0)based on the S-N curve and the Goodman curve;Secondly,three groups of different amplitudes tests are established to determine the high cycle amplitude.Finally,another three groups of the high-low combined cycle fatigue(HLCCF)tests for turbine mortise of a certain type engine are carried out.The results show that the macro and micro failure modes are identical with outfield's,which verifies the accuracy of the conversion method.Rongqiao Wang Jiaming Wei Dianyin Hu Xiuli Shen Jiang Fan 2013Propulsion and Power Research2013,2,4:4
10Flexible ITO-free organic solar cells over 10% by employing drop-coated conductive PEDOT:PSS transparent anodes显示文摘Highly conductive poly(3,4-ethylenedioxythiophene):poly(styrene-sulfonic acid)(PEDOT:PSS) has been explored to fabricate flexible and stretchable conductors. Generally, PEDOT:PSS transparent anodes are prepared by spin-coating method. In this article, we adopt a method by dropping PEDOT:PSS aqueous solution on the PET plastic substrate to fabricate flexible electrodes. Compared with spin coating, drop-coating is simple and cost-effective with large-area fabrications. Through this method, we fabricated highly transparent conductive electrodes and systematically studied their electrical, optical, morphological and mechanical properties. With dimethyl sulfoxide/methanesulfonic acid(DMSO/MSA) treated PEDOT:PSS electrode,bendable devices based on non-fullerene system displayed an open-circuit voltage of 0.925 V, a fill factor of 70.74%, and a high power conversion efficiency(PCE) of 10.23% under 100 mW cm^(-2) illumination, which retained over 80% of the initial PCE value after 1000 bending cycles. 11Highly Dispersed Pt Species with Excellent Stability and Catalytic Performance by Reducing a Perovskite-Type Oxide Precursor for CO Oxidation显示文摘A new scheme for the preparation of highly dispersed precious metal catalysts is proposed in this work. Samples of LaCo_(1-x)Pt_xO_3/SiO_2( x = 0.03, 0.05, 0.07, 0.09, and 0.10) were prepared through a simple method of citrate acid complexation combined with impregnation. In a nanocrystallite of LaCo_(1-x)Pt_xO_3, ions of lanthanum, cobalt, and platinum are evenly mixed at the atomic level and confined within the nanocrystallite. In the reduction process, platinum ions were reduced and migrated onto the surface of the nanocrystallite, and the platinum should be highly dispersed owing to the even mixing of the platinum ions in the precursor. When x = 0.05 or lower, the highest dispersion of Pt could be achieved. The highly dispersed Pt is stable, because of the strong interaction between Pt atoms and the support. The catalysts were characterized by BET surface area, temperature-programmed reduction, X-ray diff raction, transmission electron microscopy, CO temperatureprogrammed desorption, and turnover frequency. 12Integrated bioinformatics identifies the dysregulation induced by aberrant gene methylation in colorectal carcinoma显示文摘Colorectal carcinoma(CRC)is one of the most common cancers,and is associated with a poor dlinical outcome.The key genes and potential prognostic markers in colorectal carcinoma remain to be identfed and explored for clinical application.DNA expression/methyl-ation profles were downloaded from the Gene Expression Omnibus(GEO)database to identify differentially expressed/methylated genes(DEGs and DEMs).A total of 255 genes and 372 genes were identified as being up-regulated and down-regulated,respectively,in GSE113513,GSE81558,and GSE89076.There were a total of 3350 hypermethylated genes and 443 hypomethylated genes identifed in GSE 48684.Twenty genes were found to be hypermethylated as well as down-regulated,and a functional enrichment analysis revealed that these genes were mainly involved in cancer-related pathways.Among these 20 genes,GPM6A,HAND2 and C2orf40 were related to poor outcomes in cancer patients based on a survival analysis.Concurrent decreases of GPM6A,HAND2 and C2orf40 protein expression were observed in highly-dif ferentiated colorectal carcinoma tissues,and higher expression levels were found in undifferentiated or minimally-differentiated colorectal carcinoma tissues.In conclusion,20 genes were found to be downregulated and hypermethylated in CRC,among which GPM6A,HAND2 and C2orf40 were explored for their potential prognostic value.Zhenyu Ye Yecheng Li Jiaming Xie Zhenyu Feng Xiaodong Yang Yong Wu Yuwei Pu Jiawei Gao Xiangrong Xu Zhaobi Zhu Wei Li Wei Chen Chungen Xing 2021Genes & Diseases2021,8,4:2
13Low‐intensity pulsed ultrasound ameliorates angiotensin II-induced cardiac fibrosis by alleviating inflammation via a caveolin-1-dependent pathway显示文摘Objective:Cardiac hypertrophy and fibrosis are major pathological manifestations observed in left ventricular remodeling induced by angiotensin II(AngII).Low-intensity pulsed ultrasound(LIPUS)has been reported to ameliorate cardiac dysfunction and myocardial fibrosis in myocardial infarction(MI)through mechano-transduction and its downstream pathways.In this study,we aimed to investigate whether LIPUS could exert a protective effect by ameliorating AngII-induced cardiac hypertrophy and fibrosis and if so,to further elucidate the underlying molecular mechanisms.Methods:We used AngII to mimic animal and cell culture models of cardiac hypertrophy and fibrosis.LIPUS irradiation was applied in vivo for 20 min every 2 d from one week before mini-pump implantation to four weeks after mini-pump implantation,and in vitro for 20 min on each of two occasions 6 h apart.Cardiac hypertrophy and fibrosis levels were then evaluated by echocardiographic,histopathological,and molecular biological methods.Results:Our results showed that LIPUS could ameliorate left ventricular remodeling in vivo and cardiac fibrosis in vitro by reducing AngII-induced release of inflammatory cytokines,but the protective effects on cardiac hypertrophy were limited in vitro.Given that LIPUS increased the expression of caveolin-1 in response to mechanical stimulation,we inhibited caveolin-1 activity with pyrazolopyrimidine 2(pp2)in vivo and in vitro.LIPUS-induced downregulation of inflammation was reversed and the anti-fibrotic effects of LIPUS were absent.Conclusions:These results indicated that LIPUS could ameliorate AngII-induced cardiac fibrosis by alleviating inflammation via a caveolin-1-dependent pathway,providing new insights for the development of novel therapeutic apparatus in clinical practice.Kun ZHAO Jing ZHANG Tianhua XU Chuanxi YANG LiqingWENG TingtingWU XiaoguangWU Jiaming MIAO Xiasheng GUO Juan TU Dong ZHANG Bin ZHOU Wei SUN Xiangqing KONG 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,10:2
14Experimental study on improving collapsible loess with cement显示文摘The collapsibility of loess ground can directly affect stability of subgrade. Therefore, how to adopt practical technical measures to reduce or eliminate its collapse deformation is an important content in foundation design in collapsible loess zone. Selecting collapsible loess from Fuxin-Chaoyang highway in Liaoning, the authors conducted a series of tests for improving loess with cement. The loess in different water content was mixed with the cement in varying proportions, unconfined compression strength for the samples at four different curing periods were tested, and the relationships of improved soil strength among cement mixture ratio and curing periods were analyzed. When the curing periods are certain, the strength of loess increases along with the mixture ratio increases; when the cement mixture ratio is 5%-15%, the scope of increases is quite obvious; when the mixture ratio is greater than 15%, the tendency of intensity increases turns slow. 15The nucleocapsid protein of rice stripe virus in cell nuclei of vector insect regulates viral replication显示文摘Rice stripe virus(RSV)transmitted by the small brown planthopper causes severe rice yield losses in Asian countries.Although viral nuclear entry promotes viral replication in host cells,whether this phenomenon occurs in vector cells remains unknown.Therefore,in this study,we systematically evaluated the presence and roles of RSV in the nuclei of vector insect cells.We observed that the nucleocapsid protein(NP)and viral genomic RNAs were partially transported into vector cell nuclei by utilizing the importin a nuclear transport system.When blocking NP nuclear localization,cytoplasmic RSV accumulation significantly increased.In the vector cell nuclei,NP bound the transcription factor YY1 and affected its positive regulation to FAIM.Subsequently,decreased FAIM expression triggered an antiviral caspase-dependent apoptotic reaction.Our results reveal that viral nuclear entry induces completely different immune effects in vector and host cells,providing new insights into the balance between viral load and the immunity pressure in vector insects.Wan Zhao Junjie Zhu Hong Lu Jiaming Zhu Fei Jiang Wei Wang Lan Luo Le Kang Feng Cui 2022Protein & Cell2022,13,5:2

18Optimization of Extraction Process and Determination of Antioxidant Activity of Myrica ruba Leaf Pigment by Orthogonal Design显示文摘[Objectives] To determine the optimum conditions and antioxidant activity of Myrica ruba leaf pigment,and provide reference for further development and use of Myrica ruba leaves. [Methods]Pigment of Myrica ruba leaves was extracted by ethanol extraction method. Absorbance was measured at wavelength of 300 nm. Effects of ethanol volume fraction,solid-to-liquid ratio,temperature,and extraction time were studied. 20T-Splines Based Isogeometric Topology Optimization with Arbitrarily Shaped Design Domains显示文摘In this paper,a new isogeometric topology optimization(ITO)method is proposed by using T-splines based isogeometric analysis(IGA).The arbitrarily shaped design domains,directly obtained from CAD,are represented by a single T-spline surface which overcomes the topological limitations of Non-Uniform Rational B-Spline(NURBS).The coefficients correlated with control points are directly used as design variables.Therefore,the T-spline basis functions applied for geometry description and calculation of structural response are simultaneously introduced to represent the density distribution.Several numerical examples show that the proposed approach leads to a coherent workflow to handle design problems of complicated structures.The optimized results are free of checkerboard patterns without additional stabilization and filtering techniques due to the properties of T-splines,which also simplified the post-processing.In addition,through performing local refinement,we can easily achieve multiresolution optimization and infill optimization within the T-splines based framework.In general,the proposed method provides a possibility to design,analyze,and optimize engineering structures in a uniform model,which has the potential to improve design efficiency and reduce the cost of product development.Gang Zhao Jiaming Yang Wei Wang Yang Zhang Xiaoxiao Du Mayi Guo 2020Computer Modeling in Engineering & Sciences2020,,6:1
