1Pathways of China's PM;air quality 2015–2060 in the context of carbon neutrality显示文摘Clean air policies in China have substantially reduced particulate matter(PM;) air pollution in recent years,primarily by curbing end-of-pipe emissions.However,reaching the level of the World Health Organization(WHO) guidelines may instead depend upon the air quality co-benefits of ambitious climate action.Here,we assess pathways of Chinese PM;air quality from 2015 to 2060 under a combination of scenarios that link global and Chinese climate mitigation pathways(i.e.global 2℃-and 1.5℃-pathways,National Determined Contributions(NDC) pledges and carbon neutrality goals) to local clean air policies.We find that China can achieve both its near-term climate goals(peak emissions) and PM;air quality annual standard(35 μg/m;) by 2030 by fulfilling its NDC pledges and continuing air pollution control policies.However,the benefits of end-of-pipe control reductions are mostly exhausted by 2030,and reducing PM;exposure of the majority of the Chinese population to below 10 μg/m;by 2060 will likely require more ambitious climate mitigation efforts such as China’ s carbon neutrality goals and global1.5℃-pathways.Our results thus highlight that China’s carbon neutrality goals will play a critical role in reducing air pollution exposure to the level of the WHO guidelines and protecting public health.Jing Cheng Dan Tong Qiang Zhang Yang Liu Yu Lei Gang Yan Liu Yan Sha Yu Ryna Yiyun Cui Leon Clarke Guannan Geng Bo Zheng Xiaoye Zhang Steven JDavis Kebin He 2021National Science Review2021,8,12:23
2On-the-fly active learning of interpretable Bayesian force fields for atomistic rare events显示文摘Machine learned force fields typically require manual construction of training sets consisting of thousands of first principles calculations,which can result in low training efficiency and unpredictable errors when applied to structures not represented in the training set of the model.This severely limits the practical application of these models in systems with dynamics governed by important rare events,such as chemical reactions and diffusion.We present an adaptive Bayesian inference method for automating the training of interpretable,low-dimensional,and multi-element interatomic force fields using structures drawn on the fly from molecular dynamics simulations.Within an active learning framework,the internal uncertainty of a Gaussian process regression model is used to decide whether to accept the model prediction or to perform a first principles calculation to augment the training set of the model.The method is applied to a range of single-and multi-element systems and shown to achieve a favorable balance of accuracy and computational efficiency,while requiring a minimal amount of ab initio training data.We provide a fully opensource implementation of our method,as well as a procedure to map trained models to computationally efficient tabulated force fields.Jonathan Vandermause Steven B.Torrisi Simon Batzner Yu Xie Lixin Sun Alexie M.Kolpak Boris Kozinsky 2020npj Computational Materials2020,,1:17
3Chinese Tuina Downregulates the Elevated Levels of Tissue Plasminogen Activator in Sciatic Nerve Injured Sprague-Dawley Rats显示文摘Objective:To elucidate the mechanism of Chinese tuina in treating sciatic nerve crush injury,and to detect the levels of tissue plasminogen activator(t PA)and plasminogen activator inhibitor-1(PAI-1),which is thought to play an important role in nerve regeneration.Methods:Thirty-two adult male Sprague-Dawley rats were subjected to sciatic nerve crush injury and 16 rats(sham-operated group)went through a sham operation.Control group was given no treatment while tuina group received tuina therapy since day 7 post-surgery.Tuina treatment was performed once a day and lasted for 20 days.The sciatic functional index was examined every 5days during the treatment session.The rats'gastrocnemius muscles were evaluated for changes in mass and immunohistochemistry techniques were performed to detect the levels of tP A and PAI-1.Results:Tuina therapy improved the motor function of sciatic nerve injured rats(P<0.05),however,it did not increase muscle volume(P<0.05).Tuina downregulated the levels of t PA and PAI-1(P<0.05).Conclusions:The present study implies that tuina treatment could accelerate rehabilitation of peripheral nerve injury.PAN Fan YU Tian-yuan Steven Wong XIAN Si-tong LU Meng-qian WU Jian-cong GAO Yu-feng LI Xiao-qin GENG Nan YAO Bin-bin 2017Chinese Journal of Integrative Medicine2017,23,8:16
4“Three Methods and Three Points” regulates p38 mitogen-activated protein kinase in the dorsal horn of the spinal cord in a rat model of sciatic nerve injury显示文摘Tuina is a traditional Chinese treatment for sensory disturbances caused by peripheral nerve injury and related diseases. Our previous studies showed that tuina regulates relevant regions and indices of the spinal dorsal horn using the Dian, Bo, and Rou method in Yinmen(BL37), Yanglingquan(GB34), and Weizhong(BL40). Treatment prevents muscle atrophy, protects spinal cord neurons, and promotes sciatic nerve repair. The mechanisms of action of tuina for treating peripheral nerve injury remain poorly understood. This study established rat models of sciatic nerve injury using the crushing method. Rats received Chinese tuina in accordance with the principle of 'Three Methods and Three Points,' once daily for 20 days. Tuina intervention reduced paw withdrawal latency and improved wet weight of the gastrocnemius muscle, as well as promoting morphological recovery of sciatic nerve fibers, Schwann cells, and axons. The protein expression levels of phospho-p38 mitogen-activated protein kinase, tumor necrosis factor-α, and interleukin-1β also decreased. 5Parallel transjugular intrahepatic portosystemic shunt with Viatorr^(®)? stents for primary TIPS insufficiency:Case series and review of literature显示文摘BACKGROUND Transjugular intrahepatic portosystemic shunts(TIPS) can alleviate complications of portal hypertension such as ascites and variceal bleeding by decreasing the portosystemic gradient.In limited clinical situations,parallel TIPS may be only solution to alleviate either variceal bleeding or ascites secondary to portal hypertension when the primary TIPS fails to do so.Data specifically addressing the use of this partially polytetrafluoroethylene covered nitinol stent(Viatorr~?) is largely lacking despite Viatorr~? being the current gold standard for modern TIPS placement.CASE SUMMARY All three patients had portal hypertension and already had a primary Viatorr~?TIPS placed previously.All patients have undergone failed endoscopy to manage acute variceal bleeding before referral for a parallel stent(PS).PS were placed in patients presenting with recurrent variceal bleeding despite existence of a widely patent primary TIPS.Primary stent patency was verified with either Doppler ultrasound or intra-procedural TIPS stent venography.Doppler ultrasound follow-up imaging demonstrated complete patency of both primary and parallel TIPS.All three patients did well on clinical follow-up of up to six months and no major complications were recorded.A review of existing literature on the role of PS in the management of portal hypertension complications is discussed.There are three case reports of use of primary and PS Viatorr~? stents placement,only one of which is in a patient with gastrointestinal variceal bleeding despite a patent primary Viatorr~? TIPS.CONCLUSION Viatorr~? 8Evaluation of a point of care ultrasound curriculum for Indonesian physicians taught by fi rst-year medical students显示文摘BACKGROUND: The purpose of this study was to assess the short-term efficacy of a 4-week ultrasound curriculum taught by American first-year medical students to general practitioners working in public health care clinics, or puskesmas, in Bandung, Indonesia.METHODS: We performed a prospective, observational study of Indonesian health care practitioners from public clinics in Bandung, Indonesia. These practitioners were enrolled in a 4-week ultrasound training course taught by first-year American medical students. A total of six sessions were held comprising of 38 ultrasound milestones. A pre-course and post-course written exam and practical exam was taken by each participant.RESULTS: We enrolled 41 clinicians in the course. The average pre-course exam score was 35.2% with a 2.4% pass rate, whereas the average post-course exam score was 82.0% with a 92.7% pass rate. The average practical score at the completion of the course was 83.2%(SD=0.145) with 82.9% of the class passing(score above 75.0%).CONCLUSION: Our data suggests that first-year medical students can effectively teach ultrasound to physicians in Indonesia using a 4-week intensive ultrasound training course. 9Epigenetic states and expression of imprinted genes in human embryonic stem cells显示文摘AIM: To investigate the epigenetic states and expres- sion of imprinted genes in five human embryonic stem cell (hESC) lines derived in Taiwan. METHODS: The heterozygous alleles of single nucleo- tide polymorphisms (SNPs) at imprinted genes were analyzed by sequencing genomic DNAs of hESC lines and the monoallelic expression of the imprinted genes were confirmed by sequencing the cDNAs. The expres- sion profiles of 32 known imprinted genes of five hESC lines were determined using Affymetrix human genome U133 plus 2.0 DNA microarray. RESULTS: The heterozygous alleles of SNPs at seven imprinted genes, IPW , PEG10 , NESP55 , KCNQ1 , ATP10A ,TCEB3C and IGF2 , were identified and the monoallelic expression of these imprinted genes except IGF2 were confirmed. The IGF2 gene was found to be imprinted in hESC line T2 but partially imprinted in line T3 and not imprinted in line T4 embryoid bodies. Ten imprinted genes, namely GRB10 , PEG10 , SGCE, MEST , SDHD , SN- RPN , SNURF , NDN , IPW and NESP55 , were found to be highly expressed in the undifferentiated hESC lines and down-regulated in differentiated derivatives. The UBE3A gene abundantly expressed in undifferentiated hESC lines and further up-regulated in differentiated tissues. The expression levels of other 21 imprinted genes were relatively low in undifferentiated hESC lines and five of these genes (TP73 , COPG2 , OSBPL5 , IGF2 and ATP10A ) were found to be up-regulated in differentiated tissues. 12Development of a CLDN18.2-targeting immuno-PET probe for non-invasive imaging in gastrointestinal tumors显示文摘Claudin18.2(CLDN18.2)is a tight junction protein that is overexpressed in a variety of solid tumors such as gastrointestinal cancer and oesophageal cancer.It has been identified as a promising target and a potential biomarker to diagnose tumor,evaluate efficacy,and determine patient prognosis.TST001 is a recombinant humanized CLDN18.2 antibody that selectively binds to the extracellular loop of human Claudin18.2.In this study,we constructed a solid target radionuclide zirconium-^(89)(^(89)Zr)labled-TST001 to detect the expression of in the human stomach cancer BGC823CLDN18.2 cell lines.The[^(89)Zr]Zr-desferrioxamine(DFO)-TST001 showed high radiochemical purity(RCP,>99%)and specific activity(24.15±1.34 GBq/mmol),and was stable in 5%human serum albumin,and phosphate buffer saline(>85%RCP at 96 h).The EC_(50) values of TST001 and DFO-TST001 were as high as 0.413±0.055 and 0.361±0.058 nM(P>0.05),respectively.The radiotracer had a significantly higher average standard uptake values in CLDN18.2-positive tumors than in CLDN18.2-negative tumors(1.11±0.02 vs.0.49±0.03,P=0.0016)2 days post injection(p.i.).BGC823CLDN18.2 mice models showed high tumor/muscle ratios 96 h p.i.with[^(89)Zr]Zr-DFO-TST001 was much higher than those of the other imaging groups.Immunohistochemistry results showed that BGC823CLDN18.2 tumors were highly positive(+++)for CLDN18.2,while those in the BGC823 group did not express CLDN18.2().The results of ex vivo biodistribution studies showed that there was a higher distribution in the BGC823CLDN18.2 tumor bearing mice(2.05±0.16%ID/g)than BGC823 mice(0.69±0.02%ID/g)and blocking group(0.72±0.02%ID/g).A dosimetry estimation study showed that the effective dose of[^(89)Zr]Zr-DFO-TST001 was 0.0705 mSv/MBq,which is within the range of acceptable doses for nuclear medicine research.Taken together,these results suggest that Good Manufacturing Practices produced by this immuno-positron emission tomography probe can detect CLDN18.2-overexpressing tumors.Yan Chen Xingguo Hou Dapeng Li Jin Ding Jiayue Liu Zilei Wang Fei Teng Hongjun Li Fan Zhang Yi Gu Steven Yu Xueming Qian Zhi Yang Hua Zhu 2023Journal of Pharmaceutical Analysis2023,13,4:2
13Precision Phenotyping Reveals Novel Loci for Quantitative Resistance to Septoria Tritici Blotch显示文摘Accurate,high-throughput phenotyping for quantitative traits is a limiting factor for progress in plant breeding.We developed an automated image analysis to measure quantitative resistance to septoria tritici blotch(STB),a globally important wheat disease,enabling identification of small chromosome intervals containing plausible candidate genes for STB resistance.335 winter wheat cultivars were included in a replicated field experiment that experienced natural epidemic development by a highly diverse but fungicide-resistant pathogen population.More than 5.4 million automatically generated phenotypes were associated with 13,648 SNP markers to perform the GWAS.We identified 26 chromosome intervals explaining 1.9-10.6%of the variance associated with four independent resistance traits.Sixteen of the intervals overlapped with known STB resistance intervals,suggesting that our phenotyping approach can identify simultaneously(i.e.,in a single experiment)many previously defined STB resistance intervals.Seventeen of the intervals were less than 5 Mbp in size and encoded only 173 genes,including many genes associated with disease resistance.Five intervals contained four or fewer genes,providing high priority targets for functional validation.Ten chromosome intervals were not previously associated with STB resistance,perhaps representing resistance to pathogen strains that had not been tested in earlier experiments.The SNP markers associated with these chromosome intervals can be used to recombine different forms of quantitative STB resistance that are likely to be more durable than pyramids of major resistance genes.Our experiment illustrates how high-throughput automated phenotyping can accelerate breeding for quantitative disease resistance.Steven Yates Alexey Mikaberidze Simon GKrattinger Michael Abrouk Andreas Hund Kang Yu Bruno Studer Simone Fouche Lukas Meile Danilo Pereira Petteri Karisto Bruce A.McDonald 2019Plant Phenomics2019,1,1:2
