1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 4A Method for Detection of Main Metabolites in Aromatic Rice Seeds显示文摘[Objective] A method was developed for the analysis of main metabolites in aromatic rice seeds by gas chromatography-mass spectrometry( GC-MS).[Method]The main metabolites in seeds of 9 different aromatic rice varieties were compared and analyzed with two methods. Method 1: the compound was extracted by the mixture of anhydrous ethanol and methylene chloride( V/V = 1∶ 1) at 60 ℃ for 3 h in water bath; the extract was isolated and detected by gas chromatography-mass spectrometry using HP-5 capillary column with scan monitoring mode. Method 2: the main metabolites in aromatic rice seeds were detected using headspace solid phase micro-extraction and gas chromatography mass spectrometry( GC-MS). [Result]Fifteen types of common metabolites were found in 9 different aromatic rice varieties. Method 1 was more effective than method 2 in detecting the main metabolites in aromatic rice seeds. 5Pertuzumab in combination with trastuzumab and chemotherapy for Chinese patients with HER2-positive metastatic gastric or gastroesophageal junction cancer:a subpopulation analysis of the JACOB trial显示文摘Background:The JACOB trial(NCT01774786)was a double-blinded,placebo-controlled,randomized,multicenter,international,phase III trial evaluating the efficacy and safety of adding pertuzumab to trastuzumab and chemo-therapy in first-line treatment of human epidermal growth factor receptor 2(HER2)-positive metastatic gastric cancer/gastroesophageal junction cancer(GEJC).The aim of this analysis was to investigate efficacy and safety outcomes in the Chinese subpopulation from the JACOB trial.Methods:This post hoc subpopulation analysis included all patients recruited in China's Mainland(n=163;20.9%)between June 2013 and January 2016.The patients were randomly assigned in a 1:1 ratio to receive pertuzumab plus trastuzumab and chemotherapy(pertuzumab group;n=82)or placebo plus trastuzumab and chemotherapy(con-trol group;n=81).Intravenous pertuzumab(840 mg)and trastuzumab(8 mg/kg loading and 6 mg/kg maintenance doses)were given every 3 weeks until disease progression or unacceptable toxicity.Chemotherapy was given as per standard regimens/doses of capecitabine or 5-fluorouracil plus cisplatin.The primary endpoint was overall survival(OS);secondary efficacy endpoints included progression-free survival(PFS),and overall objective response rate(ORR).Results:The median OS was 18.7 months in the pertuzumab group and 16.1 months in the control group(hazard ratio[HR]0.75;95%confidence interval[CI]0.49 to 1.14).The median PFS was 10.5 and 8.6 months in the pertuzumab and control groups,respectively(HR 0.85;95%CI 0.60 to 1.21),and the median ORRs were 68.9%and 55.7%,respectively.The treatment effect in this Chinese subpopulation showed consistency with that in the global ITT population with numerically lower HR for OS and PFS compared with the control group.The safety profiles of the pertuzumab and control groups in this Chinese subpopulation analysis were generally comparable.The most common grade 3-5adverse events were neutropenia,anemia,and leukopenia.However,due to the nature of being a post hoc subgroup analysis,the results presented here are descriptive only and need to be interpreted with caution.Conclusions:OS and PFS were numerically improved by adding pertuzumab to trastuzumab and chemotherapy as first-line treatment in Chinese HER2-positive gastric cancer/GEJC patients,and this regimen demonstrated an acceptable safety profile.Tianshu Liu Yanru Qin Jin Li Ruihua Xu Jianming Xu Shujun Yang Shukui Qin Yuxian Bai Changping Wu Yixiang Mao Haiyan Wu Yilin Ge Lin Shen 2019Cancer Communications2019,39,1:6

7Study protocol of the Asian XELIRI ProjecT(AXEPT):a multinational,randomized,non-inferiority,phase Ⅲ trial of second-line chemotherapy for metastatic colorectal cancer, comparing the eicacy and safety of XELIRI with or without bevacizumab versus FOLFIRI w显示文摘Background: Capecitabine and irinotecan combination therapy(XELIRI) has been examined at various dose levels to treat metastatic colorectal cancer(m CRC). Recently, in the Association of Medical Oncology of the German Cancer Society(AIO) 0604 trial, tri?weekly XELIRI plus bevacizumab, with reduced doses of irinotecan(200 mg/m^2 on day 1) and capecitabine(1600 mg/m^2 on days 1–14), repeated every 3 weeks, has shown favorable tolerability and eicacy which were comparable to those of capecitabine and oxaliplatin(XELOX) plus bevacizumab. The doses of capecit?abine and irinotecan in the AIO trial are considered optimal. In a phase I/II study, XELIRI plus bevacizumab(BIX) as second?line chemotherapy was well tolerated and had promising eicacy in Japanese patients.Methods: The Asian XELIRI Projec T(AXEPT) is an East Asian collaborative, open?labelled, randomized, phase Ⅲ clinical trial which was designed to demonstrate the non?inferiority of XELIRI with or without bevacizumab versus standard FOLFIRI(5?fluorouracil, leucovorin, and irinotecan combination) with or without bevacizumab as second?line chemo?therapy for patients with m CRC. Patients with 20 years of age or older, histologically conirmed m CRC, Eastern Coop?erative Oncology Group performance status 0–2, adequate organ function, and disease progression or intolerance of the irst?line regimen will be eligible. Patients will be randomized(1:1) to receive standard FOLFIRI with or with?out bevacizumab(5 mg/kg on day 1), repeated every 2 weeks(FOLIRI arm) or XELIRI with or without bevacizumab(7.5 mg/kg on day 1), repeated every 3 weeks(XELIRI arm). A total of 464 events were estimated as necessary to show non?inferiority with a power of 80% at a one?sided α of 0.025, requiring a target sample size of 600 patients. 8The shallow Longmenshan crustal S-velocity structure of the fault zone using ambient noise tomography of a seismic dense array显示文摘The Longmenshan fault zone(LMSF),characterized by complex structures and strong seismicity,is located at the junction between the eastern margin of the Tibetan Plateau and the north-western Sichuan basin.Since the Wenchuan earthquake on May 12,2008,abundant studies of the formation mechanism of earthquakes along the LMSF were performed.In this study,a short-period dense seismic array deployed across the LMSF was applied by ambient noise tomography.Fifty-two 3-D seismic instruments were used for data acquisition for 26 days.We calculated the empirical Green's functions(EGFs)between different station-pairs and extracted 776 Rayleigh-wave dispersion curves between 2 and 7 s.And then,we used the direct-inversion method to obtain the fine shallow crustal S-wave velocity structure within 6 km depth in the middle section of the Longmenshan fault zone and nearby areas.Our results show that the sedimentary layer(>5 km)exists in the northwest margin of Sichuan Basin with a low S-wave velocity(~1.5-2.5 km/s)which is much thicker than that beneath the Longmenshan fault zone and the Songpan-Garze block.The high-velocity structures with clear boundaries below the middle of Longmenshan fault zone(~2-4 km)and the Songpan-Garze block(~4.5-6 km)probably reveal the NW-SE distribution patterns of both the Pengguan complex and the high-density belt hidden in the northwest of the Pengguan complex.And the obviously high-velocity anomalies observed at the depth of^1-2 km in the southeastern margin of the Songpan-Garze block can be considered as the Laojungou granites.Our results provide a high-resolution shallow velocity structure for detailed studies of the Longmenshan fault zone.Dandan Li Gaochun Wang Ruihua Lin Kai Deng Xiaobo Tian 2019Earthquake Science2019,32,5:3
9Aberrantly expressed GFRα-1/RET in patients with lacrimal adenoid cystic carcinoma is associated with high recurrence risk:a retrospective study of 51 LACC cases显示文摘Objective:Because of the poor prognosis of lacrimal adenoid cystic carcinoma(LACC),we aimed to investigate the effects of perineural invasion(PNI)and consequent aberrations in GDNF/GFRα-1/RET protein expression on LACC recurrence.Methods:Clinicopathological data for 51 histologically confirmed patients with LACC enrolled between 2001 and 2017 were retrospectively analyzed.Hematoxylin and eosin staining was applied to assess PNI.Tissue-based immunohistochemistry(IHC)detection of GDNF,GFRα-1,and RET proteins was performed on LACC formalin-fixed,paraffin-embedded specimens.We generated semi-quantitative data of the IHC results and compared them with the clinicopathological data for the 51 patients.Results:Of the 51 patients,19(37.3%)were PNI positive.Recurrence was more common for LACC with than without PNI(73.7%vs.37.5%,P=0.01).GDNF,GFRα-1,and RET proteins were expressed in 62.7%,62.7%,and 54.9%of the 51 patients with LACC,respectively.The expression of all 3 proteins was more common in patients with than without PNI.In agreement with previous findings,PNI-associated GFRα-1 and RET positivity,as detected by IHC,remained significantly associated with recurrence,whereas GDNF expression,as detected by IHC,was not correlated with LACC recurrence.Specifically,patients with concurrent GFRα-1 and RET expression may have a high risk of PNI(89.5%positivity rate)and recurrence(84.2%positivity rate).Conclusions:PNI may contribute to LACC recurrence.The concurrent expression of GFRα-1 and RET proteins,as detected by IHC,may potentially be associated with LACC PNI and recurrence.Lin Liu Liqiong Zhao Jie Zhang Guoxiang Song Carol LShields Ruihua Wei 2021Cancer Biology & Medicine2021,18,1:3
10Awareness intervention for Beijing neurologists regarding secondary prevention of cerebral infarction/transient ischemia Cross-sectional investigation显示文摘BACKGROUND: Stroke prevention guidelines should be made available to neurologists for clinical application to aid in uniformity, timing, preciseness, and acceptance of disease. OBJECTIVE: To investigate the awareness of neurologists in some Beijing hospitals of intervention in secondary prevention of cerebral infarction/transient ischemia. DESIGN: Cross-sectional study. SETTING: Beijing Tiantan Hospital, Beijing Anzhen Hospital, General Hospital of Beijing Military Area, Command of Chinese PLA, Beijing Chuiyangliu Hospital, Beijing 6th Hospital, Beijing Hepingli Hospital, and Beijing Daxing District Hospital. PARTICIPANTS: A total of 28 (associate) chief physicians, 58 attending physicians, and 54 resident physicians who engaged in clinical treatment of cerebrovascular diseases were selected from 8 hospitals in Beijing from March to April 2007. All physicians provided informed consent. METHODS: Self-made closed questionnaires were provided for data collection, consisting of 16 questions that were single choice or multiple choice. Specifically, questions 1-7 focused on awareness of blood pressure regulation in different patients and first choice of decompression drug; questions 8-12 focused on awareness of lipid regulation; and questions 13-16 focused on awareness of anti-blood platelet drugs applied in secondary prevention. The scores ranged from 0-100 points, and each question was worth 6.25 points. The scores positively correlated with the awareness rate. To test leveling real-time, the survey lasted for a maximum of 20 minutes. One questionnaire was independently finished by each subject in the survey. MAIN OUTCOME MEASURES: Awareness intervention among neurologists during secondary prevention of cerebral infarction/transient ischemia and questionnaire scores. RESULTS: 140 subjects were included in the final analysis. ① The awareness rate among neurologists for intervention during secondary prevention of cerebral infarction/transient ischemia ranged from 0.7-57.9%, the scores ranged between 0-56 points, and the mean score was 26 points. ② Scores of resident physicians were 0-56 points with a mean score of 26 points; attending physicians scored 6-50 points, with a mean score of 26 points; and chief physicians scored 6-50 points, with a mean score of 23 points. There were no significant differences among the various physicians (F = 0.771, t = 0.465, P > 0.05). CONCLUSION: Awareness among neurologists of intervention during secondary prevention of cerebral infarction/transient ischemia is not ideal. 11Investigation on Host Species and Damage Degree of Bemisia tabaci (Gennadius) in Shaanxi Area显示文摘The host plants of Bemisia tabaci( Gennadius) in Shaanxi were investigated,and its damage degree was evaluated by hazard index in 2013 and 2014.There were 73 species( variants) of host plants of B. tabaci belonging to 28 families,mainly concentrated in Cucurbitaceae,Cruciferae,Solanaceae,Laguminosae and Malvaceae. There were seven species of vegetables that suffered the damage of grade 4. B. tabaci was more serious in southern Shaanxi and lighter in northern Shaanxi. 12Oxaliplatin,Fluorouracil and Leucovorin (FOLFOX) as First-line Chemotherapy for Metastatic or Recurrent Colorectal Cancer Patients显示文摘OBJECTIVE To investigate the efficiency and safety of the oxaliplatin, fluorouracil(5-FU)and leucovorin regimen(FOLFOX)in previously untreated patients with metastatic or recurrent colorectal cancer. METHODS Previously untreated patients with metastatic or recurrent colorectal cancer received 100 mg/m2 of oxaliplatin intravenously(IV)over 2 h on day 1,and IV 400 mg/m2 of leucovorin over 2 h followed by a bolus of 400 mg/m2 of 5-FU.Then 2,600~3,000 mg/m2 of 5-FU was administered by continuous infusion over 46 h. RESULTS An evaluated response rate was determined for 97 of 105 treated patients.The overal response rate was 35.1%,9 patients(9.3%) had a complete response and 25 patients(25.8%)a partial response.Thirty-two patients(33.0%)developed stable disease and 32.0%of the patients progressed.The median time to progression(TTP)was 7.7 months and the median overal survival 20.5 months.One and 2-year survival rates were 68%and 32%.Toxic effects based on the National Cancer Institute-Common Toxicity Criteria(NCI-CTC),reaching grade 3/4 were:neutropenia 12.3%, anemia 11.3%,vomiting 4.1%and diarrhea 7.2%.Grade 3 neuropathy was 5.1%.The overall survival rate of patients who had received a radical resection was superior to the patients who had not received a operation,or had received a pal iative resection(P=0.0658).The serum levels of CEA,ALP and LDH had no relationship with survival(P>0.05). 15The formation kinetics and control of biofilms by three dominant fungi species isolated from groundwater显示文摘Filamentous fungi can enter drinking water supply systems in various ways,and exist in suspended or sessile states which threatens the health of individuals by posing a high risk of invasive infections.In this study,the biofilms formation kinetics of the three genera of fungal spores,Aspergillus niger(A.niger),Penicillium polonicum(P.polonicum)and Trichoderma harzianum(T.harzianum)isolated fromthe groundwater were reported,as well as the effects of water quality parameters were evaluated.In addition,the efficiency of low-concentrations of chlorine-based disinfectants(chlorine,chlorine dioxide and chloramine)on controlling the formation of fungal biofilms was assessed.The results showed that the biofilms formation of the three genera of fungi could be divided into the following four phases:induction,exponential,stationary and sloughing off.The optimum conditions for fungal biofilms formation were found to be neutral or weakly acidic at 28°C with rich nutrition.In fact,A.niger,P.polonicum,and T.harzianum were not observed to form mature biofilms in actual groundwater within 120 hr.Carbon was found to have the maximum effect on the fungal biofilms formation in actual groundwater,followed by nitrogen and phosphorus.The resistance of fungal species to disinfectants during the formation of biofilms decreased in the order:A.niger>T.harzianum>P.polonicum.Chlorine dioxide was observed to control the biofilms formation with maximum efficiency,followed by chlorine and chloramine.Consequently,the results of this study will provide a beneficial understanding for the formation and control of fungal biofilms.Xinyu Luo Xiangqian Xu Ruihua Cao Qiqi Wan Jingyi Wang Huining Xu Yingzi Lin Gang Wen Tinglin Huang 2021Journal of Environmental Sciences2021,33,11:1

17Ultrafast photocarrier and coherent phonon dynamics in type-II Dirac semimetal PtTe2 thin films probed by optical spectroscopy显示文摘We report the ultrafast photocarrier dynamics and coherent phonon excitation in type-II Dirac semimetal platinum ditelluride(PtTe2)thin films via femtosecond(fs)pump-probe spectroscopy at room temperature.Quantitative analysis revealed that the incoherent electronic relaxation consists of two components:a subpicosecond fast relaxation process and a slow component with a time constant of hundreds of picoseconds(ps).Furthermore,the launch of a coherent acoustic phonon(CAP)in the 20 nm film but absence in the 6.8 nm film uncovers the dominant role of temperature gradient in producing a strain wave.The sound velocity and Young’s modulus in the thick PtTe_(2) are determined to be 1.736 km/s and 29.5 GPa,respectively.In addition,the coherent optical phonon(COP)with a frequency of 4.7 THz corresponding to Te atoms out-of-plane A1g vibration has been well resolved in all films,which is ascribed to displacive excitation of coherent phonon(DECP).The observation of a strong probe-wavelength dependent COP amplitude reveals the resonant feature of the optical excitation-induced atomic displacement in PtTe2.Our findings provide deep insight into the excitation and dynamics of CAP and COP as well as the photocarriers’recovery pathway and lifetimes in PtTe_(2).Our study also demonstrates that the COP spectroscopy is a powerful tool to reveal the modulation of frequency-dependent optical constants induced by atomic vibrations,which may find applications in the fields of optoelectronics and ultrafast photonics.Peng Suo Shengnan Yan Ruihua Pu Wenjie Zhang Di Li Jiaming Chen Jibo Fu Xian Lin Feng Miao Shi-Jun Liang Weimin Liu Guohong Ma 2022Photonics Research2022,10,3:0
18Clinical characteristics of hospitalised patients with schizophrenia who were suspected to have coronavirus disease(COVID-19)in Hubei Province,China显示文摘Background Since the outbreak of C0VID-19,no data have been available for hospitalised psychiatric patients who are suspected to have COVID-19.We performed a comprehensive investigation of the clinical features of hospitalised patients with schizophrenia with or without suspected COVID-19 in Hubei Province,China.Aim To explore the clinical characteristics of hospitalised patients with schizophrenia with suspected COVID-19 in Hubei Province,China.M ethods 21 hospitalised patients with schizophrenia with suspected COVID-19(COVID-19 suspected group)in the isolation ward of a mental health hospital in Wuhan and 30 hospitalised patients with schizophrenia(clean group)in the general ward of another mental health hospital in Yichang were recruited.We retrospectively reviewed their clinical characteristics,laboratory findings and chest CT results before 21 February 2020.We also compared the emotional and mental symptoms between the two groups.Results Medical records revealed that 21 COVID-19 suspected patients were transferred to the isolation ward between 30 January 2020 and 15 February 2020.The mean age(SD)of COVID-19 suspected patients was 43.1(2.6).12(57.1%)patients showed abnormalities on chest CT before onset of respiratory symptoms.14(66.7%)patients had psychiatric medications adjustment after detection of abnormal chest CT findings.By 21 February,one patient was confirmed to have COVID-19.Even though the remaining 20(95.2%)were negative for at least two reverse transcription PCR tests,11(52.4%)patients met the diagnostic criteria for clinically confirmed cases.Compared with patients in the clean group,patients in the suspected COVID-19 group showed significantly higher stress,depression and anxiety levels and poorer sleep quality.Conclusion Setting up an independent isolation ward for hospitalised psychiatric patients who are suspected to have symptoms of COVID-19 helped control the spread of the epidemic.Patients with schizophrenia suspected to have COVID-19 showed increased stress and mood and sleep disturbances,which should be appropriately managed.Xuebing Liu Hong Lin Haifeng Jiang Ruihua Li Na Zhong Hang Su Yi Li Min Zhao 2020General Psychiatry2020,33,2:0
19Identification of Prognostic Related Hub Genes in Clear-Cell Renal Cell Carcinoma via Bioinformatical Analysis显示文摘Objective To identify new genes that correlate with prognosis of clear-cell renal cell carcinoma(ccRCC)via bioinformatics analysis.Methods The gene expression profiles of 62 ccRCC and 54 normal kidney tissues were available from the Gene Expression Omnibus database:GSE12606,GSE36895 and GSE66272.The differentially expressed genes were screened with GEO2R and J Venn online tools.Functional annotation including Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)was applied to identify the possible function of the hub genes involved in prognosis of ccRCC.In protein protein interaction network(PPI network),the STRING online tool was used to visualize the network of the differentially expressed genes,and the core gene was selected by MCODE App in Cytoscape software.Finally,GEPIA Survival Plot was performed to assess genes associated with worse survival.Results We totally found 648 diflerentially expressed genes,including 222 up-regulated genes and 426 down-regulated genes.PPI network showed that in 28 up-regulated genes 7(CCNE2,CDK1,CDC6,CCNB2,BUB1,TTK and PTTG1)enriched in cell cycle and 4 genes(CCNE2,CDK1,CCNB2 and RRM2)enriched in p53 signaling pathway.GEPIA Survival Plot assay revealed that ccRCC patients carrying CDK1,CCNB2,RRM2t BUB1,and PTTG1 had a worse survival.GEPIA Box Plot showed that BUB1,CCNB2,PTTG1,and RRM2 were over expressed in the ccRCC tissues in contrast to the normal tissues(PPing Jiang Taotao Sun Cunwu Chen Renshu Huang Zhimei Zhong Xinjian Lou Gang Liu Lin Wang Ruihua Zuo 2021Chinese Medical Sciences Journal2021,36,2:0
