| 3 | Very Low-Level Heteroplasmy mtDNA Variations Are Inherited in Humans显示文摘Little is known about the inheritance of very low heteroplasmy mitochondria DNA(mtDNA)variations.Even with the development of new next-generation sequencing methods,the practical lower limit of measured heteroplasmy is still about 1%due to the inherent noise level of the sequencing.In this study,we sequenced the mitochondrial genome of 44 individuals using Illumina high-throughput sequencing technology and obtained high-coverage mitochondria sequencing data.Our study population contains many mothere offspring pairs.This unique study design allows us to bypass the usual heteroplasmy limitation by analyzing the correlation of mutation levels at each position in the mtDNA sequence between maternally related pairs and non-related pairs.The study showed that very low heteroplasmy variants,down to almost 0.1%,are inherited maternally and that this inheritance begins to decrease at about 0.5%,corresponding to a bottleneck of about 200 mtDNA. | Yan Guo Chung-I Li Quanhu Sheng Jeanette F.Winther Qiuyin Cai John D.Boice Yu Shyr | 2013 | Journal of Genetics and Genomics2013，40，12： | 4 |
| 4 | A comparative study of pancreas transplantation between type 1 and 2 diabetes mellitus显示文摘Background:Pancreas transplantation remains the best long-term treatment option to achieve physiological euglycemia and insulin independence in patients with labile diabetes mellitus(DM).It is widely accepted as an optimal procedure for type 1 DM(T1DM),but its application in type 2 DM(T2DM)is not unanimously acknowledged.Methods:In total,146 diabetes patients undergoing pancreas transplantation were included in this study.Clinical data and outcomes were compared between the T1DM and T2DM groups.Results:Majority(93%)of the pancreas transplantations in T2DM were for uremic recipients.Complications occurred in 106(73%)patients,including 70(48%)with early complications before discharge and 79(54%)with late complications during follow-up period.Overall,rejection of pancreas graft occurred in 37(25%)patients.Total rejection rate in T2DM recipients was significantly lower than that in T1DM.The short-and long-term outcomes for endocrine function in terms of fasting blood sugar and hemoglobin A1c levels and graft survival rates are comparable between the T2DM and T1DM groups.Conclusions:T2DM is not inferior to T1DM after pancreas transplantation in terms of surgical risks,immunological and endocrine outcomes,and graft survival rates.Therefore,pancreas transplantation could be an effective option to treat selected uremic T2DM patients without significant insulin resistance. | Bor-Shiuan Shyr Bor-Uei Shyr Shih-Chin Chen Che-Chuan Loong Yi-Ming Shyr Shin-E Wang | 2021 | Hepatobiliary Surgery and Nutrition2021，10，4： | 4 |
| 5 | Lymph node ratio and preoperative CA 19-9 levels predict overall survival and recurrence-free survival in patients with resected pancreatic adenocarcinoma显示文摘AIM:Clinicopathologic factors predicting overall survival (OS) would help identify a subset to benefit from adjuvant therapy. METHODS: One hundred and sixty-nine patients patients from 1984 to 2009 with curative resections for pancreatic adenocarcinoma were included. Tumors were staged by American Joint Committee on Cancer 7th edition criteria. Univariate and multivariable analyses were performed using Kaplan-Meier methodology or Cox proportional hazard models. Log-rank tests were performed. Statistical inferences were assessed by two-sided 5% significance level. RESULTS: Median age was 67.1 (57.2-73.0) years with equal gender distribution. Tumors were in the head (89.3%) or body/tail (10.7%). On univariate analysis, adjuvant therapy, lymph node (LN) ratio, histologic grade, negative margin status, absence of peripancreatic extension, and T stage were associated with improved OS. Adjuvant therapy, LN ratio, histologic grade, number of nodes examined, negative LN status, and absence of peripancreatic extension were associated with improved recurrence-free survival (RFS). On multivariable analysis, LN ratio and carbohydrate antigen (CA) 19-9 levels were associated with OS. LN ratio was associated with RFS.
