1Arabidopsis Chloroplast FtsH,var2 and Suppressors of var2 Leaf Variegation:a Review显示文摘Variegation mutants are ideal model systems to study chloroplast biogenesis. We are interested in variegations whose green and whitesectored leaves arise as a consequence of the action of nuclear recessive genes. In this review, we focus on the Arabidopsis var2 variegation mutant, and discuss recent progress toward understanding the function of VAR2 and the mechanism of var2-mediated variegation. VAR2 is a subunit of the chloroplast FtsH complex, which is involved in turnover of the Photosystem II reaction center D1 protein, as well as in other processes required for the development and maintenance of the photosynthetic apparatus. The cells in green sectors of var2 have normal-appearing chloroplasts whereas cells in the white sectors have abnormal plastids that lack pigments and organized lamellae. To explain the mechanism of var2 variegation, we have proposed a threshold model in which the formation of chloroplasts is due to the presence of activities/processes that are able to compensate for a lack of VAR2. To gain insight into these activities, second-site suppressor screens have been carried out to obtain mutants with nonvariegation phenotypes. 4Three-dimensional properties of mesoscale cyclonic warm-core and anticyclonic cold-core eddies in the South China Sea显示文摘In general,a mesoscale cyclonic(anticyclonic)eddy has a colder(warmer)core,and it is considered as a cold(warm)eddy.However,recently research found that there are a number of'abnormal'mesoscale cyclonic(anticyclonic)eddies associated with warm(cold)cores in the South China Sea(SCS).These'abnormal'eddies pose a challenge to previous works on eddy detection,characteristic analysis,eddy-induced heat and salt transports,and even on mesoscale eddy dynamics.Based on a 9-year(2000–2008)numerical modelling data,the cyclonic warm-core eddies(CWEs)and anticyclonic cold-core eddies(ACEs)in the SCS are analyzed.This study found that the highest incidence area of the'abnormal'eddies is the northwest of Luzon Strait.In terms of the eddy snapshot counting method,8620 CWEs and 9879 ACEs are detected,accounting for 14.6%and 15.8%of the total eddy number,respectively.The size of the'abnormal'eddies is usually smaller than that of the'normal'eddies,with the radius only around 50 km.In the generation time aspect,they usually appear within the 0.1–0.3 interval in the normalized eddy lifespan.The survival time of CWEs(ACEs)occupies 16.3%(17.1%)of the total eddy lifespan.Based on two case studies,the intrusion of Kuroshio warm water is considered as a key mechanism for the generation of these'abnormal'eddies near the northeastern SCS.Wenjin Sun Yu Liu Gengxin Chen Wei Tan Xiayan Lin Yuping Guan Changming Dong 2021Acta Oceanologica Sinica2021,40,10:3
5Genetic Interactions Reveal that Speci c Defects of Chloroplast Translation are Associated with the Suppression of var2-Mediated Leaf Variegation显示文摘Arabidopsis thaliana L.yellow variegated(var2)mutant is defective in a chloroplast FtsH family metalloprotease,AtFtsH2/VAR2,and displays an intriguing green and white leaf variegation.This unique var2 mediated leaf variegation offers a simple yet powerful tool for dissecting the genetic regulation of chloroplast development.Here,we report the isolation and characterization of a new var2suppressor gene,SUPPRESSOR OF VARIEGATION8(SVR8),which encodes a putative chloroplast ribosomal large subunit protein,L24.Mutations in SVR8suppress var2leaf variegation at ambient temperature and partially suppress the cold induced chlorosis phenotype of var2.Loss of SVR8 causes unique chloroplast rRNA processing defects,particularly the 23S–4.5S dicistronic precursor.The recovery of the major abnormal processing site in svr823S–4.5S precursor indicate that it does not lie in the same position where SVR8/L24 binds on the ribosome.Surprisingly,we found that the loss of a chloroplast ribosomal small subunit protein,S21,results in aberrant chloroplast rRNA processing but not suppression of var2variegation.These ndings suggest that the disruption of speci c aspects of chloroplast translation,rather than a general impairment in chloroplast translation,suppress var2variegation and the existence of complex genetic interactions in chloroplast development.Xiayan Liu Mengdi Zheng Rui Wang Ruijuan Wang Lijun An Steve R.Rodermel Fei Yu 2013Journal of Integrative Plant Biology2013,55,10:3
6Surface hydrophobic modification enhanced catalytic performance of electrochemical nitrogen reduction reaction显示文摘Electrocatalytic nitrogen reduction reaction(NRR)is a sustainable approach for NH_(3)production with low energy consumption.However,competing hydrogen reduction reaction(HER)in aqueous solution results in low NH_(3)production and Faraday efficiency(FE).Here,MoS_(2)nanostructures with a hydrophobic surface are synthesized by alkyl thiols modification.Aerophilic and hydrophobic surface facilitates an efficient three-phase contact of N_(2),H_(2)O,and catalyst.Thus,localized concentrated N_(2)molecules can overcome the mass transfer limitation of N2 and depress the HER due to lowering the proton contacts.Although the active-sites decrease with the increase of the alkyl chain since the thiol may cover the active site,the optimized electrocatalyst achieves NH_(3)yield of 12.86×10^(-11)mol·cm^(-2)·s^(-1)at-0.25 V and 22.23%FE,which are 4.3 and 24 times higher than those of MoS2-CP electrocatalyst,respectively.The increased catalytic performance is attributed to the high N_(2)adsorption and depressed HER.Lijuan Niu Ziwen Liu Guohua Liu Mengxuan Li Xupeng Zong Dandan Wang Li An Dan Qu Xiaoming Sun Xiayan Wang Zaicheng Sun 2022Nano Research2022,15,5:2
7ABNORMAL SHOOT 6 interacts with KATANIN 1 and SHADE AVOIDANCE 4 to promote cortical microtubule severing and ordering in Arabidopsis显示文摘Plant interphase cortical microtubules(cMTs)mediate anisotropic cell expansion in response to environmental and developmental cues.In Arabidopsis thaliana,KATANIN 1(KTN1),the p60 catalytic subunit of the conserved MT-severing enzyme katanin,is essential for cMT ordering and anisotropic cell expansion.However,the regulation of KTN1-mediated cMT severing and ordering remains unclear.In this work,we report that the Arabidopsis IQ67 DOMAIN(IQD)family gene ABNORMAL SHOOT 6(ABS6)encodes a MT-associated protein.Overexpression of ABS6 leads to elongated cotyledons,directional pavement cell expansion,and highly ordered transverse cMT arrays.Genetic suppressor analysis revealed that ABS6-mediated cMT ordering is dependent on KTN1 and SHADE AVOIDANCE 4(SAV4).Live imaging of cMT dynamics showed that both ABS6 and SAV4 function as positive regulators of cMT severing.Furthermore,ABS6 directly interacts with KTN1 and SAV4 and promotes their recruitment to the cMTs.Finally,analysis of loss-of-function mutant combinations showed that ABS6,SAV4,and KTN1 work together to ensure the robust ethylene response in the apical hook of dark-grown seedlings.Together,our findings establish ABS6 and SAV4 as positive regulators of cMT severing and ordering,and highlight the role of cMT dynamics in fine-tuning differential growth in plants.Yuanfeng Li Meng Deng Haofeng Liu Yan Li Yu Chen Min Jia Hui Xue Jingxia Shao Jun Zhao Yafei Qi Lijun An Fei Yu Xiayan Liu 2021Journal of Integrative Plant Biology2021,63,4:2
8Three-dimensional characteristics of mesoscale eddies simulated by a regional model in the northwestern Pacific Ocean during 2000–2008显示文摘Mesoscale eddies play vital roles in ocean processes.Although previous studies focused on eddy surface features and individual three-dimensional(3D)eddy cases in the northwestern Pacific Ocean,the analysis of unique eddy3D regional characteristics is still lacking.A 3D eddy detection scheme is applied to 9 years(2000-2008)of eddyresolving Regional Ocean Modeling System(ROMS)output to obtain a 3D eddy dataset from the surface to a depth of 1000 m in the northwestern Pacific Ocean(15°-35°N,120°-145°E).The 3D characteristics of mesoscale eddies are analyzed in two regions,namely,Box1(Subtropical Countercurrent,15°-25°N,120°-145°E)and Box2(Southern Kuroshio Extension,25°-35°N,120°-145°E).In Box1,the current is characterized by strong vertical shear and weak horizontal shear.In Box2,the current is characterized by the strong Kuroshio,topographic effect,and the westward propagation of Rossby waves.The results indicate the importance of baroclinic instability in Box1,whereas in Box2,both the barotropic and baroclinic instability are important.Moreover,the mesoscale eddies’properties in Box1 and Box2 are distinct.The eddies in Box1 have larger number and radius but a shorter lifetime.By contrast,Box2 has fewer eddies,which have smaller radius but longer lifetime.Vertically,more eddies are detected at the subsurface than at the surface in both regions;the depth of 650 m is the turning point in Box1.Above this depth,the number of cyclonic eddies(CEs)is larger than that of anticyclonic eddies(AEs).In Box2,the number of CEs is dominant vertically.Eddy kinetic energy(EKE)and mean normalized relative vorticity in Box2are significantly higher than those in Box1.With increasing depth,the attenuation trend of EKE and relative vorticity of Box1 become greater than those of Box2.Furthermore,the upper ocean(about 300 m in depth)contains 68.6%of the eddies(instantaneous eddy).Only 16.6%of the eddies extend to 1000 m.In addition,about87%of the eddies are bowl-shaped eddies in the two regions.Only about 3%are cone-shaped eddies.With increasing depth of the eddies,the proportion of bowl-shaped eddies gradually decreases.Conversely,the coneand lens-shaped eddies are equal in number at 700-1000 m,accounting for about 30%each.Studying the 3D characteristics of eddies in two different regions of the northwestern Pacific Ocean is an important stepping stone for discussing the different eddy generation mechanisms.Guijing Yang Xiayan Lin Guoqing Han Yu Liu Gengxin Chen Jianhui Wang 2022Acta Oceanologica Sinica2022,41,10:1
9Influence of elution conditions on DNA transport behavior in free solution by hydrodynamic chromatography显示文摘We have previously developed bare narrow-bore capillary chromatography. In this work, high-performance DNA separation was realized for a size range of 10–800 base pairs(bp) utilizing bare narrow-bore capillary chromatography with 750 nm- radius capillaries. Separation behavior of double-stranded DNA(ds DNA) fragments was investigated over a range of eluent concentrations and elution pressures. DNA molecules were hydrodynamically separated in a size-dependent manner in free solution without any sieving matrices, with the longer fragments being eluted out from the capillary earlier. It was found that the eluent concentration variously influenced the transport behavior for different-sized DNA fragments depending upon the configuration of DNA molecules and the association of counterions. Ionic strength of the solutions strongly impacted DNA persistence length. Enhanced elution pressure could shorten analysis time with a slight loss in resolution. Excellent efficiency of two million theoretical plates per meter was achieved, which indicates the enormous potential of bare narrow-bore capillary chromatography for the analysis of DNA fragments. 10Fe_(2)Mo_(3)O_(8)/XC-72 electrocatalyst for enhanced electrocatalytic nitrogen reduction reaction under ambient conditions显示文摘To perform the electrochemical nitrogen reduction reaction(NRR)under milder conditions for sustainable ammonia production,electrocatalysts should exhibit high selectivity,activity,and durability.However,the key restrictions are the highly stable N≡N triple bond and the competitive hydrogen evolution reaction(HER),which make it difficult to adsorb and activate N2 on the surface of electrocatalysts,leading to a low ammonia yield and Faraday efficiency.Inspired by the enzymatic nitrogenase process and using the Fe-Mo as the active center,here we report supported Fe_(2)Mo_(3)O_(8)/XC-72 as an effective and durable electrocatalyst for the NRR.Fe_(2)Mo_(3)O_(8)/XC-72 exhibited NRR activity with an NH3 yield of 30.4μg·h^(−1)·mg^(−1)(−0.3 V)and a Faraday efficiency of 8.2%(−0.3 V).Theoretical calculations demonstrated that the electrocatalytic nitrogen fixation mechanism involved the Fe atom in the Fe_(2)Mo_(3)O_(8)/XC-72 electrocatalyst acting as the main active site in the enzymatic pathway(*NH2→*NH3),which activated nitrogen molecules and promoted the NRR performance.Guohua Liu Lijuan Niu Zhixue Ma Li An Dan Qu Dandan Wang Xiayan Wang Zaicheng Sun 2022Nano Research2022,15,7:0
11The critical factors of photocatalytic H2 production from seawater by using TiO_(2)as photocatalyst显示文摘The solar H_(2)generation directly from natural seawater is a sustainable way of green energy.However,it is limited by a low H_(2)generation rate even compared to fresh water.In this report,TiO_(2)is chosen as a model photocatalyst to disclose the critical factor to deteriorate the H_(2)generation rate from seawater.The simulated seawater(SSW),which is composed of eight ions(Na^(+),K^(+),Ca^(2+),Mg^(2+),Cl^(−),Br^(−),SO_(4)^(2−),and CO_(3)^(2−)),is investigated the effect of each ion on the H_(2)production.The results indicate that all ions have a negative effect at the same concentration as in the seawater except Br−.The CO_(3)^(2−)has the most serious deterioration,and the H_(2)production rate lowers near 40%even at[CO_(3)^(2−)]of 1.5 mmol·L^(−1).The H_(2)production rate can be recovered to 85%if the CO_(3)^(2−)is excluded from the SSW.To understand the reason,the zeta potential of the TiO_(2)treated with different ions aqueous solution reveals that the zeta potential decreases when it is treated with CO_(3)^(2−)and SO_(4)^(2−)due to they can adsorb on the surface of TiO_(2)nanoparticles.Fourier transform infrared(FTIR)and thermogravimetric analysis-mass spectroscopy(TGA-MS)further confirm that the adsorbed ion is mainly from CO_(3)^(2−).Since the pH of seawater is about 8.9 between pKa1(6.37)and pKa2(10.3)of H_(2)CO3,the CO_(3)^(2−)should exist in the form of HCO3−in the seawater.We proposed a simple method to remove the adsorbed HCO3−from the TiO_(2)surface by adjusting the pH below the pKa1.The results indicate that if a trace amount of HCl(adjusting pH~6.0)is added to the SSW,the H_(2)production rate can be recovered to 85%of that in pure water.Yaqian He Pengfei Li Wenning Liu Li An Dan Qu Xiayan Wang Zaicheng Sun 2023Nano Research2023,16,4:0
12Single-cell mass spectrometry studies of drug metabolism heterogeneity and primary resistance to gefitinib in non-small cell lung cancer cells显示文摘Patients with epidermal growth factor receptor(EGFR)wild-type non-small cell lung cancer(NSCLC)often show primary resistance to gefitinib therapy.It is thus necessary to study the metabolism of gefitinib in NSCLC cells to comprehensively reveal the reasons for the primary resistance of tumors.Herein,we develop a platform for studying drug metabolism heterogeneity based on single-cell mass spectrometry(sDMH-scMS)by integrating living-cell electrolaunching ionization MS(ILCEI-MS)and high-performance liquid chromatography-MS(HPLC-MS)analysis,and the primary resistance of NSCLC cells to gefitinib was studied using this platform.The ILCEI-MS analysis showed that approximately 11.9%of NSCLC single cells contained the gefitinib metabolite M11;HPLC-MS detection diluted the intensity of M11 in subpopulations and concealed the heterogeneity of drug metabolism in tumor single cells.The intensity of gefitinib in EGFR wild-type A549 cells was markedly lower than in mutant PC9 cells,and the intensity of gefitinib metabolites was significantly higher than in PC9 cells,suggesting that the primary resistance of NSCLC cells is related to gefitinib metabolism.Moreover,the combination of gefitinib and the drug-metabolizing enzyme inhibitorα-naphthoflavone was shown to overcome the primary resistance of the NSCLC cells.Overall,the results of this study are expected to be applicable for clinical drug resistance diagnosis and treatment at the single-cell level.Guizhen Zhu Yaoyao Zhao Wenmei Zhang Yuanyuan Wu Yuanxing Liu Guangsheng Guo Xiayan Wang Zhihong Liu 2024Chinese Chemical Letters2024,35,2:0
13UvSMEK1,a Suppressor of MEK Null,Regulates Pathogenicity,Conidiation and Conidial Germination in Rice False Smut Fungus Ustilaginoidea virens显示文摘Rice false smut,which is caused by Ustilaginoidea virens,is an emerging disease of rice spikelets in rice-growing areas worldwide.However,the infection mechanism of U.virens on rice spikelets is still unclear.Here,we characterized a suppressor of mitogen-activated protein kinase kinase or ERK kinase(MEK)null(UvSMEKI)in U.virens that is conserved among filamentous fungi.Compared with wild type U.virens strain P-1,UvSMEKI deletion mutants were defective in pathogenicity and conidial germination.In addition,conidiation of UvSMEKI deletion mutants was significantly reduced on yeast extract tryptone(YT)plates,but inc「eased in YT broth compared with the wild type.Compared with UvSMEKI expression level during the vegetative mycelia and conidiation stages,UvSMEKI dramatically increased during infection of rice florets.Surprisingly,the UvSMEKI deletion mutants exhibited higher tolerance to H_(2)O_(2) and NaCl.In summary,presented evidence suggested that UvSMEKI positively regulated pathogenicity,conidial germination and conidiation in YT broth,and negatively regulated conidiation on YT medium and tolerance to oxidative and osmotic stresses.The results enhanee our understanding of the regulatory mechanism of pathogenicity of U.virens,and present a potential molecular target for blocking rice infection by U.virens.Yu Junjie Yu Mina SONG Tianqiao CAO Huijuan Yong Mingli Pan Xiayan Qi Zhongqiang Du Yan Zhang Rongsheng Yin Xiaole Liang Dong Liu Yongfeng 2021Rice science2021,28,5:0
